Study on the Propagation of Vitex rotundifolia for Establishment of Natural Aromatic Resources (향료자원 조성을 위한 순비기나무의 증식에 관한 연구)

  • This study was to investigate the rational propagation of the Vitex rotundifolia, which is distributed as a community naturally at coastal area in Korea. The germination rate of ordinary temperature stored fruits was the highest with 67%. In the matter of the number of seedlings per fruit, 1 seed germinated fruit was the most with 40.3 % and the mean was 1.4 seedlings per fruit. The best rooting rate of both greenwood and hardwood cutting was 96.7% in the case of cuttings soaked in IBA 2500ppm for 1 minute on the Compost+Sand bed-soils. Generally the root development of cuttings was higher in the high IBA concentration with soaking in a moment than in the low IBA concentration with soaking in long time, and bed-soils mixed with sands and composts. And in the rooting rate, greenwood cutting was better about 10% than hardwood cutting of Vitex rotundifolia.

Interaction Between time of Nodal Explant Collection and Growth Regulators Determines the Efficiency of Morus alba Micropropagation

  • The hormonal requirement suiting micropropagation of Morus alba during any season throughout the year was studied. Sprouting frequency from axillary buds of M. alba was greatly influenced by the time of explant collection, the highest value was achieved when nodal explants were collected at the end of bud dormancy period (late in March) and cultured on Murashige and Skoog (MS) medium supplemented with low concentration (0.5 mg/L) of BAP, kinetin or IBA (85-68%). In addition, they showed higher axillary bud sprouting on growth-regulators-free medium (49%) than others collected in autumn or winter and cultured on medium supplemented with various growth regulators (47-48%). Regardless of that period, young explants with greenish buds collected in summer exhibiting high sprouting frequency (66%) on MS medium supplemented with 0.5 mg/L kinetin and 0.5 mg/L GA3. Shoot multiplication via adventitious bud formation was achieved when the nodal explants were cultured on MS medium supplemented with 2 mg/L BAP and 0.2 mg/L IBA. Further multiplication via nodal explants of in vitro grown shoots was obtained on MS medium supplemented with 0.5 mglL BAP and 0.5 mg/L GA3. While half strength MS medium supplemented with low concentration (0.5 mg/L) of IBA, IAA or 2,4-D stimulated adventitious root formation, IBA was the best. After transfer the plantlets to the soil, acclimatization for three weeks was essential prerequisite for survival in high frequency (92%). Peroxidase activity is related to break of bud dormancy where maximum enzyme activity was detected when the lateral buds were induced to commence growth under field condition (early in spring) or in vitro.

  • This study was conducted to establish practical micropropagation of jujube cultivars ('Geumsumg', 'Bokjo') by axillary bud culture. The results are summerized as follows : 1. Addition of activated charcoal to half-strength Murashige and Skoog(MS) medium supplemented with 0.5mg/l benzylaminopurine(BAP) enhanced shoot and root growth. At 500mg/l activated charcoal level 'Geumsung' showed best result, and shoot length and the number of multiple shoot were 6.4cm and 10.0, respectively. At 1,000mg/l activated charcoal level 'Bokjo showed best result, and shoot length and the number of multiple shoot were 7.5cm and 12.4, respectively. 2. As indole-3-butyric acid(IBA) concentration increased, rooting and callus growth of microshoot were enhanced. The optimum IBA concentration for shoot elongation and multiplication was 1.0mg/l. 3. Growth responses of shoot-tip and axillary bud segments between two jujube cultivars were different. 'Geumsung' showed that axillary bud explants were about twice better than shoot-tip explants for shoot multiplication, but 'Bokjo' showed that shoot-tip explants mere better than axillary bud explants for shoot elongation and multiplication. 4. In acclimatization processes of plantlets produced in vitro, the survival of plantlets with only root primordia in soil medium was better than that of plantlets with several routs resulting in 97.8%. 5. In cutting of in vitro-derived microshoot, paclobutrazol was more effective than IBA, naphth-aleneacetic acid(NAA) and $Rooton^{(R)}$ in rooting and root growth.

Control effect of isobutyric acid on rice blast (벼 도열병에 대한 indole butyric acid(IBA)의 방제 효과)

  • Nine plant growth regulators (PGRs) were tested for in vivo antifungal activities against on rice blast. They showed higher in vivo antifungal activities when they were applied on rice plants by soil drench rather than foliar spray. Except for 2,4-D at $500\;{\mu}g\;mL^{-1}$, the others showed a very low or no activity against the disease in foliar spray applications. In contrast, 2,4-D, indole butyric acid (IBA) and triiodobenzoic acid, at $500\;{\mu}g\;mL^{-1}$, showed control values of 98.9, 97.8 and 88.9% in soil drench applications. Furthermore, the control activity of IBA was dependent on its concentration against rice blast; IBA suppressed the development of rice blast by 71.7% at $125\;{\mu}g\;mL^{-1}$ and 85.8% at $250\;{\mu}g\;mL^{-1}$. IBA also controlled the development of rice blast on adult plants by 63.9% at a dosage of 2.56 kg/10a. The results revealed that IBA has a good activity against rice blast when it is applied by soil drench.

Effect of Calcium Application on Fruit Mineral Nutrients, Quality, and Browning in 'Fuyu' Sweet Persimmon (칼슘제 처리가 '부유' 단감의 무기성분, 품질 및 갈변율에 미치는 영향)

  • This study was established to investigate the effects of Ca application on fruit mineral nutrients, quality, and browning of 'Fuyu' sweet persimmon (Diospyros kaki). Ca foliar application at 40ml/20L/tree had a greater fruit Ca concentration than did control, Ca fertigation (40ml/20L/tree), and Ca foliar application (40ml/20L/tree) coupled with IBA fertigation (40ml/20L/tree). Fruit mineral nutrient concentrations for Mg, B, and Mn were similar or lower in the foliar treatment compared to other treatments. Fruit color was not affected. Ca treatment, whether foliar applied or fertigated, was effective in maintaining fruit firmness and in decreasing the browning symptom in fruit after MA storage at $0^{\circ}C$ for 60 and 100 days. Although Ca foliar application + IBA fertigation treatment improved fruit firmness, the positive effect on the browning and decay occurrences in fruit was not shown.

Efficient Micropropagation of Pear Germplasm Using Soot Tips and Nodal Explants

  • We micropropagated pear (Pyrus species) using shoot tips and nodal explants from three pear genotypes. The ability to establish shoot tip cultures, proliferate shoots, induce rooting, and acclimatize the resulting plantlets are all elements of in vitro micropropagation. Shoots were induced from shoot tips on Murashige and Skoog medium (MS) with five different plant growth regulator combinations. The highest shoot formation rates were achieved for the three genotypes using MS supplemented with 1.0 mg/L N6-benzyladenine (BA) and 0.1 mg/L gibberellic acid (GA3). The maximum shoot number and shoot length for the three cultivars were recorded with 2.0 mg/L BA and 0.2 mg/L indole-3-butyric acid (IBA) in multiplication medium using nodal explants produced from microshoots. Nodal explants with one or two axillary buds cultured for three weeks initiated roots on medium supplemented with various concentrations of 1-naphthaleneacetic acid (NAA) or/and IBA in half-strength MS medium for adventitious rooting. The highest rooting response was with the combination of 0.2 mg/L NAA and 0.2 mg/L IBA. A combination of NAA and IBA resulted in a significant increase in the rooting ratio over NAA or IBA alone. In this medium, the root formation rate according to ranged from 68.9% for the BaeYun No. 3 genotype to 51.8% for the Hwanggeum genotype. We also investigated the influence of the concentration the polyamine phloroglucinol in rooting medium. For all three genotypes, the highest rooting ratio, longest root length, and greatest root number were observed in the treatments with 75-150 mg/L phloroglucinol. Most rooted plants were acclimatized successfully.

Auxin Affects on Production of Adventitious Roots and Secondary Metabolites in Echinacea angustifolia (오옥신 처리가 에키네시아의 부정근 및 생리활성물질 생산에 미치는 영향)

  • The production of adventitious roots derived from root explant of Echinacea angustifolia and its secondary metabolite content were assessed in different types and levels of auxin. The induction of adventitious roots from root explant cultured in Murashige and Skoog solid medium supplemented with 1.0 mg/L indole -3-butyric acid (IBA) attained highest as 20.87 mg fresh weight and 3.07 mg dry weight per culture but root suspension culture at the same concentration of IBA enhanced biomass production as 3.07 g fresh weight and 0.38 g per culture after 4 weeks in culture. 3.0 mg/L ${\alpha}$-naphthalene acetic acid (NAA) treatment had similar effect on root biomass production as 3.07 g fresh weight and 0.38 g per culture with liquid suspension culture, whereas adventitious roots exposed to over 3.0-5.0 mg/L IBA or 5.0 mg/L NAA were less responsive by reducing the number of adventitious roots and/or changing root morphology such as short and thick. The content of secondary metabolites such as phenolic, flavonoids and total caffeic acid in adventitious roots cultured on MS medium supplemented with 1.0 mg/L IBA were attained highest as 27.20, 9.60. 10.67 mg/g dry weight, respectively. Overall, the best production of root biomass and secondary metabolites were given by 1.0 mg/L IBA.

Induction and growth of adventitious roots and bioreactor culture in Codonopsis lanceolata (더덕 (Codonopsis lanceolata)의 부정근 유도 및 생장에 미치는 배양조건과 생물반응기 배양)

  • This paper reported the establishment of mass production system of adventitious roots of Codonopsis lanceolata through shake flask and bioreactor culture. Induction of adventitious roots was started from the explants of leaf, stem and root on 1/2 strength Murashing and Skoog (MS) solid medium. Stem segments were the best explants for induction of adventitious roots compared to leaf and root segments. Among the different auxins tested (NAA, IBA and IAA), number of adventitious root per explant was highest on solid medium with 1.0 mg/L IBA, and produced $9.9{\pm}1.2$ roots per explant. However, growth of adventitious roots was fast in the presence of IBA at low concentration (0.1 mg/L). In shake flask culture, maximum production of adventitious roots (fresh weight) was obtained in half-strength MS medium compared to full-strength and one-third MS medium. When the adventitious roots produced in shake-flask culture were transferred to 5 L air-lift bioreactor, 16 times of fresh weight increase was gained after one month of culture. These results indicate that this protocol for the production of C. lanceolata adventitious roots can be applied to large scale culture for practical application.

In vitro micropropagation of Philodendron cannifolium (기내배양에 의한 Philodendron cannifolium의 대량번식)

  • In order to micropropagate uniform plantlets of Philodendron cannifolium in vitro, the shoot tips were cultured on MS media supplemented with $0.5{\sim}10.0$ mg/L BA or $0.05{\sim}0.1$ mg/L thidiazuron(TDZ). The adventitious multi-bud clusters from basal part of shoots were formed on MS media containing $2.0{\sim}5.0$ mg/L BA or $0.05{\sim}0.1$ mg/L TDZ. But the shoots grown on MS media with TDZ showed necrosis by the lack of chlorophyll. The adventitious multi-bus clusters were cut into $5{\sim}7$ mm sections and cultured on MS media containing BA and TDZ for shoot proliferation. Shoots were proliferated vigorously on MS medium supplemented with $1.0{\sim}3.0$ mg/L BA with up to 30 shoots. But abnormally swollen hard calli were formed from basal parts of shoots on MS media with TDZ and high concentration of BA(10.0 mg/L). The proliferated shoots on same media also showed necrosis by the lack of chlorophyll. The shoot growth and rooting were favorable on MS media containing $0.5{\sim}2.0$ mg/L IBA. The rooted plantlets were acclimatizated effectively in soil mixed with perlite 1:vermiculite 1 or vermiculite alone. Fifteen mL of liquid medium containing 10 g/L activated charcoal and 30 g/L sucrose were added in same vessels after small shoots were proliferated to stimulate shoot growth and rooting. After 8 weeks in culture, the shoots were dipped into high concentration of IBA solution. and planted in soil mexed with perlite 1:vermiculite 1. The shoot growth and rooting were favorable in dipping treatments of $500{\sim}2,000$ ppm IBA solutions for 10 sec.

Differentiation of Winter Buds of Prunus yedoensis Matsumura from Jeju Island Depending on the Collection Time and Media Conditions (제주(濟州) 자생(自生) 왕벚나무(Prunus yedoensis Matsumura) 동아(冬芽)의 채취시기(採取時期)와 배지(培地)의 조건(條件)에 따른 기관유도(器官誘導))

  • We tried to mass-propagate Prunus yedoensis from Jeju through tissue culture. We investigated the effect of bud collection time, the concentration of $NH_4NO_3$ in media and plant growth regulators(BAP, $GA_3$ and IBA) on the differentiation of winter buds. Buds, taken in February, flushed well regardless of various in vitro conditions. Bud flushing rate was significantly different depending on the collection time. BAP appeared to be effective on bud flushing. Sixty percent of buds taken in October flushed on the media containing $3.0mg/{\ell}$ BAR. No buds flushed on the medium supplemented with IBA. Buds, after flushing in BAP media, grew as foliated shoots and showed a rosette of leaves. When $GA_3$ supplemented to the BAP-containing media as a higher concentration than that of BAP, shoots elongated and developed into normal shoots. The combination of BAP $1.0mg/{\ell}$ and $GA_3$ $2.0mg/{\ell}$ is most recommendable for shoot elongation.
