Cytokine Release from Organotypic Human Oral Tissue Following Exposure to Oral Care Product Chemicals

  • Purpose: Measuring viability of a three-dimensional in vitro organotypic human oral tissue model has been suggested as an alternative test method to the oral mucosa irritation test of oral care products. The aim of this study was to investigate the production of two different cytokines using organotypic human oral tissue model following exposure to chemicals that are commonly used in oral care products. Materials and Methods: The organotypic human oral tissues were exposed to ethanol, sodium lauryl sulphate or hydrogen peroxide for 90 minutes. Following exposure, interleukin (IL)-1α and IL-8 productions were assessed and correlated with cell viability testing as well as histology of the organotypic human oral tissues. Result: High levels of IL-8 were released from organotypic human oral tissues in all of the test and control groups without any significant differences between them. In contrast, differences were found in IL-1α release between the test and control groups. Additionally, the trend of IL-1α release corresponded to the phenotypes observed in histological analysis while different trend existed between IL-1α release and cell viability. Conclusion: The study concluded the non-specific release of IL-8 for the assessment of oral care product chemicals' toxicity, while potential of measuring IL-1α cytokine level as the possible alternative test method.

Inward Leakage Test for Particulate Filtering Respirators in Korea (방진마스크의 안면부 누설률에 관한 연구)

  • Korean certification regulation for particulate filtering respirators requires inward leakage (IL) test as European Standards (EN) and the standard levels of regulation are the same as those of EN. This study was conducted to evaluate particulate filtering respirators being commercially used in the market by using IL and assess the characteristics of IL. The study began with discussing the concept of IL, comparison of IL with fit test, and IL measurement method. Three brands of half masks and 10 brands of filtering facepieces (two top class, four 1st class and four 2nd class), total 13 brands respirators, and 10 test panels (subjects) who were classified in 9 facial grids in accordance with face length and lip length, were selected for IL test. IL tests were conducted in the laboratory of 3M Innovation Center which was established by EN standard. As expected ILs of half masks were lower than those of filtering facepiece mask. ILs of half masks and some filtering facepieces were significantly different in manufacturers. ILs of 1st class filtering facepieces were found to be much more than those of 2nd class and thus the result would cause wearers to get confused to select a mask. Four of six brands being no compliance with standards were thought that they should be tested again for certification because of a lot of differences from standards. There were no significant differences among ILs of five exercises. In 6 out of 13 brands lognormal distribution of ILs may be a better fit distribution and in 7 brands both lognormal and normal distributions were rejected. The result indicates that geometric mean may be better than arithmetic mean to establish standard.

A Study on the Performance Prediction and Evaluation of Scale Down Noise Reducing Device on the Top of Noise Barrier (축소모형 방음벽 상단장치의 성능예측 및 평가에 관한 연구)

  • The purpose of this study is to set up an acoustic prediction technique and to perform the IL test of scale down noise reducing device for the development of the noise reducing device as the development of 400km/h class high speed train. First of all, the IL prediction of noise reducing device was performed with the 2D BEM method. And the noise test of scale down noise reducing device in anechoic chamber was performed for the verification of acoustic prediction technique and IL performance evaluation. As the results, the acoustic prediction technique for the development of noise reducing device was verified because the averaged IL difference between prediction and test is in 2dB(A). And the measured IL value of noise reducing device is less than 2dB(A), and additional IL with polyester absorption material is increased about 0.5dB(A).

Noninvasive Detection of Specific Diagnostic Biomarkers for Atopic Dermatitis

  • The diagnosis of atopic dermatitis (AD) includes a test that checks allergen-mediated skin reactions and a method of measuring the total IgE and allergen-specific IgE in blood. However, these test methods are performed directly on the patient, which cause some pain or discomfort. In addition, the skin response test or IgE may result in false negative in about 20% of patients. In the present study, to identify specific biomarkers, HaCaT cells were used as a human keratinocyte that make up the skin, were treated IL-4 and IL-13 for 24 hours to induce a situation similar to keratinocytes in AD patients. In the HaCaT cells, pro-inflammatory cytokine such as IL-5, IL-6, and MCP-1 were increased by IL-4 and IL-13 and skin barrier proteins was reduced by IL-4 and L-13. This results showed that a situation similar to the stratum corneum of an actual patient is induced in HaCaT cells. And then the secretions of Kallikrein (KLK) 5 and KLK7 protease were checked by enzyme-linked immunosorbent assay (ELISA). It was specifically increased by IL-4 and IL-13. This showed that AD-related protease can be detected at the protein level using keratinocytes that can be taken in a non-invasive manner and suggested the possibility of applying it to AD diagnosis.

Anti-microbial, Anti-inflammatory and Anti-oxidative Effects of Herbal Medicine Extracts as Anti-gingivitis Ingredients (치주 질환 예방 및 치료용 소재로서 수종의 생약성분 추출물에 관한 항균, 항염, 항산화 효능 연구)

  • This study was to estimate the effect of herbal medicines on periodontal disease. To screen effective materials for periodontal disease, we performed a series of test for 15 herbal medicine extracts. MIC test for P. gingivalis, IL-$1{\alpha}$ inhibition test, MMP-1 inhibition test and SOD activation test were carried out for herbal medicine extracts, respectively. The results are as followings : 1) Eunhang, Youkdoogu, Daewhang, Hoobak, Goojulcho, Yongacho, Mokhyang, Sesin, Sancho and Hoehyang extracts were effective for reduce P. gingivalis, 2) Eunhang, Jacyak, Youkdoogu, Hagocho, Hoobak, Guaru and Sanyak extracts showed high IL-$1{\alpha}$ inhibition rates, 3) Eunhang and Youkdoogu extracts have effects on MMP-1 inhibition, 4) Eunhang, Sanyak and Youkdoogu extracts effectively activate SOD. Especially, Eunhang extract has high performance through all estimations and can effectively prevent and treat periodontal disease.

  • Multivariate unit root tests for the VAR(p) model have been commonly used in time series analysis. Several unit root tests were developed and recently Shin(2004) suggested a cointegration test based on weighted symmetric estimator. In this paper, we suggest a multivariate unit root test statistic based on the weighted symmetric estimator. Using a small simulation study, we compare the powers of the new test statistic with the statistics suggested in Shin(2004) and Fuller(1996).

  • Fiber Reinforced Plastic (FRP) insulator has a higher performance than porcelain. It is only used in domestic AC 25 kV electric railway system. Seoul Metro has developed DC 1500 V polymer insulator since 2008. The test of moisture infiltration is carried out. The test results showed that all result values satisfy the test standard.

Hyaluronic acid reduces inflammation and crevicular fluid IL-1β concentrations in peri-implantitis: a randomized controlled clinical trial

  • Purpose: This study investigated the effects of hyaluronic acid (HA) on peri-implant clinical variables and crevicular concentrations of the proinflammatory biomarkers interleukin (IL)-1β and tumor necrosis factor (TNF)-α in patients with peri-implantitis. Methods: A randomized controlled trial was conducted in peri-implantitis patients. Patients were randomized to receive a 0.8% HA gel (test group), an excipient-based gel (control group 1), or no gel (control group 2). Clinical periodontal variables and marginal bone loss after 0, 45, and 90 days of treatment were assessed. IL-1β and TNF-α levels in crevicular fluid were measured by enzyme-linked immunosorbent assays at baseline and after 45 days of treatment. Clustering analysis was performed, considering the possibility of multiple implants in a single patient. Results: Sixty-one patients with 100 dental implants were assigned to the test group, control group 1, or control group 2. Probing pocket depth (PPD) was significantly lower in the test group than in both control groups at 45 days (control 1: 95% CI, -1.66, -0.40 mm; control 2: 95% CI, -1.07, -0.01 mm) and 90 days (control 1: 95% CI, -1.72, -0.54 mm; control 2: 95% CI, -1.13, -0.15 mm). There was a trend towards less bleeding on probing in the test group than in control group 2 at 90 days (P=0.07). Implants with a PPD ≥5 mm showed higher levels of IL-1β in the control group 2 at 45 days than in the test group (P=0.04). Conclusions: This study demonstrates for the first time that the topical application of a HA gel in the peri-implant pocket and around implants with peri-implantitis may reduce inflammation and crevicular fluid IL-1β levels.

Intra-articular Injection of $IL-1{\beta}$ Facilitated Formalin-induced Temporomandibular Joint Pain in Freely Moving Rats

  • The present study was performed to investigate the effects of intra-articular injection of interleukin-1${\beta}$ (IL-1${\beta}$) on the formalin-induced temporomandibular joint (TMJ) pain. Under anesthesia, a 30-gauge needle was introduced into the right TMJ region for injection of formalin. Microinjection of 50 ${\mu}l$ of 5% formalin significantly produced noxious scratching behavioral response, and the scratching behavior lasted for 40 min. Although the responses produced by formalin injection were divided into two phases, the response of 1st phase did not significantly differ from the scratching behavior response in the saline-treated group. We examined the effects of intra-articular injection of IL-1${\beta}$ on the number of noxious behavioral responses produced by 50${\mu}l$ of 5% formalin injection. Intra-articular injection of 100 pg and 1 ng of IL-1${\beta}$ significantly increased the number of behavioral responses of the 2nd phase, while 10 pg of IL-1${\beta}$ did not change the formalin-induced behavioral responses. To investigate whether IL-1 receptor was involved in the intra-articular administration of IL-1${\beta}$-induced hyperalgesic response, IL-1 receptor antagonist (IL- ra, 50 ng) was administrated together with IL-1${\beta}$ injection. IL-1${\beta}$ receptor antagonist blocked IL-1${\beta}$- induced hyperalgesic response in the TMJ formalin test. These results suggest that intra-articular injection of IL-1${\beta}$ facilitated the transmission of nociceptive information in the TMJ area.
