Effects of Perinatal Nutrition on Metabolic and Hormonal Profiles of Goat Kids (Capra hircus) during Their First Day of Life

  • The aim of the present work was to monitor metabolic and hormonal profiles in newborn kids, born from dams fed diets with low or high levels of energy requirements. Starting from the last month of pregnancy, 14 goats were randomly allocated to two groups: Group LD (low diet) and Group HD (high diet) that received a diet that covered 80% and 140% of their energy requirement, respectively. At delivery, the kids were weighed and a blood sample was taken before they suckled colostrum (Time 0) and 1, 2, 3, 12 and 24 h after they started suckling. Plasma insulin, IGF-I, glucose, fT3 and fT4 concentrations were not influenced by the dietary treatments, but a significant effect of time was observed as they progressively increased during the first 12 h of life. Plasma cholesterol, triglyceride, albumin, globulin and total protein plasma concentrations were significantly higher in Group HD than those of Group LD. In Group HD, cortisol concentrations were significantly lower than those of Group LD. Positive correlations were observed between LW and IGF-I (r = 0.71; p<0.05), plasma insulin and glucose (r = 0.79; p<0.05) and total protein and globulin concentrations (r = 0.97; p<0.001). Our results show that perinatal nutrition affects newborn goat kids' metabolic and hormonal profile.

Glucosamine Hydrochloride and N-Acetylglucosamine Influence the Response of Bovine Chondrocytes to TGF-β3 and IGF in Monolayer and Three-Dimensional Tissue Culture

  • BACKGROUND: Glucosamine hydrochloride (GlcN HCl) has been shown to inhibit cell growth and matrix synthesis, but not with N-acetyl-glucosamine (GlcNAc) supplementation. This effect might be related to an inhibition of critical growth factors (GF), or to a different metabolization of the two glucosamine derivatives. The aim of the present study was to evaluate the synergy between GlcN HCl, GlcNAc, and GF on proliferation and cartilage matrix synthesis. METHOD: Bovine chondrocytes were cultivated in monolayers for 48 h and in three-dimensional (3D) chitosan scaffolds for 30 days in perfusion bioreactors. Serum-free (SF) medium was supplemented with either growth factors (GF) $TGF-{\beta}$ ($5ng\;mL^{-1}$) and IGF-I ($10ng\;mL^{-1}$), GlcN HCl or GlcNAc at 1mM each or both. Six groups were compared according to medium supplementation: (a) SF control; (b) SF + GlcN HCl; (c) SF + GlcNAc; (d) SF + GF; (e) SF + GF + GlcN HCl; and (f) SF + GF + GlcNAc. Cell proliferation, proteoglycan, collagen I (COL1), and collagen II (COL2) synthesis were evaluated. RESULTS: The two glucosamines showed opposite effects in monolayer culture: GlcN HCl significantly reduced proliferation and GlcNAc significantly augmented cellular metabolism. In the 30 days 3D culture, the GlcN HCl added to GF stimulated cell proliferation more than when compared to GF only, but the proteoglycan synthesis was smaller than GF. However, GlcNAc added to GF improved the cell proliferation and proteoglycan synthesis more than when compared to GF and GF/GlcN HCl. The synthesis of COL1 and COL2 was observed in all groups containing GF. CONCLUSION: GlcN HCl and GlcNAc increased cell growth and stimulated COL2 synthesis in long-time 3D culture. However, only GlcNAc added to GF improved proteoglycan synthesis.

Association of Insulin Receptor Substrate-1 G972R Variant with Non-small Cell Lung Cancer Risk

  • Background: The insulin receptor substrate-1 (IRS-1) is the primary docking molecule for the insulin-like growth factor I receptor (IGF-IR), and is required for activation of the phosphatidylinositol 3'-kinase (PI3K) pathway. IRS-1 activation of the (PI3K) pathway regulates IGF-mediated survival, enhancement of cellular motility and apoptosis. Therefore, we attempted to ascertain whether IRS-1 genetic variations affect an individual's risk for non-small cell lung cancer (NSCLC). Methods: Two-hundred and eighteen subjects, either diagnosed with NSCLC or control subjects, were matched by age, gender and smoking status. Genomic DNA from each subject was amplified by PCR and analyzed according to the restriction fragment length polymorphism (RFLP) profile to detect the IRS-1 G972R polymorphism. Results: The frequencies of each polymorphic variation, in the control population, were as follows: GG=103 (94.5%) and GR=6 (5.5%); for the NSCLC subjects, the genotypic frequencies were as follows: GG=106 (97.2%) and GR=3 (2.8%). We could not demonstrate statistically significant differences in the genotypic distribution between the NSCLC and the control subjects (p=0.499, Fisher's Exact test). The relative risk of NSCLC, associated with the IRS-1 G972R polymorphic variation, was 1.028 (95% CI; 0.63~9.90). In addition, we found no differences between polymorphic variants with regard to the histological subtype of NSCLC. Conclusion: We did not observe any noteworthy differences in the frequency of the IRS-1 G972R polymorphism in NSCLC patients, compared to control subjects. These results suggest suggesting that, in our study population, the IRS-1 G972R polymorphism does may not appear to be associated with an increased risk of NSCLC.

초저열량식이로 인한 체력 저하에서 산야초추출물의 지구력 증진 효과 (Effect of Wild Plant Extracts Based on Korea Traditional Prescription on Endurance Exercise Performance during Calorie Restriction Period)

  • 본 연구에서는 저열량식이요법의 부작용 증세중 체력저하를 개선할 수 있도록 동의보감의 처방중 체력 증진과 연관된 보음(補陰), 보혈(補血) 기능을 갖는 사물탕(四物楊)을 기반으로 하여 개발된 생약 조성물의 체력 증진 기능을 규명하고자 실시하였다. 저열량식이요법의 모델로서 섭취 식이량의 50%를 제한하는 제한식이를 2주간 실시한 결과 제한식이를 섭취한 군은 강제 수영 모델에서 총 수영 시간이 감소하고 근육내 ATP 함량의 감소, 혈액중 젖산 함량의 증가가 나타나 식이 제한에 의해서 체력 감소가 나타남을 확인할 수 있었다. 본 연구에서 사용된 산야초 추출물을 동일하게 식이제한을 한 실험동물에게 섭취시킨 결과 총수영시간의 증가와 근육내 ATP의 증가, 젖산 함량의 감소가 나타나 저열량식이에 의한 부작용인 체력 저하를 산야초 추출물이 완화시킬 수 있는 기능이 있음을 확인 할 수 있었다. 또한, 산야초 추출물의 투여는 체력과 높은 연관성이 있는 것으로 알려진 혈중 IGF-1의 농도를 증가시키는 것이 관찰되어 체력 향상에 도움을 주는 것으로 추측된다. 식이 제한 환경에서 나타날 수 있는 천연물 추출물의 독성을 확인하기 위하여 간기능 지표인 GOT, GPT를 측정한 결과 특별한 변화가 나타나지 않아 산야초 추줄물의 안전성을 확인할 수 있었다.

미역부산물의 첨가가 홀스타인 비유우의 비유성적과 내분비생리에 미치는 영향 (Effect of Brown Seaweed Waste Supplementation on Lactational Performance and Endocrine Physiology in Holstein Lactating Cows)

  • 본 시험은 미역분말의 첨가가 홀스타인 젖소의 유생산과 내분비계에 미치는 영향을 규명하기 위하여 실시하였다. 기초사료에 미역분말을 1일 두당 800g 수준으로 급여하여 대조구와 비교하였다. 사료건물섭취량은 처리에 의한 영향을 받지 않았고 비유말기 산유량은 대조구에 비해 일일 6.25kg 이나 많게 유의하게 증가하였다(p<0.05). 하지만 유성분은 처리구간에 유의한 차이를 나타내지 않았다. 대조구의 체세포수는 유동적이었지만 처리구의 체세포수는 시험 종료 시 $10^5$개/ml로 시험개시에 비해 무려 53.77%의 감소효과를 보여 탁월한 체세포 감소효과를 나타내었다. 미역분말의 첨가가 혈중 면역글로부린의 함량에는 영향을 주지 않은 것으로 나타났다. 비유에 직접적인 영향을 미치는 IGF-1의 혈장농도는 미역처리구에서 유의성 있게 증가하였으며 비유와 직접적인 관련이 있는 또 다른 호르몬인 $T_3$$T_4$의 농도도 유의적으로 증가하는 양상을 나타내었다. 지질대사산물인 TC, HDL-C와 LDL-C의 농도는 모두 처리구에서 증가하는 양상을 나타냈다. 이와 같이 미역부산물의 사료 중 첨가는 유생산량의 증가를 보였는데 이는 혈중 비유관련 호르몬의 분비에 영향을 준 결과로 확인된 바 젖소사료자원으로서의 응용가치가 클 것으로 사료된다.

뇌종양세포주에서의 성장인자수용체의 발현 : 면역독소 치료의 연구를 위한 예비실험 (Growth Factor Receptor Expression on Brain Tumor Cell Lines : Preliminary Study for in vitro and in vivo Experiments of Immunotoxin Therapy)

  • 목 적 : 종양세포의 세포면에서는 성장인자 수용체들이 과발현되고 이들이 성장인자들과 결합함으로서 세포의 성장을 촉진하게 된다. 원발성 악성 뇌종양에 대한 새로운 치료법 중 하나가 면역독소를 사용하는 방법이다. 독소는 대상 세포의 세포면에 있는 수용체를 통해 세포질로 이행되며, 면역독소가 선택적인 항종양작용을 나타내기 위해서는 수용체가 과발현되어야 한다. 강력한 세포독성의 효과가 있음에도 불구하고 면역독소를 환자에 투여하였을 때 종양을 완치시키지는 못하였는데, 면역독소의 효과에 영향을 미치는 여러 인자들에 대한 더 많은 연구가 요구된다. 이러한 연구에 사용되어질 뇌종양세포주들에서 성장인자수용체들의 발현 여부를 알아보기 위해 본 연구를 시행하였다. 대상 및 방법 : 한 개의 수모세포종 세포주(Daoy)와 두 개의 교모세포종 세포주(U373 MG, T98 G)에서, transferrin 수용체, insulin-like growth factor-1 수용체, 그리고 interleukin-4 수용체들의 발현을 flow cytometric analysis를 이용하여 조사하였다. 결 과 : Transferrin 수용체와 interleukin-4 수용체는 Daoy, U373 MG, 그리고 T98 G 모두에서 발현되었다. Insulin-like growth factor-1 수용체는 Daoy와 U373 MG에서는 발현되었지만 T98 G에서는 발현되지 않았다. 결 론 : Transferrin 수용체와 interleukin-4 수용체는 면역독소 치료에 적합한 것으로 보인다. 본 실험의 결과는 상기의 세포주를 사용하여 면역독소와 관련된 실험을 하는데 참고가 되어야하며, 면역독소 치료에 있어서 적절한 면역독소를 선택하는 문제 등 치료 모델을 확립하는데 고려되어야 할 것이다.

Growth hormone and receptor gene mutations in Chinese Banna miniature pig

  • The Banna miniature pig (BNMP) is a representative miniature pig breed in China. Even though BNMP dwarfism is obvious, its underlying causative mutations remain unknown. In this study, the BNMP and Large White pig (LWP) serum growth hormone (GH) and insulin-like growth factor (IGF-1) levels were detected by ELISA and compared. BNMP serum IGF-1 levels were significantly lower than LWP levels (P<0.05). The miniature condition may arise from mutations in the GH and GH receptor (GHR) genes. Therefore, GH and GHR cDNA from the BNMP were cloned into a pMD18-T vector by RT-PCR using the total RNA obtained from the BNMP's pituitary and liver tissues. Sequencing results indicated that the open reading frame of the BNMP GH gene is composed of a 26-residue signal peptide and a 191-residue mature peptide. The coding sequence of the BNMP GHR gene contained 639 amino acids, including a signal peptide that is 18 amino acids long. Two amino acid substitutions, A09V and R22Q, were found in the signal peptide of the GH gene. Additionally, the S104P mutation was found in the BNMP's mature GH protein. Four mutations in the cytoplasmic domain of GHR may influence the downstream signal transduction of GHR, which needs further experimental evidence.

Differential effects of type 1 diabetes mellitus and subsequent osteoblastic β-catenin activation on trabecular and cortical bone in a mouse mode

  • Type 1 diabetes mellitus (T1DM) is a pathological condition associated with osteopenia. $WNT/{\beta}$-catenin signaling is implicated in this process. Trabecular and cortical bone respond differently to $WNT/{\beta}$-catenin signaling in healthy mice. We investigated whether this signaling has different effects on trabecular and cortical bone in T1DM. We first established a streptozotocin-induced T1DM mouse model and then constitutively activated ${\beta}$-catenin in osteoblasts in the setting of T1DM (T1-CA). The extent of bone loss was greater in trabecular bone than that in cortical bone in T1DM mice, and this difference was consistent with the reduction in the expression of ${\beta}$-catenin signaling in the two bone compartments. Further experiments demonstrated that in T1DM mice, trabecular bone showed lower levels of insulin-like growth factor-1 receptor (IGF-1R) than the levels in cortical bone, leading to lower $WNT/{\beta}$-catenin signaling activity through the inhibition of the IGF-1R/Akt/glycogen synthase kinase $3{\beta}$ ($GSK3{\beta}$) pathway. After ${\beta}$-catenin was activated in T1-CA mice, the bone mass and bone strength increased to substantially greater extents in trabecular bone than those in cortical bone. In addition, the cortical bone of the T1-CA mice displayed an unexpected increase in bone porosity, with increased bone resorption. The downregulated expression of WNT16 might be responsible for these cortical bone changes. In conclusion, we found that although the activation of $WNT/{\beta}$-catenin signaling increased the trabecular bone mass and bone strength in T1DM mice, it also increased the cortical bone porosity, impairing the bone strength. These findings should be considered in the future treatment of T1DM-related osteopenia.

Four Dammarane Triterpenes and Their Inhibitory Properties Against Eight Receptor Tyrosine Kinases

  • In recent years, tyrosine kinases (TKs) have been the target to combat cancers, and most of the developed inhibitors are of synthetic origin. Natural compounds that have the properties as the TK's inhibitors are very limited. This paper described the isolation of a new dammarane triterpene from the tree bark of Sandoricum koetjape, along with three known related dammaranes from the damar resin of Shorea javanica, as well as their inhibitory properties against eight receptor TKs (RTKs: EGFR, HER2, HER4, IGF1R, InsR, KDR, PDGFRα, and PDGFRβ). Based on the NMR and mass spectral data the new compound was identified as (12β,20S)-12,20-dihydroxy-3,4-seco-dammaran-4,24-dien-3-oic acid (12β-hydroxydammarenolic acid) (1), while the three known compounds were identified as (20S)-20-hydroxy-3,4-seco-dammaran-4,24-dien-3-oic acid (dammarenolic acid) (2), (3β,20S)-3,20-dihydroxydammaran-24-ene (3), and (20S)-3-oxo-20-hydroxydammaran-24-ene (4). The tyrosine kinase assay of the four compounds resulted only 1 and 2 at concentration of 10 μM that had weak activity against EGFR and InsR, with their % inhibitory were 30%, 27% (1), 45%, and 32% (2), respectively. The results suggested that the presence of a linear carboxylic acid group in both compounds could be of significance to the inhibitory properties against the two RTKs.
