Two Unrecorded Endophytic Fungi Isolated from Funaria hygrometrica in Korea (표주박이끼(Funaria hygrometrica)에서 분리된 2종의 국내 미기록 내생균)

  • In the present study, we isolated endophytic fungal strains from the rhizoids of the moss Funaria hygrometrica. The isolated strains were identified based on morphological characteristics and analysis of the internal transcribed spacer (ITS) and large subunit (LSU) rDNA sequence regions. Consequently, we confirmed the presence of two endophytic fungal species, Curvularia protuberata and Didymella anserina, which have not been reported in Korea previously. Here, we describe the morphological characteristics and molecular analysis results of these fungal species.

A New Report on Phialocephala piceae Isolated from Leaf of Diospyros kaki in Korea (감나무 잎에서 분리한 Phialocephala piceae에 대한 보고)

  • A previously unrecorded fungus was isolated from the persimmon (Diospyros kaki) leaf phyllosphere in Korea. The isolated fungus was characterized by morphological and phylogenetic analyses. The typical morphological characteristics of Phialocephala piceae, including dark brown colonies and short, thick conidiophores, were observed on the isolated fungus. A phylogenetic analysis based on the internal transcribed spacer (ITS) region and RNA polymerase II largest subunit (RPB1) also confirmed the identification of the isolated fungal species as P. piceae. Therefore, this is the first report of P. piceae in Korea.

  • We isolated endophytic fungal strains from the leaves of Abies koreana growing in Korea. The fungal strains were identified as Coleophoma parafusiformis and Coniochaeta ligniaria, based on morphological characteristics and sequence analysis of the internal transcribed spacer region and large subunit region of ribosomal DNA. These species have not been previously reported in Korea. In this study, we report the morphological characteristics and results of phylogenetic analysis of the two novel fungal strains.

  • Three endophytic fungal strains were isolated from aquatic plants collected in Jeju. The fungi were identified by sequencing and phylogenetically analyzing their internal transcribed spacer (ITS) regions. Consequently, the three strains NNIBRFG2982, NNIBRFG2984 and NNIBRFG 3040 were confirmed as the fungal species Acremonium tubakii, Colletotrichum cliviae and Cylindrocarpon pauciseptatum, respectively, all of which are new to Korea.

  • We isolated endophytic fungi from Smilax china and Cryptomeria japonica. These fungal strains were identified based on their morphological characteristics and phylogenetic analyses of their internal transcribed spacer, large subunit rDNA, and translation elongation factor 1-α DNA. Among them, we discovered two fungi belonging to the genus Phyllosticta, which have not been previously recorded in Korea. We have described these two fungal strains, Phyllosticta ericarum 19E458 and Phyllosticta philoprina 19E012 in this study.

  • The polymerase chain reaction (PCR) technique was used to distinguish from two morphologically similar red algal species; Acrosorium polyneurum and A. yendoi. Total DNA was extracted by the LiCl method. The extracted DNA (15 ng) in a $25{\mu}\ell$ reaction volume was amplified by the PCR technique using primers covering with mitochondrial D-loop gene, nuclear rDNA internal transcribed spacer (ITS), and nuclear rDNA external transcribed spacer. A. yendoi could be distinguished from A. polyneurum on the producible basis of amplified ITS fragment of 650 bp.

  • A simple method for identifying of ectomycorrhizal fungi was presented, using the polymerase chain reaction (PCR) to amplify the ITS (Internal transcribed spacer) regions of the nuclear ribosormal repeat. The sequences analyzed 6 species, Pisolithus tinctorius, Chroogomphus rutilus, Leucogyrophana pinastri, Suillus granulatus, Lactarius laeticolorus, and Suillus bovinus at hongreung forest, and analysed 10 species, Craterellus lutescens, Thelephoroid mycorrhizal, Lactarius quieticolor, Tricholoma matsutake, Lactarius chrysorrheus, Sarcodon aspratus, Russula versicolor, Suillus luteus, Tricholoma terreum, and Amanita vaginata at hongcheon forest. Finally, the amplification by PCR with ITS1-ITS4 primers offers good results over classical identification for ectomycorrhizal fungi species.

  • The family Ulvaceae species are difficult to distinguish from one another on the basis of morphological and cytological criteria alone. ITS2 sequences are hewn to evolve quickly and have been reported to be useful for the study of intraspecific and interspecific variation and biogeography in algae (Bakker et al., 1992). We will here describe the basic characteristics of the ITS2 sequences in Ulva and Effteromorpha to compare our result with the above previous studies. (omitted)

Characterization of Rhizodermea veluwensis Isolated from the Roots of Rhododendron mucronulatum in Korea

  • A fungal strain was isolated from surface-sterilized roots of Rhododendron mucronulatum, a plant species belonging to the Ericaceae family, collected from Mt. Minjujisan, Korea. This fungal strain was identified as Rhizodermea veluwensis based on its morphological characteristics and based on phylogenetic analysis of its internal transcribed spacer regions and large-subunit rDNA. R. veluwensis has not been previously reported in Korea, and for the first time, we report and describe it herein.

  • We report on an unrecorded endophytic fungus, Grovesiella abieticola (Zeller & Goodd.) M. Morelet & Gremmen isolated from Abies koreana of Mt. Halla in Jeju. In this study, we used a culture method to determine its conidia and compared relative species using an internal transcribed spacer (ITS) 1, 2, and 5.8S ribosomal DNA sequence. Thus, we present the cultural characteristics and morphology of G. abieticola in this paper.
