뇌(腦) 성상세포(星狀細胞)를 대상으로 한 Cobrotoxin의 염증(炎症) 치료(治療) 기전(機轉) 연구(硏究) (The Study of anti-inflammatory Mechanism with Cobra Venom on Astrocytes of Rats)

  • Cobrotoxin의 항염증 치료 기전에 대해 연구하기 위하여 성상세포에 LPS 및 SNP로 염증을 유도한 후 NF-${\kappa}B$와 DNA의 결합 능력, NF-kB Dependent Luciferase 발현, Astrocyte의 세포활성, NF-${\kappa}B$ 구성 단백질인 P50, P-$1{\kappa}B$, $1{\kappa}BB$ 및 염증(炎症)관련 유전자(遺傳子)인 Cox-2, iNOS, cPLA2의 발현과 GSH와 DTT로 sulf-hydryl기를 환원시 NF-${\kappa}B$와 DNA의 결합 능력, NF-${\kappa}B$ 구성 단백질인 P50 발현에 미치는 영향과 Cobrotoxin의 성상세포 내 유입 등을 관찰하여 다음과 같은 결론을 얻었다. 1. LPS 로 염증을 유발한 후 NF-${\kappa}B$와 DNA의 결합 능력을 관찰한 결과 Cobrotoxin 0.1${\mu}g/m{\ell}$ 처리군, Astrocyte 내에서의 Cobrotoxin 0.1, 0.5${\mu}g/m{\ell}$ 처리군에서 모두 대조군에 비하여 유의한 억제를 나타내었다. 2. LPS로 염증을 유발한 후 Astrocyte 내에서 NF-${\kappa}B$ Dependent Luciferase 발현을 살펴본 결과 Cobrotoxin 모든 처리군에서 대조군에 비하여 유의한 억제를 나타내었다. 3. SNP로 염증을 유발한 후 Cobrotoxin이 NF-${\kappa}B$ 구성 단백질인 P50, P-$1{\kappa}B$, $1{\kappa}B$ 발현에 미치는 영향을 살펴본 결과 P50와 $1{\kappa}B$는 Cobrotoxin 0.1, 0.5 및 $1{\mu}g/m{\ell}$ 모든 처리군에서 대조군에 비하여 유의한 억제를 나타내었고, P-$1{\kappa}B$는 Cobrotoxin $0.1{\mu}g/m{\ell}$ 처리군에서 대조군에 비하여 억제를, Cobrotoxin 0.5, $1{\mu}g/m{\ell}$ 처리군에서 각각 대조군에 비하여 유의한 억제를 나타내었다. 4. LPS로 염증을 유발한 후 Cobrotoxin이 NF-${\kappa}B$ 구성 단백질인 P50, P-$1{\kappa}B$, $1{\kappa}B$ 발현에 미치는 영향을 살펴본 결과 P50와 $1{\kappa}B$는 Cobrotoxin 0.5, $1{\mu}g/m{\ell}$ 처리군에서 각각 대조군에 비하여 유의한 억제를 나타내었다. 5. SNP로 염증을 유발한 후 Cobrotoxin이 염증(炎症) 관련 유전자(遺傳子)인 Cox-2, iNOS, cPLA2 발현에 미치는 영향을 살펴본 결과 Cox-2, iNOS, cPLA2 모두 Cobrotoxin $1{\mu}g/m{\ell}$ 처리군에서 대조군에 비하여 유의한 억제를 나타내었다. 6. LPS로 염증을 유발한 후 Cobrotoxin이 염증(炎症) 관련 견전자(遣傳子)인 Cox-2, iNOS, cPLA2 발현에 미치는 영향을 살펴본 결과 Cox-2와 cPLA2의 경우 Cobrotoxin 0.1, 0.5 및 $1{\mu}g/m{\ell}$ 모든 처리군에서, iNOS의 경우 Cobrotoxin 0.5, $1{\mu}g/m{\ell}$ 처리군에서 대조군에 비하여 유의한 억제를 나타내었다. 7. Astrocyte내에서 SNP로 염증을 유발한 후 GSH와 DTT로 sulf-hydryl기를 환원하여 NF-${\kappa}B$와 BNA의 결합 능력을 관찰한 결과 Cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 1mM Cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 5mM의 동시처리군은 각각 Cobrotoxin $0.5{\mu}g/m{\ell}$ 처리군에 비하여 유의한 증가를 나타내었다. 8. Astrocyte 내에서 LPS로 염증을 유발한 후 GSH와 DTT로 sulf-hydryl기를 환원하여 NF-${\kappa}B$와 DNA의 결합 능력을 관찰한 결과 cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 1mM Cobrotoxin $0.5{\mu}g/m{\ell}$과 DTT 5mM의 동시처리군과 Cobrotoxin $0.5{\mu}g/m{\ell}$과 GSH 1mM Cobrotoxin $0.5{\mu}g/m{\ell}$과 GSH 5mM의 동시처리군 모두에서 Cobrotoxin $0.5{\mu}g/m{\ell}$ 처리군에 비하여 유의한 증가를 나타내었다. 9. Astrocyte 내에서 SNP로 염증을 유발한 후 GSH와 DTT로 sulf-hydryl기 환원시 NF-${\kappa}B$ 구성 단백질인 P50 발현에 미치는 영향을 관찰한 결과 SNP, Cobrotoxin $1{\mu}g/m{\ell}$와 DTT 1 또는 5mM 동시처리군과 SNP, Cobrotoxin $1{\mu}g/m{\ell}$와 GSH 1 또는 5mM 동시처리군 모두에서 Cobrotoxin $1{\mu}g/m{\ell}$ 처리군에 비하여 발현의 유의한 증가를 나타내었다. 10. Cobrotoxin의 세포 내 유입 확인을 살펴본 결과 Astrocyte 내에서 Cobrotoxin이 세포내로 유입되는 것으로 나타났다. 이상의 결과로 보아 Cobrotoxin 처리는 성상 세포 들을 대상을 LPS 및 SNP로 유도된 NF-${\kappa}B$ 관련 염증 기전과 iNOS, COX-2, cPLA2와 같은 염증관련 유전자의 발현 및 NO, PGE2,에 유의한 변동을 나타내었고, 이들 결과는 Cobrotoxin의 항염증 효과 및 그 치료기전에 대하여 입증한 것이며, 향후 안전성 연구를 바탕으로 신경계 및 심혈관계 염증치료에 약침개발과 같은 적극적인 활용이 기대된다.

Celastrol ameliorates cytokine toxicity and pro-inflammatory immune responses by suppressing NF-κB activation in RINm5F beta cells

  • Upregulation of pro-inflammatory mediators contributes to ${\beta}$-cell destruction and enhanced infiltration of immune cells into pancreatic islets during development of type 1 diabetes mellitus. In this study, we examined the regulatory effects and the mechanisms of action of celastrol against cytotoxicity and pro-inflammatory immune responses in the RINm5F rat pancreatic ${\beta}$-cell line stimulated with a combination of interleukin-1 beta, tumor necrosis factor-alpha, and interferon-${\gamma}$. Celastrol significantly restored cytokine-induced cell death and significantly inhibited cytokine-induced nitric oxide production. In addition, the protective effect of celastrol was correlated with a reduction in pro-inflammatory mediators, such as inducible nitric oxide synthase, cyclooxygenase-2, and CC chemokine ligand 2. Furthermore, celastrol significantly suppressed cytokine-induced signaling cascades leading to nuclear factor kappa B (NF-${\kappa}B$) activation, including $I{\kappa}B$-kinase (IKK) activation, $I{\kappa}B$ degradation, p65 phosphorylation, and p65 DNA binding activity. These results suggest that celastrol may exert its cytoprotective activity by suppressing cytokine-induced expression of pro-inflammatory mediators by inhibiting activation of NF-${\kappa}B$ in RINm5F cells.

Salicortin suppresses lipopolysaccharide-stimulated inflammatory responses via blockade of NF-κB and JNK activation in RAW 264.7 macrophages

  • We isolated the phenolic glucoside salicortin from a Populus euramericana bark extract, and examined its ability to suppress inflammatory responses as well as the molecular mechanisms underlying these abilities, using lipopolysaccharide (LPS)-stimulated RAW264.7 cells. Salicortin inhibited iNOS expression and the subsequent production of NO in a dose-dependent manner in the LPS-stimulated RAW 264.7 cells. Salicortin significantly suppressed LPS-induced signal cascades of NF-${\kappa}B$ activation, such as IKK activation, $I{\kappa}B{\alpha}$ phosphorylation and p65 phosphorylation in RAW 264.7 cells. In addition, salicortin inhibited the LPS-induced activation of JNK, but not ERK or p38 MAPK. Furthermore, salicortin significantly inhibited production of pro-inflammatory cytokines, such as TNF-${\alpha}$, IL-$1{\beta}$ and IL-6 in the LPS-stimulated RAW 264.7 cells. These findings suggest that salicortin may show its anti-inflammatory activity by suppressing the LPS-induced expression of pro-inflammatory mediators through inhibition of NF-${\kappa}B$ and JNK MAPK signaling cascades in macrophages.

Toll-like receptors 신호전달체계 조절을 통한 resveratrol, (-)-epigallocatechin-3-gallate, curcumin의 항염증 효과 (Anti-inflammatory Effects of Resveratrol, (-)-Epigallocatechin-3-gallate and Curcumin by the Modulation of Toll-like Receptor Signaling Pathways)

  • Toll-like receptors (TLRs) induce innate immune responses that are essential for host defenses against invading microbial pathogens, thus leading to the activation of adaptive immune responses. In general, TLRs have two major downstream signaling pathways: the MyD88- and TRIF-dependent pathways, which lead to the activation of $NF-{\kappa}B$ and IRF3. Numerous studies have demonstrated that certain phytochemicals possessing anti-inflammatory effects inhibit $NF-{\kappa}B$ activation induced by pro-inflammatory stimuli, including lipopolysaccharides and $TNF{\alpha}$. However, the direct molecular targets for such anti-inflammatory phytochemicals have not been fully identified. Identifying the direct targets of phytochemicals within the TLR pathways is important because the activation of TLRs by pro-inflammatory stimuli can induce inflammatory responses that are the key etiological conditions in the development of many chronic inflammatory diseases. In this paper we discuss the molecular targets of resveratrol, (-)-epigallocatechin-3-gallate (EGCG), and curcumin in the TLR signaling pathways. Resveratrol specifically inhibited the TRIF pathway in TLR3 and TLR4 signaling, by targetting TBK1 and RIP1 in the TRIF complex. Furthermore, EGCG suppressed the activation of IRF3 by targetting TBK1 in the TRIF-dependent signaling pathways. In contrast, the molecular target of curcumin within the TLR signaling pathways is the receptor itself, in addition to $IKK{\beta}$. Together, certain dietary phytochemicals can modulate TLR-derived signaling and inflammatory target gene expression, and in turn, alter susceptibility to microbial infection and chronic inflammatory diseases.

Inhibition of the Expression of Matrix Metalloproteinases in Articular Chondrocytes by Resveratrol through Affecting Nuclear Factor-Kappa B Signaling Pathway

  • In the present study, we tried to examine whether resveratrol regulates the expression of matrix metalloproteinases (MMPs) through affecting nuclear factor-kappa B ($NF-{\kappa}B$) in articular chondrocytes. Rabbit articular chondrocytes were cultured in a monolayer, and reverse transcription-polymerase chain reaction (RT-PCR) was used to measure interleukin-${\beta}$ ($IL-1{\beta}$)-induced gene expression of MMP-3, MMP-1, MMP-13, a disintegrin and metalloproteinase with thrombospondin motifs-4 (ADAMTS-4), ADAMTS-5 and type II collagen. Effect of resveratrol on $IL-1{\beta}$-induced secretion of MMP-3 was investigated in rabbit articular chondrocytes using western blot analysis. To elucidate the action mechanism of resveratrol, effect of resveratrol on $IL-1{\beta}$-induced $NF-{\kappa}B$ signaling pathway was investigated in SW1353, a human chondrosarcoma cell line, by western blot analysis. The results were as follows: (1) resveratrol inhibited the gene expression of MMP-3, MMP-1, MMP-13, ADAMTS-4, and ADAMTS-5, but increased the gene expression of type II collagen; (2) resveratrol reduced the secretion of MMP-3; (3) resveratrol inhibited $IL-1{\beta}$induced activation (phosphorylation) of inhibitory kappa B kinase (IKK), and thus phosphorylation and degradation of inhibitory kappa $B{\alpha}$ ($I{\kappa}B{\alpha}$); (4) resveratrol inhibited $IL-1{\beta}$-induced phosphorylation and nuclear translocation of $NF-{\kappa}B$ p65. This, in turn, led to the down-regulation of gene expression of MMPs in SW1353 cells. These results suggest that resveratrol can regulate the expression of MMPs through affecting $NF-{\kappa}B$ by directly acting on articular chondrocytes.

봉약침액(蜂藥鍼液)이 Scopolamine으로 유발(誘發)된 기억(記憶) 장애(障碍)에 미치는 영향(影響) (The Effect of Bee Venom on Scopolamine Induced Memorial Impairment)

  • Alzheimer's disease (AD) is the most prevalent form of neurodegenerative disease associated with aging in the human population. This disease is characterized by the following 4 structural changes : Atrophy of the Cortex, Parasympathetic, and other neural cells, the existence of Neurofibrillary tangles (NFTs), and the accumulation of Senile plaques. NFTs and Senile plaques is known to be the index of this disease. Senile plaques disturbs the neutro transmission and depletes of Acetylcholine. So, Recovery of Acetylcholine is the primal objective for treating Alzheimer's disease. So, Inhibiting the activity of Acetylcholine Esterase (AChE), which causes the hydrolysus of acetylcholine into choline and acetate, can be seen as a key role for treating Alzheimer's disease. Increasing body of evidence has been demonstrated that Bee Venom Acupuncture (BV) could compete with complex protein involving in multiple step of $NF-_{\kappa}B$ activation and exert the anti -inflammatory potential of combined inhibition of the prostanoid and nitric oxide synthesis systems by inhibition of IKK and $NF-_{\kappa}B$. The effect of BV through behavioral tests after memory impairment induced by Scopolamine. We examined the improving effect of BV on the Scopolamine (1 mg/Kg, i.p.)-induced memorial impairment using passive avoidance response and water maze tests in the mice. BV (0.84, $1.67\;{\mu}g/ml$) reversed the Scopolamine-induced memorial impairment in dose dependent manner. This study therefore suggests that BV acupuncture method may be useful for prevention of development or progression of AD.

Heme oxygenase-1 (HO-1)/carbon monoxide (CO) axis suppresses RANKL-induced osteoclastic differentiation by inhibiting redox-sensitive NF-κB activation

  • Heme oxygenase (HO-1) catalyzes heme to carbon monoxide (CO), biliverdin/bilirubin, and iron and is known to prevent the pathogenesis of several human diseases. We assessed the beneficial effect of heme degradation products on osteoclastogenesis induced by receptor activator of NF-${\kappa}B$ ligand (RANKL). Treatment of RAW264.7 cells with CORM-2 (a CO donor) and bilirubin, but not with iron, decreased RANKL-induced osteoclastogenesis, with CORM-2 having a more potent anti-osteogenic effect. CORM-2 also inhibited RANKL-induced osteoclastogenesis and osteoclastic resorption activity in marrow-derived macrophages. Treatment with hemin, a HO-1 inducer, strongly inhibited RANKL-induced osteoclastogenesis in wild-type macrophages, but was ineffective in $HO-1^{+/-}$ cells. CORM-2 reduced RANKL-induced NFATc1 expression by inhibiting IKK-dependent NF-${\kappa}B$ activation and reactive oxygen species production. These results suggest that CO potently inhibits RANKL-induced osteoclastogenesis by inhibiting redox-sensitive NF-${\kappa}B$-mediated NFATc1 expression. Our findings indicate that HO-1/CO can act as an anti-resorption agent and reduce bone loss by blocking osteoclast differentiation.

Smilax guianensis Vitman Extract Prevents LPS-Induced Inflammation by Inhibiting the NF-κB Pathway in RAW 264.7 Cells

  • Nutraceutical treatments can reduce inflammation and prevent the development of inflammatory diseases. In this study, the anti-inflammatory effects of Smilax guianensis Vitman extract (SGE) were examined. SGE suppressed lipopolysaccharide (LPS)-mediated nitrite production in RAW 264.7 cells. SGE also prevented the LPS-induced expression of inducible nitric oxide synthase (iNOS) but not cyclooxygenase (COX)-2. Western blot analysis showed that SGE attenuated LPS-induced phosphorylation of IκB kinase (IKK), inhibitor of kappa B (IκB), and p65. Additionally, SGE inhibited LPS-induced IκB degradation in RAW 264.7 cells. Western blot analysis of the cytosolic and nuclear fractions, as well as immunofluorescence assay results, revealed that SGE suppressed LPS-induced p65 nuclear translocation in RAW 264.7 cells. Moreover, SGE reduced LPS-induced interleukin (IL)-1β, IL-6, and tumor necrosis factor-α (TNF-α) mRNA expression and IL-1β and IL-6 protein expression in RAW 264.7 cells. Collectively, these results indicate that SGE suppresses the NF-κB signaling pathway and thereby inhibits the production of NO, IL-1β, and IL-6.

Tussilagone suppressed the production and gene expression of MUC5AC mucin via regulating nuclear factor-kappa B signaling pathway in airway epithelial cells

  • In the present study, we investigated whether tussilagone, a natural product derived from Tussilago farfara, significantly affects the production and gene expression of airway MUC5AC mucin. Confluent NCI-H292 cells were pretreated with tussilagone for 30 min and then stimulated with EGF (epidermal growth factor) or PMA (phorbol 12-myristate 13-acetate) for 24 h or the indicated periods. The MUC5AC mucin gene expression was measured by RT-PCR. Production of MUC5AC mucin protein was measured by ELISA. To elucidate the action mechanism of tussilagone, effect of tussilagone on PMA-induced $NF-{\kappa}B$ signaling pathway was investigated by western blot analysis. Tussilagone significantly inhibited the production of MUC5AC mucin protein and down-regulated the expression of MUC5AC mucin gene, induced by EGF or PMA. Tussilagone inhibited PMA-induced activation (phosphorylation) of inhibitory kappa B kinase (IKK), and thus phosphorylation and degradation of inhibitory kappa Ba ($I{\kappa}B{\alpha}$). Tussilagone inhibited PMA-induced phosphorylation and nuclear translocation of nuclear factor kappa B ($NF-{\kappa}B$) p65. This, in turn, led to the down-regulation of MUC5AC protein production in NCI-H292 cells. These results suggest that tussilagone can regulate the production and gene expression of mucin by acting on airway epithelial cells through regulation of $NF-{\kappa}B$ signaling pathway.

A Novel Synthetic Compound, YH-1118, Inhibited LPS-Induced Inflammatory Response by Suppressing IκB Kinase/NF-κB Pathway in Raw 264.7 Cells

  • For the search of a potent first-in-class compound to inactivate macrophages responsible for inflammatory responses, in the present study, we investigated the anti-nflammatory effects of YH-1118, a novel synthetic compound, in a lipopolysaccharide (LPS)-stimulated mouse macrophage cell line, Raw 264.7. YH-1118 inhibited LPS-induced nitric oxide (NO) production and inducible NO synthase (iNOS) expression at both the protein and mRNA levels. The suppression of LPS-induced iNOS expression by YH-1118 was mediated via nuclear factor kappa B (NF-κB), but not activator protein-1 (AP-1) transcription factor. This was supported by the finding that YH-1118 attenuated the phosphorylation of inhibitor of κBα (IκBα) and nuclear translocation and DNA binding activity of NF-κB. Through the mechanisms that YH-1118 inhibited the activation of IκB kinases (IKKs), upstream activators of NF-κB, or p38 MAPK, YH-1118 significantly suppressed LPS-induced production of pro-inflammatory cytokines, tumor necrosis factor-α, interleukin-1β (IL-1β), and IL-6 (p < 0.05). In conclusion, our results suggest that YH-1118 inhibits LPS-induced inflammatory responses by blocking IKK and NF-κB activation in macrophages, and may be a therapeutic candidate for the treatment of various inflammatory diseases.
