The Effects of Hwanso-dan(Huanshaodan) Hot Water Extract & Ultra-fine Powder on Cytokine and Memory Deficit Model (환소단(還少丹)이 microglia 염증반응 cytokine과 건망증 생쥐모델에 미치는 영향)

  • Objectives : This experiment was designed to investigate the effect of the Hwanso-dan hot water extract & ultra-fine powder on microglia and memory deficit model Methods : The effects of the Hwanso-dan hot water extract on expression of IL-l${\beta}$, IL-6, TNF-${\alpha}$ mRNA and production of IL-l${\beta}$, IL-6, TNF-${\alpha}$ in BV2 microglial cell line treated by lipopolysacchaide were investigated. The effects of the Hwanso-dan hot water extract & ultra-fine powder on the behavior of the memory deficit mice induced by scopolamine and uric acid & AChE in serum of the memory deficit mice induced by scopolamine were investigated. Results : 1. The Hwanso-dan hot water extract suppressed the expression of IL-l${\beta}$, IL-6, TNF-${\alpha}$ mRNA in BV2 microglial cell line treated by lipopolysacchaide. 2. The Hwanso-dan hot water extract suppressed the production of IL-l${\beta}$, IL-6, TNF-${\alpha}$ in BV2 microglial cell line. 3. The Hwanso-dan hot water extract & ultra-fine powder decreased uric acid and AChE significantly in the serum of the memory deficit mice induced by scopolamine. 4. The Hwanso-dan hot water extract & ultra-fine powder groups showed significantly inhibitory effect on the scopolamine${\sim}$induced impairment of memory in the experiment of Morris water maze. Conclusions : This experiment shows that the Hwanso-dan hot water extract & ultra-fine powder might be effective for the prevention and treatment of Memory deficit disease. Investigation into the clinical use of the Hwanso-dan hot water extract & ultra-fine powder for Alzheimer's disease is suggested for future research.

Anti-inflammatory Effect of Artemisia Capillaris Thunberg in Lipopolysaccharide-exposed Rats (인진호(茵蔯蒿)가 LPS 염증유발 흰쥐의 전염증성 cytokine 생산 및 혈액성상에 미치는 영향)

  • Objectives : The present study investigated anti-inflammatory effect of Artemisia Capilaris Thunberg in lipopolysaccharide-exposed rats. Methods : We divided lipopolysaccharide-exposed Sprague-Dawley rats into 4 groups. They were normal group, feed with 100 mg/kg Artemisia Capillaris Thunberg group, feed with 200 mg/kg Artemisia Capillaris Thunberg group and feed with 300 mg/kg Artemisia capilaris Thunberg group. They were administered for 6 weeks. We measured counts of red blood cell(RBC), the values of hemoglobin(Hb) and packed cell volume(PCV), plasma total protein concentration, albumin concentration, the ratio of albumin/globulin, the activities of plasma glutamic oxaloacetic transaminase(GOT), glutamic pyruvic transaminase(GPT), lactate dehydrogenase(LDH), the counts of white blood cell(WBC), the ratio of neutrophils, lymphocytes, monocytes, basophils, eosinophils, the concentration of plasma interleukin-$1{\beta}$($IL-1{\beta}$), plasma interleukin-6(IL-6), plasma tumor necrosis factor-$\alpha$($TNF-{\alpha}$), plasma interleukin-10(IL-10), the concentration of liver $IL-1{\beta}$ and IL-6, $TNF-{\alpha}$, IL-10. Results : Counts of RBC and the values of Hb and PCV, plasma total protein concentration and albumin concentration, the activities of plasma GOT, GPT and LDH showed no significant difference in the treatment groups. and the ratio of albumin/globulin was increased in Artemisia Capillaris Thunberg groups. The counts of WBC showed lower values in Artemisia Capillaris Thunberg groups than those of control group, In the ratio of neutrophils Thunberg groups. The ration of monocytes, basophils and eosinophils were below 5%, and showed no characteristic trend. The concentration of plasma interleukin-$1{\beta}$($IL-1{\beta}$), plasma inerleukin-6(IL-6) and plasma tumor necrosis factor-$\alpha$($TNF-{\alpha}$) showed a lower values in the Artemisia Capillaris Thunberg groups than those of control group, and the concentration of plasma interleukin-10(IL-10) showed no significant difference in the treatment groups. The concentration of liver $IL-1{\beta}$ and IL-6 showed a lower values in the Artemisia Capillaris Thunberg groups than those of control group, however the concentration of liver $TNF-{\alpha}$ and IL-10 showed no significant difference in the treatment groups. Conclusions : The Artemisia Capillaris Thunberg groups gives positive results of anti-inflammatory response by lipopolysaccharide(LPS) derivation.

The Effect of Dohongsamul-tang on Cytokine Production in Peripheral Blood Monocular Cells of Patients with Acute Cerebral Infarction

  • Objectives : We investigated the effect of Dohongsamul-tang (DHSMT) on the production of various cytokines in lipopolysaccaride (LPS) stimulated peripheral mononuclear cells (PBMCs) from CI patients. Methods: Cell viability was determined using MTT assay. ELISA was carried out for investigating $TNF-{\alpha}$, $IL-1{\beta}$, IL-6, IL-8, IL-4, and $TGF-{\beta}$ 1 Results : The amount of tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-$1{\beta}$, IL-6, IL-8, IL-4, and transforming growth factor(TGF)-${\beta}$ 1 in PBMC culture supernatant significantly increased in the LPS treated cells compared to unstimulated cells. We show that DHSMT inhibited the production of TNF-${\alpha}$, IL-1${\beta}$, IL-6, IL-8, and IL-4 induced by LPS in a dose dependent manner. The maximal inhibition rate of the TNF-${\alpha}$, IL-1${\beta}$, IL-6, IL-8, and IL-4 production by pretreatment of DHSMT (1.0mg/ml) was 38.52 ${\pm}$ 2.5% (P < 0.01), 44.02 ${\pm}$ 3.5% (P < 0.05), 45.32 ${\pm}$ 2.3% (P < 0.01), 42.30 ${\pm}$ 3.1% (P < 0.05), and 49.70 ${\pm}$ 3.1%(P &lt; 0.05), respectively. On the other hand, DMSMT significantly increased the LPS-induced TGF-${\beta}$ 1 production(P<0.05). Conclusions : Taken together. these results suggest that DHSMT might have regulatory effects on cytokine production, which might explain its beneficial effect in the treatment of CI.

Effects of Woobangja on Anti-allergic Inflammation (우방자(牛蒡子)가 항알러지 염증반응에 미치는 영향)

  • Objective : Allergic Inflammation is related with secretion of Cytokine. This study was performed to examine the effects of Woobangja on anti-allergic inflammation. Method : While macrophage 264.7cells was chosen as a normal group a control group was classified into three groups. One was stimulated with LPS. and another was pretreated with Woobangja for 1 hour. The third was pretreated with gydrocortisone for 1 hour. After the pretreatment, macrophage were incubated with lipopolysaccharide(LPS) 100 ng/ml for 12h and media collected and $TNF-{\alpha}$, IL-6, $IL-1{\beta}$, IL-10 concentration in supernatants were measured each by Enzyme linked immuno-sorbent assay. Woobangja were used $50\;{\mu}g/ml$, $100\;{\mu}g/ml$, $250\;{\mu}g/ml$, $500\;{\mu}g/ml$, 1 mg/ml. Hydrocortisones were used respectively $10^{-8}\;M$,$10^{-7}\;M$,$10^{-6}\;M$,$10^{-5}\;M$,$10^{-4}\;M$. Results : Woobangja showed inhibitory effect on $TNF-{\alpha}$ by LPS-stimulated macrophage 264.7. The inhibitory effect was most significant in 1mg/ml(p<0.01), and has increased according to the number of doses. Woobangja also showed inhibitory effect on IL-10 by LPS-stimulated macrophasg 264.7. The inhibitory effect was most significant in $100\;{\mu}g/ml$, and was not in a dose-dependent manner as Hydrocortisone group. Woobangja and Hydrocortison showed contrary effect on $IL-1{\beta}$ in al five concentration(p<0.01), and at the lowest concentration ($50\;{\mu}g/ml$) the level of $IL-1{\beta}$ was the lowest. On the other hand hydrocortison was observed to have inhibitory effect on $IL-1{\beta}$ in all five concentration(p<0.01). IL-6 was inhibited by hydrocortison in a roughly dose-dependent manner, but was not inhibited by Woobangja. On the contrary Woobangja obviously increased the expression of $IL-1{\beta}$ in all five concentration(p<0.01), but it was not related with concentrations. Conclusion : 1. Woobangja does significantly inhibit the expression of $TNF-{\alpha}$ by LPS-stimulated macrophage 264.7. 2. Woobangja does significantly increse the expression of IL-6 by LPS-stimulated macrophage 264.7. 3. Woobangja does significantly increse the expression of $IL-1{\beta}$ by LPS-stimulated macrophage 264.7. 4. Woobangja does significantly inhibit the expression of IL-10 by LPS-stimulated macrophage 264.7. 5. Woobangja is observer to have anti-allergic inflammatory effect through inhibiting inflammatory cytokine.

Effects of Wikyung-Tang on the Lipopolysaccharide-Induced Acute Lung Injury in Mice (위경탕(葦莖湯)이 LPS로 유발된 급성 폐손상에 대한 영향)

  • Wikyung-Tang(WKT) is herbal medication used in abcess-causing respiratory disease. Previous in vitro study demonstrates that WKY presents anti-proliferative effects in A549 cells. Here we show that WKY protects mice against lipopolysaccharide(LPS)-induced acute lung injury (ALI). We pretreated mice orally with WKY(2.34 and 5.85 g/kg body weight) 1, 24 and 48 hours before intratracheal administration of LPS. For same condition, control group was pretaken orally distilled water before LPS administration. 24 hours after LPS intratracheal instillation, bronchoalveolar lavege fluids(BALF) was obtained to measure protein and proinflammatory cytokines(TNF-${\alpha}$, IL-$1{\beta}$, IL-6). Protein and proinflammatory cytokines in BALF of WKT treated groups were totally decreased. Statistically, Protein, TNF-${\alpha}$ and IL-$1{\beta}$ of high concentrate WKT treated group decreased significantly compared with control group. In conclusion, WKY had some anti-inflammatory effect in a clinically relevant model of ALI. these results indicated that WKY was effective in inhibiting ALI and might act as a potential therapeutic reagent for treating ALI in the future.

Anti-Inflammatory Effect of Fermented Artemisia princeps Pamp in Mice

  • Essential oil-excluded Artemisia princeps Pamp var Ssajuarissuk (AP) was fermented with Lactobacillus brevis K-1, which was isolated from cabbage Kimchi, and the anti-inflammatory effects of AP and fermented AP (FAP) on lipopolysaccharide (LPS)-induced inflammatory response in peritoneal macrophages were investigated. AP and FAP inhibited LPS-induced TNF-$\alpha$, IL-$1{\beta}$, COX-2, iNOS and COX-2 expression, as well as NF-${\kappa}B$ activation. AP and FAP also reduced ear thickness, inflammatory cytokine (TNF-$\alpha$, IL-$1{\beta}$ and IL-6) expression and NF-${\kappa}B$ activation with 12-O-tetradecanoylphorbol-13-acetate (TPA) induced dermatitis in mice. Furthermore, AP and FAP also reduced exudate volume, cell number, protein amount, inflammatory cytokines (TNF-$\alpha$, IL-$1{\beta}$ and IL-6) expression and NF-${\kappa}B$ activation in carrageenan-induced air pouch inflammation in mice. The inhibitory effects of FAP were more potent than those of non-fermented AP. Based on these findings, we propose that FAP can improve inflammatory diseases, such as dermatitis, by inhibiting the NF-${\kappa}B$ pathway.

The Effects of Pro-inflammatory Cytokines by Cisplatin on the Death of Sensory Hair Cells. (시스플라틴에 의한 염증성 사이토카인의 청각유모세포 사멸 효과)

  • Cisplatin (cis-diamminedichloroplatinum II : CDDP) is the most widely used anticancer drug against a variety of human neoplasms. However, its clinical use is limited by the onset of severe side effects, including ototoxicity and nephrotoxicity. Even though a number of evidences in cytotoxic mechanism of cisplatin have been suggested, the role of pro-inflammatory cytokines in cisplatin cytotoxicity of auditory cells has not yet been demonstrated. Herein our data clearly demonstrated that cisplatin decreased the viability of HEI-OC1 auditory cells, which was inhibited by the addition of neutralizing $anti-TNF-{\alpha}$, $anti-IL-1{\beta}$ and anti-IL-6 antibodies. Consistently, Neutralization with antibodies against pro-inflammatory cytokines ameliorated the cell death and disarrangement of cochlea hair cell layers in the rat primary cochlear explants which were treated with cisplatin. Furthermore, exogeneous supplementation with free radical scavengers, including GSH and NAC, significantly prevented the cytotoxicity of cisplatin in the rat primary cochlea explants. We also observed that $TNF-{\alpha}$ was predominantly expressed in Deiters and Hensen's cells located in hair cell zone of cisplatin-treated cochlear explants. These findings suggest that pro-inflammatory cytokines, including $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6, may play a pivotal role in the pathophysiology of hair cell damages caused by ototoxic drug cisplatin.

Anti-inflammatory effects of Hwangnyeonhaedok-tang and Fermented Hwangnyeonhaedok-tang (황련해독탕 및 발효황련해독탕의 항염증 효과)

  • Objective : Recently interest of fermented herb medicine has increased in Oriental medicine. Herb medicine fermented in a proper condition resultingly has better or new virtue by microbe. This study was designed to compare the anti-inflammatory effects of herbal decoction and fermented herbal decoction in Hwangnyeonhaedok-tang (Huanglian fiedu-tang; HHT). Methods : The effects of HHT and FHHT on anti-inflammation were measured by production of NO and pro-inflammatory cytokine productions including TNF-${\alpha}$ and IL-1${\beta}$, IL-6 in LPS induced RAW 264.7 cells. Results : 1. Cell viability in three doses of HHT extract (0.01, 0.05 and 0.10 mg/ml) and two doses of FHHT extract (0.01, 0.05 mg/ml) was more than 70% and 80% each. 2. In RAW 264.7 cells, LPS displayed significant stimulatory effects on the production of NO. However, both HHT, FHHT showed significant inhibitory effects on the production of NO. 3. Pre-treatment by HHT in a various concentrations (0.01, 0.05, 0.10, 0.50mg/ml) reduced the elevated production of TNF-${\alpha}$, IL-1${\beta}$, IL-1${\beta}$ mRNA, IL-6 mRNA. More than 0.05mg/ml concentrations of HHT reduced the production of IL-6. And TNF-${\alpha}$, IL-1${\beta}$ level of mouse serum reduced in a various dose(0.1, 0.5, 1.0g/kg). 4. Pre-treatment by FHHT in a various concentrations (0.01, 0.05, 0.10, 0.50mg/ml) reduced the elevated production of TNF-${\alpha}$. More than 0.05mg/ml concentrations of FHHT reduced the expression of TNF-${\alpha}$ mRNA. 0.1, 0.50mg/ml concentrations of FHHT reduced the production of IL-6. And IL-1${\beta}$ level of mouse serum reduced in a various dose(0.1, 0.5, 1.0g/kg). Conclusions : These results indicated that both HHT and FHHT could have anti-inflammatory capacity by inhibiting the production of NO and cytokines in vitro. But HHT has more strong anti-inflammatory effects than that of FHHT in broad outlines.

Effects of Ginseng Radix plus Crataegi Fructus on the Gene Expression in Relation to Alzheimer's Disease. (인삼산사복합방(人蔘山査複合方)이 Alzheimer성 치매와 관련된 유전자 발현(發顯)에 미치는 영향)

  • Objectives : This research was investigated the effect of the Ginseng Radix plus Crataegi Fructus on the gene expression in relation to Alzheimer's disease. Methods : Observed gene expression of the Ginseng Radix plus Crataegi Fructus extract on $IL-1{\beta}$, IL-6, $TNF-{\alpha}$, COX-2, and NOS-II mRNA of BV2 microglia cell line treated with lipopolysacchride. Results : The Ginseng Radix plus Crataegi Fructus extract suppressed the gene expression of $IL-1{\beta}$, IL-6, $TNF-{\alpha}$, COX-2, NOS-II mRNA in BV2 microglia cell line treated with lipopolysacchride. Conclusion : These results suggest that the Ginseng Radix plus Crataegi Fructus extract may be effective for the prevention and treatment of Alzheimer's disease. Investigation into the clinical use of the Ginseng Radix plus Crataegi Fructus extract for Alzheimer's disease is suggested for future research.

Anti-inflammatory Effects of Achyranthes japonica Nakai and Aralia continentalis Kitagawa Complex Fermented Extracts on LPS-stimulated RAW264.7 Macrophage (LPS로 자극된 RAW264.7 대식세포에서 우슬 및 땅두릅 복합 발효추출물의 항염증 효과)

  • This study investigated the anti-inflammatory effects of mixed extracts of Achyranthes japonica Nakai (Aj) and Aralia continentalis Kitagawa (Ac) (ratios of 1 : 2, 1 : 3, 1 : 5, 2 : 1, 3 : 1 and 5 : 1) on RAW264.7 macrophages. Cell toxicity was determined using a cell counting kit (CCK) assay. We evaluated anti-inflammatory effects of the mixed extracts of Aj and Ac by measuring interleukin $(IL)-1{\beta}$, IL-6, and tumor necrosis factor $(TNF){\alpha}$ using an enzyme-linked immunosorbent assay (ELISA) kit assay. The mixed extracts of Aj and Ac inhibited lipopolysaccharide (LPS)-induced $IL-1{\beta}$ and $TNF{\alpha}$ in LPS-stimulated macrophages. Comparing different ratios of the mixed extracts, the 2 : 1 ratio of Aj and Ac has much more potency and inhibited the production of $TNF{\alpha}$ in LPS-induced RAW264.7 cells. The results of the present study showed that the mixed extracts of Aj and Ac have potential anti-inflammatory effects on RAW264.7 macrophages. Therefore, these extracts may be used as a good source of functional foods for the protection against inflammatory diseases.
