ACUTE INFECTIOUS BURSAL DISEASE IN CHICKENS : PATHOLOGICAL OBSERVATION AND VIRUS ISOLATION

  • Pathological and virological investigations were conducted on suspected outbreaks of infectious bursal disease (IBD) in a broiler farm and five pullet-raising poultry farms of Mymensingh and Tangail districts of Bangladesh. About 80 to 100 percent chicks were affected at the age of 26 to 45 days and mortality varied from 20 to 30 percent in broilers and 40 to 80 percent in layer chicks. Signs, symptoms, gross and microscopic lesions were typical of acute IBD. Several isolates of virus could be obtained by embryo inoculation and the virus was diagnosed as infectious bursal disease virus (IBDV) by agar gel immunodiffusion test (AGID). The virus isolate belonged to the very virulent pathotype of IBDV causing 100 percent mortality in three weeks old chicks on experimental infection.

  • The incidence of inflammatory bowel disease (IBD), especially Crohn disease, in children is remarkably increasing in Korea. Therefore, it is necessary for pediatrician to be aware of the initial presentation of Crohn disease and ulcerative colitis. Laboratory tests, radiologic studies, and endoscopic procedures are helpful in differentiating between them. At the time of presentation, most of children with IBD have abdominal pain, diarrhea, hematochezia and/or weight loss. However, atypical presentation of these diseases may contribute to a delay in diagnosis.

  • The Efficacy of PSB-2061, was investigated in comparision with predinisolone in acetic acid and Picrylsulfonic acid solution (TNBS)-induced rat inflammatory bowel disease (IBD) for 5 days. 5 % TNBS solution were administered with polyethylene (P.E) tube inserted to rats intracolon, which causing colitis to the rats. The TNBS control group (the saline treated colitic rat) exhibited ulceration and inflammation of the distal colon with formation of granuloma and pathologic connections. We checked the inflammatory parameters like rat's weight, food intake quantity change during administration. After 5 days, we sacrificed the rats and checked the colon's length, ulcer and pathologic condition. Oral treatment with PSB-2061 resulted in significant recovery of macroscopic parameters like weight and diet intake change. Especially, PSB-2061 extract had a more potent effect than $mesalazine^{(R)}$ on macroscopic colonic damage score. We can suggest that PSB extract could be a promising drug in the treatment of IBD.

  • Crohn's disease, a major form of inflammatory bowel disease (IBD), is a chronic inflammatory condition that is characterized by microvascular and macrovascular involvement. Some extraintestinal complications can occur due to chronic systemic inflammation in IBD. Among them, a pulmonary thromboembolism is a rare manifestation of IBD but is associated with a high morbidity and mortality. To our best of knowledge, there is only one case report of a pulmonary thromboembolism as a complication of Crohn's disease in Korea. We present another rare case of pulmonary thromboembolism as complication in a 25-year-old man with underlying Crohn's disease.

Sulfoquinovosylmonoacylglycerols regulating intestinal inflammation in co-culture system from the brown alga Turbinaria ornata

  • The inflammatory bowel diseases (IBD) including ulcerative colitis and Crohn disease are characterized by chronic inflammation throughout the gastrointestinal tract. The prevalence of IBD has been increasing worldwide, and has sometimes led to irreversible impairment of gastrointestinal structure and functions. In the present study, we identified a new sulfoquinovosylmonoacylglycerols (SQMG) (1) together with two known SQMGs (2 and 3) regulating intestinal inflammation from the brown alga Turbinaria ornata. The anti-inflammatory properties of two bioactive SQMGs, 1 and 2 were evaluated using an in vitro co-culture system consisting of human epithelial Caco-2 cells and PMA (phorbol 12-myristate 12-acetate)-differentiated THP-1 macrophages. Treatment with 1 or 2 inhibited the production nitric oxide and prostaglandin E2 induced by lipopolysaccharide and interferon γ challenge. The expressions of inducible nitric oxide synthase and cyclooxygenase 2 were markedly down-regulated in response to inhibition of nuclear factor κB translocation to nucleus. These findings suggest the potential use of the brown alga T. ornata and its biologically active metabolites SQMGs as pharmaceutical adjuvants in the treatment of inflammation-related diseases, including IBD.

종계의 생산성 향상을 위한 질병.위생 관리

  • 본 연구에서는 국내 종계의 생산성 저하요인들을 평가·분석하였으며, 국내사육 종계의 실질적인 생산성을 극대화 시킬 수 있는 방안을 제시하고자 시험을 수행하여 다음과 같은 결론을 얻었다. 1. 혈청검색에 의한 품종별 종게군 질병 모니터링을 실시 한 결과, SE, MS 등 2종의 난계대성 세균성 질병과 REV, APV, CAV, ORT, REO 등 5종의 만성소모성 질병을 유발하는 질병이 종계군의 산란기에 감염되어 이들이 단독 또는 중복감염되어 나타나는 피해가 예상되었으며, 이들의 감염을 효과적으로 예방 할 수 있는 백신의 개발 및 도입의 필요성이 대두되었다. 2. 집중관리 모니터링 대상농장에 대한 ND, IB, IBD 등 3종의 생독백신에 대한 접종효능을 평가해 본 결과 산란종계, 육용종계를 막론하고 전반적으로 니플급수기를 이용한 음수접종법 적용시 5주령 이하 계군에 2내지 3회의 음수접종을 실시하였음에도 불구하고 항체가 거의 형성되지 못하거나 매우 불균일한 면역반응이 나타나는 것으로 조사되어 국내 야외농장에 대한 분무접종 유도 등 기존의 백신접종법에 대한 시급한 개선대책 마련이 요구되었다. 3. 육성기 및 산란기 종계 질병·위생관리 프로그램 확립을 위하여 ND, IB, IBD에 대한 개선된 백신접종 프로그램을 모델계군에 적용한 후, 주령별로 생독백신 접종효능을 조사한 결과 백신접종 후 ND, IB, IBD에 대한 평균 항체가와 항체의 균일도 (%CV)가 표준 목표치 이상 상승하여 개선의 효과를 볼 수 있었다. 4. 산란기 종계 질병·위생관리 프로그램 확립을 위하여 실시된 변이형 IB에 대한 방어능 시험결과 정기적인 Mass type IB 생독백신의 분무접종은 국내 유행 호흡기형 IBV 유전형 VI에 대한 방어효과는 다소 떨어지는 것으로 조사되어 국내분리주를 이용한 IB 생독백신의 개발이 요구되었다. 5. 종계장 살모넬라 및 마이코플라즈마 조기 검출을 위하여 실시한 종계장 살모넬라 및 마이코플라즈마 환경 모니터링 실시 결과 기존의 평판응집반응이나 ELISA에 의한 혈청검색법에 비하여 검출효율이 매우 높은 것으로 조사되어 향후 국내 종게군에 대한 살모넬라 및 마이코플라즈마 조기검출과 방제연구에 큰 도움이 될 것으로 사료되었다.

Anti-inflammatory Effect of p-Hydroxycinnamic Acid on HT-29 Intestinal Cells and Its Therapeutic Effect of Immune Bowel Disease (대장 상피세포에서 p-Hydroxycinnamic Acid의 항염증 효과와 염증성 장질환에 대한 치료 효과)

  • Inflammatory bowel disease (IBD) is a chronic inflammatory disorder on the large intestine that has been considered as an incurable not only in Western society but also in Eastern Asia in recent years. Despite enormous efforts to develop novel therapeutics for this disease, strategy using bioactive compounds from natural product is still considered as important. p-hydroxycinnamic acid (HCA) is an intermediate substance found in several plants and has been known to possess anti-inflammation but little evidence is reported whether HCA has an inhibitory effect on intestinal inflammation. In the present study, we observed HCA does not show cytotoxic and apoptotic in HT-29 cells. Quantitative PCR analysis revealed that HCA effectively blocks the activity of HT-29 cells stimulated with TNF-α treatment. HCA inhibits translocation of p65 and MAPK pathways in activated HT-29 cells by TNF-α treatment. Besides, oral administration of HCA attenuates manifestation of DSS-induced inflammatory disease in vivo. Histological analysis exhibited that oral administration of HCA recovers IBD symptoms. The expression of pro-inflammatory cytokines were reduced by oral administration of HCA on intestinal tissues. Therefore, these results suggest that HCA has a potent anti-inflammatory effect on intestinal cells as well as show a therapeutic potential for treating IBD in vivo.
