Anti-inflammatory and Antioxidant Effects of Zizyphus jujuba Miller var. inermis extracts (대추 추출물의 항산화 및 아토피 피부염 관련 항염증 효과)

  • In the present study, the effect of Z. jujuba Miller var. inermis Rehder extracts on the secretion of atopic dermatitis (AD)-related cytokines and hyaluronidase activity was investigated. We prepared four fractions, butylene glycol (JB), ethanol (JE), and water (JW), with Z. jujuba Miller var. inermis extracts. JW significantly reduced the secretion of interleukin-8 and JE reduced the secretion of tumor necrosis factor-alpha in human keratinocyte HaCaT cells. Also, hyaluronidase activity was measured by enzyme assay and the fractions inhibited the activity in a dose-dependent manner. In addition, the human dermal fibroblast, HDF-n cells were treated with the extracts and antioxidant activities were measured. The results showed that the extracts increased the free radical scavenging activity and the superoxide dismutase activity. Taken together, Z. jujuba Miller var. inermis extracts reduced the secretion of AD-related cytokines and inhibited the hyaluronidase. In addition, the extracts showed antioxidant activity.

Screening of Hyaluronidase Inhibitory and Free Radical Scavenging Activity in vitro of Traditional Herbal Medicine Extracts (생약재 추출물의 hyaluronidase 저해 및 라디칼 소거 활성 검색)

  • For the screening of anti-inflammation and antioxidative activities, ethanolic extract of 40 species of traditional herbal medicines were examined their hyaluronidase inhibitory effect and radical scavenging activity in vitro. From the result of the hyaluronidase inhibitory activity using a Morgan-Elson assay, Astragali Radix, Eucommia Cortex, Schizandrae Fructus, Scutellaria Radix, Acanthopanacis Cortex, Chaenomelis Fructus, Amomum xanthioides Wallich and Moutan Radicis Cortex showed more than 50% hyaluronidase inhibitory effects at the concentration of 10 mg/mL. In the various solvent fractions (n-hexane, chloroform, ethyl acetate, water) prepared from ethanolic extracts, the ethyl acetate fraction of all extracts tested showed strong activity. Antioxidative activity was evaluated by assaying electron-donating ability to DPPH free radical and scavenging of hydroxyl radical (ㆍOH) generated through Fenton reaction, respectively. Rubus coreanus Miq, Moutan Radicis Cortex, Paeoniae Radix Alba, Plantaginis Semen and Sorbus commixta Hedl. showed high activity more than 90%, yet similar activity to $\alpha$-tocopherol and BHA at the concentration of 1 mg/mL in electron donating activity. The scavenging effects of ethanolic extracts on hydroxyl radical were investigated using a 2-deoxyribose oxidation method and tested all extracts showed significant radical scavenging activity. The experiment was also performed to examine whether herbal medicines having significant lipid peroxidation inhibitory activity, Schizandrae-Fructus is the strongest inhibitory activity in both linoleic acid and liposome peroxidation.

Lipoxygenases, Hyaluronidase, and Xanthine Oxidase Inhibitory Effects Extracted from Five Hydrocotyle Species

  • Hydrocotyle is a genus of prostrate, perennial aquatic or semi-aquatic plants formerly classified in the family Apiaceae, now in the family Araliaceae. Lipoxygenases (LOX) are present in the human body and play an important role in the stimulation of inflammatory reactions. Ethanolic extracts of five Hydrocotyle species (H. ramiflora, H. maritima, H. nepalensis, H. sibthorpioides, and H. yabei) showed inhibition of 23.5~50.6% at 2.0 mg/mL. Their extracts showed LOX inhibition in half maximal effective concentration (EC50) range 15.1~15.7 ㎍/mL. Hyaluronic acid is a glycosaminoglycan, a major component of the extracellular matrix Five extracts of these species inhibited less than 23.0% of Hyaluronidase (HAase) activity at a concentration of 2.0 mg/mL Xanthine oxidase (XO) is a form of xanthine oxidoreductase, a type of enzyme that generates reactive oxygen species. Five Hydrocotyle species were found to have inhibitory activity of XO at 2.0 mg/ml, with 65% having greater than 50% inhibition. H. ramiflora exhibited the highest activity with an inhibition of 80.0%. The results suggested that Lipoxygenases, Hyaluronidase, and Busan 47340, Republic of Korea from five Hydrocotyle species might be multifunctional and prevent the degradation of allergic reactions and inflammation.

Screening of the Tyrosinase Inhibition and Hyaluronidase Inhibition Activities, and Radical Scavenging Effects Using Plants in Cheju (제주산 식물을 이용한 Tyrosinase 억제 활성, Hyaluronidase 억제 활성, 라디칼 소거 활성 검색)

  • Solvent extracts of 17 plants collected in Cheju Island were investigated for their biological properties related to cosmeceuticals such as tyrosinase and hyaluronidase inhibition and also radical scavenging effects. The chloroform fraction of Phytolacca esculenta root exhibited strong inhibition against tyrosinase activity. No fraction showed significant hyaluronidase inhibition. Some solvent extracts of plants such as Achyranthes japonica and Artemisia princeps showed considerable radical scavenging activities.

Analysis of Human O-GlcNAcase Gene and the Expression of the Recombinant Gene. (사람의 O-linked N-acetyl-$\beta$-D-glucosaminidase 유전자의 분석과 재조합 발현)

  • Dynamic modification of cytoplasmic and nuclear proteins by O-linked N-acetylglucosamine (O-GlcNAc) on Ser and Thr residues is ubiquitous in higher eukaryotes. And this modification may serve as a signaling mod-ification analogous to protein phosphorylation. Addition and cleavage of O-GlcNAc are catalyzed by O-linked GlcNAc transferase (OGT) and O-linked N-acety1glucosaminidase (O-GlcNAcase), respectively. Two types of human O-GlcNAcase gene were cloned and expressed as three fusion proteins in Escherichia coli. O-GlcNA-case activity showed in the order of thioredoxin fusion> $6{\times}His$ tag> GST fusion. O-GlcNAcase had enzy-matic activity against only ${\rho}$NP-GlcNAc of seven tested substrate analogs. Blast search revealed that O-GlcNAcase has two conserved domains, amino terminal hyaluronidase-like domain and carboxy terminal N-acetyltransferase domain. Extensive deletion studies were done to define catalytically important domains. The deletions of hyaluronidase-like domain and N-acetyltransferase domain abolished enzyme activity. But, N-ter-minal 55 amino acid deletion and C-terminal truncation showed lower activity. Based on deletion analysis, we suggest that hyaluronidase-like domain is essential for enzyme activity and carboxy terminal N-acetyltrans-ferase domain may be modulatory function.

Industrialization possibilities of purified pig sperm hyaluronidase

  • The goals of the present study were to develop a simple method for obtain highly purified pig sperm hyaluronidase (pHyase) and to assess its activity, function, and safety. In mammals, sperm-specific glycophosphatidylinositol (GPI)-anchored Hyase assists sperm penetration through the cumulus mass surrounding the egg and aids in the dispersal of the cumulus-oocyte complex. Recently, Purified bovine sperm hyaluronidase (bHyase) has been shown to enhance therapeutic drug transport by breaking down the hyaluronan barrier to the lymphatic and capillary vessels, thereby facilitating tissue absorption. Commercially available Hyase is typically isolated from bovine or ovine; which have several disadvantages, including the risk of bovine spongiform encephalopathy, low homology with human Hyase, and the requirement for relatively complex isolation procedures. This study successfully isolated highly purified pHyase in only two steps, using ammonium sulfate precipitation and fast protein liquid chromatography. The isolated Hyase had activity equal to that of commercial bHyase, facilitated in vitro fertilization, and effectively dissolved high molecule hyaluronic acid. This simple, effective isolation method could improve the availability of pHyase for research and clinical applications.

Effects of Phenylpropanoid Compounds on Melanin Production in B16 Melanoma Cells (B16 Melanoma 세포에서 Phenylprlopanoid 화합물이 Melanin 생성에 미치는 영향)

  • To investigate the relationship between structure and biological activity of phenylpropanoids, we measured effects of phenylpropanoids on anti-oxidant and whitening activity, In DPPH radical scavenging activity, caffeic acid analogues showed the significant anti-oxidant activity. Although phenylpropanoids did not inhibit purified-tyrosinase activity, they significantly inhibited tyrosinase activity and melanin production in MSH-stimulated B16 melanoma cells. However, phenylpropanoids did not affect tyrosinase expression in MSH-stimulated B16 melanoma cells, which suggest that inhibition of MSH-induced melanin production was due to tyrosinase inhibition mediated via other signal pathways but not expression of tyrosinase. Phenylpropanoids also significantly inhibited both hyaluronidase and elastase activity, suggesting that phenylpropanoids may be used as whitening, hydration and anti-wrinkling agents. Hydroxyl residue of aromatic ring in phenylpropanoids plays an important role in anti-oxidant and whitening activity.

Anti-skin aging activities of ethanol extract from Echinodorus cordifolius L. in human keratinocytes (물수선화 에탄올 추출물의 피부 노화 억제 효과)

  • Echinodorus cordifolius (L.) is an aquatic plant in the family Alismataceae. The anti-skin aging activity of E. cordifolius (L.) has not been yet reported. Therefore, the objective of the present study was to prepare 70% ethanol extract (ECEE) from E. cordifolius (L.) and investigate their antioxidant and anti-hyaluronidase activities for confirm the potential of anti-skin aging. ECEE showed good activities of DPPH, hydrogen peroxide scavenging, and hyaluronidase inhibition, with EC50 and IC50 values of 31.4, 300, and 450 ㎍/mL, respectively. ECEE also significantly improved cell viability and inhibited intracellular reactive oxygen species dose-dependently against 1 mM hydrogen peroxide-induced oxidative stress in immortalized human keratinocytes (HaCaT cells). Furthermore, ECEE upregulated hyaluronic acid (HA)-synthesizing enzyme hyaluronan synthase 2 (HAS2) expression level, but downregulated expression level of HA-degrading enzyme hyaluronidase 2, resulting in increased HA production in HaCaT cells. Taken together, these results suggest that ECEE shows antioxidant and anti-hyaluronidase potential and could be a functional cosmetic ingredients for anti-skin aging.

Identification and Characterization of Bull Sperm Hyaluronidase Hyal5

  • During mammalian fertilization, germ cell-specific hyaluronidases, such as sperm adhesion molecule 1 (SPAM1) and hyaluronoglucosaminidase 5 (Hyal5), are important for the dispersal of the cumulus mass. In this study, we demonstrated that bull Hyal5 is a single copy gene on chromosome 4 that is expressed specifically in the testis. In addition, we expressed recombinant bull SPAM1 and Hyal5 in human embryonic kidney 293T cells and showed that these enzymes possessed hyaluronidase activity. We also demonstrated that a polyclonal antibody against bull sperm hyaluronidase inhibits sperm-egg interactions in an in vitro fertilization (IVF) assay. Our results suggested that bull Hyal5 may have a critical role in bull fertilization.

Inhibition Effect Against Elastase, Collagenase, Hyaluronidase and Anti-oxidant Activity of Thinning Green Ball Apple

  • The thinning Green ball apple was extracted using water and ethanol and a phenolic concentration of thinning Green ball apple was $50-200{\mu}g/mL$. The water and ethanol extracts of thinning Green ball apple showed 94.69% and 92.24% 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity and 100.30% and 99.16% 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity at phenolic concentration of $200{\mu}g/mL$, respectively. The water and ethanol extracts of thinning Green ball apple showed antioxidant protection factor of 1.76 antioxidant protection factor and 1.76 antioxidant protection factor, respectively. The water and ethanol extracts showed 101.46% and 99.64% anti-oxidative effect on thiobarbituric acid reactive substances at phenolic concentration of $200{\mu}g/mL$. Hence, the water and ethanol extracts of thinning Green ball apple can be considered a potential anti-oxidant. The water and ethanol extracts showed 33.28% and 32.14% hyaluronidase inhibition, respectively, at phenolic concentration of $150{\mu}g/mL$. The water and ethanol extracts showed 47.33% and 40.92% elastase inhibition and 46.19% and 65.58% collagenase inhibition at phenolic concentration of $200{\mu}g/mL$, respectively. About these experiments, thinning Green ball apple was found to exhibit anti-oxidation activity as well as hyaluronidase, elastase and collagenase inhibitory activities. Therefore, thinning Green ball apple can be considered a potential source for functional food.
