Panax ginseng C. A. Meyer is well known as traditional herbal medicine, and ginseng berries are known to exhibit potential immune-enhancing functions. However, little is known about the in vivo immunomodulatory activity of Korean ginseng berries. In this study, crude Korean ginseng berries polysaccharides (GBP) were isolated and their immunomodulatory activities were investigated using cyclophosphamide (CY)-induced immunosuppressive BALB/c mice. In CY-treated mice, oral administration of GBP (50-500 mg/kg BW) remarkably increased their spleen sizes and spleen indices and activated NK cell activities. GBP also resulted in the proliferation of splenic lymphocytes (coordinating with ConA: plant mitogen which is known to stimulate T-cell or LPS: endotoxin which binds receptor complex in B cells to promote the secretion of pro-inflammatory cytokines) in a dose-dependent manner. In addition, GBP significantly stimulated mRNA expression levels of immune-associated genes including interleukin-1β (IL-1β), IL-2, IL-4, IL-6, tumor necrosis factor-α (TNF-α), interferon-γ (IFN-γ), toll-like receptor 4 (TLR-4), and cyclooxygenase-2 (COX-2) in CY-treated mice. These results indicate that GBP is involved in immune effects against CY-induced immunosuppression. Thus, GBP could be developed as an immunomodulation agent for medicinal or functional food application.
Objectives : The purpose of this study was to evaluate the anti-Inflammatory effect of Hwadokdan(HDD) extracted with water. Methods : We pretreated LPS-activated Raw 264.7 cell with HDD and anti-inflammatory effect of HDD is investigated by measuring NO production, relative iNOS, and TNF-α, IL-6, IL-1β, PGE2 productions level, p-IκBα, IκBα, NFκB. Results : HDD reduced NO production and iNOS protein, TNF-α, IL-6 level at concentration of 100㎍/㎖ and 300㎍/㎖ in LPS-activated Raw 264.7 cell significantly. There wasn't a significant cytotoxicity in MTT assay. And HDD reduced IL-1β level at concentration of 300㎍/㎖ in LPS-activated Raw 264.7 cell significantly. HDD reduced p-IκBα and NFκB protein level at concentration of 300㎍/㎖ in LPS-activated Raw 264.7 cell significantly. And HDD enhanced IκBα protein level at concentration of 100㎍/㎖ and 300㎍/㎖ in LPS-activated Raw 264.7 cell significantly. Conclusions : These suggests that HDD can be used as a therapeutic drug for various inflammatory diseases.

Objectives: Gout is an inflammatory arthritis of the joints and soft tissues occurring due to deposition of monosodium urate (MSU) crystals, which are caused by persistent hyperuricemia. Soyeom pharmacopuncture is one treatment method that has been traditionally used for pain management in Oriental medicine. However, studies on its effect in reducing gout pain have been insufficient. Therefore, we selected Soyeom pharmacopuncture among natural products used in Korea as the new target of our study. Methods: The effects of Soyeom pharmacopuncture were examined in mouse models of acute gout induced by injection of MSU crystals into footpads. IL-1β, IL-6, and TNF-α production were examined by immunoblotting and enzyme-linked immunosorbent assay as hallmarks of NLRP3 inflammasome and cytokine activation. Results: Soyeom pharmacopuncture reduced foot edema in gout-induced mice, as well as IL-1β, nitrite, IL-6, and TNF-α production. Moreover, Soyeom pharmacopuncture also reduced MSU-induced gout inflammatory gene expressions, specifically those in the NF-kB pathway. Conclusion: Pharmacopuncture may serve as a new solution for other inflammatory diseases as well. Through active follow-up studies, we could thoroughly understand the clinical value of Soyeom pharmacopuncture.
Phospholipase C is a key enzyme of signaling pathways hydrolyzed phosphatidylinositol 4,5-bisphosphate to generate 2 second messengers. Among the PLC, $PLC-{\beta}$ subfamily consisted of 4 isoforms, $PLC-{\beta}$ 1~4. Here, we studied the tissue specific expression of $PLC-{\beta}3$ in olive flounder (Paralichthys olivaceus) following external stimulation like lipopolysaccharide (LPS), concanavalin A (ConA) and environmental stress compared with the inflammatory cytokines IL-1b. $PoPLC-{\beta}3$ gene transcripts has the effect in stimulated tissue compared to control. These results provide what we sure to be a important role for $PLC-{\beta}3$ activity in tissue and verify $PLC-{\beta}3$ as potential immune enzyme for signal transduction.
Objective: The aims of this study were to analyze deficiency-excess pattern identification and to compare the blood cytokines in patients with asthma. Methods: A total of 112 patients with asthma who met the inclusion and exclusion criteria were divided into deficiency syndrome and excess syndrome groups. Blood was examined for eotaxin, interleukin (IL)-1β, IL-4, IL-5, IL-6, IL-13, and tumor necrosis factor (TNF)-α. The Quality of Life Questionnaire for Adult Korean Asthmatics (QLQAKA), a Visual Analogue Scale (VAS), and heart rate variability (HRV) tests were administered to both groups. Results: Pattern identification divided the 112 patients into two categories: a deficiency syndrome group (N=52) and an excess syndrome group (N=60). Analysis of blood cytokines showed higher levels of IL-4, IL-5, and IL-13 in the deficient pattern than in the excess pattern group, but the difference was not statistically significant. Analysis of the HRV revealed a significantly higher mean value for the very-low-frequency (VLF) and high-frequency (HF) bands in the deficiency than in the excess syndrome group. The morbidity duration was longer in the deficiency than in the excess syndrome group, but the difference was not statistically significant. Analysis of the QLQAKA and VAS scores showed a negative correlation, whereas BMI and VAS showed a positive correlation. Conclusions: Levels of blood cytokines, including eotaxin, IL-1β, IL-4, IL-5, IL-6, IL-13, and TNF-α, did not differ statistically between the deficiency and excess syndrome groups. The development of a more accurate asthma-specific pattern identification tool would be useful in asthma control.
This study aimed to investigate changes in the activity and mRNA expression of plasminogen activators (PAs) induced by $17{\beta}$-estradiol ($E_2$), human chorionic gonadotropin (hCG), and interleukin-$1{\beta}$ ($IL-1{\beta}$) in porcine endometrial cells. Endometrial cells were isolated from the epithelium and cultured to 80% confluence. They were then treated for 24 h with $E_2$ (0.2, 2, 20, and 200 ng/mL), $IL-1{\beta}$ (0.1, 1, 10, and 100 ng/mL), and hCG (0.5, 1, 1.5 and 2 IU/mL). mRNA expressions of urokinase-type (uPA) and tissue-type (tPA) PAs were analyzed using reverse transcription PCR, and activities were measured using a PA activity assay. mRNA expressions of uPA and tPA increased with $E_2$ treatment; however, this was not significant. Similarly, treatment with hCG did not influence the mRNA expressions of PAs. Interestingly, treatment with 0.1 ng/mL $IL-1{\beta}$ significantly reduced the mRNA expression of uPA, but did not affect that of tPA. Treatment with 2, 20, and 200 ng/mL $E_2$ increased PA activity compared with the control group; treatment with 0.1 and 1 ng/mL $IL-1{\beta}$ significantly increased PA activity compared with the other $IL-1{\beta}$ treatment groups, whereas treatment with 10 and 100 ng/mL $IL-1{\beta}$ decreased. Treatment with 2 IU/mL hCG increased PA activity compared with the other treatment groups, although there were no significant differences between the hCG and control groups. In conclusion, the activity and mRNA expression of PAs were differently regulated by the hormone/cytokine and its concentration in porcine endometrial cells. Therefore, understanding PA regulatory mechanisms may help to improve the reproductive potential of domestic animals.
본 연구는 COHEE의 항염증 효과를 확인하기 위한 실험으로써 pro-inflammatory cytokine의 분비량 및 iNOS, COX-2, NF-κB와 MAPKs의 발현량을 관찰하였다. 먼저 MTT assay를 통해 COHEE가 세포 생존율에 있어 독성을 나타내지 않음을 확인한 후 동일한 농도로 추후실험을 진행하였다. COHEE에 의해 NO 분비량이 농도 의존적으로 감소하였으며 IL-6, TNF-α 및 IL-1β의 분비량 또한 유의적으로 감소하였다. 특히 100 μg/ml의 농도에서 LPS 단독처리구인 대조구에 비하여 IL-6, TNF-α 및 IL-1β의 분비량을 각각 66%, 43% 및 93% 억제시켰다. 이러한 결과가 전염증성 매개인자의 전사인자인 NF-κB와 MAPKs 경로에 의한 것인지 확인하기 위하여 발현량을 관찰한 결과, COHEE가 LPS 처리에 의해 현저히 증가한 단백질의 발현을 농도 의존적으로 유의성 있게 억제하였다. 또한 COHEE는 croton oil로 부종을 유발한 마우스모델에서 귀부종 억제효과를 나타내었고 250 mg/kg 농도에서 조직의 경피 및 진피 두께의 발달을 prednisolone 10 mg/kg 처리구와 유사한 정도까지 현저히 억제시키고 염증성 세포인 mast cell의 침윤 억제 효과도 확인하였다. 이를 통해 COHEE는 염증 반응의 전사인자인 NF-κB의 발현을 조절함으로써 iNOS와 COX-2의 발현을 억제하고 그에 따라 전염증성 매개인자인 NO, IL-6, TNF-α 및 IL-1β의 분비를 억제하여 항염증 활성을 가지는 것을 확인하였으며, 이 결과를 종합해볼 때, COHEE가 염증 치료제로써의 소재로 이용될 가치가 충분할 것으로 사료된다.
산수유 나무는 그 열매인 산수유와 함께 동아시아의 많은 국가에서 식용 및 약용으로 오랫동안 널리 사용되고 있다. 선행 연구들에 의하면 산수유 나무 추출물 및 구성 성분은 다양한 약리학적 잠재력을 가지고 있음에도 불구하고 부위별 추출물에 대한 체계적인 성분 분석은 아직 충분하지 않은 실정이다. 본 연구에서는 산수유 나무의 각 부위(청산수유, 적산수유, 씨앗, 줄기, 잎, 건피)의 유효 성분 함량을 분석하여 항산화 활성을 검증하였다. 본 연구의 결과에 따르면 조사된 3가지 지표 성분 중 morroniside은 청산수유와 적산수유에 가장 많이 함유하고 있었으며, loganin은 청산수유와 씨앗에 많았다. 잎, 적산수유, 건피의 함량은 비슷한 수준이었다. Cornin은 잎에 가장 많았으며, 청산수유, 적산수유 건피 순이었다. 또한 총 폴리페놀 함량은 적산수유, 청산수유, 잎 순으로 높았으며, 총 플라보노이드 함량은 잎이 가장 높았다. 그리고, DPPH radical 소거능은 잎, 씨앗, 적산수유 추출물 순으로 높은 소거 활성을 나타냈다. 따라서 항산화 활성이 가장 높은 잎 추출물의 항염증 효능을 조사하였으며, 이를 위하여 LPS로 자극된 RAW 264.7 대식세포에서 NO와 같은 염증성 매개 인자와 IL-1β와 IL-6를 포함한 염증성 사이토카인의 생성에 미치는 잎 추출물의 영향을 평가하였다. 본 연구의 결과에 의하면 세포독성이 없는 조건에서 잎 추출물은 LPS에 의하여 증가된 iNOS의 발현과 NO의 생성을 유의적으로 억제하였다. 또한 잎 추출물은 LPS에 의한 IL-1β 및 IL-6의 생성 및 발현을 현저하게 감소시켰다. 따라서 산수유 나무의 잎 추출물은 항산화제와 항염증제의 개발을 위한 천연 재료로서 잠재력이 있음을 시사하며, 관련 기전 연구가 추가로 진행되어야 할 것이다.
This study was performed to evaluate the effect of Sorghum bicolor L. Moench(Sorghum, su-su) extracts on mouse immune cell activation. As ex vivo experiment, different concentrations(0, 50, 500mg/kg B.W.) of Sorghum bicolor L. Moench water extracts were orally administrated into mouse every other day for four weeks. The proliferation of mouse splenocytes, the number of plaque forming cells(PFC) and the cytokine IL-1β production by activated macrophage were used as indices for immunocompetence. Splenocyte proliferation was enhanced in mouse orally administrated with 50mg/kg B.W./day concentration compared to that of control group. Especially, the highest proliferation of spleoncyte was seen in the mouse orally administrated at the concentration of 50mg/kg B.W./day. The number of plaque forming cells(PFC) to SRBC were significantly enhanced when compared with control group. Also, the mouse of Sorghum bicolor L. Moench water extracts 50mg/kg B.W./day supplementation group with LPS stimulation enhanced level of IL-1$\beta$ cytokine production. This study suggest that supplementation of Sorghum bicolor L. Moench water extracts may enhance the immune function by regulating the splenocytes proliferation, increasing the number of PFC and enhancing the cytokine production by activated macrophage.
뇌에서 과량의 reactive oxygen species (ROS) 생성에 의해 유발되는 산화적 스트레스는 알츠하이머 질환과 같은 신경퇴행성 질환의 원인으로 알려져 있다. 본 연구에서는 kaempferol, kaempferol-3-O-glucoside, quercetin, quercetin-3-β-D-glucoside와 같은 flavonoid와 그 배당체의 H2O2 유도 산화적 스트레스에 대한 C6 신경교세포보호 효과를 확인하였다. H2O2만을 처리한 control군은 아무것도 처리하지 않은 normal군에 비해 세포 생존율 감소와 ROS 생성 증가를 통해 C6 신경교세포의 산화적 손상이 유도되었음을 확인하였다. 반면 4가지 flavonoid를 각각 처리한 군의 경우, H2O2를 처리한 control군에 비해 세포 생존율 증가와 ROS 생성 감소를 통해 산화적 손상 억제를 통한 신경교세포 보호 효과를 확인하였다. Flavonoid의 신경교세포 보호 작용 메커니즘을 규명하기 위해, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), and interleukine (IL)-1β 등의 염증 관련 단백질 발현을 측정하였다. H2O2를 처리한 control군은 normal군에 비해 iNOS, COX-2, IL-1β 단백질 발현 증가와 IκB-α 발현 감소를 통해 신경교세포의 산화적 손상으로 인한 염증 반응을 확인하였다. 반면, 4가지 flavonoid를 각각 처리한 군의 경우 iNOS, COX-2, IL-1β 단백질 발현 감소와 IκB-α 발현 증가를 나타내어, 염증 반응 개선을 통한 신경교세포 보호 효과를 확인하였다. 특히, quercetin과 그 배당체인 quercetin-3-β-D-glucoside를 처리한 군은 kaempferol과 그 배당체인 kaempferol-3-O-glucoside를 처리한 군에 비해 우수한 신경교세포 보호 효과를 나타내었다. 본 연구는 4가지 flavonoid가 신경교세포에서 산화적 스트레스 억제를 통해 신경퇴행성 질환을 예방 및 치료할 수 있을 것으로 사료된다.
