생지화학적 환원조건에서 우라늄의 침철석 및 몬모릴로나이트에 대한 수착 특성 (Sorption Characteristics of Uranium on Goethite and Montmorillonite under Biogeochemical Reducing Conditions)

  • 산화/환원 반응에 매우 민감한 우라늄의 침철석 및 몬모릴로나이트에 대한 수착 특성을 알아보기 위해 산화우라늄(VI)과 환원우라늄(IV)를 준비하였다. 환원우라늄은 황산염환원박테리아에 의해 황산염이 환원되는 과정에서 같이 환원된 우라늄(IV)를 희석하여 사용하였다. 광물에 대한 우라늄의 수착량은 우라늄(IV)가 우라늄(VI)에 비해 상대적으로 낮았으며, 이러한 원인 중의 하나는 용액상의 우라늄(IV)가 미세한 콜로이드 형태로 존재하여 광물 표면에 대한 수착력이 약했기 때문이다. 투과전자현미경을 사용하여 우라늄(IV)가 나노 콜로이드의 특징을 가지고 있음을 확인하였고, 이러한 결과는 심부 자연계의 지하수를 따라 이동 가능한 우라늄종이 이온성 우라늄(VI)뿐만 아니라 콜로이드성 우라늄(IV)도 포함될 수 있음을 의미한다.

ESP 프로토콜에서의 문제점 보완 알고리즘 (The problem resolution algorithm in ESP protocol)

  • IPSec은 공용 네트워크 상에서 암호화와 인증, 무결성을 제공하기 위해 사용되는 프로토콜이다. IPSec에서는 전송되는 패킷에 암호화와 인증, 무결성을 추가해 전달하기 위해 ESP프로토콜을 사용한다. ESP는 패킷 암호화를 위해 DES-CBC 알고리즘을 이용하는데 이 모드에서는 ESP프로토콜로 보호되어 전송되는 데이터를 암호화하기 위한 초기 값으로 IV(Initialization Vector) 값을 사용한다. 이 값은 수신 측의 패킷 복호화를 위해 공개적으로 전달되므로 중간에 공격자에 의해 공격당할 위험이 많다. IV의 값이 조금이라도 변경되면 ESP의 모든 데이터의 복호화가 이루어지지 않고 상위 레벨의 정보가 변경되는 통 심각한 문제가 발생한다. 이것은 보안을 목적으로 하는 IPSec에서는 치명적인 약점이 된 수 있다. 따라서 본 논문에서는 ESP프로토콜에서의 문제점인 IV를 안전하게 전송하기 위한 새로운 알고리즘을 제시한다. 이 방법은 IV 값을 DES-ECB 알고리즘을 이용하여 암호화하여 IV 공격을 방어하고 메시지 인증 함수를 추가 적용하여 ESP 전체 데이터의 무결성 체크도 가능하게 한다. 제안된 알고리즘으로 IV와 데이터에 대한 공격을 방어할 수 있고 안전한 전송을 보장한다.

Astragaloside IV Prevents Obesity-Associated Hypertension by Improving Pro-Inflammatory Reaction and Leptin Resistance

  • Low-grade pro-inflammatory state and leptin resistance are important underlying mechanisms that contribute to obesity-associated hypertension. We tested the hypothesis that Astragaloside IV (As IV), known to counteract obesity and hypertension, could prevent obesity-associated hypertension by inhibiting pro-inflammatory reaction and leptin resistance. High-fat diet (HFD) induced obese rats were randomly assigned to three groups: the HFD control group (HF con group), As IV group, and the As IV + ${\alpha}$-bungaratoxin (${\alpha}-BGT$) group (As IV+${\alpha}-BGT$ group). As IV ($20mg{\cdot}Kg^{-1}{\cdot}d^{-1}$) was administrated to rats for 6 weeks via daily oral gavage. Body weight and blood pressure were continuously measured, and NE levels in the plasma and renal cortex was evaluated to reflect the sympathetic activity. The expressions of leptin receptor (LepRb) mRNA, phosphorylated signal transducer and activator of transcription-3 (p-STAT3), phosphorylated phosphatidylinositol 3-kinase (p-PI3K), suppressor of cytokine signaling 3 (SOCS3) mRNA, and protein-tyrosine phosphatase 1B (PTP1B) mRNA, pro-opiomelanocortin (POMC) mRNA and neuropeptide Y (NPY) mRNA were measured by Western blot or qRT-PCR to evaluate the hypothalamic leptin sensitivity. Additionally, we measured the protein or mRNA levels of ${\alpha}7nAChR$, inhibitor of nuclear factor ${\kappa}B$ kinase subunit ${\beta}/nuclear$ factor ${\kappa}B$ ($IKK{\beta}/NF-KB$) and pro-inflammatory cytokines ($IL-1{\beta}$ and $TNF-{\alpha}$) in hypothalamus and adipose tissue to reflect the anti-inflammatory effects of As IV through upregulating expression of ${\alpha}7nAChR$. We found that As IV prevented body weight gain and adipose accumulation, and also improved metabolic disorders in HFD rats. Furthermore, As IV decreased BP and HR, as well as NE levels in blood and renal tissue. In the hypothalamus, As IV alleviated leptin resistance as evidenced by the increased p-STAT3, LepRb mRNA and POMC mRNA, and decreased p-PI3K, SOCS3 mRNA, and PTP1B mRNA. The effects of As IV on leptin sensitivity were related in part to the up-regulated ${\alpha}7nAchR$ and suppressed $IKK{\beta}/NF-KB$ signaling and pro-inflammatory cytokines in the hypothalamus and adipose tissue, since co-administration of ${\alpha}7nAChR$ selective antagonist ${\alpha}-BGT$ could weaken the improved effect of As IV on central leptin resistance. Our study suggested that As IV could efficiently prevent obesityassociated hypertension through inhibiting inflammatory reaction and improving leptin resistance; furthermore, these effects of As IV was partly related to the increased ${\alpha}7nAchR$ expression.

  • [M(L/L')$(NO_3)_n$].$mH_2O$ 및 [VO(L/L')($SO_4)$].$2H_2O$ (여기서 L/L'는 Schiff 염기로 thiocarbohydrazide (TCH), benzilmonohydrazone (BMH)/diacetylmonohydrazone (DMH) 및 carbon disulphide에서 유도된 "3,4,10,11-tetraphenyl/tetramethyl-1,2,5,6,8,9,12,13-octaazacyclotetradeca-2,4,9,11-tetraene-7,14-dithione")의 분자식을 갖는 일련의 거대고리 착물을 (M = $UO_2$(VI), Th(IV) and ZrO(IV), n = 2, 4, m = 2, 3)을 금속이온 주형법으로 합성하였다. 이들 착물의 특성을 원소분석, 열분석, 몰 전기전도도, 자기모멘트 그리고 전자, 적외선 및 $^1H$-NMR로 조사하였다. 바나딜 착물의 경우 ESR 및 순환 전압전류법을 사용하였다. 그 결과, VO(IV) 이온은 5배위의 상자기성 착물을 이루며, $UO_2$(VI) 및 ZrO(IV)는 6배위 그리고 Th(IV)는 8배위를 가지나 공통적으로 위의 조성을 갖는 반자기성임을 알았다.

XAD-16-Chromotropic Acid 킬레이트 수지를 이용한 몇 가지 금속이온의 흡착거동에 관한 연구 (Studies on the Sorption Behavior of Some Metal Ions using XAD-16-Chromotropic Acid Chelating Resin)

  • XAD-16-Chromotropic acid (CTA)형 킬레이트 수지에 대한 몇 가지 금속이온의 흡착특성을 뱃치법과 용리법으로 조사하였다. pH 변화에 따른 금속 이온의 흡착율을 조사한 결과 pH 3.0 ~ 6.0에서 Hf(IV), Zr(IV), Th(IV) 이온이 다른 금속 이온들에 비해 높은 선택성을 보였으며, Zr(IV)의 최대흡착용량은 0.81 mmol/g이었다. 가리움제로서 CDTA, EDTA, NTA 및 $NH_4F$ 등을 사용하여 흡착에 미치는 영향을 조사한 결과 혼합 금속 용액으로부터 몇 가지 희토류 금속 이온들의 분리가 가능함을 알 수 있었다. 킬레이트 수지에 대한 몇 가지 금속 이온들의 흡착거동을 용리법으로 조사 검토한 결과 pH 4에서 돌파점 용량과 총괄 용량으로부터 얻은 금속 이온의 용리 순서는 Zr(IV)>Th(IV)>Hf(IV)>U(VI)>Cu(II)>In(III)>Pb(II) 이었다. 한편 HCl, $HNO_3$, $HClO_4$ 등의 탈착제에 의한 탈착 특성을 조사한 결과 2 M HCl에서 높은 탈착효율을 나타내었다. XAD-16-CTA 킬레이트 수지는 혼합 금속이온 중에 함유된 Th(IV) 이온의 선택적 분리 및 회수에 유용함을 알 수 있었다.

  • 염산 산성(1∼4N) 용액에서 Zr(IV)-XO 착물의 흡광도가 옥살산의 첨가량에 반비례하는 현상을 이용하여 미량의 옥살산을 정량하는 방법을 확립하였다. 말산, 말론산, 말레산, 후마르산, 숙신산, 엽산, 글루타민산은 옥살산량의 100배가 공존하여도 미량의 옥살산을 정량하는 데 방해하지 않지만 타르타르산과 시트르산은 옥살산 보다 많으면 방해한다.

Anti-effects of Photodynamic Therapy in Peroxiredoxin IV-induced AMC-HN3 Cell Lines

  • Photodynamic therapy (PDT) is a treatment utilizing the generation of singlet oxygen and other reactive oxygen species (ROS), which selectively accumulate in target cells. Peroxiredoxin (prx) plays an important role in eliminating peroxides generated during metabolism. Prx exert protective antioxidant role in cells though peroxidase activity. The aim of present work is to investigate the cytotoxicity of photofrin-mediated PDT in prx IV-transfectant AMC-HN3 cell lines. We confirmed that PDT has an effect on ROS generation in prx IV-induced cell lines. Treatment of PDT in prx IV-HN3 cell lines inhibits cytotoxic effects. Prx IV-induced HN3 cell lines resists in cell death during PDT. Also, prx IV-HN3 cell lines treated PDT inhibited ROS generation in contrast with vector control. We indicated that prx IV-induced AMC-HN3 cell lines have a function as inhibitors during PDT.

한국 웩슬러 유아지능검사 4판(K-WPPSI-IV)의 지능구조에 관한 연구 (A Study on the Structure of Intelligence Measured by the K-WPPSI-IV)

  • Objective: This study examined the construct validity of K-WPPSI-IV. Factor structures of the structures of the K-WPPSI-IV full scale as well as primary index scales for two age ranges (2 years, 6 months to 3 years, 11 months; 4 years to 7 years, 7 months) were examined. Methods: Data were collected from 1,700 children aged 2 years, 6 months to 7 years, 7 months during the K-WPPSI-IV standardization. Confirmatory factor analyses were conducted using the K-WPPSI-IV subtest performances with maximum likelihood estimation using Amos 18. Results: First, the three-factor model (verbal comprehension, visual spatial, and working memory) fitted best for the younger age range. However, the five-factor model (verbal comprehension, visual spatial, fluid reasoning, working memory, and processing speed) fitted best for the older age range. Residuals suggest the presence of two nested subfactors within the verbal comprehension factor (broad/expressive and focused/simple). Second, the confirmatory factor analysis on primary index subtests identified factors that account for the intercorrelations among the reduced sets of primary index subtests. Conclusion: The findings showed that the theoretical structures of WPPSI-IV subtests were confirmed within K-WPPSI-IV.

Cloning, Expression, and Characterization of Endoglucanase Gene egIV from Trichoderma viride AS 3.3711

  • Endoglucanase gene egIV was cloned from Trichoderma viride AS 3.3711, an important cellulose-producing fungus, by using an RT-PCR protocol. The egIV cDNA is 1,297 bp in length and contains a 1,035 bp open reading frame encoding a 344 amino acid protein with an estimated molecular mass of 35.5 kDa and isoelectronic point (pI) of 5.29. The expression of gene egIV in T. viride AS 3.3711 could be induced by sucrose, corn straw, carboxymethylcellulose (CMC), or microcrystalline cellulose, but especially by CMC. The transcripts of egIV were regulated under these substrates, but the expression level of the egIV gene could be inhibited by glucose and fructose. Three recombinant vectors, pYES2-xegIV, $pYES2M{\alpha}$-egIV, and $pYES2M{\alpha}$-xegIV, were constructed to express the egIV gene in Saccharomyces cerevisiae H158. The CMCase activity of yeast transformants $IpYES2M{\alpha}$-xegIV was higher than that of transformant IpYES2-xegIV or $IpYES2M{\alpha}$-egIV, with the highest activity of 0.13 U/ml at induction for 48 h, illustrating that the modified egIV gene could enhance CMCase activity and that $MF{\alpha}$ signal peptide from S. cerevisiae could regulate exogenous gene expression more effectively in S. cerevisiae. The recombinant EGIV enzyme was stable at pH 3.5 to 7.5 and temperature of $35^{\circ}C$ to $65^{\circ}C$. The optimal reaction condition for EGIV enzyme activity was at the temperature of $55^{\circ}C$, pH of 5.0, 0.75 mM $Ba^{2+}$, and using CMC as substrate. Under these conditions, the highest activity of EGIV enzyme in transformant $IpYES2M{\alpha}$-xegIV was 0.18 U/ml. These properties would provide technical parameters for utilizing cellulose in industrial bioethanol production.

Recanalization Rate and Clinical Outcomes of Intravenous Tissue Plasminogen Activator Administration for Large Vessel Occlusion Stroke Patients

  • Objective : Stroke caused from large vessel occlusion (LVO) has emerged as the most common stroke subtype worldwide. Intravenous tissue plasminogen activator administration (IV-tPA) and additional intraarterial thrombectomy (IA-Tx) is regarded as standard treatment. In this study, the authors try to find the early recanalization rate of IV-tPA in LVO stroke patients. Methods : Total 300 patients undertook IA-Tx with confirmed anterior circulation LVO, were analyzed retrospectively. Brain computed tomography angiography (CTA) was the initial imaging study and acute stroke magnetic resonance angiography (MRA) followed after finished IV-tPA. Early recanalization rate was evaluated by acute stroke MRA within 2 hours after the IV-tPA. In 167 patients undertook IV-tPA only and 133 non-recanalized patients by IV-tPA, additional IA-Tx tried (IV-tPA + IA-Tx group). And 131 patients, non-recanalized by IV-tPA (IV-tPA group) additional IA-Tx recommend and tried according to the patient condition and compliance. Results : Early recanalization rate of LVO after IV-tPA was 12.0% (36/300). In recanalized patients, favorable outcome (modified Rankin Scale, 0-2) was 69.4% (25/36) while it was 32.1% (42/131, p<0.001) in non-recanalized patients. Among 133 patients, non-recanalized after intravenous recombinant tissue plasminogen activator and undertook additional IA-Tx, the clinical outcome was better than not undertaken additional IA-Tx (favorable outcome was 42.9% vs. 32.1%, p=0.046). Analysis according to the perfusion/diffusion (P/D)-mismatching or not, in patient with IV-tPA with IA-Tx (133 patients), favorable outcome was higher in P/D-mismatching patient (52/104; 50.0%) than P/D-matching patients (5/29; 17.2%; p=0.001). Which treatment tired, P/D-mismatching was favored in clinical outcome (iv-tPA only, p=0.008 and IV-tPA with IA-Tx, p=0.001). Conclusion : The P/D-mismatching influences on the recanalization and clinical outcomes of IV-tPA and IA-Tx. The authors would like to propose that we had better prepare IA-Tx when LVO is diagnosed on initial diagnostic imaging. Furthermore, if the patient shows P/D-mismatching on MRA after IV-tPA, additional IA-Tx improves treatment results and lessen the futile recanalization.
