Error-immune Algorithm for Absolute Testing of Rotationally Asymmetric Surface Deviation

  • Based on Zernike polynomial fitting, we propose an algorithm believed to be new for interferometric measurements of rotationally asymmetric surface deviation of optics. This method tests and calculates each angular surface by choosing specified rotation angles with lowest error. The entire figure can be obtained by superimposing these sub-surfaces. Simulation and experiment studies for verifying the proposed algorithm are presented. The results show that the accuracy of the proposed method is higher than single-rotation algorithm and almost comparable to the rotation-averaging algorithm with fewer rotation measurements. The new algorithm can achieve a balance between the efficiency and accuracy.

Human CD8+ T-Cell Populations That Express Natural Killer Receptors

  • CD8+ T cells are activated by TCRs that recognize specific cognate Ags, while NK-cell activation is regulated by a balance between signals from germline-encoded activating and inhibitory NK receptors. Through these different processes of Ag recognition, CD8+ T cells and NK cells play distinct roles as adaptive and innate immune cells, respectively. However, some human CD8+ T cells have been found to express activating or inhibitory NK receptors. CD8+ T-cell populations expressing NK receptors straddle the innate-adaptive boundary with their innate-like features. Recent breakthrough technical advances in multi-omics analysis have enabled elucidation of the unique immunologic characteristics of these populations. However, studies have not yet fully clarified the heterogeneity and immunological characteristics of each CD8+ T-cell population expressing NK receptors. Here we aimed to review the current knowledge of various CD8+ T-cell populations expressing NK receptors, and to pave the way for delineating the landscape and identifying the various roles of these T-cell populations.

  • The world has witnessed unimaginable damage from the coronavirus disease-19 (COVID-19) pandemic. Because the pandemic is growing rapidly, it is important to consider diverse treatment options to effectively treat people worldwide. Since the immune system is at the hub of the infection, it is essential to regulate the dynamic balance in order to prevent the overexaggerated immune responses that subsequently result in multiorgan damage. The use of stem cells as treatment options has gained tremendous momentum in the past decade. The revolutionary measures in science have brought to the world mesenchymal stem cells (MSCs) and MSC-derived exosomes (MSC-Exo) as therapeutic opportunities for various diseases. The MSCs and MSC-Exos have immunomodulatory functions; they can be used as therapy to strike a balance in the immune cells of patients with COVID-19. In this review, we discuss the basics of the cytokine storm in COVID-19, MSCs, and MSC-derived exosomes and the potential and stem-cell-based ongoing clinical trials for COVID-19.

Effects of Chronic Inflammation on Energy Metabolism and Growth Performance in Weanling Piglets

  • The effect of a chronic inflammation (cell-mediated immune response) on energy metabolism and growth performance was assessed in weanling piglets. Twenty four barrows of 4 wk of age were assigned to one of two immunization treatments : Control group [CON: immunized with Incomplete Freund's Adjuvant (lFA)] or Immunization group [IMMU: immunized with Complete Freund's Adjuvant (CFA)]. On d0, piglets were weaned and subcutaneously immunized at the medial side of the femur with 2 ml of IFA or CFA, respectively. Energy and nitrogen balances were measured per group during 13-d balance period, and total $(HP_{tot})$, activity-related ($(HP_{act})$) and non-activity-related $(HP_{cor})$ heat production were determined every 9-min by indirect calorimetry. Ig total titers to Mycobacterium butyricum, which is present in CFA, were higher (p<0.01) in IMMU than in CON on d13 (2.5 vs 1.8) and d20 (2.9 vs 1.8) after immunization. There were no differences (p>0.10) between treatments in rectal temperature, performance, feed intake, and availability and partitioning of energy during the balance period. Average daily feed intake was numerically higher in IMMU than in CON (0.34 vs 0.32 kg/d), but there was no difference (p>0.10) in metabolizability of the dietary energy between treatments. $HP_{act}/HP_{tot}$ was 16.24 and 16.89%, and retained energy was 251 and 268 $268\;kJ{\cdot}kg^{0.75}{\cdot}d^{-1}$ for CON and IMMU, respectively. Numerically, maintenance requirement of IMMU was even lower than that of CON $(419\;vs\;427\;kJ{\cdot}kg^{0.75}{\cdot}d^{-1})$. The present study suggests that a chronic inflammation has no effect on energy metabolism and growth performance, in spite of the difference in systemic antibody responses. The reason was considered to be due to locally induced immune response, resulting from the possible encapsulation at the site of injection, and/or to a low systemic immune stress which is within a functionally acceptable physiological range for the piglets.

  • Allergy 질환은 지난 십여년 동안 개발도상국을 포함해서 전 세계적으로 증가하고 있다. Allergy 염증 반응은 수지상 세포와 같은 항원제시 세포에 의한 allergy 항원섭취를 시작으로 하여 Th2 면역 반응에 의해서 일어난다. 장내 미생물은 신체의 대사나 생리적 기능을 조절하고, 생애 초기의 면역 체계 성숙과 일생 동안 면역 체계 항상성 및 상피세포 총체성에 기여한다. Bifidobacteria는 Th1/Th2 balance에 strain-specific 한 면역 자극 특성을 가지며, TSLP와 IgE 발현을 억제 시키고 Flg과 FoxP3 발현을 촉진 시켜 allergy를 완화시킨다. 또한 Unmethylated CpG motif ODN은 B 세포와 수지상 세포의 TLR9에 의해 인식 되어 선천성과 적응성 면역 반응을 유도하고, Clostridium butyricum에 의해서 생산된 butyrate는 수지상 세포의 anti-inflammatory 유전자의 발현을 유도하기 위해 GPR109a signaling pathway를 활성화시키고, GPR43 활성화를 통하여 tTreg 세포 proliferation을 직접 자극하거나 HADC 활성을 억제시켜 Foxp3 gene intronic enhancer의 histone H3 acetylation을 통해 naive $CD4^+$ T 세포를 pTreg 세포로 분화시킨다.

Molecular Characterization and Expression Pattern of Na+-K+-2Cl- Cotransporter 2 (NKCC2) in the Intestine of Starry Flounder Platichthys stellatus after Bacterial Challenge

  • We identified the $Na^+-K^+-2Cl^-$ cotransporter 2 (NKCC2) cDNA isoform from starry flounder, Platichthys stellate. The NKCC2 cDNA encoded a polypeptide of 1,043 amino acids representing 12 putative transmembrane domains based on the bioinformatic topology prediction. In addition, starry flounder NKCC2 possessed highly conserved residues within transmembrane domain 4, known as an essential site for its function. End-point reverse transcription-polymerase chain reaction analysis revealed that the NKCC2 transcript was moderately expressed only in the anterior and posterior intestines and the rectum. The NKCC2 mRNA level in the rectum, but not in other segments, was significantly induced 3 days post Streptococcus parauberis challenge, indicating that excess salt may be transported into the rectum. Taken together, our data indicate that an S. parauberis infection could tip the intestinal fluid balance in favor of fluid accumulation, indicating that bacterial pathogens can interfere with intestinal osmotic balance and normal mucosal immune homeostasis.

Anti-inflammatory Effects and its Mechanisms of Hesperidin in an Asthmatic Mouse Model Induced by Ovalbumin

  • Hesperidin, a member of the flavanone group of flavonoids, can be isolated in large amounts from the rinds of some citrus species [e.g., Citrus aurantium L. (bitter orange), Citrus sinensis L. (sweet orange) and Citrus unshiu Marcov. (satsuma mandarin)], and has been reported to have anticarcinogenic, antihypotensive and antimicrobial properties. Despite the efficacy of these polyphenolic compounds as immune modulators, the effects of the flavonoids are poorly understood about allergic effect. In this study, we investigated whether hesperidin could influence on Th1 and Th2 balance. Allergic reactions included an increase in the number of eosinophils in bronchoalveolar lavage (BAL) fluid, an increase in inflammatory cell infiltration into the lung tissue around blood vessels and airways, airway luminal narrowing, the development of airway hyper-responsiveness (AHR). The administration of hesperidin before the last airway OVA challenge resulted in a significant inhibition of all asthmatic reactions. Accordingly, this study may provide evidence that hesperidin plays a critical role in the amelioration of the pathogenetic process of asthma in mice. These findings provide new insight into the immunopharmacological role of hesperidin in terms of its effects in a murine model of asthma, and also broaden current perspectives in our understanding of the immunopharmacological functions of hesperidin.

Effects of Rubus coreanus byproducts on intestinal microbiota and the immune modulation

  • Objective: Although the efficacy of Rubus coreanus (RC) byproducts as a feed additive has been recognized, its effects on intestinal microorganisms and the immune system are still unknown. Methods: Six-week-old male rats were treated with 0.5% RC (T1), 1.0% RC (T2), and 1.5% RC (T3) for 4 weeks. Results: We found that treatment with RC byproducts significantly increased the daily gain of body weight and feed intake. Treg-cell differentiation was enhanced in the mesenteric lymph nodes and spleen from the rats fed with RC byproducts. Illumina sequencing showed that bacteria in the phylum Firmicutes decreased and while those in the phylum Bacteroidetes increased in RC-treated groups. Particularly, the pathogenic microorganisms in the family Peptococcaceae decreased, and the non-pathogenic families Lachnospiraceae and S24-7 increased. Quantitative polymerase chain reaction analysis showed that the RC byproducts increased the lactic acid bacteria Bifidobacterium spp., Oscillospira spp., Leuconostoc citreum, and Weissella cibaria in a concentration-dependent manner. Conclusion: RC byproducts may be effective in immunomodulation by affecting intestinal microorganisms.

타고난 면역반응이 활성화된 육계병아리의 생산성과 혈액 항산화계에 미치는 사료중 미역제품 수준의 영향

  • 사료중 미역제품 수준이 타고난 면역반응을 활성화한 육계병아리의 생산성과 항산화계에 미치는 영향을 조사하였다. LPS를 주입하여 타고난 면역반응을 활성화하였다. 미역제품 2.0 % 사료는 뇨산태 질소의 배설량을 감소시켜 질소밸런스와 사료효율을 유의하게 증가시켰다(P<0.05), 미역제품 1.0 % 사료는 타고난 면역 활성화시의 생산성 감소를 완화시켰다. 미역제품 2.0 % 사료는 혈장 SOD의 활성을 낮추었다. 그러나 타고난 면역의 활성화는 적혈구 세포액의 SOD 활성과 혈장 peroxide 수준을 유의하게 높였다. 미역제품 사료는 과산화물분해효소의 활성을 유의하게(P<0.05)높였다. 본 성적은 육계 병아리에서 미역제품 2.0 % 사료는 단백질 분해량을 감소시킴으로써 단백질 축적량을 높여서 육계병아리의 생산성을 증가시키는 것을 나타내었다. 그리고 미역제품 2.0 % 사료 급여시 급성기 반응 및 정상 병아리의 항산화계효소 활성 감소, 생산성의 증가는 혈액 항산화계의 변화와 연계된다는 것을 나타내었다.

Association of Intracellular $T_H1-T_H2$ Balance in CD4+ T-cells and MIP-$1{\alpha}$ in CD8+ T-cells with Disease Severity in Adults with Dengue

  • Background: We tested the hypothesis that dengue haemorrhagic fever (DHF) is associated with a $T_H1$-skewed immune response as opposed to dengue fever (DF). Methods: We estimated intracellular (in T-cells) and serum levels of designate $T_H1/T_H2$ cytokines [interferon-${\gamma}$ (IFN-${\gamma}$), interleukin-4 (IL-4), and tumor necrosis factor-${\alpha}$] and macrophage inflammatory protein-$1{\alpha}$ (MIP-$1{\alpha}$) at admission, 48h, and day 5 in 20 adults with dengue (DF=10, DHF=10) and 10 dengue-naive healthy controls. Results: At admission, intracellular IFN-${\gamma}$/IL-4 ratio in CD4+ T-cells and proportion of MIP-$1{\alpha}$-positive CD8+ T-cells were significantly higher in patients with DHF [7.21 (5.36~10.81) vs. 3.04 (1.75~4.02); p=0.011 and 6.2% (3.2~8.2%) vs. 2.4% (2.0~3.6%); p=0.023]. The latter showed a significant positive correlation with IFN-${\gamma}$/IL-4 ratio in CD4+ T-cells (Spearman's rho=0.64; p=0.003), percentage-change in haematocrit (rho=0.47; p=0.048), and serum alanine amino-transferase level (rho=0.61; p=0.009). Conclusion: We conclude that DHF is associated with a $T_H1$-skewed immune response. Further, MIP-$1{\alpha}$ in CD8+ T-cells is an important immunologic correlate of disease severity in dengue.
