This study was designed to investigate the effect of garlic shoot extract administration on serum and liver tissue lipid levels in hyperlipidemic rats. The rats were fed a 45% high-fat diet to induce hyperlipidemia. They were then administered garlic shoot extract (50%) and ethanol extract (75%) at 200 mg/kg B.W./day (GSA-1, GSB-1) and 400 mg/kg B.W./day (GSA-2, GSB-2) for 5 weeks. The total lipid and triglyceride contents in serum were lowest in the GSB-2 group, and the total cholesterol content was significantly lower in the GSA-2 and GSB-2 groups than in the control group. The high density lipoprotein (HDL)-cholesterol content of the GSB-2 group was similar to that of the normal group. The low density lipoprotein (LDL) and very low density lipoprotein (VLDL)-cholesterol contents were significantly lower in the GSB-1 and GSB-2 group than in the control group. atherogenic index (AI) and cardiac risk factor (CRF) were lowest in the GSB-2 group (0.58 and 1.57, respectively). Aspartate aminotransferase (AST), alanine aminotransferase (ALT), and alkaline phosphatase (ALP) activities were significantly higher in the control group than in the normal group, and the levels of AST, ALT, and ALP were significantly lower in the GSA-2 and GSB-2 groups than in the control group. The total lipid content in liver tissue was significantly lower in all the experimental groups compared to that of the control group, but it was not significantly different among the experimental groups. The total cholesterol and triglyceride content in liver tissue was lowest in the GSB-2 group. Antioxidant activity in serum and liver tissue was highest in the GSB-2 group (40.16% and 47.41%, respectively).The thiobarbituric acid reactive substance (TBARS) content in serum and liver tissue was lowest in the GSB-2 group, with the significant difference. Our results suggest that garlic shoot extracts may improve lipid metabolism in serum and liver tissue and potentially reduce hyperlipidemia.
This study was performed to investigate the effects of ramie (Boehmeria nivea) leaf powder on improvements in lipid metabolism in serum, liver and adipose tissue, and the anti-obesity effect in rats fed a high fat/high cholesterol diet for 4 weeks to induce a hyperlipidemic and obese model rat. Male Sprague-Dawley rats weighting 210 g were divided into four groups; a normal diet group (N), a high fat/high cholesterol diet group (HFC), a high fat/high cholesterol diet with 5% ramie leaf powder group (HFC-RL), and a high fat/high cholesterol diet with 10% ramie leaf powder group (HFC-RH). The body weight gain increased with a high fat/high cholesterol diet, but gradually decreased in the ramie leaf powder fed groups compared with the HFC group. The liver index in the HFC group was highest among the four groups, although the difference was not significant compared with the ramie leaf powder fed groups. The adipose tissues weight in the HFC group was heavier than that of the N group, whereas those of groups fed ramie leaf powder decreased gradually. Alkaline phosphatase activity was not different between the HFC groups, but serum alkaline aminotransferase and lactate dehydrogenase activities decreased significantly after ramie leaf powder feeding. Levels of serum triglyceride, total cholesterol, LDL-cholesterol, atherogenic index and cardiac risk factors tended to decrease in the ramie leaf powder fed groups compared with the HFC group, whereas serum HDL-cholesterol level decreased in the HFC group and markedly increased in the ramie leaf powder fed groups. Levels of triglyceride and total cholesterol in the liver were significantly lower in the ramie leaf powder fed groups than in the HFC group. Levels of triglyceride and total cholesterol in adipose tissues were also lower in the ramie leaves powder fed groups than in the HFC group. The activities of heparinreleasable lipoprotein lipase (LPL) and total-extractable LPL in adipose tissues increased in the HFC group compared to that of the N group, but those of the ramie leaf powder fed groups decreased significantly. These results suggest that ramie leaf powder may improve lipid metabolism in serum, liver and adipose tissue and potentially reduce lipid storage.
Target herbal ingredient (THI) is an extract made from two herbs, Scutellariae Radix and Platycodi Radix. It has been developed as a treatment for metabolic diseases such as hyperlipidemia, atherosclerosis, and hypertension. One component of these two herbs has been reported to have anti-inflammatory, anti-hyperlipidemic, and anti-obesity activities. However, there have been no reports about the effects of the mixed extract of these two herbs on metabolic diseases. In this study, we investigated the metabolic effects of THI using a diet-induced obesity (DIO) mouse model. High-fat diet (HFD) mice were orally administered daily with 250 mg/kg of THI. After 10 weeks of treatment, the THI-administered HFD mice showed reduction of body weights and epididymal white adipose tissue weights as well as improved glucose tolerance. In addition, the level of total cholesterol in the serum was markedly reduced. To elucidate the molecular mechanism of the metabolic effects of THI in vitro, 3T3-L1 cells were treated with THI, after which the mRNA levels of adipogenic transcription factors, including C/$EBP{\alpha}$ and $PPAR{\gamma}$, were measured. The results show that the expression of these two transcription factors was down regulated by THI in a dose-dependent manner. We also examined the combinatorial effects of THI and swimming exercise on metabolic status. THI administration simultaneously accompanied by swimming exercise had a synergistic effect on serum cholesterol levels. These findings suggest that THI could be developed as a supplement for improving metabolic status.

We examined the effects of unripe black raspberry (UBR) and red ginseng (RG) extracts on cholesterol improvement in C57BL/6J mice fed a HCD (high cholesterol diet) for 12 weeks. Hepatic total lipid and total cholesterol contents were significantly induced in hyperlipidemic mice. However, supplementation with UBR, RG and simvastatin effectively reduced these lipid profiles. Further, UBR and co-treatment with UBR and RG increased expression of LDL receptor, SREBP2, and SR-B1 mRNA compared with HCD. The ApoB/ApoA1 ratio was reduced by co-treatment with UBR and RG compared to treatment with UBR. In addition, histopathologic evaluation showed that co-treatment with UBR and RG suppressed lipid accumulation as well as FAS and leptin expression in plasma. These results indicate that co-treatment with UBR and RG may be useful for the prevention of hypercholesterolemia.
The herb of Allium victorialis var. platyphyllum (Liliaceae) has been used as an edible wild herb and to treat heart failure and gastritis. We have already reported antihyperlipidemic anti-tumor effects of this plant. To enlarge the commercial availability of this food, it was investigated whether the extracts of A. victorialis var. platyphyllum reduce hyperlipidemia and obesity or not. The plants tested in this experiment were collected from two eco-types of Is. Ullung and Mt. Odae cultivated at Pyongchang. Extracts were prepared by extracting the fresh leaves and those dried at $36^{\circ}C$ and $90^{\circ}C$, respectively. Pretreatment with the ethanolic extracts for two weeks(p.o.) reduced serum triglyceride-, total cholesterol- and LDL-cholesterol contents in rats induced by Triton WR-1339, respectively. Furthermore, oral administration of the extracts also inhibited the hyperlipidemia induced with oral diet of 30% corn oil. In the other attempt to find to alleviate the obesity, the model rats with obesity were induced by the high fat-diet for six weeks. Post-treatment with the extracts for two weeks significantly reduced the hyperlipidemia. Retroperitoneal-, epidymal- and total abdominal fat pad weights were considerably reduced at 100 mg/kg oral administration of the extracts. Increased feces lipid contents were also found in the rat treated with the extracts. The extract may lead to the higher activity in treatment of hyperlipidemia and obesity than of the dried one.
Objectives : This study was designed to examine the effects of Polygonatum odoratum EtOH ext. on lowering lipid and anti-oxidation using hyperlipidemic rat. Methods : Male rats weighting $195.21{\pm}4.93g$ were divided into 4 groups and fed high fat diet for 8 weeks. Each of 7 rats was divided into a control and sample group. We fed a control group of rats a basal diet and administered normal saline(100 mg/kg, 1 time/1 day) for 4 weeks. And we fed each experimental group of rats basal diet and administered an extract of Polygonatum odoratum(100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 4 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. We measured lipid of plasma and liver, concentration of anti-oxidative activity and tumor necrosis factor-$\alpha$($TNF-{\alpha}$). Results : 1. Concentration of plasma free fatty acid, low density lipoprotein-cholesterol showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Concentration of plasma total cholesterol, triglyceride showed a significant decrement in all Polygonatum odoratum EtOH ext. groups than that of control group. However, concentration of plasma high density lipoprotein-cholesterol was not significantly different in all the treatment groups. 2. Concentration of liver total cholesterol showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Concentration of liver triglyceride(TG) showed a significant decrement in all Polygonatum odoratum EtOH ext. groups than that of control group. 3. Concentration of plasma thiobarbituric acid reactive substance, and liver thiobarbituric acid reactive substance showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. 4. The values of glutathione peroxidase activity showed a significant increment in all Polygonatum odoratum EtOH ext. groups than that of control group. The values of superoxide dismutase activity and catalase activity showed a significant increment in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. 5. The values of plasma aspartate aminotransferase and alanine aminotransferase activities were not significantly different in all treatment groups. 6. Concentration of liver $TNF-{\alpha}$ showed a significant decrement in the 200 mg/kg and 300 mg/kg Polygonatum odoratum EtOH ext. groups than that of control group. Conclusions : Based on the results in this study, the Polygonatum odoratum EtOH ext. showed a positive effect in lowering lipid and anti-oxidation.
The purpose of this study was to investigate the effect of germinated buckwheat on blood pressure, plasma glucose and lipid levels of spontaneously hypertensive rats(SHR). The male and female SHR were divided into two groups. After feeding the germinated buckwheat diet(50%, w/w) ad libitum for 6 weeks, the weight gain of the germinated buckwheat group in male rats was higher than those of control group. Systolic blood pressure of the germinated buckwheat group in male rats was significantly decreased at 6 weeks by 3%. The fasting glucose levels of the germinated buckwheat group had a tendency to be lower compared with those of the control group. Total cholesterol, HDL-cholesterol and HTR of female SHR fed the germinated buckwheat diet were significantly increased compared to the control diet. Atherogenic index of the germinated buckwheat group was decreased in males(ns) and females(p<0.05) than those of the control group. From these results the germinated buckwheat might be useful for anti-hypertensive and anti-hyperlipidemic agents.

We examined the effects of unripe black raspberry water extract (UBR-W) on lipid metabolism and oxidative stress in mice. C57BL/6J mice were divided into 4 groups: those administered a control diet (CTL), high-fat diet (HFD), UBR-W and simvastatin for 12 weeks. In the HFD group, LDL cholesterol were significantly higher than in the CTL group. However, the UBR-W treated group showed dose-dependent reduction of plasma LDL levels. Hepatic total lipid, TC, and malondialdehyde were significantly increased in hyperlipidemic mice. However, supplementation with either UBR-W or simvastatin effectively reduced these lipid profiles and lipid peroxidation. UBR-W increased mRNA expression of the LDL receptor, sterol regulatory element binding protein 2 (SREBP2), 3-hydroxy-3-methylglutaryl (HMG)-CoA reductase and ATP-binding cassette transporter A1 (ABCA1) compared to that observed in the HFD group. In addition, UBR-W and simvastatin showed significantly reduced oxidized LDL uptake by the scavenger receptor CD36. These results suggest that UBR-W is useful for treatment and prevention of hyperlipidemia and lipid peroxidation.
This study was carried out to investigate the effects of Plantago asiatica L. extract on anti oxidative potential, free radical generation and the lipid levels in rats. Sprague-Dawley(SD) rats were divided into two groups based on their diet, as follows: the AIN-76 diet (control group), and the modified AIN-76 diet(cholesterol 0.5%) with 0.5% P. asiatica extract, for 7 weeks. The body weight and teed efficiency ratios of the two groups did not significantly differ. The antioxidative potentials more significantly increased in the group that was fed P. asiatica extract than in the control group(p<0.05). There was no difference in the rate of free radical generation, though. The weights of the organs, such as heart, kidney, liver, and spleen, of the rats in the two groups did not differ, though. The ratio of the HDL cholesterol to the total cholesterol in the P. asiatica group was significantly higher than that in the control group, and the other serum lipid parameters (total cholesterol, HDL cholesterol, triglyceride, and phospholipids) did not differ between the two groups. These results imply that supplementation with P. asiatica extract may improve the antioxidant potential and decrease the lipid levels in the blood.

Hyperlipidemia is a major predisposing factor of atherosclerosis and cerebrovascular accidents. In effort to develope a hypolipidemic drug from medicinal herb, We tested three prescriptions of traditional medicine for the lipid lowering effect on diet-induced hyperlipidemic rats. GyejI-Bokryung-Hwan, Ohyak-Sunki-San, and Shihoga-Yongol-Moryu-Tang were selected based on the comnon prescriptions for patients with hyperlipidemia-related diseases. Water extract from each prescriptions was made by the method used in the clinical setting, and administered intragastrically once a day, for 4 weeks. Fenofibrate and lovastatin were given by the same method as the control drugs. Blood levels of total cholesterol(TC), high density lipoprotein(HDL), low density lipoprotein (LDL), and triglyceride(TG) were measured before, 1, 2, 3, and 4 weeks after starting the drug administration. All of the traditional prescriptions did not show lipid lowering effect, while fenofibrate lower the blood cholesterol levels(TC;from 215mg/dl before to 182m41 at 4 weeks-point of drug administration, and LDL ;from 203mg/dl before to aut 161, and 163mg/dl at 3 and 4 week-point respectively). Oyak-Sunki-San increased the level of TC (from 283mg/dl to over 350mg/dl starting from the first week of drug administratin). Effects on TG variable in the cases of traditional medicines and control drug In conclusion, any of the three traditional medical prescriptions did not decrease the level of blood cholesterol.
