Morphological studies on kidney in cyclophosphamide-treated NOD mice (Cyclophosphamide를 투영한 NOD 마우스의 신장에 대한 형태학적 연구)

  • This study was carried out to investigate diabetic renal changes in cyclophosphamide(CY) treated nonobese diabetic(NOD) mice and to develop animal model of diabetic human nephropathy. The 8-week-old NOD and ICR mice were injected with cyclophosphamide intraperitoneally at 200mg/kg body weight and compared the chemical effects on these mice with the non-treated NOD and ICR mice respectively. The renal glomeruli in ICR, cyclophosphamide-treated ICR(ICR+CY), NOD and cyclophosphamide-treated NOD(NOD+CY) mice were observed by the light and electron microscopes. The results obtained were summarized as follows ; 1. Spontaneous incidences of diabetes mellitus in NOD mice were significantly promoted by dosing with cyclophosphamide. 2. Glomerulohypertrophy, proliferation of mesangium, partial thickening of glomerular basement membrane, and partial fusion of pedicels of podocyte were observed in NOD mice and NOD+CY mice. These changes were not observed in both ICR mice and ICR+CY mice. 3. The diabetic nephropathy observed in NOD+CY mice was more severe than that of non-treated NOD mice.

Acute Intravenous and Oral Toxicity of DWC-751 in Rats and Mice (랫드 및 마우스에서 DWC-751의 급성정맥 및 경구 독성시험)

  • Single intravenous and oral administration to SD rats and ICR mice of both sexes were performed to investigate the acute toxicity of DWC-751, a new parenteral cephalosporin. $LD_50$ values for ICR mice and SD rats administered intravenously with DWC-751 were as follows; 1151.1 mg/kg (male SD rat), 1183.5 mg/kg (female SD rat), 2698.1 mg/kg (male ICR mouse), 2833.0 mg/kg (female ICR mouse). It is suggested that $LD_50$ values in rats and mice of both sexes would be 5000 mg/kg in oral route. Major general symptoms induced by injection intravenously with DWC-751 are decreased motor activity, increased respiratory rate, tremor and convulsion. In oral route, piloerection and soft stool are observed to 4 day after administration. No significant body weight changes were observed at any level in the groups administered with DWC-751. The gross finding of rats administered intravenously was observed cecum distension.

Embryonic Effects of Radiation on ICR Mice Depending Developmental Stages (ICR 마우스의 주요기관 형성기에 있어서 기형 발생의 시기특이성에 관한 실험적 검토)

  • The ICR pregnant mice were irradiated at 1.5Gy in every 6 hours in the period of organogenesis in order to classify the stage specificity of the embryonic effects of radiation and the stage of development differentiation of the primordium of each major organ. Intrauterine death, fetal body weight and external malformation in live fetuses were observed on day 18 of gestation. There was no statistically significant difference in the intrauterine mortality at any stage organogenesis. The fetal body weight of the mice irradiated in the intermediate stage of organogenesis showed significantly lower. There were specific highly sensitive stages in the incidences of each external malformation, that is exencephalia, open eyelid, cleft palate, anomalies of extremities and anomalies of the tail. At these stage, the primordia of the major organs are established in ICR mice.

Invasion of Metagonimus yokogawai into the submucosal layer of the small intestine of immunosuppressed mice (면역억제 마우스에 있어서 요코가와흡충의 소장 점막하 조직 침입)

  • Metosonimus yokosawai was found deeply invaded into the submucosa of the small intestine of mice (ICR) when they were immunosuppressed by prednisolone injection. Experimental groups consisted of control, fluke infection (1,800 metacercariae per mouse) and fluke infection plus immunosuppression. In fluke infection group, many worms were found sectioned in the intervillous space of the jejunum and ileum at 6 hrs, 12 hrs, and 1 day after infection, and pathological changes characterized by villous atrophy and crypt hyperplasia were observed. After 3 days, only a few worms were found in intestinal sections, and after 7 days, the pathological changes became minimal. No worm was found penetrated beyond the mucosal layer. On the other hand, in immunosuppressed mice, numerous worms were found sectioned in the duodenum and jejunum, irrespective of the infection period up to 14 days. Pathological changes of the mucosa were minimal until 3 days after infection, but at 5 days marked destruction of the mucosal layer was observed. At this time many flukes were found invaded deeply into the submucosa facing the muscular layer. Despite continuous immunosuppression, the mucosal damage was gradually recovered at 7-21 days post-infection. The results showed that immunosuppression of ICR mice can induce, for a short perid of time, severe mucosal damage, and allow deep invasion of M. yokogcuwai into the submucosa of the small intestine.

  • Hematological studies in two strains of normal laboratory mice, namely ICR and ddY which were produced in Koren were carried out. Blood analysis was performed by using different methods, a fully automated analyzer (ELT 1, 500), Coulter counter (model S plus IV) and manual technique to examine possible differences resulting from methodolgies'slight differences among the values of blood parameters were obtained. There was a slight difference noted between sexes, however, similar values were obtained from both strains.

Acute Oral Toxicity of AG-OS, Water Treatment Agent, in Rats and Mice (렛드와 마우스에서 Ag-Os(수처리제)의 급성 독성)

  • Ag-Os, water treatment agent, was administered orally to ICR mice and Sprague-Dawley rats to investigate the acute oral toxicity. $LD_{50}$ values were above 5 g/kg, 2,000 fold higher than the expected concentration in water, in both species with oral administration. There were also no differences in body weight changes, clinical signs and atopsy findings between all treated groups and control group. Therefore, it was concluded that Ag-Os is a very safe compound.

Toxicity Evaluation of Irradiated Tarakjuk for Three Months (감마선 조사 타락죽의 3개월 반복투여 독성평가)

  • This study was conducted to evaluate the possible subacute toxicity of gamma-irradiated Tarakjuk. Tarakjuk was irradiated at a dose of 30 kGy at room temperature. For the animal study, AIN-93G as a control diet and 30 kGy irradiated and non-irradiated Tarakjuk diets were administered to male and female ICR mice (10 mice per group) for 3 months. During the experimental period, the group fed 30 kGy irradiated Tarakjuk did not show any changes in appearance, behavior, mortality, body weight, organ weight, or food consumption compared to control. Further, all biochemical parameters were in normal ranges. In the histopathological examination of liver and kidney tissues of ICR mice, there were no significant differences between the control and 30 kGy irradiated Tarakjuk groups. These results indicate that Tarakjuk irradiated at 30 kGy did not cause any toxic effects under these experimental conditions.

Acute Toxicity of Leuconostoc citreum GJ7 Isolated from Kimchi in Mice (김치로부터 분리한 Leuconostoc citreum GJ7의 마우스에 대한 급성독성)

  • The present study was carried out to investigate the potential acute toxicity of Leuconostoc citreum GJ7 (Leu. citreum GJ7), a lactic acid bacterium isolated from Kimchi, in ICR male and female mice. The test article was administered to the mice orally or intraperitoneally. Mortality rates, clinical findings, and body weight changes were monitored for the 2 weeks following administration. The results showed that in 50% of the cases, lethal doses ($LD_{50}$) of Leu. citreum GJ7 were determined as >5, 000 mg/kg (p.o.) and >2500 mg/kg (i.p.) in both sexes. There were no significant changes in general conditions, body weights clinical signs and any gross lesions between vehicle control and Leu. citreum GJ7-treated groups. Hence, it is suggested that Leu. citreum GJ7 does not induce any significant acute toxicity in ICR mice.

The Long Term Effect of Buchu (Chinese chives) Diet on ROS Formation in the Liver and Skin Tissue of ICR mice (장기간의 부추식이가 ICR 마우스의 간과 피부조직의 활성산소종 생성에 미치는 영향)

  • To investigate the long term effect of buchu (Chinese chives) diet on ROS formation in the liver and stin tissue of ICR mice, one of control, 2% or 5% buchu-added diet was fed to ICR mice for 12 months. Superoxide anion (O2ㆍ), hydrogen peroxide($H_2O$$_2$) and hydroxyl radical (ㆍOH) contents were measured in cytosol, microsome, mitochondria of liver and skin of mice, respectively. Behu diet showed a significant decrease of superoxide anion, hydrogen peroxide and hydroxyl radical contents in liver and skin tissues compared to control diet, and this effect is especially higer at 5% than at 2% buchu diet level. ICR mice showed an age-dependent increase in ROS contents, while the group fed buchu diet decreased its ROS contents significantly and ROS contents of liver appeared to be 2 fold higher than skin. The results of the present study suggest that antioxidative components and sulfur-compounds in buchu diet appear to be responsible for the inhibition of ROS formation in ICR mice.

  • The present study was carried out to investigate the in vivo single-dose acute toxicity of Bacillus subtilis JNS isolated from Cheonggukjang, which is a probiotic candidate showing strong and broad antibacterial activity. The test sample was orally administrated to male and female ICR mice at a highest dose of 2,000 mg/kg for 14 days. No significant change in general conditions, mortalities, body weight changes, clinical signs, autopsy findings, or presence of gross lesions was observed in either sex of mice. The results indicate that up to 2,000 mg/kg of B. subtilis JNS had no adverse effect on ICR mice.
