Immunomodulatory Activity of Protein-Bound Polysaccharide Extracted from Gheiidonium majus

  • In the course of searching immunomodulators from natural sources, the protein-bound polysaccharide, CM-Ala, has been isolated from the water extract of Chelidonium majus L. (Papaveraceae). The immunostimulatory characteristics have been investigated in several experiments such as generation of activated killer (AK) cells, proliferation of splenocytes, activation of macrophages and granulocyte macrophage-colony forming cell (GM-CFC) assay. Of the fractions obtained using Sephacryl S200 column chromatography, CM-Ala was the most effective fraction that augmented the cytotoxicity against Yac-1 tumor cells from 0.88% to 34.18% by culturing with splenocytes for 5 days. CM-Ala also enhanced nitric oxide production by two fold in peritoneal macrophages and exhibited antitumor activity. It showed mitogenic activity on both spleen cells and bone marrow cells. CM-Ala induced proliferation of splenocytes by 84 fold and increased GM-CFC numbers by 1.48 fold over than the non-treated. On the contrary, CM-Ala had cytotoxic activity to a diverse group of tumor cells. From the above results, we proposed that CM-Ala has a possibility of an effective antitumor immunostimulator.

Anticancer Drugs at Low Concentrations Upregulate the Activity of Natural Killer Cell

  • Natural killer (NK) cells are innate cytotoxic lymphoid cells that actively prevent neoplastic development, growth, and metastatic dissemination in a process called cancer immunosurveillance. Regulation of the cytotoxic activity of NK cells relies on integrated interactions between inhibitory receptors and numerous activating receptors that act in tandem to eliminate tumor cells efficiently. Conventional chemotherapy is designed to produce an anti-proliferative or cytotoxic effect on early tumor cell division. Therapies designed to kill cancer cells and simultaneously maintain host anti-tumor immunity are attractive strategies for controlling tumor growth. Depending on the drug and dose used, several chemotherapeutic agents cause DNA damage and cancer cell death through apoptosis, immunogenic cell death, or other forms of non-killing (i.e., mitotic catastrophe, senescence, autophagy). Among stress-induced immunostimulatory proteins, changes in the expression levels of NK cell activating and inhibitory ligands and tumor cell death receptors play an important role in the detection and elimination by innate immune effectors including NK cells. Therefore, we will address how these cytotoxic lymphocytes sense and respond to high and low concentrations of drug-induced stress to the drug cisplatin, among the various types of drugs that contribute to their anticancer activity.

Effect of Korean Mistletoe (Viscum album Coloratum) on the Non-Specific Immune Responses in Japanese Eel (Anguilla japonica)

  • In the present paper, the immunostimulatory effects of Korean mistletoe (Viscum album Coloratum) on the non-specific immune responses of Japanese eel (Anguilla japonica) were examined. Eel were innoculated with mistletoe, Freund's complete adjuvant (FCA), or phosphate-buffered saline (PBS) as a control into their peritoneal cavities. The number of nitrobule tetrazolium (NBT)-positive cells in the head kidney of fish was significantly increased by the second day post-injection of mistletoe. ROI products were more enhanced in mistletoe-injected fish kidney leucocytes than in FCA-injected ones. The level of lysozyme activity detected in the serum of fish 2 days after injection with mistletoe was also significantly higher than that found in the serum of the control fish. The appropriate concentration of mistletoe to induce the highest level of serum lysozyme activity was revealed to 1000${\mu}g$/200 g of fish. In phagocytic activity assay, mistletoe-sensitized eel kidney phagocytes captured more zymosan than did the control fish. Korean mistletoe appeared to be a good activator of the non-specific immune responses of Japanese eel.

돈태반 효소 가수분해물의 cyclophosphamide에 의한 면역 저하 동물 모델에 미치는 면역 증진 효과 (Immunostimulatory effects of enzymatic porcine placental hydrolyzate against cyclophosphamide-induced immunosuppressed model)

  • 본 연구는 돈태반 효소 가수분해물의 면역증진 효과를 확인하고자 Cy를 이용한 면역 저하 모델에서 농도별 돈태반 효소 가수분해물을 투여한 실험군의 주간 체중과 조직 중량, 혈중 면역세포(백혈구, 과립구, 림프구, 중간구) 함량, 혈중 cytokine 및 immunoglobulin 함량, 자연살해세포 활성, 비장 조직 분석을 수행하였다. 체중은 대조군과 비교하여 돈태반 효소 가수분해물을 투여한 실험군 중 중농도 투여군(1.03 mg/kg BW, total nitrogen)과 고농도 투여군(2.07 mg/kg BW, total nitrogen)에서 다소 증가하는 경향을 보였고, 조직 중량은 돈태반 효소 가수분해물을 투여한 실험군이 Cy만을 단독 투여한 대조군에 비해 다소 높았으며, 이 중 고농도 투여군은 비장과 흉선 조직 중량 모두에서 대조군보다 유의적으로 높게 조사되었다. 각 실험군별 비장 조직을 이용한 자연살해세포 활성 분석 결과 정상군에 비해 대조군은 유의하게 감소하였으나 돈태반 효소 가수분해물을 고농도로 투여한 실험군과 양성대조군은 대조군에 비해 증가하는 경향을 보여 정상군과 유사한 수준을 보였다. 일반 혈액학적 분석(CBC analysis)에서 돈태반 효소 가수분해물을 투여한 실험군은 대조군에 비해 백혈구와 과립구, 림프구 및 중간구에서 모두 높은 함량을 보이는 것으로 나타났는데, 특히 고농도 투여군은 백혈구의 경우 양성 대조군인 HemoHIM 투여군과 유사한 수준으로, 과립구와 중간구의 경우 양성대조군보다 더 높은 함량을 보이는 것으로 조사되었다. 혈중 cytokine과 immunoglobulin의 함량을 분석한 결과 돈태반효소 가수분해물을 투여한 실험군에서 혈중 TNF-α와 IL-1β, IL-2, IL-12 및 IgG의 함량을 대조군과 비교하여 유의하게 증가시키거나 증가시키는 경향을 보였다. 또한 조직학적 분석에서 비장조직은 대조군에서 관찰되던 백색수질의 붕괴와 적색수질에서의 세포 응축현상은 돈태반 효소 가수분해물을 투여한 실험군에서 점차 호전되는 경향을 보였다. 이러한 결과를 바탕으로 돈태반효소 가수분해물은 Cy로 인한 세포와 조직 손상을 감소시키고 혈중 면역 관련 인자들의 함량을 증가시켜 면역력을 증진시키는데 긍정적인 영향을 미치는 것으로 판단되며, 추후 이를 활용한 건강 기능성 개발 및 의약품 개발에 따른 활용가치가 매우 높을 것으로 생각된다.

항생제 대체제로서 Clostridium butyricum IDCC 9207 백미 발효물의 항균작용과 장관 면역증진효과 (White Rice Fermented by Clostridium butyricum IDCC 9207 as an Alternative to Antibiotic having Antibacterial and Immunostimulatory Activity)

  • 이 연구는 천연소재 probiotics 발효물을 대상으로 가축의 장내 유해세균으로 알려진 S. typhimurium KCTC 2054와 E. coli O157:H7의 생장을 억제시키는 유효 발효물을 선발하여 특성을 연구하는 데 있다. 실험 샘플 444개 중 유해 세균에 대한 항균력이 가장 우수한 것으로 C. butyricum IDCC 9207 백미 발효물(FWR)을 최종 선발하였다. C. butyricum IDCC 9207이 함유된 백미 발효물 (FWRCb9207)을 S. typhimurium KCTC 2054로 감염시킨 ICR-mice에 일정기간 투여하였을 때, salmonella의 생육이 억제됨을 확인하였으며, 장관에서 분비되는 secretory immunoglobulin A (sIgA)의 양이 증가됨을 확인하였다. FWRCb9207은 기존의 생균제 보다 빠른 정장효과를 나타낼 수 있으며, 발효물보다 생균보강을 통해 정장작용의 기간을 늘릴 수 있는 것이 장점이다. 또한 FWRCb9207의 급여를 통해 사료로부터 쉽게 감염되는 S. typhimurium, S. gallinarum, S. pullorum, S. choleraesuis ssp. choleraesuis 등의 유해세균 증식을 효과적으로 억제시키고 면역력을 증진시킴으로써 가축의 증체율을 높였다. 이러한 FWRCb9207의 synbiotic effects는 궁극적으로 기존 항생제를 대체함으로써 항생제 오남용으로 발생되는 문제점을 해소할 수 있을 것으로 기대된다.

Immunostimulatory activity of hydrolyzed and fermented Platycodon grandiflorum extract occurs via the MAPK and NF-κB signaling pathway in RAW 264.7 cells

  • BACKGROUND/OBJECTIVES: Platycodon grandiflorum (PG) has long been known as a medicinal herb effective in various diseases, including bronchitis and asthma, but is still more widely used for food. Fermentation methods are being applied to increase the pharmacological composition of PG extracts and commercialize them with high added value. This study examines the hydrolyzed and fermented PG extract (HFPGE) fermented with Lactobacillus casei in RAW 264.7 cells, and investigates the effect of amplifying the immune and the probable molecular mechanism. MATERIALS/METHODS: HFPGE's total phenolic, flavonoid, saponin, and platycodin D contents were analyzed by colorimetric analysis or high-performance liquid chromatography. Cell viability was measured by the MTT assay. Phagocytic activity was analyzed by a phagocytosis assay kit, nitric oxide (NO) production by a Griess reagent system, and cytokines by enzyme-linked immunosorbent assay kits. The mRNA expressions of inducible nitric oxide synthase (iNOS) and cytokines were analyzed by reverse transcription-polymerase chain reaction, whereas MAPK and nuclear factor (NF)-κB activation were analyzed by Western blots. RESULTS: Compared to PGE, HFPGE was determined to contain 13.76 times and 6.69 times higher contents of crude saponin and platycodin D, respectively. HFPGE promoted cell proliferation and phagocytosis in RAW 264.7 cells and regulated the NO production and iNOS expression. Treatment with HFPGE also resulted in increased production of interleukin (IL)-1β, IL-6, tumor necrosis factor (TNF)-α, C-X-C motif chemokine ligand10, granulocyte-colony-stimulating factor, granulocyte-macrophage colony-stimulating factor, and monocyte chemoattractant protein-1, and the mRNA expressions of these cytokines. HFPGE also resulted in significantly increasing the phosphorylation of NF-κB p65, extracellular signal-regulated kinase, and c-Jun N-terminal kinase. CONCLUSIONS: Taken together, our results imply that fermentation and hydrolysis result in the extraction of more active ingredients of PG. Furthermore, we determined that HFPGE exerts immunostimulatory activity via the MAPK and NF-κB signaling pathways.

Immunostimulating Effect of a Well-known Thai Folkloric Remedy in Breast Cancer Patients

  • The study aimed to evaluate immune-stimulating effects of a well-known Thai folkloric remedy when used for adjuvant therapy with conventional chemotherapeutics for treatment of breast cancer. Immunostimulating influence of the remedy (215 mg/kg body weight per day) on NK cell activity and TNF-${\alpha}$ release from the monocytes/macrophages were investigated in a total of 15 healthy women and 13 female patients with breast cancer (Group 1). The effect of breast tumor surgery on NK cell activity was further investigated in 18 female patients with breast cancer (Group 2). NK cell cytotoxic activity was determined by chromium release cytotoxic assay using K562, an erythroleukemic cell line. TNF-${\alpha}$ release from monocytes/macrophages separated from blood samples was determined through a biological assay using actinomycin D-treated L929 mouse fibroblast cells in the presence and absence of LPS. Baseline NK cell activity of the monocytes/macrophages separated from Group 2 patients expressed as %cytotoxicity was significantly lower than in the healthy subjects at E:T ratios of 100:1 and 25:1. In healthy subjects, there was no change in NK cell cytotoxic activity (%cytotoxicity or LU) following 1 and 2 weeks of treatment with the remedy compared with the baseline at various E:T ratios but the binding activity (%binding) was significantly increased after 2 weeks of treatment. The addition of one or two conventional chemotherapeutic regimens did not significantly reduce the NK cytotoxic activity but did affect release of TNF-${\alpha}$ in both unstimulated and LPS-stimulated samples. Surgery produced a significant suppressive effect on NK cell activity. The use of the remedy as an adjunct therapy may improve therapeutic efficacy and safety profiles of conventional chemotherapeutic regimens through stimulation of the immune system in cancer patients.

Induction of Autophagy by Rosa acicularis Leaves Extracts in RAW264.7 Cells

  • Autophagy contributes to enhancing the immune system (innate and adaptive immune system) against foreign pathogens. Autophagy of macrophages is used as a major indicator for developing vaccine adjuvants to increase the adaptive immune response. In this study, RAL increased the production of immunostimulatory mediators and phagocytotic activity in RAW264.7 cells. RAL increased p62/SQSTM1 expression. Inhibition of TLR4, JNK, and PI3K/AKT blocked RAL-mediated increase of p62/SQSTM1. RAL activated JNK and PI3K/AKT signaling. RAL-mediated activation of JNK and PI3K/AKT signaling was reversed by TLR4 inhibition. Taken together, it is believed that RAL-mediated autophagy may be dependent on activating via TLR4-dependent activation of JNK and PI3K/AKT signaling in macrophages.

In vitro에서 Quaternary Ammonium Compounds(QACs)이 뱀장어의 면역반응에 미치는 영향 (Effect of Quaternary Ammonium Compounds(QACs) on the Immune Response of eel(Anguilla japonica) In vitro)

  • Quaternary ammonium compounds(QACs)가 뱀장어의 면역반응에 미치는 영향을 측정하기 위해서 뱀장어의 혈액으로부터 림프구를 분리하여 증식능을 측정하였으며 또한 신장으로부터 탐식세포를 분리하여 탐식능, pinocytosis 및 superoxide anion활성능 등을 측정하였다. 림프구의 증식능은 mytogen(T-cell mitogen: ConA and PHA, B-cell mitogen: LPS) 및 QACs처리농도에 따라 별다른 차이를 보이지 않았다. 한편 신장 유래의 대식세포의 세균살해능은 QACs첨가농도에 따라 유의성 있게 증가되었다. 이러한 결과는 QACs이 작동대식세포로의 분화 및 활성을 촉진시켜서 탐식능, pinocytes 및 superoxide생성 등의 분비를 증가시킨 것으로 사료된다. 이상의 결과로 미루어 QACs가 어체의 면역반응에 관여하는 작용기전은 면역작동세포의 수를 증가시키기보다는 세포의 기능활성을 증강시킴을 알 수 있다.

Heat-Killed Lactobacillus plantarum KCTC 13314BP Enhances Phagocytic Activity and Immunomodulatory Effects via Activation of MAPK and STAT3 Pathways

  • Identification of novel probiotic strains is of great interest in the field of functional foods. Specific strains of heat-killed bacteria have been reported to exert immunomodulatory effects. Herein, we investigated the immune-stimulatory function of heat-killed Lactobacillus plantarum KCTC 13314BP (LBP). Treatment with LBP significantly increased the production of $TNF-{\alpha}$ and IL-6 by macrophages. More importantly, LBP was able to enhance the phagocytic activity of macrophages against bacterial particles. Activation of p38, JNK, ERK, $NF-{\kappa}B$, and STAT3 was involved in the immunomodulatory function of LBP. LBP treatment significantly increased production of $TNF-{\alpha}$ by bone marrow-derived macrophages and splenocytes, further confirming the immunostimulatory effect of LBP in primary immune cells. Interestingly, the immunomodulatory effects of LBP were much stronger than those of Lactobacillus rhamnosus GG, a well-known probiotic strain. These results indicate that LBP can be a promising immune-enhancing functional food agent.
