Amberlite IRC-50에 의한 alkaloid의 미량검출법 (The microdetection of alkaloids with amberlite IRC-50)

  • Cation exchange resin particles amberlite IRC-50 were used as a medium for a identific test of alkaloids with dragendorff reagent. The procedures were as follows; A few particles of amberlite IRC-50 were added to the small portion of sample solutions on a spot plate or in a test tube. After 20 minutes-an hour, a drop of dragendorff reagent were added to particles of resin that were adsorbed. When alkaloid was present, the original color of the resin particles changed instantly to red or reddish orange depending on the amount and kind of alkaloid, while in the absence of alkaloid the original color of the resin particles changed light yellow. These methods were more sensitive than the ordinary spot test or paper spot test for alkaloid. The limits of identifications of nine alkaloids were tested by these methods and compared with the paper spot test method.

항공용 고속 롤러베어링의 냉각 방식에 따른 작동간극 변화 (Variation of Operating Clearance Depending on Cooling Methods of High-Speed Roller Bearings for Aerospace Applications)

  • In this study, the expansion, stress, and operating clearance of bearing elements during operation are observed using the inner/outer ring temperature test data of a 3.0×106 DN-class roller bearing. The operating clearance characteristics of inner-/outer-ring cooling (IORC) bearings are compared to those of inner-ring cooling (IRC) bearings. For IRC bearings, the thermal expansion of the outer ring is the most important factor in clearance variation. As a result, the operating clearance is less than the initial clearance of 61 ㎛, and the operating clearance decreases to 0.5 ㎛ at 25,500 rpm. Conversely, the temperature of the outer ring of IORC bearings is lower than that of IRC bearings, so the operating clearance is kept smaller. When the coolant flow rate to the outer ring is approximately 1.5 to 2.0 L/min, the temperature difference between the inner and outer rings is minimized and the operating clearance is maintained at a significantly lower level than IRC bearings. Small operating clearances are expected to be effective in reducing cage slip and skid damage in roller bearings. The results and analysis procedures of this study can be utilized to design of bearing clearance, lubricant flow rate, and assembled interference in the early design stage of aerospace roller bearings.

클러스터 분석을 위한 IRC기반 클러스터 개수 자동 결정 방법 (Systematic Determination of Number of Clusters Based on Input Representation Coverage)

  • 클러스터 분석에 있어 중요한 문제 중의 하나는 주어진 데이터에 내재된 적절한 클러스터의 수를 찾아내는 것이다. 본 논문에서는 이러한 클러스터의 개수를 체계적으로 결정하기 위하여 IRC (Input Representation Coverage) 개념을 새로이 정의하고, 이를 이용하여 주어진 데이터에 적합한 클러스터의 개수를 자동 결정하는 방법을 제시한다. 또한, 이러한 방법의 유용성 및 응용성을 알아보기 위하여 가상 데이터를 가지고 분석 실험을 하였으며, 실험을 통해 데이터에 내재된 실제 클러스터의 개수를 찾아내는 데에 제안된 방법이 매우 유용하게 사용될 수 있음을 보여준다.

안전문화 선행지표 개발을 위한 사건보고율 측정 (Measurement of Incident-reporting Rate for Developing a Leading Indicator of Safety Culture)

  • Various leading indicators of safety culture have been advocated for proactive actions as lagging indicators have limitations in reflecting the attitudes and behaviors due to their reactivity and low sensitivity. This study proposes a model of incident-reporting culture (IRC) and determines the influence of the components on incident-reporting rate (IRR) in order to develop proactive indicators of safety culture. A questionnaire survey was administered to 614 workers at a chemical company in Korea, and the internal psychological aspects were explored by using perceptions, attitude, and backgrounds. The relationship between these factors and IRR was quantitatively confirmed. The workers are more reluctant to report injury than property damage, the perception of severity is the most influencing factor, and most property damages are reported regardless of worker's willingness. These features should be prioritized when improving IRC, and the criteria of IRC need to be aligned with safety culture.

난백 및 이온교환수지의 전처리 조건이 난백 Lysozyme의 추출에 미치는 영향 (Effects of Egg White and Ion Exchange Resin Pretreatment on Separation of Egg White Lysozyme)

  • 난백으로부터 lysozyme을 효과적으로 추출하기 위하여 난백의 전처리와 이온교환수지의 선택 및 전처리 등 추출조건에 관한 연구를 실시한 결과는 다음과 같다. 난백의 점도저하를 위하여 2,000 rpm의 속도로 30분간 균질하고 45분간 초음파처리를 실시한 결과 난백의 겉보기 점도가 3 cp까지 떨어졌으나 pH의 변화에 따라서는 점도의 차이가 크지 않았다. lysozyme의 추출을 위한 이온교환수지의 선택을 위하여 Amberlite IRC-84 등 6종류의 수지에 대한 Lysozyme 흡착력을 검토한 결과 CM-Sepdex C-25>Duolte C-464>Amberlite C-50>Dowex MSC-1>Amberlite IRC-50>Amberlite IRC-84의 순으로 lysozyme 흡착력이 높았으나 난백에의 적용시 CM-Sephadex C-25의 경우에는 난백의 유출이 용이하지 않은 것으로 나타나 Duolite C-464가 가장 적합한 것으로 나타났다. 양이온교환수지인 Duolite C-464 의 양이온형태 결정을 위하여 $Na^{+}$, $NH_4$$^{+}$. $K^{+}$ form으로 만들어 lysozyme 흡착력을 검토한 결과 $Na^{+}$$NH_4$$^{+}$$K^{+}$의 순으로 나타나 $Na^{+}$ form으로 하는 것이 가장 효과적인 것으로 나타났다. Duolite C-464의 $Na^{+}$form을 이용하여 lysozyme을 추출할 경우 rinse buffer는 pH6.5의 0.1M sodium phosphate buffer, eluting solution은 10% ammoniun sulfate가 효율적이었다. Lysozyme을 추출한 전, 후 난백의 가공적성을 측정한 결과 기포력은 추출전의 난백액에 대하여 85.3%로서 다소 떨어졌으나 가열한 난백겔의 경우에는 추출전, 후 색택의 변화가 크지 않았으며 조직감의 경우에도 hardness가 다소 떨어졌으나 elasticity는 오히려 13%가량 증가되었다.

AKARI IRC SURVEY OF THE LARGE MAGELLANIC CLOUD: AN OVERVIEW OF THE SURVEY AND A BRIEF DESCRIPTION OF THE POINT SOURCE CATALOG

  • We observed an area of 10 $deg^2$ of the Large Magellanic Cloud using the Infrared Camera (IRC) onboard AKARI. The observations were carried out using five imaging filters (3, 7, 11, 15, and $24{\mu}m$) and the prism disperser ($2-5{\mu}m$, ${\lambda}/{\Delta}{\lambda}{\sim}20$) equipped in the IRC. This paper presents an outline of the survey project and also describes very briefly the newly compiled near- to mid-infrared point source catalog. The $10{\sigma}$ limiting magnitudes are 17.9, 13.8, 12.4, 9.9, and 8.6 mag at 3.2, 7, 11, 15 and $24{\mu}m$, respectively. The photometric accuracy is estimated to be about 0.1 mag at $3.2{\mu}m$ and 0.06 - 0.07 mag in the other bands. The position accuracy is 0.3" at 3.2, 7 and $11{\mu}m$ and 1.0" at 15 and $24{\mu}m$. The sensitivities at 3.2, 7, and $24{\mu}m$ are roughly comparable to those of the Spitzer SAGE LMC point source catalog, while the AKARI catalog provides the data at 11 and $15{\mu}m$, covering the near- to mid-infrared spectral range continuously.

Streptomyces 속 균주가 생성하는 Trypsin Inhibitor (제1보) 균의 분리 및 저해물질의 정제 (Trypsin Inhibitor from Streptomyces sp. ( Part 1) Isolation of microorganism and purification of the inhibitor)

  • Trypsin에 대한 강한 저해물질을 생성하는 Streptomyces속 균주 AS-707을 토양으로부터 얻어 그 배양액에서 Trypsin inhibitor를 분리정제하여 저해물질의 안정성과 여러가지의 protease에 대한 저해성 여부를 검토한 결과는 다음과 같다. 배양액을 Amberlite IRC-50에 흡착 methanol추출. 2차 Amberlite IRC-50, CM-cellulosecolumn chromatography로 정제하여 active amorphous powder를 얻었는데 이 때의 비율은 26%였다. 분리정제된 물질은 trypsin 이외에 papain, $\alpha$-chymotrypsin, Azotobacter vineiandi protease와 Bacillus subtilis protease 등에 대해서도 저해작용을 나타내었으며, 안정성은 비교적 커서 10$0^{\circ}C$에서 120분간 가열해도 잔존활성이 약90%였으며, pH처리에 대해서는 37$^{\circ}C$에서 처리하면 산에서 Alkali에 걸치는 대단히 넓은 pH범위 (pH 2.0~12.0)에서 안정하였으나 6$0^{\circ}C$에서 처리하면 산에서는 안정하였으나 Alkali에서는 불안정하였다.

Ge(Ⅳ)-Mercaptoacetic Acid 착물에 의한 흡착벗김 전압-전류법 (Adsorptive Stripping Voltammetry of Ge(IV)-Mercaptoacetic Acid Complex)

  • Mercaptoacetic acid를 이용하여 흡착벗김 전압전류법으로 미량의 게르마늄을 정량하는 방법을 연구하였다. 이때 지지전해질은 0.25 M NaCl 용액(pH 6.0)을 사용하였고, mercaptoacetic acid의 농도는 $5.0{\times}10^{-6}M$이 되도록 조절하여 실험하였다. Ge(IV)-Mercaptoacetic acid 착물의 봉우리 전위는 -1.402V vs. Ag/AgCl에서 나타났다. 착물의 봉우리 전위에 미치는 NaCl의 농도, mercaptoacetic acid의 농도, 축적 시간등에 대하여 조사하였다. 또한, Amberlite IRC-718 킬레이트 수지를 다른 이온들로 부터 Ge(IV)을 분리하는데 적용하였다.

Evolving live load criteria in bridge design code guidelines - A case study of India based on IRC 6

  • One of the instances which demand structural engineer's greatest attention and upgradation is the changing live load requirement in bridge design code. The challenge increases in developing countries as the pace of infrastructural growth is being catered by the respective country codes with bigger and heavier vehicles to be considered in the design. This paper presents the case study of India where Indian Roads Congress (IRC) codes in its revised version from 2014 to 2017 introduced massive Special vehicle (SV) around 40 m long and weighing 3850 kN to be considered in the design of road bridges. The code does not specify the minimum distance between successive special vehicles unlike other loading classes and hence the consequences of it form the motivation for this study. The effect of SV in comparison with Class 70R, Class AA, Class A, and Class B loading is studied based on the maximum bending moment with moving load applied in Autodesk Robot Structural Analysis. The spans considered in the analysis varied from 10 m to 1991 m corresponding to the span of Akashi Kaikyo Bridge (longest bridge span in the world). A total of 182 analyses for 7 types of vehicles (class B, class A, class 70R tracked, class 70R wheeled, class AA tracked, AA wheeled, and Special vehicle) on 26 different span lengths is carried out. The span corresponding to other vehicles which would equal the bending moment of a single SV is presented along with a comparison relative to Standard Uniformly Distributed Load. Further, the results are presented by introducing a new parameter named Intensity Factor which is proven to relate the effect of axle spacing of vehicle on the normalized bending moment developed.
