배암차즈기(Salvia plebeia R.) 추출물의 항염 및 항 알레르기 효과 (A Study on the Anti-inflammatory and Anti-allergic Effect of Salvia plebeia R. extracts)

  • The Salvia plebeia R. which is the biennial plant belonging to the Labiatae department, grows naturally in the Korea entire area. Presently, its extract (SPRE) is known to have an anti-inflammation and anti-allergy activity, but there are a few evidences about it. SPRE inhibits pro-inflammatory cytokine such as TNF-$\alpha$ and IL-6 as well as nitric oxide (NO) production in lipopolysaccharide (LPS) treated- macrophages. The co-administration of SPRE during OVA sensitization significantly reduced total IgE levels in mice. The mice who received SPRE co-administered with OVA showed a significant increase in serum OVA-specific IgG2a/b levels. Spleen-cell cultures harvested from OVA-sensitized mice showed a significant decrease in Th2 cytokine levels with a concomitant increase in Th1 cytokine levels only when SPRE co-administered with OVA. These results demonstrate that SPRE can control the LPS-induced inflammatory reaction and prevent antigen-induced Th2 immune responses in mice.

Response of Commercial Cotton Cultivars to Fusarium solani

  • Twenty-nine isolates of Fusarium solani, originally isolated from diseased cotton roots in Egypt, were evaluated for their ability to cause symptoms on four genetically diverse cotton cultivars. Analysis of variance showed highly significant variance among cultivars, and isolates as well as the isolate x genotype interactions were highly significant(p < 0.0001). Although most isolates showed intermediate pathogenicity, there were two groups of isolates that showed significant differences in pathogenicity on all four cultivars. None of the cultivars were found to be immune to any of the isolates. On all cultivars, there were strong significant positive correlations between dry weight and each of preemergence damping-off, survival, and plant height. Considering 75% similarity in virulence, two groups comprising a total of 29 isolates were recognized. Ninety-three percent of the isolates have the same pathogenicity patterns with consistently low pathogenicity, and narrow diversity of virulence. Isolates Fs4 and Fs5 shared the same distinct overall virulence spectrum with consistently high pathogenicity. There was no clear-cut relationship between virulence of the isolates based on reaction pattern on 4 cultivars and each of host genotype, previous crop, and geographic origin.

한우 혈림프절에 관한 형태학적 연구 (Morphological studies on hemolymph nodes in the Korean native cattle)

  • Hemolymph nodes of the Korean native cattle were observed gross anatomically and light microscopically in this study. Hemolymph nodes of the Korean native cattle were found mainly at the periphery of the thoracic and abdominal aortae although there were some other parts to observe these small organs. These organs were small in size and spherical or ovoid in shape, and color of them was dark red or gray in common with red band. The hemolymph nodes were surrounded by a relatively thick connective tissue capsules composed of dense collagen fibers with many scattered smooth muscle fibers and there was a hilus in each node. The parenchyma was divided into an outer cortex and an inner medulla. A few primary and secondary lymph nodules were observed in the cortex and there were diffuse lymphatic tissues among them. The medullary cords consisted of diffuse lymphatic tissues were separated by the medullary sinuses which filled with erythrocytes. The afferent and efferent lymph vessels were observed at the periphery of the capsule and the hilus area, respectively. The stroma of the hemolymph nodes was composed of reticular cells and fibers, and the meshwork of the stroma was filled with many lymphocytes and erythrocytes, and a few macrophages and megakaryocytes etc. These findings suggest that the hemolymph node of the Korean native cattle is involved in blood filtration and immune reaction.

The Effect of Taglisodog-eum Extract on Lipopolysaccharide-induced Otitis media

  • Objectives : It has been known that immune reaction plays an important role in the pathogenesis of otitis media (OM). We investigated the change of middle ear mucosal inflammation induced by lipopolysaccharide (LPS) following administration of Taglisodog-eum (TSE) in experimental animals. Materials and Methods : Otitis media was induced by injecting 1 mg/kg of Pseudomonas aeruginosa LPS transtympanically in 60 rats. These rats were divided into two groups, the LPS group (n=30, remained naive after OM elicitation) and the TSE group (n=30, treated with TSE after OM elicitation) and 6 additional rats were kept as a normal control group (n=6, remained naive until tissue collection). The rats were killed at the 1st, 3rd, and 7th days after challenge. The temporal bones in each group were harvested and examined histopathologically by hematoxyline-eosin stain. Middle ear mucosa were taken at the 1st, 3rd, and 7th days after challenge. The levels of spicing variants of TNF-${\alpha}$ transcription were evaluated by Realtime-PCR. Results : TSE suppressed LPS-induced TNF-${\alpha}$ mRNA expression and thickness of the submucosal layer and infiltration of inflammatory cells in rat middle ear epithelium. Conclusion : The results suggest that TSE may be effective in decreasing inflammation with particular application to mucosal metaplasia in OM.

Antinociceptive, Immunomodulatory and Antipyretic Activity of Nymphayol Isolated from Nymphaea stellata (Willd.) Flowers

  • In the present study, we aimed to analyze the antinociceptive, immunomodulatory and antipyretic activities of nymphayol were investigated in wistar rats and mice. Antinociceptive effect was evaluated by acetic acid induced writhing, formalin induced paw licking and hot-plate tests. Immunomodulatory activity was assessed by neutrophil adhesion test, humoral response to sheep red blood cells, delayed-type hypersensitivity, phagocytic activity and cyclophosphamide induced myelosuppression. Antipyretic activity was evaluated by yeast induced hyperthermia in rats. Nymphayol produced significant (p<0.05) antinociceptive activity in acetic acid induced writhing response and late phase of the formalin induced paw licking response. Pre-treatment with nymphayol (50 mg/kg, oral) evoked a significant increase in neutrophil adhesion to nylon fibres. The augmentation of humoral immune response to sheep red blood cells by nymphayol (50 mg/kg) was evidenced by increase in antibody titres in rats. Oral administration of nymphayol (50 mg/kg) to rats potentiated the delayed-type hypersensitivity reaction induced by sheep red blood cells. Treatment with nymphayol showed a significant (p<0.05) reduction in pyrexia in rats. The results suggest that nymphayol possesses potent anti-nociceptive, immunomodulatory and antipyretic activities.

In Vitro Anti-Inflammatory Effects of the Essential Oil of Artemisia iwayomogi and Its Main Component, Vulgarone B

  • The essential oil fraction was obtained from the leaves and flowers of Artemisia iwayomogi (Compositae) by steam distillation, and its main component, vulgarone B, was isolated by column chromatography. RAW 264.7 cells were used to investigate the anti-inflammatory properties of A. iwayomogi and vulgarone B. Cell viability was determined by MTT assay after treatment with various dilutions of the compounds. In addition, several assays were used to determine the effects of A. iwayomogi essential oil components on immune stimulation. Nitric oxide production in cells activated with lipopolysaccharide (LPS) was evaluated by reaction with Griess reagent. Both vulgarone B and the essential oil fraction of A. iwayomogi inhibited the production of nitric oxide. The effects on various cytokines released from the cells were also measured using ELISA. The production of prostaglandin $E_2$ was significantly decreased by treatment with A. iwayomogi oils. LPS-induced IL-$1{\beta}$ and IL-6 production were also decreased in a dose-dependent manner, but no significant effect on TNF-${\alpha}$ was observed at the concentrations tested. Finally, Western blot analysis revealed that A. iwayomogi oils reduced the levels of COX-2 and iNOS.

  • Inflammatory reaction is characterized by over-production of inflammatory mediators due to an up-regulation of inflammatory pathways, which produce pro-inflammatory mediators, such as interleukin-1beta (IL-$1{\beta}$), IL-6, tumour necrosis factor alpha (TNF-$\alpha$), prostaglantin $E_2$ ($PGE_2$), and nitric oxide (NO) in U937 cells. We investigate the anti-inflammatory effects of water extracts from Lonicerae Flos and Scutellariae Radix in lipopolysaccharide (LPS)-stimulated U937 cells. Each extract suppressed the production of inflammatory mediators (NO, IL-$1{\beta}$, TNF-$\alpha$, and $PGE_2$) and the expression of inducible NO synthase and cyclooxygenase-2 in LPS- stimulated U937 cells in a dose-dependent manner. These suppressive effects were synergistically increased by their combination. Their combination extract also inhibited NF-${\kappa}B$-DNA complex of NF-${\kappa}B$ binding activity and translocation of NF-${\kappa}B$ from cytosol to nucleus. These results suggest that the combination of water-extractable components of Lonicerae Flos and Scutellariae Radix may be useful for therapeutic drugs against inflammatory immune diseases, probably by suppressing the production of inflammatory mediators.

Gene Expression Profile in Microglia following Ischemia-Reperfusion Injury

  • Microglial activation is thought to play a role in the pathogenesis of many brain disorders. Therefore, understanding the response of microglia to noxious stimuli may provide insights into their role in disorders such as stroke and neurodegeneration. Many genes involved in this response have been identified individually, but not systematically. In this regards, the microarray system permitted to screen a large number of genes in biological or pathological processes. Therefore, we used microarray technology to evaluate the effect of oxygen glucose deprivation (OGD) and reperfusion on gene expression in microglia under ischemia-like and activating conditions. Primary microglial cultures were prepared from postnatal mice brain. The cells were exposed to 4 hrs of OGD and 1 h of reperfusion at $37^{\circ}C$. Isolated mRNA were run on GeneChips. After OGD and reperfusion, >2-fold increases of 90 genes and >2-fold decrease of 41 genes were found. Among the genes differentially increased by OGD and reperfusion in microglia were inflammatory and immune related genes such as prostaglandin E synthase, $IL-1{\beta}$, and $TNF-{\alpha}$. Microarray analysis of gene expression may be useful for elucidating novel molecular mediators of microglial reaction to reperfusion injury and provide insights into the molecular basis of brain disorders.

하부식도에서 발생한 GIST 1예 (Esophageal GIST : case report)

  • Currently gastrointestinal mesenchymal tumors are divided into three major categories: myogenic tumors(leiomyoma, leiomyosarcoma), neurogenic tumors (schwannomas) and neoplasms that belong to neither group, which are known by GIST(gastrointestinal stromal tumors). The stromal tumors are hetrogenous, so that they may show myogenic or neurogenic differentiation or both, or no differentiation at all in some patients. The best defining feature for GIST is their expression of KIT-protein(CD117). Leiomyomas are the most common mesenchymal tumor in esophagus. Esophageal GISTS are very rare in comparision to those of the stomach and intestine. Recently we experieneced one case of the esophageal GIST, so that we describe an esophageal GIST on immunohistochemical analysis. A 70 years old woman complained of dysphagia and nausea for 3 days. FGS showed a huge elevated lesion in lower esophagus 33cm distal to incisor, which was covered with normal mucosa. CT and UGI showed the intramural tumor of lower third of the esophagus. The distal esophagectomy and esophago-gastrostomy were performed. The tumor was located in lower third of esophagus and measured as $6{\times}3.7$cm in size. Immunohistochemically, it showed weakly positive CD117 and diffusely positive S-100. SMA, desmin, NES and chromogranin showed negative immune-reaction. The patient was followed for 15 month after operation. There was no recurrence.

Immunomodulating Activity of Fungal ${\beta}-Glucan$ through Dectin-1 and Toll-like Receptor on Murine Macrophage

  • [ ${\beta}-Glucan$ ] is a glucose polymer that has linkage of ${\beta}-(1,3)$, -(1,4) and -(1,6). As exclusively found in fungal and bacterial cell wall, not in animal, ${\beta}-glucans$ are recognized by innate immune system. Dendritic cells (DC) or macrophages possesses pattern recognition molecule (PRM) for binding ${\beta}-glucans$ as pathogen-associated molecular pattern (PAMP). Recently ${\beta}-glucans$ receptor was cloned from DC and named as dectin-l which belongs to type II C-type lectin family. Human dectin-l is consisted of 7 exons and 6 introns. The polypeptide of dectin-l has 247 amino acids and has cytoplasmic, transmembrane, stalk and carbohydrate recognition domains. Dectin-l could recognize variety of beta-l,3 and/or beta-l,6 glucan linkages, but not alpha-glucans. In our macrophage cell line culture system, dectin-l mRNA was detected in RA W264.7 cells by reverse transcription-polymerase chain reaction (RT-PCR). Dectin-l was also detected in the murine organs of spleen, thymus, lung and intestines. Treatment of RA W264.7 cells with ${\beta}-glucans$ of Ganoderma lucidum (GLG) resulted in increased expression of IL-6 and $TNF-{\alpha}$ in the presence of LPS. However, GLG alone did not increase IL-6 nor $TNF-{\alpha}$ These results suggest that receptor dectin-l cooperate with CD14 to activate signal transduction that is very critical in immunoresponse.
