Effects of Culture Condition on Callus Formation from Anther Culture of Petunia hybrida (Petunia hybrida의 약배양(約培養)으로부터 callus 형성(形成)에 미치는 배양조건(培養條件)의 영향(影響))

  • The study was conducted to get basic information for haploid production of Petunia hybrida through anther culture by investigating several factors, namely anther stage, culture medium, cold treatment, and genotypes. The results are summarized as follows; Four genotypes of Petunia hybrida were used in a study of anther culture. Plants of each genotype were grown in controlled environments at $20-30^{\circ}C$ with a 16h photoperiod. Equal numbers were harvested from each genotype. The anthers were taken from buds which had received the 14 days' cold treatment at $4^{\circ}C$. Anthers were dissected out aseptically and plated on 1/2 strength MS agar medium containing 5.0mg/${\ell}$ 6-Benzylaminopurine(BAP). Four weeks after culture, light green callus was formed. From these calli, plantlets were regenerated on MS medium containing 2.0mg/${\ell}$ 2-isopentenyl adenine(2ip) after 3 weeks.

Identification of Oocyte-Specific Diva-Associated Proteins using Mass Spectrometry (Mass Spectrometry를 이용한 난자 특이적인 Diva와 상호작용하는 단백질의 동정)

  • Objective: We previously described that Diva is highly expressed in matured metaphase II (MII) oocytes compared to immature germinal vesicle (GV) oocytes in mouse. We report here that the expression of Diva transcript as well as protein is oocyte-specific. To elucidate its physiological role in oocyte, the binding partner(s) of Diva has been identified by using immunoprecipitation (IP) followed by Mass Spectrometry. Methods: NIH/3T3 cells were transiently transfected for 24 h with either empty vector for control or FLAG-tagged mouse Diva construct, and IP was performed with anti-FLAG antibody. The immuno-isolated complexes were resolved by SDS-PAGE on a 12% gel followed by Coomassie Blue staining. For in-gel digestion, 15 bands of interest were excised manually and digested with trypsin. All mass spectra were acquired at a positive reflector mode by a 4700 Proteomics Analyzer (Applied Biosystems, Framingham, MA). Proteins were identified by searching the NCBI nonredundant database using MASCOT Peptide Mass Fingerprint software (Matrixscience, London). Results: Diva-associated complexes were formed in FLAG-tagged mouse Diva-overexpressed NIH/3T3 cells via IP using anti-FLAG-conjugated beads. Among the excised 15 bands, actin and actin-binding proteins such as tropomyosin, tropomodulin 3, and ${\alpha}$-actinin were identified. Binding between Diva and actin or tropomyosin was confirmed by IP followed by Western blot analysis. Both bindings were also detected endogenously in mouse ovaries, indicating that Diva works with actin and tropomyosin. Conclusions: This is the first report that immuno-isolated Diva-associated complexes are related to actin filament of the cytoskeletal system. When we consider the association of Diva with actin and tropomyosin, oocyte-specific Diva may play a role in modulating the cytoskeletal system during oocyte maturation.

In Vitro Regeneration of Lycium chinense Miller and Detection of Silent Somaclones with RAPD Polymorphisms

  • An efficient system for the regeneration of adventitious shoots from in vitro cultured leaf sections of Lycium chinense Miller was developed and silent somaclones from the regenerants detected with RAPD method. Among the eight media tested (B5, SH, N&N, 1/2MS, MS, 3/2MS, GD and WPM), and four cytokinins (BA, kinetin, 2ip and zeatin) with different concentrations (1, 5, 10, 20, 30 and 40 $\mu{M}$), 1/2 MS medium supplemented with 20 and 30 $\mu{M}$ zeatin showed the best regeneration frequency (100% and 93.7%) and higher average number of shoots (9.0 and 9.4). All regenerants easily elongated after subculturing on 1/4MS without growth stimulants and produced spontaneous adventitious roots from their basal parts. With phenotypically normal 40 regenerants, RAPD analysis with 15 different random primers was performed to examine the cryptic somaclonal variants. No substantial differences in banding patterns were found in the amplified polymorphic DNAs implying no DNA changes during dedifferentiation into adventitious shoots. However, one (OPF-4) of the 15 primers detected silent somaclonal variation in one regenerant in which two different polymorphic bands did not appear when compared with the rest regenerants. The results indicate that regenerantion via intervening callus phase can be used to establish true-to-type planting stocks for homogeneous population.

Optimization of the packet size to enhance the voice quality of the VOIP system (VOIP 음질 개선을 위한 패킷 크기의 최적화)

  • In this paper we discuss the effect of the delay limit and the packet size related to the quality of service on a VoIP system using the Internet. We also provide a guideline to determining the optimal packet size of the voice data for a given delay limit. Empirical studies are done with two personal computers connected through the packet switched public IP network. The sender encodes the voice signal from the microphone to get PCM and ADPCM data and sends the data to the receiver using UDP packets. The receiver plays the reconstructed voice from the stream with lost and delayed packets. The quality of the reconstructed voice is evaluated offline by the MNB (Measuring Normal Block) method using the data acquired from the both sides. The result shows that under the delay limit of 100ms for 40Kbps, 32Kbps and l6Kbps of ADPCM data, the minimum packet size should be 300bytes, 400bytes and 600bytes respectively and the maximum packet size should be l200bytes commonly for the best quality of voice.

  • There are lots of researches for mobility of MS(mobile station) in All IP based network. Specially, NEMO(NEwork MObility) is not supporting mobility of each MS but supporting mobility of network that include group of MS. Some research try to overcome limitation of wireless with the protocol in wired state and it maintains the performance such as wire environment. There are no researches about multicast with reliability in NEMO. Therefore, this paper suggests efficient algorithm to solve problems when RMTP(Reliable Multicast Transport Protocol) apply to NEMO environment to support high reliability with multicast. And this paper shows the better performance of proposed algorithm for delay and transmission rate between AR and TLMR comparing with RMTP in NEMO.

  • Network on a chip(NoC) is a communication subsystem between intellectual property(IP) cores in a SoC and improves high performance in the scalability and the power efficiency compared with conventional buses and crossbar switches. NoC needs a synchronizer to overcome the metastability problem between data links. This paper presents a new mesochronous synchronizer(MS) which is composed of selection window generator, selection signal generator, and data buffer. A delay line circuit is used to build selection window in selection window generator based on the delayed clock cycle of transmitted clock and the transmitted clock is compared with local clock to generate a selection signal in the SW(selection window). This MS gets rid of the restriction of metastability by choosing a rising edge or a falling edge of local clock according to the value of selection signal. The simulation results show that the proposed MS operates correctly for all phase differences between a transmitted clock and a local clock.

Determination of Heterocyclic Amines and Acrylamide in Agricultural Products with Liquid Chromatography-Tandem Mass Spectrometry

  • Heterocyclic amines (HCAs) and acrylamide are unintended hazardous substances generated by heating or processing of foods and are known as carcinogenic and mutagenic agents by the animal experiments. A simple method was established for a rapid and accurate determination of 12 types of HCAs (IQ, MeIQ, Glu-P-1, Glu-P-2, MeIQx, Trp-P-1, Trp-P-2, PhIP, $A{\alpha}C$, $MeA{\alpha}C$, Harman and Norharman) and acrylamide in three food matrices (non-fat liquid, non-fat solid and fat solid) by isotope dilution liquid chromatography-tandem mass spectrometry (LC-MS/MS). In every sample, a mixture of internal standards including $IQ-d_3$, $MeIQx-d_3$, $PhIP-d_3$, $Trp-P-2-^{13}C_2-^{15}N$ and $MeA{\alpha}C-d_3$ was spiked for quantification of HCAs and $^{13}C_3$-acrylamide was also spiked for the analysis of acrylamide. HCAs and acrylamide in sample were extracted with acetonitrile and water, respectively, and then two solid-phase extraction cartridges, ChemElut: HLB for HCAs and Accucat: HLB for acrylamide, were used for efficiently removing interferences such as pigment, lipid, polar, nonpolar and ionic compounds. Established method was validated in terms of recovery, accuracy, precision, limit of detection, limit of quantitation, and linearity. This method showed good precision (RSD < 20%), accuracy (71.8~119.1%) and recovery (66.0~118.9%). The detection limits were < 3.1 ng/g for all analytes. The correlation coefficients for all the HCAs and acrylamide were > 0.995, showing excellent linearity. These methods for the detection of HCAs and acrylamide by LC-MS/MS were applied to real samples and were successfully used for quantitative monitoring in the total diet study and this can be applied to risk assessment in various food matrices.

  • Mobility management is one of the most important research issues in 6LoWPAN, Since the IP-WSN application domain is expanded to real-time applications such as healthcare and surveillance systems, a fast and seamless handover becomes an important criterion in 6LoWPAN. However, a draft of IETF 6LoWPAN WG for mobility support does not decrease handover delay. Although LoWMob supports a fast intra-PAN handover. it can be supported when the infrastructure node has the location information of the other nodes in the mobile node's moving direction. In this paper, we propose a fast and seamless mobility protocol to support intra-PAN handover, named intra-MARIO. In intra-MARIO, a parent node of the mobile node preconfigures its handover to the PAN when the parent node detects its movement, thereby reducing the handover delay. Since intra-MARIO also supports route optimization, the mobile node can communicate with its corresponding nodes through the optimal route. In this paper, we analysis the signaling cost and evaluates that the handover can be completed in 20ms by simulation.

Structures and some Properties of the Antimicrobial Compounds in the Red Alga, Symphyocladia latiuscula (참보라색우무에서 추출한 항균물질의 구조 및 특성)

  • Three antimicrobial compounds (SL-l, SL-2 and SL-3) were isolated and identified from the marine red alga, Symphyocladia latiuscula. In addition, their biological functionalities such as cytotoxicity and desmutagenic activity were investigated. From the cryophyllized S. JatiuscuJa, SL-l, SL-2 and SL-3 were purified by solvent extractions and HPLC.SL-2 was crystallized in benzene-diethyl ether solvent. On the EI-MS spectra, it was found that they had three bromines in their structure which showed typical signal strength ratios at $M^+, [M+2]^+, [M+4]^+, [M+6]^+ (13: 38: 37: 12)$. $SL-l$ was identified as 2,3,6-tribromo-4,5-dihydroxybenzyl alcohol ($C_8H_7Br_3O_3, MW=374$) by NMR and MS spectra. SL-2 was assigned as 2,3,6-tribromo-4,5-dihydroxybenzyl methyl ether ($C_8H_7Br_3O_3, MW=388$) and confirmed by X-ray crystallographic analysis. SL-3 was presumed as an isomer of SL-2. Methanol extract of the S. latiuscula showed antimicrobial activities against all strains tested (bacteria, 15 strains; yeasts, 17 strains; fungi, 4 strains), much or less. The strongest inhibition activity of the methanol extract was to the Vibrio mimicus ($50 {\mu}g/ml$) and V. vulnificus ($50 {\mu}g/ml$). The mice injected intraperitoneally with 3 mg of SL-l and 5 mg of 5L-2 showed no acute toxicity response. SL-2 showed higher desmutagenic activity than SL-l against PhIP and MeIQx.

  • MMI system for strip caster 2 is described. It has been developed as a PC-based MMI system whose data is collected from the VME-based control system by ETHERNET. MMI elements that mimic the caster and its utilities are designed by using commercial program. In order to implement the UDP/IP communication in MMI, UDP/IP manager program with DDE that is a intercommunication protocol in MS-window is developed and connected with MMI mimics. Operator commands in MMI system are carried out immediately in VME system and the responses are displayed on MMI system. The developed MMI system has been proven to be convenient and stable through the operation of the past 5 years.
