Organ Formation-The Formation of Adventitious Roots, Trichomes and Calli from Leaf Segments of Arabidopsis thaliana by Naphthaleneacetic acid Concentrations, and Their Determination times (애기장대 (Arabidopsis thaliana) 잎 절편에서의 기관형성 특히 Naphthaleneacetic acid의 농도에 따른 부정근, 모용 및 캘러스 형성과 Determination Time)

  • The effect of auxins and cytokinins on the formation of adventitious shoots, adventitious roots, trichomes, and calli in MS basal medium was investigated in leaf segments from ecotype Columbia of Arabidopsis thaliana. Adventitious shoots, adventitious roots, trichome, and calli were formed from leaf segments by a wide range of hormone concentrations and combinations. Adventitious shoots were formed respectively in treatment with 0.1mg/L IAA and 10 mg/L BA. Adventitious roots were formed in treatments with low concentration of IAA and NAA. Trichomes and calli were formed by increasing the concentration of IAA and NAA. The optimal combination was 0.5mg/L NAA and 0.1mg/L BA for trichome formation, 10mg/L NAA and 10mg/L BA for calli formation. When NAA was treated alone in culture media, adventitious roots were formed in 0.1mg/L, trichomes were formed in 2.0mg/L, and calli were formed in 10mg/L. Inductive time for formation of adventitious roots, trichomes and calli were determined at 6,7 and 18 days respectively by periodical transfer of leaf segments from NAA containing medium to NAA free medium.

Interaction between Brassinolide and Auxins on Bioassays (Brassinolide와 Auxin과의 상호작용의 생물검정에 의한 평가)

  • This study was conducted to investigate the interaction effect of a new plant growth regulator brassinolide and auxins using several bioassay methods such as rice lamina inclination test, oat coleoptile segment growth test and radish hypocotyl elongation test. For rice lamina inclination test, the antagonistic response showed at low concentration combinations of homobrassinolide and auxins (IAA and 2,4-D), while the combinations of high concentration responsed mostly synergistic or additive effects, IAA was generally higher than 2,4-D for combination effect with homobrassinolide. For oat coleoptile segment growth test, the synergistic effect showed at IAA concentrations less than 0.1ppm while additive response exhibited above 0.3ppm regardless of homobrassinolide concentrations. In radish hypocotyl elongation test, the interaction response varied with cotyledon. The sections removed cotyledon showed mostly antagonistic effects, except for combination of homobrassinolide with IAA 3ppm. Interaction responses were Quite different from bioassay methods, particularly using experimental materials: antagonistic responses were usually shown at the section that attached growing point while these for sections that removed growing point were responded synergistic or additive effect due to unstable endogenous hormones.

Isolation of Photosynthetic Bacterium, Rhodopseudomonas palustris JK-1 and Researches on IAA and Carotenoid Production (광합성세균 Rhodopseudomonas palustis 분리 및 IAA와 Carotenoid 생성에 관한 연구)

  • The JK-1 isolate which was the best producer of indole-3-acetic acid and carotenoid among the 388 strains isolated from 28 wetlands in Jeju, was identified to be Rhodopseudomonas palustirs belongs to a typical group of non sulfur purple bacteria based on 16S sRNA sequencing. This study investigated the effect of different cultural conditions of pH, temperature, agitation, light and aeration on growth, IAA and carotenoid production of photosynthetic bacterium JK-1 for optimization of IAA and carotenoid production. It was found that growth, IAA, carotenoid, and bacteriochlorophyll production with light (3,000~3,500 Lux) and agitation (100 rpm) showed better results than those with dark/static or dark/agitation (100 rpm) in anaerobic conditions. The optimal pH, temperature and agitation speed for cell growth were 7, $30^{\circ}C$, 150 rpm, for IAA production were 9, $30^{\circ}C$, 150rpm and for carotenoid production were 6, $25^{\circ}C$, 50 rpm, cultured for 72 h under anaerobic light, respectively. The growth and IAA production were high in aerobic culture compared with anaerocic culture, whereas carotenoid and bacteriochlorophyll content were decreased extremely in aerobic condition (0.5~1 vvm). Subsequently, the optimal culture conditions for JK-1 were selected with pH 7, $30^{\circ}C$ and 100 rpm under anaerobic light and the effect on plant growth was tested by pot assay. Inoculation of JK-1 with 3% (v/v) level caused increase in shoot and root dry weigh that varied from 20%~58% to 40%~28% in young radish in camparison to uninoculated treatment at 50 days of growth. The study suggests that the JK-1 isolate may serve as efficient biofertilizer inoculants to promote plant growth.

In Vitro Propagation of Guzmania cv. Cherry by Axillary Shoot Culture (측지배양에 의한 Guzmania cv. Cherry의 기내 대량번식)

  • Guzmania was propagated through in vitro culture of lateral shoots. When new shoots grown in greenhouse were cut and cultured in vitro, contamination rate was very high at about 80% in the first stage of in vitro culture. Among cytokinin treatments for agar medium, 2.0 mg/L BA was most effective for shoot multiplication, and those with 0.5 mg/L kinetin and 0.5~1.0 mg/L BA were favorable for shoot multiplication. BA was more effective for shoot multiplication than kinetin, and shoot multiplication was more enhanced when 2.0 mg/L BA was combined with 0.1~0.5 mg/L IAA than 2.0 mg/L BA alone. The medium with 2.0 mg/L BA and 0.1 mg/L IAA showed the highest rate of shoot multiplication with about 8.7 in shoot number, and those with 2.0 mg/L BA and 0.5~1.0 mg/L IAA also resulted in high multiplication of shoots. Shoots were multiplicated more in liquid rotation culture(80 rpm) with the medium containing 0.5 mg/L BA and 0.1 mg/L IAA than liquid stagnating and solid cultures. Regenerated shoots formed roots very favorably in the medium supplemented with 2.0 mg/L IBA.

Isolation and Characterization of ACC Synthase Gene Family in Mung Bean (Vigna radiata L.): Differential Expression of the Three ACC Synthase enes in Response to Auxin and Brassinosteroid

  • By screening a cDNA library of auxin-treated mung bean (Vigna radiata L.) hypocotyls, we have isolated two full-length cDNA clones, pVR-ACS6 and pVR-ACS7, for 1-aminocyclopropane-1-carboxylate (ACC) synthase, the rate-limiting enzyme in the ethylene biosynthetic pathway. While PVR-ACS6 corresponds to the previously identified PCR fragment pMBA1, pVR-ACS7 is a new cDNA clone. A comparison of deduced amino acid sequences among auxin-induced ACC synthases reveal that these enzymes share a high degree of homology (65-75%) to VR-ACS6 and VR-ACS7 polypeptides, but only about 50% to VR-ACS1 polypeptide. ACS6 and ACS7 are specifically induced by auxin, while ACS1 is induced by cycloheximide, and to lesser extent by excision and auxin treatment. Results from nuclear run-on transcription assay and RNA gel blot studies revealed that all three genes were transcriptionally active displaying unique patterns of induction by IAA and various hormones in etiolated hypocotyls. Particularly, 24-epibrassinolide (BR), an active brassinosteroid, specifically enhanced the expression of VR-ACS7 by distinct temporal induction mechanism compared to that of IAA. In addition, BR synergistically increased the IAA-induced VR-ACS6 and VR-ACS7 transcript levels, while it effectively abolished both the IAA- and kinetin-induced accumulation of VR-ACS1 mRNA. In light-grown plants, VR-ACS1 was induced by IAA in roots, whereas W-ACS6 in epicotyls. IAA- and BR-treatments were not able to increase the VR-ACS7 transcript in the light-grown tissues. These results indicate that the expression of ACC synthase multigene family is regulated by complex hormonal and developmental networks in a gene- and tissue-specific manner in mung bean plants. The VR-ACS7 gene was isolated, and chimeric fusion between the 2.4 kb 5'-upstream region and the $\beta$-glucuronidase (GUS) reporter gene was constructed and introduced into Nicotiana tobacum. Analysis of transgenic tobacco plants revealed the VR-ACS7 promoter-driven GUS activity at a highly localized region of the hypocotyl-root junction of control seedlings, while a marked induction of GUS activity was detected only in the hypocotyl region of the IAA-treated transgenic seedlings where rapid cell elongation occurs. Although there was a modest synergistic effect of BR on the IAA-induced GUS activity, BR alone failed to increase the GUS activity, suggesting that induction of VR-ACS7 occurs via separate signaling pathways in response to IAA and BR.

Effects of Growth Regulators on Plant Regeneration from the Cotyledon Explant in Oriental Melon (Cucumis melo L.) (참외 (Cucumis melo L.)자엽절편으로부터 식물체 재분화에 미치는 생장조절물질의 영향)

  • This study was carried out to find the effects of plant growth regulators on callus formation, rooting and shooting from cotyledon explant in oriental me]on. Various combinations of 0.1 mg/L auxins (IAA, NAA) and 0.5, 1.0. 1.5, 2.0 mg/L cytokinins (BA, kinetin, zeatin) were treated to the MS basal medium, respectively. Callus was induced mort effectively as 2,437.0 mg (FW)/explant in MS medium supplemented with 0.1 mg/L NAA and 2.0 mg/L BA, but that was non-embryogenic callus as colored yellow white and broke easily. Root was induced most effectively at a frequency of 98.0% in MS medium supplemented with 0.1 mg/L NAA and 0-5 mg/L kinetin. Shoots formed on cut part of vein at a frequency of 98.0% in MS medium supplemented with 0.1 mg/L IAA and 2.0 mg/L BA, that were multiple shoots. in case of its concentration, BA and lower concentration of IAA and NAA (0.01 and 0.05 mg/L). respectively. shooting ratio was not increased. The result of treatment with BA 0-5 mg/L and IAA 0.1 mg/L, callus induced at a week, and shoot start to form multiple shoots about 3 weeks after inoculation. After 2 times subculture as 2 weeks intervals, divided shoots rooted and developed into intact plantlets at 10 weeks and then that grown normally on pots after acclimatization.

Interaction of Brassinolide with Other Known Plant Growth Regulators (Brassinolide와 기존 식물생장조절제(植物生長調節劑)와의 상호작용(相互作用))

  • This study was attempted to evaluate the combining effect of HBR (homobrassinolide) with the known growth regulators such as GA (gibberellic acid), BA(6-benzyl aminopurine), IAA (indole-3-acetic acid), B-9 (N-dimethylamino succinamic acid) and CCC (2-chloroethyl-trimethylammonium chloride) on the growth of radish hypocotyl. A single application of HBR increased hypocotyl growth as its rates increased from 0.1 to 1.0 ppm, showing a maximum increase at 1.0 ppm. GA and BA had no direct effects on hyopcotyl growth, but IAA showed some effect as its concentration increased. However, the mixed application of HBR with GA, BA and IAA increased the length of radish hypocotyl as the concentration of HBR became higher. The mixture of HBR with GA and BA showed antagonistic reaction on radish hypocotyl growth, but synergistic effect was shown in the higher rate mixture of HBR with IAA in the range of HBR at 0.03 to 0.30 ppm with IAA at 3.0 to 10.0 ppm, but antagonistic or additive response at the mixture of low rates. An increased growth of hypocotyl by HBR was ified by CCC, showing the strong antagonistic reaction, but B-9 was not able to ify HBR's effect on hypocotyl growth.

The Effects of Auxin and Casein Hydrolysate on the Growth of Ginseng Hairy Root (인삼모상근의 생장에 미치는 Auxin과 Casein Hydrolysate의 영향)

  • In this study, the effects of auxin and casein hydrolysate (CH) on the growth of ginseng hairy root was elucidated. Ginseng hairy root was cultured under light and dark conditions in MS solid and liquid medium with various concentrations of auxin and CH for fifty days. After harvesting the cultures, the fresh and dry weight of cultures were examined, respectively. In the MS solid culture, 1 mg/L of IBA was most effective on the growth of ginseng hairy root under the dark condition, whereas IAA and CH did not affect on the growth of ginseng hairy root. In the MS liquid culture, the growth was maintained regularly by the treatments of IAA and NAA. IBA and CH restrained the growth of ginseng hairy root.

Phytohormones Responses to NaCl Treatment in Rice Seedling (벼 유묘에서 NaCl처리에 따른 식물 호르몬의 변동)

  • Ten days old rice seedlings were treated with NaCl and the contents of endogenous hormones such as GA$_3$, ABA, IAA and zeatin were measured by ELISA. Water content and seedling growth were decreased as the salt concentration was increased. GA$_3$ increased up to 24 hours after NaCl treatment and decreased thereafter. ABA was raised by four times in 48 hours after NaCl treatment. IAA and zeatin decreased as the NaCl concentration and duration of treatment increased. GA$_3$ and ABA showed positive and negative correlation with the water content in the plant tissue, respectively.

Dynamics of the mobile insert helix in the domain III-IV of Aux/IAA17 probed by site-directed spin labeling and paramagnetic NMR spectroscopy

  • The plant hormone auxin is involved in all stages of plant development. Aux/IAAs are the transcriptional repressors that bind to the Auxin Response Factors (ARFs) to regulate the gene expression upon auxin release. Aux/IAA have highly conserved C-terminal domains (domains III-IV) that mediate both homotypic and heterotypic interactions between Aux/IAA and ARF family proteins. Recent studies revealed that the conserved domains III-IV share a common ${\beta}$-grasp fold that oligomerizes in a front-to-back manner. In particular, Aux/IAA contains a mobile insert helix in the domain III-IV, whereas ARFs do not. Here, we investigated the dynamics of the insert helix using paramagnetic NMR spectroscopy. The insert helix exhibited fast motions in the ps-ns time scale from $^{15}N$ relaxation data, but the amplitude of the motion is likely limited to the local neighborhood. Our result suggests that the motion of the helix may have functional implications in protein-protein interactions for transcriptional regulations.
