The Wine Yeast Strain-Dependent Expression of Genes Implicated in Sulfide Production in Response to Nitrogen Availability

  • Sulfur metabolism in S. cerevisiae is well established, but the mechanisms underlying the formation of sulfide remain obscure. Here, we investigated by real-time RT-PCR the dependence of expression levels of MET3, MET5/ECM17, MET10, MET16, and MET17 along with SSU1 on nitrogen availability in two wine yeast strains that produce divergent sulfide profiles. MET3 was the most highly expressed of the genes studied in strain PYCC4072, and SSU1 in strain UCD522. The strains behaved differently according to the sampling times, with UCD522 and PYCC4072 showing the highest expression levels at 120 h and 72 h, respectively. In the presence of 267 mg assimilable N/l, the genes were more highly expressed in strain UCD522 than in PYCC4072. MET5/ECM17 and MET17 were only weakly expressed in both strains under any condition tested. MET10 and SSU1 in both strains, but MET16 only in PYCC4072, were consistently upregulated when sulfide production was inhibited. This study illustrates that strain genotype could be important in determining enzyme activities and therefore the rate of sulfide liberation. This linkage, for some yeast strains, of sulfide production to expression levels of genes associated with sulfate assimilation and sulfur amino acid biosynthesis could be relevant for defining new strategies for the genetic improvement of wine yeasts.

  • Until now, most of the studies about the impulse buying have focused mainly on the product of off-line shop, while there have relatively been very few studies about the product in internet shopping mall. The goal of this study was to understand Impulse Buying Behavior(IBB) for digital contents in the internet. Data has been collected from 278 respondents. The questionnaire method was adopted to collect the data for this study. The research was conducted by using SPSS 12.0 and AMOS 5.0. It was found that as follows: First, marketing stimulation factors, such as price, product, promotion was related positively to FLOW. Second, marketing stimulation factors was related partly positively to IBB. Third, FLOW has mediating effects in the relations between marketing stimulation factors and IBB. Finally, Self-Behavior Control and Sensation Seeking Tendancy has moderating effect in the IBB process. Thus, It also provides a core strategic Implication with Digital Contents Marketers.

  • Hybrid broadcasts utilizing heterogeneous networks can provide not only uniform broadcasting services but also various services using broadcast networks and communication networks. In particular, as content is consumed in various countries and regions, demands for personalized services continue to increase, and research on content insertion technology utilizing heterogeneous networks has been actively conducted. The most important technical challenge when inserting content based on heterogeneous networks is that the start of the inserted content, which replaces the original broadcast content at the time of content insertion, should proceed smoothly, and it must be able to accurately return to the original broadcast content. Currently, UHD broadcasting is converted to digital. However, since there is a system that supports the frame rate used in the analog method, when content insertion occurs in a conventional UHD broadcasting service, there is a problem in decoding the broadcast and inserted content. Since the replacement cost of the broadcasting system is astronomical, this paper proposes a content insertion method using by frame control that can support analog methods without replacing transmission equipment.

  • In this paper, we consider cooperative elliptic systems under conjugation conditions. We first prove the existence of the state for 2 × 2 cooperative elliptic systems with Dirichlet and Neumann conditions, then we find the set of equations and inequalities that characterizes the optimal control of distributed type for these systems. The case of n × n cooperative systems is also established.

  • The Low Voltage DC-DC converters (LDCs) of the Electric Vehicles require high power density and high efficiency operation over the wide range of load and input voltage variations. This paper introduces a novel topology which combines three 1 MHz Half-Bridge (HB) LLC resonant converters and an Inverting Buck-Boost (IBB) converter to adjust the output voltage without frequency modulation. The switching frequency of the proposed converter is fixed at 1MHz to achieve a constant frequency operation for the resonant converter. In the proposed topology GaN FETs and planar transformers are employed to optimize the converter operation at high frequency. A 1 MHz/1.8 kW prototype converter is built to verify the feasibility and the validity of the proposed LDC topology.

  • Let R be a ring with characteristic different from 2. An additive mapping F : R → R is called a generalized derivation on R if there exists a derivation d : R → R such that F(xy) = F(x)y + xd(y) holds for all x, y ∈ R. In the present paper, we show that if R is a prime ring satisfying certain identities involving a generalized derivation F associated with a derivation d, then R becomes commutative and in some cases d comes out to be zero (i.e., F becomes a left centralizer). We provide some counter examples to justify that the restrictions imposed in the hypotheses of our theorems are not superfluous.

Analysis of the Relationship between MHC-DRB1 Gene Polymorphism and Hydatidosis in Kazakh Sheep

  • The objective of this work was to analyze the relationship between ovine major histocompatibility complex (MHC) DRB1 gene polymorphism and genetic resistance to hydatidosis in Kazakh sheep. The Ovar (ovine MHC) class II DRB1 second exon was amplified by polymerase chain reaction (PCR) from DNA samples of 702 Kazakh sheep, including 302 sheep with hydatidosis and 400 health controls. PCR products were characterized by the restriction fragment length polymorphism (RFLP) technique using five restriction enzymes, i.e., MvaI, HaeIII, SacI, SacII and Hin1I, yielding 14 alleles and 28 genotypes. Comparing the frequency of genotypes in hydatidosis sheep with the control group, it was found that the genotype frequencies of MvaIbc, Hin1Iab, SacIIab, HaeIIIde, HaeIIIdf and HaeIIIdd in control sheep were significantly (p<0.01) higher than in hydatidosis sheep, indicating that a significant correlation existed between these genotypes and resistance to hydatidosis. Genotype frequencies of MvaIbb, SacIIaa, Hin1Ibb and HaeIIIef in sheep with hydatidosis were extremely significantly (p<0.01) higher than in the control group, and the genotype frequency of HaeIIIab was significantly higher (p<0.05), indicating that a marked correlation existed between these genotypes and susceptibility to hydatidosis. By way of analyzing haplotype with these resistant genotypes, the hydatidosis resistant haplotype MvaIbc-SacIIab-Hin1Iab of Kazakh sheep was screened out, and then verified through artificial hydatid infection in sheep. The results indicated that the infection rate of sheep with the resistant haplotype of hydatidosis was significantly lower (p<0.01) than without this resistant haplotype. It showed that the genic haplotype MvaIbc-SacIIab-Hin1Iab of Ovar-DRB1 exon 2 was the resistant haplotype of hydatidosis in Kazakh sheep.

A New Protein of ${\alpha}$-Amylase Activity from Lactococcus lactis

  • An extracellular ${\alpha}$-amylase from Lactococcus lactis IBB500 was purified and characterized. The optimum conditions for the enzyme activity were a pH of 4.5, temperature of $35^{\circ}C$, and enzyme molecular mass of 121 kDa. The genome analysis and a plasmid curing experiment indicated that $amy^+$ genes were located in a plasmid of 30 kb. An analysis of the phylogenetic relationships strongly supported a hypothesis of horizontal gene transfer. A strong homology was found for the peptides with the sequence of ${\alpha}$-amylases from Ralstonia pikettii and Ralstonia solanacearum. The protein with ${\alpha}$-amylase activity purified in this study is the first one described for the Lactococcus lactis species, and this paper is the first report on a Lactococcus lactis strain belonging to the amylolytic lactic acid bacteria (ALAB).

  • 최근 UHDTV 방송의 대두와 함께 방송 부가 서비스에 대한 관심이 높아지고 있다. ATSC 3.0, HbbTV, IBB 등 각 방송 표준 기구로부터 부가서비스에 대한 표준이 공표되고 있으며, 이 표준들은 공히 Companion Screen에 대한 내용을 포함하고 있다. Companion Screen은 사용자가 TV를 통하여 시청하고 있는 컨텐츠에 대한 부가 정보를 사용자의 스마트폰/태블릿으로 표시할 수 있어 방송 기반의 개인화 서비스 구현방법으로 각광받고 있다. 본 논문에서는 이러한 Companion Screen과 IoT 기기의 연동을 통한 서비스 구성 방법을 제시한다. IoT는 최근 급격한 발전을 거듭하고 있으며 다양한 제품군의 출시도 지속적으로 이루어지고 있다. 이러한 IoT를 Companion Screen과 연계하여 활용함으로써, 방송과 통신을 연계하는 신규 서비스의 구성이 가능할 것으로 예상된다. 본 논문에서 제시하는 서비스 구성 기법은 방송에 내제된 부가 정보 기반의 IoT 제어를 Companion Screen을 통하여 구성할 수 있도록 한다.

Seed collection strategies for plant restoration with the aid of neutral genetic diversity

  • One key step in the plant restoration process is the collection of seeds from the field. For the selection of source populations of target plant species for translocation purposes (reintroduction or reinforcements), several approaches are possible. A practical method involves the use of data from reciprocal transplant studies. If no direct data are available, knowledge of population genetics and the phylogeography of the target species can serve as an alternative. In this short review, we briefly propose guidelines for those collecting seeds for plant species restoration based on population genetics theory, focusing on two main questions: Where does the plant material come from and how are sources designated, and how are seeds efficiently collected from local populations? While genetic data on a larger scale (phylogeography and population genetics) are needed to form a reply to the first question, similar data on a smaller scale (fine-scale genetic structures within populations) are necessary to shed light on the second issue.
