Yeast Two-hybrid System을 이용한 cTPx II 결합단백질 탐색 및 분석 (Screening and Analysis for cTPx II-Interacting Protein Using Yeast Wo-hybrid System)

  • 효모에는 여러 가지 종류의 thiol peroxid se 동위 효소들인 cytoplasmic TPx I, cTPx II, cTPx III, mitochodrial TPx (mTPx), 및 nuclear TPx (nTPx)가 존재하고 있다. 특히 cTPx II는 다른 효모TPx와 비교해 볼 때 매우 낮은 peroxidase 활성을 보이나, cTPx II를 제거한 cTPx II mutant균주는 심하게 성장이 저해되는 특징을 보인다. 본 연구에서는 효모에서의 cTPx II의 생리학적 기능을 밝히는 연구의 첫 과정으로 cTPx II와 상호 작용하는 단백질을 탐색하였다. Sacchromyces cerevisiae genomic DNA library에서 yeast two-hybrid system을 이용하여 cTPx II와 상호 결합하는 단백질을 탐색하여 그 단백질들의 기능을 연구하여, 궁극적으로 cTPx II의 생리기능을 밝히는데 이 연구의 목적을 두었다.

Selective Cytotoxicity of Novel Platinum(II) Coordination Complexes on Human Bladder Cancer Cell-Lines and Normal Kidney Cells

  • Cisplatin is often effective in cancer treatment, but its clinical use is limited because of its nephrotoxicity. We have synthesized new platinum(II) coordination complexes (PC-1 & PC-2) containing trans-${\iota}$ and cis-1,2-diaminocyclohexane (DACH) as carrier ligands and L-3 -phenyllactic acid (PLA) as a leaving group with the aim of reducing nephrotoxicity but maintaining its anticancer activity. In this study, new platinum(II) complex compounds were evaluated for selective cytotoxicity on cancer cell-lines and normal kidney cells. The new platinum complexes have demonstrated high efficacy in the cytotoxicity against human bladder carcinoma cell-lines (T-24/HT-1376). The cytotoxicity of these compounds against rabbit proximal renal tubular cells and human renal cortical tissues, was determined by MTT assay, the [3H]-thymidine uptake and glucose consumption test, and found to be quite less than those of cisplatin. Based on our results, these novel platinum compounds appear to be valuable lead compounds with high efficacy and low nephrotoxicity.

Hematopoietic Cell Transplantation in Patients with Mucopolysaccharidosis Type II

  • Mucopolysaccharidosis type II (MPS II, Hunter syndrome) is an X-linked lysosomal storage disorder caused by deficiency of the enzyme iduronate-2-sulfatase, leading to the accumulation of glycosaminoglycans (GAGs), which affects multiple organs and systems. Current treatments for MPS II include enzyme replacement therapy (ERT) and hematopoietic cell transplantation (HCT) to reduce the accumulation of GAGs. HCT has the potential advantage that donor-derived enzyme-competent cells can provide a continuous secreting source of the enzyme. However, HCT as a treatment for MPS II remains controversial because its effectiveness is unclear, particularly in terms of neurological symptoms. To date, several clinical experiences with HCT in MPS II have been reported. In this paper, we review post-HCT outcomes in the previously published literature and discuss the effects of HCT on each of the clinical signs and symptoms of MPS II.

Pharmacophore Modeling of Angiotensin-Ⅱ from Study of Its Nonpeptidic Antagonists

  • Early attempts to identify plausible conformations of a linear octapeptide hormone, angiotensin-II (Asp-Arg-Val-Tyr-Ile-His-Pro-Phe), using various theoretical and experimental methods, have led to various conformational models. So far, no consensus has been made about the solution phase structure and the receptor binding structure of angiotensin-II. The ultimate goal for the conformation study of the peptide hormone is to develop a new potent drug. Therefore, we have devised a strategy for designing the pharmacophore by studying thermodynamically possible conformations of various kinds of angiotensin-II antagonists and angiotensin-II.

피리딘을 포함하는 여섯자리 $N_6$ 리간드와 전이금속 이온 착물의 안정도상수 (Stability Constants for Transition Metal ions of Hexadentate $N_6$ Ligands Containing Two Pyridyl Groups)

  • 두 개의 피리딘기를 포함하는 열린고리 형태의 여섯자리 질소계 리간드 1,15-bis(2-pyridyl)-2,6,10,14-tetraazapentadecane tetrahydroehloride (bpyped 4HCl)을 합성하고 원소분석, 적외선 분광법, 핵자기 공명법 및 질량 스펙트럼으로 분석하였다. 합성된 리간드의 산 해리상수($logK{^n}_H$)와 Cu(II), Ni(II), Co(II) 및 Zn(II) 에 대한 안정도 상수값을 수용액에서 전위차 적정법으로 구하였고, 이때 온도는 $25^{\circ}C$ 및 이온강도 0.10 M($KNO_3$)으로 고정시켰다. 지방족 아민의 질소 원자들 사이에 한 개 또는 두 개의 프로필기를 포함하는 bpyped와 유사한 리간드들의 지방족 아민 고리 길이 효과에 따른 산 해리 상수 및 전이금속(II)에 대한 안정도 상수 영향을 알아보았다.

Electrocatalytic Reduction of CO2 by Copper (II) Cyclam Derivatives

  • This study investigates Cu(II) complexes of cyclam, propylene cross-bridged cyclam (PCB-cyclam), and propylene cross-bridged cyclam diacetate (PCB-TE2A) as homogeneous electrocatalysts for CO2 reduction in comparison with Ni(II)-cyclam. It is found that Cu(II)-cyclam can catalyze CO2 reduction at the potential close to its thermodynamic value (0.75 V vs. Ag/AgCl) in tris-HCl buffer (pH 8.45) on a glassy carbon electrode. Cu(II)-cyclam, however, suffers from severe demetalation due to the insufficient stability of Cu(I)-cyclam. Cu(II)-PCB-cyclam and Cu(II)-PCB-TE2A are revealed to exhibit much less demetalation behavior, but poor CO2 reduction activities as well. The inferior electrocatalytic ability of Cu(II)-PCB-cyclam is ascribed to its redox potential that is too high for CO2 reduction, and that of Cu(II)-PCB-TE2A to the steric hindrance preventing facile contact with CO2 molecules. This study suggests that in addition to the redox potential and chemical stability, the stereochemical aspect has to be considered in designing efficient electrocatalysts for CO2 reduction.

TiO2 광측매를 이용한 Cu(II)-EDTA의 산화에서 용존산소와 Cu(II)와 EDTA 초기 당량의 영향 (The effect of dissolved oxygen and initial complextation of Cu(II) and EDTA on photooxidation of Cu(II)-EDTA by TiO2)

  • The effects of initial concentration of dissolved oxygen content, Cu(II) and EDTA in an aqueous Cu(II)-EDTA solution on $TiO_2$ photo-oxidation of EDTA were investigated using $TiO_2$ (Degussa P-25) and UV irradiation at $20{\circ}C$. In the presence of dissolved oxygen and/or Cu(II) the photo-oxidation rates of EDTA were enhanced. The rates linearly increased in the range of initial Cu(II) concentration below 1.79 mM, while abode this concentration those were kept constant. The trend or the EDTA photo-oxidation rates appeared to be akin to the Langmuir-Hinshelwood equation farm and the k values calculated were 0.05 mM/min for the free-EDTA system, and 0.17 mM/min far the Cu(II)-EDTA system. These meant the aqueous EDTA decomposition was enhanced due to weakening of the intra-molecular bond strength of EDTA by complexation with Cu(II) added. It was concluded the decomposition of aqueous EDTA by $TiO_2$ photo-oxidation was maximum in the presence of dissolved oxygen supplied by air purging and of Cu(II) with its concentration for 1:1 Cu(II)-EDTA complexation ratio.

양이온교환수지에 대한 Co(II), Ni(II), Cr(III), Fe(III) 이온의 흡착 특성 - 원자로 일차 냉각재 계통내 탈염 공정에의 적용 (Adsorption Characteristics of Co(II), Ni(II), Cr(III) and Fe(III) Ions onto Cation Exchange Resin - Application to the Demineralizing Process in a Primary Coolant System of PWR)

  • 가압경수로 일차냉각수내 탈염공정의 효과적 운용을 위하여 Amberlite IRN-77 양이온 교환수지의 특성 및 Co(II), Ni(II), Cr(III), Fe(III) 이온의 흡착거동을 연구하였다. 용존 금속이온 농도가 약 $200\;mgL^{-1}$인 용액 100 mL에 대하여 이온교환수지 투입량은 약 0.6 g이 가장 적절하였으며, 이온교환 반응 속도 측정 결과 대부분의 흡착은 반응 1 시간 이내에 신속히 발생하였다. 수지에서의 양이온 교환은 Langmuir 흡착등온선을 잘 따르는 것으로 나타났으나, Fe(III)의 경우, 다른 금속과는 달리 쉽게 평형에 이르지 못하였으며 이는 공기와 오래 접촉한 결과 철산화물 또는 수산화물을 형성하며 용액 중으로부터 침전되기 때문인 것으로 관찰되었다. 전자가가 동일한 Co(II)와 Ni(II)가 동시에 포함된 모의 조제용액에 대하여 흡착실험을 한 결과, 각 이온은 거의 동일한 흡착거동을 보였다. 그러나 전자가가 큰 Cr(III)이 첨가된 경우, 이들은 기존에 수지에 흡착되어 있던 전자가가 낮은 금속들을 탈착시키고 대신 흡착하는 경향을 보였다. 따라서 이온교환수지를 이용한 효과적인 탈염 공정을 위해서는 용존 금속 상호간의 경쟁적 흡착관계의 규명이 필요한 것으로 여겨진다.

Synthesis of Co(II), Ni(II) and Cu(II) Complexes from Schiff base Ligand and Reactivity Studies with Thermosetting Epoxy Resin

  • A hybrid thermosetting maleimido epoxy compound 4-(N-maleimidophenyl) glycidylether (N-MPGE) containing Co(II), Ni(II) and Cu(II) ions was prepared by curing N-MPGE and tetradentate Schiff base Co(II), Ni(II) and Cu(II) complexes. The curing polymerization reaction of N-MPGE with metal complexes as curing agents was studied. The cured samples were studied for thermal stability, chemical (acid/alkali/solvent) and water absorption resistance and homogeneity of the cured systems. The tetradentate Schiff base, 3-[(Z)-2-piperazin-1-yl-ethylimino]-1,3-dihydro indol-2-one was synthesized by the condensation of Isatin (Indole-2, 3-dione) with 1-(2-aminoethyl)piperazine (AEP). Its complexes with Co(II), Ni(II) and Cu(II) have been synthesized and characterized by microanalysis, conductivity, Uv-Visible, FT-IR, TGA and magnetic susceptibility measurements. The spectral data revealed that the ligand acts as a neutral tetradentate Schiff base and coordinating through the azomethine nitrogen, two piperazine nitrogen atoms and carbonyl oxygen.
