The effect of hyaluronic acid on anti-inflammatory action in mouse (마우스에서 히알우론산 나트륨이 항염효과에 미치는 영향에 관한 연구)

  • Purpose: The purpose of this study was not only to evaluate the relative mRNA expression of interleukin-$1{\beta}$(IL-$1{\beta}$), cyclooxygenase2 (COX-2) and prostaglandin E2 (PGE2) by RT-PCR analysis but to observe pattern of edema by light microscopic and electron microscope after topical apply of hyaluronic acid in inflammation-guided mouse. Material and methods: Mice of this study were devided into 4 groups: Control group (no inflammation guided), Positive control (inflammation guided + vaselin apply), Protopic group (inflammation guided + protopic apply), Hyaluronic group (inflammation guided + hyaluronic acid apply). Results: Hyaluronic group showed less expressions of IL-$1{\beta}$, COX-2, PGE2 than those of positive control & protopic group. Hyaluronic group revealed a decreased inflammation than positive control & protopic group in Light Microscope. Hyaluronic group appeared decreased edema of ear compare to positive control & protopic group in Elecron Microscope. Conclusion: It was considered that hyaluronic acid has an antiinflammatory effect for intercepting the gene expression of cytokines related to inflammation.

Development of tablets and evaluation of ingredient content and pharmacological effects of Yukgunja-tang (육군자탕의 정제 개발과 성분함량 및 약리효과 평가)

  • Objective : Yukgunja-tang is one of the herbal prescriptions widely used for functional indigestion. In this study, we evaluated the pharmacological effect through the Yukgunja-tang formulation development. Methods : The RAW 264.7 cells were pretreated with Yukgunja-tang tablet (YGJT-T : 50, 100 and 200 ㎍/㎖) and then stimulated with lipopolysaccharide (LPS : 500 ng/㎖). Cell viability, inflammatory mediators such as nitric oxide (NO) and prostaglandin E2 (PGE2) and inflammatory cytokines (IL-1β, IL-6, and TNF-α) were measured. Also, ICR mice induced acute gastritis by oral administration of 150 mM HCl in 60% ethanol. The YGJT-T (30 mg/kg) was pretreated for 3 days, and 150 mM HCl in 60% ethanol was orally administered 1 hour after the last drug treatment. Mice were sacrificed 1 hour after oral administration of 150 mM HCl in 60% ethanol. The gastric mucosa was observed, and inflammatory cytokines in the gastric tissue were measured. Results : The marker components of YGJT-T were determined by simultaneous analysis using HPLC. In RAW 264.7 cells, pretreatment of YGJT-T was non-toxic and inhibited the production of inflammatory mediators such as NO and PGE2 and suppressed inflammatory cytokines. In addition, pretreatment of YGJT-T improved bleeding and edema due to gastric lesions caused by acute gastritis and suppressed inflammatory cytokines. Conclusion : In summary, our results confirmed that treatment with YGJT-T has anti-inflammatory and anti-gastritis effects in vitro and in vivo. Therefore, in this study, YGJT-T could support a potential strategy for the prevention and treatment of gastritis.

  • pH는 두 처리구 모두 저장기간이 길어짐에 따라 유의적으로 감소하였다(P<0.05). T1의 염도는 저장 전 기간 동안 유의적인 차이가 없었고(P>0.05), T2는 저장 28일에는 1.33%로 유의적으로 높았다(P<0.05). 당도는 T1이 T2 보다 저장 전 기간 동안 유의적으로 높았다(P<0.05). 저장기간이 길어짐에 따라 T1에서 $L^*$값과 $b^*$값은 증가하는 반면 T2는 감소하였다. $a^*$값과 전단가(g/$cm^2$)는 저장기간이 경과함에 따라 모두 증가하였다. 관능검사결과, 저장 28일차까지 전체적인 기호도는 6,75(T1)${\sim}$7.00(T2) 수준으로 비교적 높은 점수를 얻었다. 본 연구에서 양념으로 사전 숙성 처리하여 진공포장한 양념 사슴육은 새로운 메뉴로서의 가치가 있을 것으로 여겨진다. 본 연구에서 양념으로 사전 숙성 처리하여 진공포장한 양념 사슴육은 새로운 메뉴로 유용할 것으로 여겨진다.

ANALYSIS OF 67 MALIGNANT SALIVARY GLAND TUMORS IN KOREAN POPULATION (한국인에서 발생한 67례의 악성타액선종양에 대한 연구)

  • Malignant salivary gland tumor is rare neoplasm. In Korean population, retrospective study of malignant salivary gland tumor has not been performed. We analyzed 67 cases of malignant salivary gland tumors from 2001 to 2005 in Seoul National University Dental Hospital in Seoul, Korea. The mean age is 51.7 and the male to female ratio is 1:1.39. The most affected site is the palate. Histologically, the tumors were classified as adenoid cystic carcinoma(34.4%), mucoepidermoid carcinoma(31.3%), adenocarcinoma, NOS(11.9%), polymorphous low grade adenocarcinoma(3.0%), salivary duct carcionoma(6.0%), carcinoma ex pleomorphic adenoma(4.5%), myoepithelial carcinoma(4.5%), epithelial-myoepithelial carcinoma(1.5%), cyatadenocarcinoma(1.5%) and adenosquamous carcinoma(1.5%).

  • To elucidate the involvement of interleukin-$1{\beta}$ converting enzyme (ICE) in the course of experimental autoimmune encephalomyelitis (EAE), we induced EAE by immunizing rats with an emulsion of rat spinal cord homogenate with complete Freund's adjuvant supplemented with Mycobacterium tuberculosis (H37Ra, 5mg/ml) and then examined the expression of ICE in the spinal cord of rats with EAE. In normal rat spinal cords, ICE is constitutively, but weakly, expressed in ependymal cells, neurons, and some neuroglial cells. In EAE, many inflammatory cells are positive for ICE, and the majority of ICE+ cells were identified as ED1+ macrophages. During this stage of EAE, the number of ICE+ cells in brain cells, including neurons and astrocytes, increased and these cells also had increased ICE immunoreactivity. These findings suggest that the upregulation of ICE in both brain cells and invading hematogenous cells is stimulated by a secretory product from inflammatory cells, and that this enzyme is involved in the pathogenesis of EAE via the production of IL-1 beta.

  • Kawasaki disease (KD) is an acute pediatric vasculitis that affects genetically susceptible infants and children. To identify coding variants that influence susceptibility to KD, we conducted whole exome sequencing of 159 patients with KD and 902 controls, and performed a replication study in an independent 586 cases and 732 controls. We identified five rare coding variants in five genes (FCRLA, PTGER4, IL17F, CARD11, and SIGLEC10) associated with KD (odds ratio [OR], 1.18 to 4.41; p = 0.0027-0.031). We also performed association analysis in 26 KD patients with coronary artery aneurysms (CAAs; diameter > 5 mm) and 124 patients without CAAs (diameter < 3 mm), and identified another five rare coding variants in five genes (FGFR4, IL31RA, FNDC1, MMP8, and FOXN1), which may be associated with CAA (OR, 3.89 to 37.3; p = 0.0058- 0.0261). These results provide insights into new candidate genes and genetic variants potentially involved in the development of KD and CAA.

Properties of fermented soybean meal by kefir and its biological function

  • Yeast strains are capable of hydrolyzing non-digestible saccharides, such as melibiose, raffinose, and stachyose, found in soy meal components. This study revealed the biochemical properties of fermented soybean meal during 72 hours with kefir. Starchyose and raffinose, non-digestible components, were almost digested in kefir 150 mL + soybean meal 500 g + water 70 mL and galactose was produced. Proteolysis of the soybean meal produced most of the small molecule peptides in kefir 150 mL + soybean meal 500 g + water 70 mL. The production of the vitamin B group and C were the highest in kefir 250 mL + soybean meal 500 g. The yeast number of the fermented soybean meal was 7.0 × 106 CFU·mL-1 which was the highest in kefir 250 mL + soybean meal 500 g. The lactic acid bacteria of the fermented soybean meal was the highest at 3.5 × 109 CFU·mL-1 in kefir 70 mL + soybean meal 500 g. The antioxidant effect was the highest at 57% in kefir 250 mL + soybean meal 500 g. Expression of inflammation-related cytokine (interleukin [IL]-1β, tumor necrosis factor [TNF]-α, and interleukin [IL]-6) was significantly inhibited in fermented soybean meals with different treatments. These results suggest that fermented soybean meal by kefir has an antiinflammatory and anti-oxidation activity and could be utilized in feed manufacturing, and inhydrolyzing non-digestible soy meal components.

Effect of black chokeberry on skeletal muscle damage and neuronal cell death

  • [Purpose] Numerous epidemiological studies have shown that it is possible to prescribe exercise for neurodegenerative disease, such as Alzheimer's disease and Parkinson's disease. However, despite the availability of diverse scientific knowledge, the effects of exercise in this regard are still unclear. Therefore, this study attempted to investigate a substance, such as black chokeberry (Aronia melanocapa L.) that could improve the ability of the treatment and enhance the benefits of exercising in neurodegenerative diseases. [Methods] The cell viability was tested with 2,3-bis[2-methyloxy-4-nitro-5-sulfophenyl]-2H-tetrazolim-5-carboxanilide and the cells were stained with ethidium homodimer-1 solution. The mRNA expression levels were evaluated by microarray. The active compounds of black chokeberry ethanolic extract (BCE) were analyzed by gas chromatography. The chemical shift analysis in the brain was performed using magnetic resonance spectroscopy. [Results] BCE treatment decreased hydrogen peroxide-induced L6 cell death and beta amyloid induced primary neuronal cell death. Furthermore, BCE treatment significantly reduced the mRNA levels of the inflammatory factors, such as IL-1α, Cxcl13, IL36rn, Itgb2, Epha2, Slamf8, Itgb6, Kdm6b, Acvr1, Cd6, Adora3, Cd27, Gata3, Tnfrsf25, Cd40lg, Clec10a, and Slc11a1, in the primary neuronal cells. Next, we identified 16 active compounds from BCE, including D-mannitol. In vivo, BCE (administered orally at a dosage of 50 mg/kg) significantly regulated chemical shift in the brain. [Conclusion] Our findings suggest that BCE can serve as a candidate for neurodegenerative disease therapy owing to its cyto-protective and anti-inflammatory effects. Therefore, BCE treatment is expected to prevent damage to the muscles and neurons of the athletes who continue high intensity exercise. In future studies, it would be necessary to elucidate the effects of combined BCE intake and exercise.

  • Nano-sized cathode materials for high power lithium ion polymer battery are easily and economically prepared using united eutectic self-mixing method without any artificial mixing procedures of reactants and ultra-miniaturization of products. While the micro-sized $LiNi_{0.7}Co_{0.3}O_2$ exhibits the discharge capacities of 167.8 mAh/g at 0.1C and 142.5 mAh/g at 3.0C, those of the nano-sized $LiNi_{0.7}Co_{0.3}O_2$ are 170.8 mAh/g at 0.1C and 159.3 mAh/g at 3.0C. In the case of $LiCoO_2$, the micro-sized $LiCoO_2$ exhibits the discharge capacities of 134.8 mAh/g at 0.1C and 118.6 mAh/g at 5.0C. Differently, the nano-sized $LiCoO_2$ exhibits the discharge capacities of 137.2 mAh/g at 0.1C and 131.7 mAh/g at 5.0C.

Quality Evaluation of Low-fat Pork Loaf Containing Silkworm Powder and Vegetable Worm (Paecilomyces japonica) During Cold Storage

  • This study was performed to determine physicochemical and sensory quality of low-fat pork loaf containing silkworm powder and vegetable worm (Paecilomyces japonica). Total 2% of fat replacer (soy protein isolate:maltodextrin:$\kappa$-carrageenan:water=1:0.5:0.5:10, w/v) was added. The loaf was separated into CTL (control), T1 (0.2% silkworm powder), T2 (0.2% vegetable worm), T3 (0.4% silkworm powder), T4 (0.4% vegetable worm), T5 (0.1% silkworm powder+0.1% vegetable worm), and T6 (0.2% silkworm powder+0.2% vegetable worm). Pork loaf of T3 showed the highest pH value and cooking loss of pork loaves containing silkworm and vegetable worm was higher than control at day 5. 2-Thiobarbituric acid reactive substances of T2 and T5 showed significantly lower values than control and those additives may reduce lipid oxidation of meat. Overall acceptability was not adversely influenced by silkworm powder and vegetable worm at day 0 and 5. These results indicated that those silkworm powder and vegetable worm could be utilized for pork product industry.
