Lactosylceramide Mediates the Expression of Adhesion Molecules in TNF-${\alpha}$ and IFN ${\gamma}$-stimulated Primary Cultured Astrocytes

  • Here we have investigated how lactosylceramide (LacCer) modulates gene expression of adhesion molecules in TNF-${\alpha}$ and IFN ${\gamma}$ (CM)-stimulated astrocytes. We have observed that stimulation of astrocytes with CM increased the gene expression of ICAM-1 and VCAM-1. D-Threo-1-phenyl-2-decanoylamino-3-morpholino-1-propanol (PDMP) and N-butyldeoxynojirimycin (NBDNJ), inhibitors of glucosylceramide synthase (GLS) and LacCer synthase (galactosyltransferase, GalT-2), inhibited the gene expression of ICAM-1 and VCAM-1 and activation of their gene promoter induced by CM, which were reversed by exogenously supplied LacCer. Silencing of GalT-2 gene using its antisense oligonucleotides also attenuated CM-induced ICAM-1 and VCAM-1 expression, which were reversed by LacCer. PDMP treatment and silencing of GalT-2 gene significantly reduced CM-induced luciferase activities in NF-${\kappa}B$, AP-1, GAS, and STAT-3 luciferase vectors-transfected cells. In addition, LacCer reversed the inhibition of NF-${\kappa}B$ and STAT-1 luciferase activities by PDMP. Taken together, our results suggest that LacCer may play a crucial role in the expression of ICAM-1 and VCAM-1 via modulating transcription factors, such as NF-${\kappa}B$, AP-1, STAT-1, and STAT-3 in CM-stimulated astrocytes.

디젤분진 및 미세분진이 천식마우스의 폐조직에서 ICAM-1과 VCAM-1의 발현에 미치는 효과 (The Effects of Diesel Exhaust Particulates and Particulate Matters on the ICAM-1 and VCAM-1 Expression in the Lung of Asthma-incuced Mouse)

  • 천식이 유발된 Balb/c마우스와 동일한 조건의 IL-10 KO 마우스에 천식의 원인으로 알려진 DEP와 지하철역내에서 채집한 PM (10 ${\mu}g/m^3$)을 inhalation chamber,에 넣고 하루 4시간씩 흡입시킨 후 시료들을 채취하여 ICAM-1, VCAM-1의발현을 살펴 천식증상의 악화에 DEP와 PM이 어데한 영향을 미치는지 확인하였다. 본 실험의 결과 천식이 유발된 일반 Balb/c 마우스에 있어서는 DEP와 PM의 노출에 의하여 ICAM-1 및 VCAM-1의 발현이 세기관지 주위 조직들에서 미약하게 증가하였다. 그러나 IL-10 KO 마우스의 경우 DEP와 PM을 노출시켰을 때 ICAM-1 및 VCAM-1의 발현이 아주 강하게 증가하였다. 따라서, 본 결과는 IL-10에 대한 항체요법이 천식증상의 완화에 쓰일 수 있는 가능성을 암시하며, 한편 자동차 배기가스와 지하철 미세분진의 발생을 예방할 경우 천식과 관련한 세기관지의 염증을 완화시킬 수 있음을 간접적으로 증명한 것이라 할 수 있다.

Modulation of $TNF-{\alpha}-induced$ ICAM-1 Expression, NO and $H_2O_2$ Production by Alginate, Allicin and Ascorbic Acid in Human Endothelial Cells

  • Plant nutrients are believed to provide protection against various diseases including inflammation. Since interactions of the cell adhesion molecules are known to play important roles in mediating inflammation, inhibiting adhesion protein upregulation is a possible therapeutic target. In this study, the interacellular adhesion molecule-1 (ICAM-1) was induced in human umbilical endothelial cells (HUVECs) after stimulation with $TNF-{\alpha}$. In addition, alginate, ascorbic acid and allicin were demonstrated to inhibit the $TNF-{\alpha}$ induced expression of ICAM-1 on the HUVECs in a dose-dependent manner. These compounds also inhibited the production of NO and $H_2O_2$ induced by $TNF-{\alpha}$, which suggests that the inhibition of ICAM-1 expression by the three compounds may be due to the modulated production of the reactive oxygen/nitrogen components. Overall, these results indicate that these dietary components have a therapeutic potential in the treatment of various inflammatory disorders associated with an increase in endothelial leukocyte adhesion molecules.

Cyclooxygenase-2 Can Modulate ICAM-1 Expression in Aorta or Heart Tissues of Rats Treated with Synthetic Estrogen or Soy-isoflavones

  • The identification of COX-2 (cyclooxygenase-2) has led to potential novel insights on disease pathogenesis (atherosclerosis, cancer, Alzheimer's disease) and the regulation of normal organ function. The present in vivo study with estrogen or soy-isoflavones has provided evidence for the association between COX-2 and ICAM-1 (Intercellular adhersion molecule-1). In the system of mature female rats, soy-isoflavones exerted more pronounced effect on ICAM-1 inhibitory and COX-2 stimulatory effect than estrogen. In the system of ovariectomized estrogen deficient rats, the down-regulatory properties of soy-isoflavones on ICAM-1 was less evident, whereas estrogen exerted the inhibitory activity. These results demonstrate that COX-2 limits adhersion molecule expression on rat aorta cells and suggest that COX-2 may play a protective role in cardiovascular system in mature female rats. Soy-isoflavones appear to have beneficial effect on vascular systems through modulation of ICAM-1 and COX-2, and these molecules appeared to be closely associated.

간질성 폐질환환자들의 기관지 폐포세척액내 폐포 대식세포와 임파구의 접착분자 발현 및 Soluble ICAM-1 농도에 관한 연구 (The Expression of Adhesion Molecules on Alveolar Macrophages and Lymphocytes and Soluble ICAM-1 Level in Serum and Bronchoalveolar Lavge(BAL) Fluid of Patients with Diffuse Interstitial Lung Diseases(DILD))

  • 연구배경: 간질성 폐질환은 여러가지 다양한 원인에 의해, 또는 아직 밝혀지지 않은 원인에 의해 폐포염 및 폐간질내 염증으로 시작해 종국에는 폐섬유증으로 진행되는 질환들로 염증세포들의 이동에 필요한 접착분자들이 이들 질환의 발병기전에 중요한 역할을 할 것으로 추정되며, 현재까지는 뚜렷이 적당한 지표가 없었던 질환의 활동성을 판정하는 지표로 사용할 수 있을 가능성이 많다. 그러나 이제까지는 BAL내 AM에서 면역염색법으로 측정한 결과들로 방법 자체가 음성과 양성세포를 감별하기가 쉽지 않을 뿐 아니라 발현도 증가를 정량적으로 측정하지 못하는 단점이 있고, 따라서 발표된 결과들이 차이가 많다. 저자들은 간질성 폐질환 환자들에서 기관지 폐포세척액(BAL)내 AM과 임파구에서의 ${\beta}_2$-integrin(CD18)과 ICAM-1의 발현도를 FACS를 이용한 relative median fluorescence intenity(RMFI)를 측정하여 정상 대조군과 비교 관찰하고, 또한 혈청 및 BAL액내 가용정 ICAM-1(sICAM-1) 농도를 측정하고, BAL액내 다른 지표들과 비교하여 이들을 간질성 폐질환의 활성도를 반영할 수 있는 지표로 사용할 가능성을 알아 보았다. 방법: 대상은 조직학적으로 확인 된 미만성 간질성 폐섬유증(IPF) 환자 11명, 교원성 폐질환과 연관된 폐섬유증환자(CVD-PF) 6명, 폐유육종증 9명, 과민성 폐장염환자 2명과 대조군으후 건강한 정상인 9명이었고 BAL은 통상적인 방법을 사용하였다. ICAM-1 및 CD18은 단일항체를 사용하여 FACScan으로 median fluorescence intenity를 측정하여 RMFI를 구하고, sICAM-1농도는 ELISA법으로 측정하였다. 결과: AM에서 ICAM-1의 발현도는 $3.30{\pm}1.16$으로 대조군($0.93{\pm}0.18$)보다 증가되었고 임파구의 ICAM-1 발현도 $5.39{\pm}2.70$으로 대조군($1.06{\pm}0.21$)보다 높았다. CD18의 발현도는 임파구에서 $24.9{\pm}14.9$로 대조군($4.69{\pm}3.77$) 보다 향진되었으며, 이들 ICAM-1의 발현도는 AM 백분률과 역상관관계륜 보였고(r=-0.66, p=0.0001) 임파구의 백분율과는 정상관관계를 보였다(r=0.447, p=0.0116). 임파구가 증가되는 유육종증, 교원성질환, 과민성폐렴에서는 임파구의 ICAM-1 발현도와 BAL액내 임파구의 %(r=0.747, p=0.0006) 및 임파구농도(r=0.832, p=0.0002)와 좋은 상관관계를 보였고, 임파구의 IL-2수용체발현과도 연관관계를 나타내었다(r=0.539, p=0.0075). 또한 유육종증에서는 혈중 ACE 농도와 임파구의 ICAM-1 발현도가(r=0.905, p=0.0132) 높은 상관관계를 보여, 이들 접착분자 발현도가 병소의 활동성과 연관이 있음을 시사 하였다. 혈청내 sICAM-1농도는 환자군에서 $499.7{\pm}222.2\;ng/ml$로 대조군의 $199.0{\pm}38.9\;ng/ml$보다 높았으며(p=0.0097), BAL액내 sICAM-1농도도 환자군에서 $41.8{\pm}23.0\;ng/ml$로 대조군의 $20.1{\pm}13.6\;ng/ml$보다 증가를 보였다. 또한 혈청내 sICAM-1 농도는 AM의 ICAM-1 발현도 및 (r=0.554, p=0.0259) BAL액내 sICAM-1농도와 상관관계를 보였다. 또한 혈청 및 BAL액내의 albumin과 sICAM-1의 비율을 비교한 결과 BAL액내의 sICAM-1 농도가 훨씬 높은 것으로 미루어 대부분의 sICAM-1은 병소내에서 생성된 것임을 짐작할 수 있었다. 결론: 간질성폐질환환자들의 AM및 BAL-임파구에서 접착분자들의 발현도가 증가되었고, 혈청 및 BAL액내 sICAM-1 농도가 상승되어 있어 이들 접착분자들이 발병기전에 중요한 역할을 한다고 생각되며, 앞으로 이 활동성 판정의 지표로 사용할 기능성을 시사하였다.

과루인 및 과루인청혈플러스가 인간 탯줄 정맥 내피 세포(Human umbilicl vein endothelial cells, HUVECs)에서 염증인자 발현 억제에 미치는 영향 (The Effect of Trichosanthes Kirilowii Maximowicz Extract and Trichosanthes Kirilowii Maximowicz Cheonghyeol Plus on Anti-Inflammatory Factor Expression in Human Umbilical Vein Endothelial Cells (HUVECs))

  • Objective: To examine the effects of Trichosanthes kirilowii Maximowicz extract (TE) and Trichosanthes kirilowii Maxi mowicz Cheonghyeol Plus Phellinus linteus Cheonghyeol plus (TCP) on anti-inflammatory factor expression in human umbilical vein endothelial cells (HUVECs). Methods: HUVECs were activated with TNF-α and then treated with TE and TCP. The expression levels were then measured for intracellular genes (KLF2, eNOS, MCP-1, ICAM-1, and VCAM-1), proteins (KLF2, eNOS, MCP-1, ICAM-1, VCAM-1, ERK, and JNK, p38), and extracellular biomarkers (ICAM-1, VCAM-1, and MCP-1). Results: 1. TCP at concentrations of 100 ㎍/mL or greater significantly increased the expression of KLF2 and eNOS intracellular genes and significantly decreased the expression of ICAM-1, VCAM-1, and MCP-1 genes compared to the control group. 2. TCP at concentrations of 100 ㎍/mL or greater significantly increased the expression of KLF2, eNOS proteins compared to the control group, and significantly reduced the expression of VCAM-1, ICAM-1, MCP-1, ERK, and p38 proteins. However, JNK protein phosphorylation showed no significant change compared to the control group. 3. TCP at concentrations of 100 ㎍/mL or more significantly decreased the production of MCP-1, ICAM-1, and VCAM-1 extracellular biomarkers compared to the control group. 4. TE at a concentration of 100 ㎍/mL did not cause any significant change in the expression of intracellular genes or proteins, in the production of the extracellular biomarker MCP-1, or in the amount of JNK protein compared to the control group. Other intracellular genes, proteins, and extracellular biomarker expression showed the same trend as observed with TCP exposure. Conclusion: This study experimentally confirmed that TE and TCP could be effective in preventing or inhibiting various inflammatory vascular diseases due to their anti-inflammatory effects.

금은화가 HUVEC에서 죽상동맥경화증 관련 염증인자 발현 억제에 미치는 영향 (The Effect of Lonicera Japonica Thunberg on Inflammatory Factor Expression Associated with Atherosclerosis)

  • Objective: The purpose of this study was to investigate the effect of Lonicera Japonica Thunberg (LJT) on the inflammatory factor expression associated with atherosclerosis in human umbilical vein endothelial cells (HUVECs). Methods: After treatment with LJT in HUVEC which is treated with TNF-α, we measured the expression levels of biomarkers (MCP-1, ICAM-1, VCAM-1, KLF2, and eNOS), mRNA (CCL2, ICAM1, VCAM1, KLF2, and NOS3), and the proteins (MCP-1, ICAM-1, VCAM-1, KLF2, eNOS, ERK, JNK, and p38). Results: 1. Compared to the control, LJT significantly reduced MCP-1 and VCAM-1 levels at concentrations of 100, 200, and 400 ㎍/ml and ICAM-1 expression at 200 and 400 ㎍/ml compared to the control. It increased KLF2 levels at all three concentrations, but not significantly, while eNOS expression was significantly increased at 400 ㎍/ml. 2. LJT was seen to significantly reduce the expression of CCL2, ICAM1, and VCAM1 mRNA at concentrations of 100, 200, and 400 ㎍/ml compared to the control. In contrast, significantly increased KLF2 and NOS3 mRNA levels were observed at 400 ㎍/ml and at 200 and 400 ㎍/ml, respectively. 3. Compared to the control, LJT significantly reduced the protein expression of MCP-1 and VCAM-1 at 200 and 400 ㎍/ml and of ICAM-1 at 400 ㎍/ml. In addition, it increased both KLF2 and eNOS protein levels at 200 and 400 ㎍/ml. Although LJT did not have an effect on ERK expression in comparison with the control, it significantly reduced JNK levels at 200 and 400 ㎍/ml and p38 levels at 400 ㎍/ml. Conclusions: These results suggest that LJT has an effect on the inhibition of inflammatory factor expression associated with atherosclerosis in HUVECs which could contribute to the prevention of cardiovascular and cerebrovascular diseases.

γ-Irradiation Induced Adhesion Molecules are Reduced by Vitamin C in Human Endothelial Cells

  • Inflammation is a frequent radiation-induced reaction following therapeutic irradiation. Treatment of human umbilical endothelial cells (HUVEC) with ${\gamma}$-irradiation (${\gamma}$IR) induces the expression of adhesion proteins such as intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and E-selectin. Since the upregulation of these proteins on endothelial cell Surface has been known to be associated with inflammation, interfering with the expression of adhesion molecules is an important therapeutic target. In the present study, we demonstrate that vitamin C inhibits ${\gamma}$IR induced expression of ICAM-1, VCAM-1, and E-selectin on HUVEC in a dose- and time dependent manner. Vitamin C a1so inhibited the production of Nitric oxide (NO) induced by ${\gamma}$IR. These data suggest that vitamin C has therapeutic potential for the treatment of various inflammatory disorder associated with an increase of endothelial leukocyte adhesion molecules.

한탄바이러스 감염 내피세포에서 부착분자의 발현 (II) -In Situ Hybridization- (Expression of Some Adhesion Molecules on the Cultured Endothelial Cells of Human Umbilical Vein Infected with Hantaan Virus)

  • Histopathological vascular changes in hemorrhagic fever with renal syndrome (HFRS) caused by Hantaan virus include increased vascular permeability, disseminated intravascular coagulation, thrombocytopenia and changes in coagulation activity. Although vascular endothelial cells of main target organs such as kidney infected with Hantaan virus are not damaged but swelling of endothelial cells, perivascular exudates and infiltration of mononuclear cells and fresh interstitial hemorrhages are common. However, the pathogenesis of cell infiltration and hemorrhages around vascular endothelial cells are not well understood. Some endothelial cell molecules or vascular adhesins that acts as adhesion moleulces for leukocyte are expressed on endothelial cells close to site of inflammation. However, whether the expression of endothelial adhesion molecules such as vascular cell adhesion molecule-1 (VCAM-1), intercellular adhesion molecule (ICAM-1) and endothelial leukocyte adhesion molecule (ELAM) on vascular endothelial cells are increased by infection with Hantaan virus has not been studied. In this study, the relationship between the expression of VCAM-1, ICAM-1 and ELAM and adhesion of mononuclear cells on endothelial cells of human blood vessels infected with Hantaan virus was investigated. The endothelial cells of umbilical vein was passaged three times in culture medium and the monolayered cells were infected with $10^5\;pfu/ml$ of Hantaan virus grown in Vera E6 cell cultures. The multiplication of virus in cultured endothelial cells was monitored by immunohistochemistry and the expression of adhesion molecules was demonstrated by immunohistochemistry using monoclonal antibodies against VCAM-1, ICAM-1 and ELAM. And in situ hybriditation against ICAM-1 was also performed. The endothelial adhesion molecules, VCAM and ICAM, were expressed after 6 hours postinfection, respectively, and their expressions lasted for 72 hours. Similar expression of VCAM and ICAM appeared on endothelial cells by infection with virus, but the expression of ELAM was not recognized up to 72 hours postinfection. Microscopically, it was noted that many monocuclear cells adhered on endothelial cells infected with viruses. In an electronmicroscopic study, the transendothelial migration of mononuclear cells was observed on monolayered endothelial cells infected with virus. This results suggested that the endothelial adhesion molecules, particulary VCAM and ICAM, might be expressed on endothelial cells by infection with Hantaan virus and these molecules play a key role in the adhesion and extravasation of inflammatory cells around blood vessels.

방사선 치료후 기관지-폐포세척액내 폐포대식세포 및 임파구의 접착분자발현 변화와 방사선에 의한 폐렴 및 폐섬유증발생의 예측인자로서의 의의 (The Expression of Adhesion Molecules on BAL Cells and Serum Soluble ICAM-1 Level after the Radiotherapy for the Lung Cancer and Its Relationship to the Development of of Radiation Pneumonitis and Fibrosis)

  • 연구배경: 폐암은 우리나라 남자들의 악성종양중에서 2번째로 많으며, 수술요법이 가장 좋은 치료이나 대부분의 환자들이 진단당시 수술을 받을 수 없을 정도로 진행되어서 방사선치료(RT)나 항암치료를 동반한 RT를 받게되는데, 방사선-폐렴 및 폐섬유증의 발생이 제일 중요한 부작용이다. 방사선-폐렴은 대개 치료부위에 국한되어 발생하나 때로는 반대쪽에서까지 생기기도하며, Gibson 등은 RT후 양쪽폐에서 BAL액내 세포수 및 임파구가 증가한다고 보고하였다. 방사선-폐렴이 RT의 가장 중요한 제한요소이기 때문에 이의 발생을 조기에 진단하는 것이 환자의 예후에 매우 중요한 영향을 미친다. 이에 연구자들은 염증반응을 야기하는데 중요한 역할을 한다고 알려진 접착분자(ICAM-1, CD18)들이 방사선-폐렴의 발생에도 작용을 하는지, 만일 한다면 이를 방사선-폐렴의 진단 및 예측인자로 사용할 수 있을 가를 살펴보고, 또 방사선-폐렴이 양측성으로 오는 가를 보기위하여 본 연구를 시행하였다. 대상: 조직학적으로 확인된 폐암으로 RT를 받을 환자 29예를 대상으로 하였는데 RT 전에는 16예에서, RT후 1~2개월에는 18예에서 BAL및 혈중 sICAM 농도를 측정하였다. 5예에서는 RT전 및 후에 BAL을 시행하였다. 결과: 7예에서 임상적으로 유의한 방사선-폐렴이 발생하였다. 전 환자군에서 보면, BAL 액내 총 세포수가 $20.2{\pm}10.2{\times}10^6\;cells/L$에서 $35.3{\pm}21.6{\times}10^6\;cells/L$(p=0.0344)로 증가하였으며, 임파구도 $5.3{\pm}4.2%$에서 $39.6{\pm}23.4%$로 유의하게 증가하였다(p=0.0001). 이러한 변화는 RT를 받은 쪽 뿐 아니라 받지 않은 쪽에서도 같이 일어났으며, 방사선-폐렴이 생긴 환자와 생기지 않은 환자 사이에서도 차이가 없었다. 혈중 sICAM농도와 BAL애내 농도는 RT전후로 유의한 차이가 없었으나(혈청: $378{\pm}148$, $411{\pm}150\;ng/ml$, BALF: $20.2{\pm}12.2$, $45.1{\pm}34.8\;ng/ml$) 방사선-폐렴이 발생한 환자들에서는 방사선-폐렴이 생기지 않은 환자에 비해 sICAM 유의하게 증가되었다(혈청: $505{\pm}164$ vs $345{\pm}102\;ng/ml$, p=0.0253, BALF: $67.9{\pm}36.3$ vs $25.2{\pm}17.9\;ng/ml$, p=0.0112). 또한 폐포대식세포(AM)에서의 ICAM-1 발현도도 RT후에 증가하는 추세를 보였으나 통계적 유의성은 없었는데(RMFI: from $1.28{\pm}0.479$ to $1.63{\pm}0.539$, p=0.0605), 방사선-폐렴이 생긴 환자들에서($2.10{\pm}0.390$) 방사선-폐렴이 생기지 않은 환자에 비해($1.28{\pm}0.31$, p=0.0002) 유의하게 높았다. 이상의 BAL 결과로 미루어 RT후에 대부분의 환자들에서 양측성으로 subclinical alveolitis가 발생한 확인하였다. 그러나 임상적인 방사선-폐렴은 훨씬 드물게 발생하고, AM의 ICAM발현도 및 혈중 sICAM농도는 이러한 임상적인 방사선-폐렴발생의 좋은 지표가 될 수 있다고 사료된다.
