  • Oxidative damage to DNA can be caused by excited oxygen species, which are produced by radiation or are by-products of aerobic metabolism. Endogenous evels of 8-hydroxy-2'deoxyguanosine (8-OH-dG), an adduct that results from the damage of DNA caused by hydroxyl radical, have been detected in E. coli and S. typhimurium. Treatment of bacterial cells with various concentrations of hydrogen peroxide caused a moderate increase in the 8-OH-dG content. The enzymatic release of 8-OH-dG from asocorbate/Cu(II)-treated DNA was effected by an extract of E. coli cells. These results indicate that 8-OH-dG is formed in vivo inbacterial DNA through endogenous oxidative mechanisms and on treatment with an oxygen radical-producing agent and that it is repairable.

  • The conventional development of multi-component electrodes is based on the researcher's experience and is based on trial and error. Therefore, there is a need for a scientific method to reduce the time and economic losses thereof and systematize the mixing of electrode components. In this study, we use design of mixture experiments (DOME)- in particular a simplex lattice design with Design Expert program- to attempt to find an optimum mixing ratio for a three-component electrode for the high RNO degradation; RNO is an indictor of OH radical formation. The experiment included 12 experimental points with 2 center replicates for 3 different independent variables (with the molar ratio of Ru, Ti, Ir). As the Prob > F value of the 'Quadratic' model is 0.0026, the secondary model was found to be suitable. Applying the molar ratio of the electrode components to the corrected response model results is an RNO removal efficiency (%) = 59.89 × [Ru] + 9.78 × [Ti] + 67.03 × [Ir] + 66.38 × [Ru] × [Ir] + 132.86 × [Ti] × [Ir]. The R2 value of the equation is 0.9374 after the error term is excluded. The optimized formulation of the ternary electrode for an high RNO degradation was acquired when the molar ratio of Ru 0.100, Ti 0.200, Ir 0.700 (desirability d value, 1).

  • The objective of this study is to evaluate antioxidant activities of Rehmanniae Radix Preparata (RRP) prepared from dried root through nine repeated steaming-drying cycles. The total phenolic content of the final cycle of RRP increased to 165.2% compared with that of the 1st cycling product. Antioxidant activities which were determined by ferric-reducing antioxidant potential (FRAP), 2,2'-azinobis(3 ethybenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging, 1,1-diphenyl-2-picrydrazyl (DPPH), and hydroxyl radical scavenging activities, increased remarkably as the number of steaming-drying cycles increased. Especially, FRAP value increased to 89.1%. Further, $IC_{50}$ values for DPPH and hydroxyl radical scavenging activities of the final 9th-cycling product decreased to 58.3% and 67%, respectively. Based on these results, it was observed that the antioxidant activities of RRP improve according to an increased number of steaming-drying cycles.

  • Bupleury radix has been used for the treatment of fever, liver disease, inflammation in traditional medicine. The present study was carried out to evaluate the antioxidant effects of Bupleury radix aqua-acupuncture solution (BRAS) in vitro. Oxygen derived free radicals produced in the course of normal aerobic life, such as superoxide anion radical($O_2^-$ ), hydroxyl radicaI( OH), hydrogen peroxide($H_2O_2$) and singlet oxygen($^1O_2$) can attack polyunsaturated fatty acid in cell membranes, enzymes, other cell compounds, and give rise to lipid peroxidation, DNA damage, lipofuscin accumulation, structure alteration of cell membrane and cell death. In this study, antioxidant effects of BRAS on lipid peroxidation were determined according to the method of TBA. BRAS inhibited markedly peroxidation of linoleic acid during the autoxidation, and also inhibited lipid peroxidation induced by hydroxyl radical derived from $H_2O_2-Fe^{2+}$ in rat liver homogenate. And BRAS showed 30% scavenging effect on DPPH radical, also exhibited a 30% inhibitory effect on superoxide generation from xanthine-xanthine oxidase system. In addition, BRAS protected the cell death induced by tert-butyl hydroperoxide(t-BHP) and significantly increased cell viability in the normal rat liver cell(Ac2F).

  • Chemical composition of the Eucommia ulmoides bark, including extractives, proximate, mineral, fatty acid and monosaccharide compositions, was studied The most abundant mineral was calcium (533.17 mg/l00 g). $\alpha-linolenic$ acid (24.7%) and linoleic acid (24.3%), showed higher contents among the fatty acids. Major monosaccharides of E. ulmoides balk were arabinose (13.94 mg/g), xylose (18.91 mg/g) and glucose (119.7 mg/g). From the bark of E ulmoides, four compounds were isolated and their structures were elucidated as caffeic acid (I), kaempferol (II), quercetin (III) and isoquercitrin (IV) by spectroscopic analysis such as NMR and MS, including cellulose TLC and other chemical evidence such as hydrolyzation and acetylation. The antioxidant activities of four isolated compounds were evaluated by DPPH free radical scavenging, hydroxyl scavenging and reducing power assays. The results indicated that all the isolated compounds showed higher DPPH radical scavenging activity than $\alpha-tocopherol$ and BHT that were used as positive controls and these four compounds exhibited considerable reducing power and hydroxyl radical (OH) scavenging activity. Considering from the results above, it suggests that the E. ulmoides bark is a potential natural source of antioxidant material.

  • The sonolytic decomposition of NHCs(Nitrogen Heterocyclic Compounds), such as atrazine[6-chloro-N-ethyl-N'-(1-methylethyl)-1,3,5-triazine-2,4-diamine], simazine(6-chloro-N,N'-diethyl-1,3,5-triazine-2,4-diamine), trietazine(6-chloro-N,N,N'-triethyl-1,3, 5-triazine-2,4-diamine), in water was investigated at a ultrasound frequency of 200kHz with an acoustic intensity of 200W under argon and air atmospheres. The concentration of NHCs decreased with irradiation, indicating pseudo-first-order kinetics. The rates were in the range $1.06{\sim}2.07({\times}10^{-2}min^{-1})$ under air and $1.30{\sim}2.59({\times}10^{-2}min^{-1})$ under argon at a concentration of $200{\mu}M$ of NHCs. The rate of hydroxyl radicals(${\bullet}{OH}$) formation from water is $19.8{\mu}M\;min^{-1}$ under argon and $14.7{\mu}M\;min^{-1}$ under air in the same sonolysis conditions. The sonolysis of NHCs is effectively inhibited, but not completely, by the addition of t-BuOH(2-methyl-2-propanol), which is known to be an efficient ${\bullet}{OH}$ radical scavenger in aqueous sonolysis. This suggests that the main decomposition of NHCs proceeds via reaction with ${\bullet}{OH}$ radical; a thermal reaction also occurs, although its contribution is small. The addition of appropriate amounts of Fenton's reagent $[Fe^{2+}]$ accelerates the decomposition. This is probably due to the regeneration of ${\bullet}{OH}$ radicals from hydrogen peroxide, which would be formed from recombination of ${\bullet}{OH}$ radicals and which may contribute a little to the decomposition.

  • This study investigated the antioxidant activity of methanol (MeOH) and water extracts from roots of Cirsium japonicum in vitro, MeOH extract showed a stronger tree radical scavenging activity than water extract. However, both of extracts showed a concentration dependent hydroxyl radical scavenging activity, reducing power and metal chelating ability, MeOH extract had greater phenolic and flavonoid contents than water extract. The antidiabetic activity of these two extracts was evaluated by the a-glucosidase inhibition assay, The water extract showed a considerable a-glucosidase inhibitory activity. To our knowledge, this may be the first time to report the antioxidant and antidiabetic activities in Cirsium japonicum roots.
