Linkage between US Financial Uncertainty and Stock Markets of SAARC Countries

  • The primary purpose of the study is to investigate the volatility spillover from financial uncertainty (FU) of the United States (US) to the stock markets of SAARC member countries including India, Sri-Lanka, Pakistan, and Bangladesh. The empirical literature overlooked SAARC countries and the FU index. Based on the estimation method, the data of FU is available for three different forecast horizons including 1-month, 3-months, and 12-months. For empirical analysis, monthly data is used from February 2013 to September 2019. EGARCH model is employed to investigate the volatility spillover effects. The findings of the study show that the spillover effect of FU varies with the forecast horizon. The FU with a higher forecast horizon has a significant spillover effect on more countries. The spillover effect of US financial uncertainty is negative in most of the SAARC countries. Bangladesh stock market is influenced by FU with all three forecast horizons whereas the volatility of the Pakistan stock market is not influenced by FU with any forecast horizon. The findings are consistent with the concept of "limited trade openness" in the financial markets of emerging economies. The emerging economies avoid financial market openness to minimize the risk of spillover of other countries.

In vitro micropropagation of Philodendron cannifolium (기내배양에 의한 Philodendron cannifolium의 대량번식)

  • In order to micropropagate uniform plantlets of Philodendron cannifolium in vitro, the shoot tips were cultured on MS media supplemented with $0.5{\sim}10.0$ mg/L BA or $0.05{\sim}0.1$ mg/L thidiazuron(TDZ). The adventitious multi-bud clusters from basal part of shoots were formed on MS media containing $2.0{\sim}5.0$ mg/L BA or $0.05{\sim}0.1$ mg/L TDZ. But the shoots grown on MS media with TDZ showed necrosis by the lack of chlorophyll. The adventitious multi-bus clusters were cut into $5{\sim}7$ mm sections and cultured on MS media containing BA and TDZ for shoot proliferation. Shoots were proliferated vigorously on MS medium supplemented with $1.0{\sim}3.0$ mg/L BA with up to 30 shoots. But abnormally swollen hard calli were formed from basal parts of shoots on MS media with TDZ and high concentration of BA(10.0 mg/L). The proliferated shoots on same media also showed necrosis by the lack of chlorophyll. The shoot growth and rooting were favorable on MS media containing $0.5{\sim}2.0$ mg/L IBA. The rooted plantlets were acclimatizated effectively in soil mixed with perlite 1:vermiculite 1 or vermiculite alone. Fifteen mL of liquid medium containing 10 g/L activated charcoal and 30 g/L sucrose were added in same vessels after small shoots were proliferated to stimulate shoot growth and rooting. After 8 weeks in culture, the shoots were dipped into high concentration of IBA solution. and planted in soil mexed with perlite 1:vermiculite 1. The shoot growth and rooting were favorable in dipping treatments of $500{\sim}2,000$ ppm IBA solutions for 10 sec.

  • We tried to mass-propagate Prunus yedoensis from Jeju through tissue culture. We investigated the effect of bud collection time, the concentration of $NH_4NO_3$ in media and plant growth regulators(BAP, $GA_3$ and IBA) on the differentiation of winter buds. Buds, taken in February, flushed well regardless of various in vitro conditions. Bud flushing rate was significantly different depending on the collection time. BAP appeared to be effective on bud flushing. Sixty percent of buds taken in October flushed on the media containing $3.0mg/{\ell}$ BAR. No buds flushed on the medium supplemented with IBA. Buds, after flushing in BAP media, grew as foliated shoots and showed a rosette of leaves. When $GA_3$ supplemented to the BAP-containing media as a higher concentration than that of BAP, shoots elongated and developed into normal shoots. The combination of BAP $1.0mg/{\ell}$ and $GA_3$ $2.0mg/{\ell}$ is most recommendable for shoot elongation.

Plant Regeneration from Adventitious Roots of Rehmannia glutinosa Liboschitz and Bioreactor Culture (지황 부정근을 이용한 식물체 재분화 및 생물반응기 배양)

  • This experiment was carried out to develop rapid mass propagation via shoot organogenesis system from adventitious roots of Rehmannia glutinosa. The induction of adventitious roots from leaf explants was most favorable to MS solid medium supplemented with 2mg/L IBA. However, the growth of adventitious roots was highest when they were cultured on 1/3 strength MS liquid medium supplemented with 2mg/L IBA. When the adventitious roots were grown in 10L bioreactor, 10g roots as initial inoculum was increased to 225g after 6 weeks of culture. The harvested roots were cultured onto solid medium to induce plant regeneration. The optimal adventitious shoot formation was observed on MS medium supplemented with 2mg/L BA. Rooting of individual shoots was induced after transfer to half strength MS medium without growth regulators. Plantlets after acclimatization were successfully transplanted in the field and no phenotypic variation was observed among them.

The Study of the Root Cutting Propagation of Kalopanax pictus Nakai (음나무 삽목 번석에 관한 연구)

  • This study officially selected Kalopanax pictus. The root cutting and investigation conducted how the production process of the rooting ratio according to its age and its length gives some influence to the rooting ratio and what is the usage value in the effect of rooting ratio of an accelerant of plant growth and IBA treatment. The ages and ratio of rooting are one year $92.0\%$, two years $80.0\%$, and three years $67.0\%$. As shown above, one year cutting showed the highest growth. The rooting ratio by the length of the cutting-tree is $75.0\%$ in 5cm, $92.0\%$ in 10cm and $89.0\%$ in 15cm. There is a little differences according to the result of each analysis in the rooting ratio by the length of the cutting tree. Two groups having the length 10cm and 15cm showed the similar rooting ratio, while the group having the length 5cm showed that the rooting ratio is very low. The rooting ratio treated by the hormone, IBA showed that the treated group was $94\%$ and the untreated group was $92\%$. Although the treated group showed a little high rooting growth ratio, there was no significant difference. Thus, it is said that the treatment of IBA in the cutting tree is insignificant.

Effect of Plant Growth Regulators and Media on Regeneration of Sorghum bicolor (L.) Moench (바이오에너지용 수수 품종의 재분화율 증진을 위한 배지와 생장조절제 효과)

  • This study was carried out to optimize the embryogenic callus induction and plant regeneration from mature seeds of Sorghum bicolor. The effect of growth regulators was investigated on formation of embryogenic callus. The highest frequency of embryogenic callus was observed when the mature seeds were cultured on B5 medium supplemented with 2 mg/L 2,4-Dichlorophenoxyacetic acid (2,4-D). The highest frequency of plant regeneration from embryogenic callus was observed on MS medium with 0.5 $mg\;l^{-1}$ 6-benzyl amino purine (BAP) and 0.25 $mg\;l^{-1}$ indole-3-butyric acid (IBA) to optimize the shoot regeneration. High concentration of BAP (1 $mg\;l^{-1}$) supplemented with IBA (0.25 $mg\;l^{-1}$) was effective combination for shoot multiplication. MS medium supplemented with 1 $mg\;l^{-1}$ IBA was found to be the most effective for inducing roots. Normal rooted plantlets were transferred to the greenhouse for hardening with over 90% survival rate. Hence, this reproducible protocol could be useful for mass propagation and genetic transformation of S. bicolor.

Effect of Medium Components and Culture Methods on Shoots Regeneration from Athyrium niponicum (개고사리의 기내 포자체 재생에 미치는 배지구성물질 및 배양방법의 영향)

  • Present studies are carried out to find media components and culture methods for in vitro propagation of Athyrium niponicum and to establish the optimal economic masspropagation systems. Among pinnae, petiole and rhizome segments only rhizome segments produced young plants. Rhizome segments showed vigorous plant regeneration on 1/2MS medium and supplement to 1% sucrose and 50 $mg{\cdot}L^{-1}$ $NaH_2PO_4$ were promoted the plant regeneration from rhizome segments. Kinetin was better than BA for plant regeneration and combination with 2 ${\mu}M$ kinetin and 5 ${\mu}M$ IBA was most efficient for plant regeneration. Solid or liquid medium with or without 0.1% qactivated charcoal in modified 1/2MS medium (1% sucrose, 50 $mg{\cdot}L^{-1}$ $NaH_2PO_4$, 2 ${\mu}M$ kinetin, 5 ${\mu}M$ IBA, pH 5.8) were used to find the optimal culture methods. The plant regeneration from rhizome segments were most vigorous on solid medium without activated charcoal. The addition of activated charcoal were inhibited the plant regeneration from rhizome segments not only on solid medium but also liquid stationary or suspension culture.

Mass Propagation of Vitex negundo L., in vitro

  • Shoot proliferation was obtained from shoot tips and nodal explants of Vitex negundo L. on MS medium supplemented with either BAP or KIN (0.1-2.0 mg/L) alone or in combination with NAA (0.1 mg/L). The concentrations of cytokinins combined with NAA produced multiple shoots from shoot tips and nodal explants. The highest mean percentage (84.3$\pm$8.0) of shoot multiplication's were observed on nodal explants in the presence of BAP (1.5 mg/L) and NAA (0.1 mg/L) followed by shoot tips (65.0$\pm$5.0). The regenerated shootlets were rooted on MS basal medium IAA, IBA, NAA (0.1-1.5 mg/L). The maximum number of roots (51.0$\pm$2.6) was achieved on the medium containing IBA (1.0 mg/L) followed by other auxins (NAA, IAA). The regenerated plants were successfully transferred to a mixture of vermiculate and soil. About 95% of the plantlets survived when transferred to the field.

Practical Factors Controlling in vitro Multiplication and Rooting in Empetrum nigrum var. japonicum, an Endangered Woody Species

  • The plant Empetrum nigrum, valued in the traditional system of medicine, is well known for its antibacterial, antifungal, and antioxidant properties. In the present work, the effect of removal of shoot apical meristem (SAM) on shoot proliferation was studied. It was observed that removal of SAM promoted shoot proliferation whereas intact tip resulted in higher survival percentage. Further, the effect of different concentrations of BA on above was also studied. During root formation the effect of light quality after treatment with IBA was investigated. For rooting, continuous red light without IBA resulted in maximum rooting percentage. The above factors when taken into consideration during micropropagation of this endangered plant can result in healthier plantlets. The results show that the species could be successfully conserved by in vitro propagation system.
