Effects of Hwangryunhaedoktang Herbal-acupuncture at G21(Kyonjong: 肩井) on the Heart Rate Variability (견정혈(肩井穴) 황련해독탕약침(黃連解毒湯藥鍼)이 심박변이율(HRV)에 미치는 영향)

  • Objective : In this study, we investigated the effects of Hwangryunhaedoktang Herbal-acupuncture(HHT) at a acupoint, $G_{21}$(Kyonjong) on the Heart Rate Variability(HRV) in adults, as well as we tried to observe how HHT affects on the balance of the autonomic nervous system. Methods : The subject were comprised of 20 healthy adults, who didn't take any drugs which affected on the autonomic nervous system and had normal sinus rhythm in electrocardiogram. The control group was comprised of 9 subjects, who were injected normal saline at $G_{21}$(Kyonjong). The experiment group consisted of 10 subjects, who were injected HHT at $G_{21}$(Kyonjong). We measured HRV by CANS 3000 on three times : before, immediately after, and 20 minutes after injection. The SPSS 10.0 for windows was used to analyze the date and the Wilcoxon signed rank test (in group) and Mann-Whitney U-test(between two groups) were used to verify the results. Results : The following results were obtained ; 1. In each group, Mean-HRV and Mean-RR showed a significant change during 20 minutes after injection. But there were no significant difference between two groups(p>0.05). 2. In experiment group, 20 minutes after injection, norm LF and LHR showed a significant decrease and norm HF showed a significant increase. There were significant difference between two groups(p<0.05). Conclusions : The findings suggest that HHT at $G_{21}$(Kyonjong) in adults relatively activate parasympathetic nervous system and inactivate sympathetic nervous system.

Antioxidant, Anti-wrinkle and Whitening Effect of Fermented Extracts of Hwangryunhaedoktang (황련해독탕 발효물의 항산화, 항주름 및 미백 효과)

  • Hwangryunhaedoktang (HHT) has been traditionally used as a preventive and therapeutic medicine to treat enervation and diverse chronic diseases. This study was designed to compare the antioxidant, anti-wrinkle and whitening effects of HHT extract and its fermented extract by Leuconostoc mesenteroides (FHHT). FHHT was prepared by inoculation of L. mesenteroides after the extraction procedure with 70% ethanol. HHT and FHHT was investigated via high-performance liquid chromatography (HPLC). Simultaneous qualitative analysis of two bioacitive components, berberine and palmatine. was achieved by comparing their retention times ($t_R$) and UV spectra with those of the standard components. Cell viability test results indicated that both HHT and FHHT were non-toxic. In DPPH radical scavenging ability, $SC_{50}$ values of the FHHT was $68.85{\mu}g/mL$, which is more effective than HHT. Moreover, FHHT showed higher expression in production of procollagen type I than HHT. In nontoxic concentration range, FHHT showed strong melanin production inhibitory effect in ${\alpha}-melanocyte$ stimulating hormone (${\alpha}-MSH$)-stimulated B16F10 cell ($IC_{50}=9.82{\mu}g/mL$). These results suggested that fermented extracts of hwangryunhaedoktang had considerable potential as a cosmetics ingredient with an antioxidant and anti-wrinkle and whitening effects.

  • Objective : This study was designed to find out whether NS(normal saline) is able to be constituted as an appropriate control group for HHT(Hwangryunhaedoktang herbal acupuncture) and HP(Homnis Placenta herbal acupuncture). Methods : NS and two herbal acupuncture were inserted into Quchi(LI 11) of the subjects. After 5 minutes the subjects completed a questionnaire rating the intensity of 21 kinds of acupuncture sensation; hurting, penetrating, sharp, aching, intense, spreading, radiating, tingling, pricking, stinging, pulling, heavy, dull, numb, electric, shocking, hot, burning, cool, pulsing, and throbbing. We compared the subjective acupuncture sensation between the two groups. Results : As for HHT seven items of the acupuncture sensation were significantly different from NS. In case of HP just two items were significantly different from NS. Conclusion : We found that NS may be able to be an placebo herbal acupuncture for HP. Further study is needed for the appropriate placebo herbal acupuncture.

The Effect of Hwangryunhaedoktang on Proliferations of Various Human Cancer Cells (황련해독탕이 수종의 인간 암세포 증식에 미치는 영향)

  • Objectives The aim of this study is to investigate whether hwang-ryun-haedok-tang (HDT) affect proliferations of androgen-dependent LNCaP prostate cancer cells, androgen-independent PC-3, DU-145 prostate cancer cells, MCF-7 human breast cancer cells, A549, NCI-H292 human pulmonary cancer cells and K-562 human chronic myelogenous leukemia cells. Materials and Methods Effects of HDT on proliferations of each cancer cell line were investigated. 20,000 cells/well were plated in each well of 96-well culture plate. After 24 hrs, 0.01-10% of HDT in culture medium was added to cancer cells. The number of cells was counted by using SRB assay or direct cell counting method after 72 hours from drug treatment. Effect of baicalein or berebrine on proliferation was assessed according to the same method. Results (1) HDT inhibited proliferations of LNCaP, PC-3 and DU-145 prostate cancer cells. (2) HDT inhibited proliferation of MCF-7 breast cancer cells. (3) HDT also inhibited proliferations of A549, NCI-H292 pulmonary cancer cells and K-562 chronic myelogenous leukemia cells. (4) Baicalein and berberine also showed inhibitory effects on proliferations of prostate and breast cancer cells. Conclusion : HDT inhibited proliferations of human prostate, breast, pulmonary and blood cancer cells. These results suggest us the potential use of HDT as a chemopreventive or chemotherapeutic agent. Effect of HDT on human cancer should be further investigated using in vivo experimental models that can reflect pathophysiology of human cancer through another studies.

Antioxidative and Antimicrobial Activities of Herbal Complex Extract from Hwangryunhaedok-tang and Essential Oil of Chamaecyparis obtusa (황련해독탕(黃連解毒湯)·편백(扁柏) 정유 복합추출물의 항산화 및 항균활성에 관한 연구)

  • Objectives : The present study was designed to investigate effects of mixed extracts (9:1, v/v) of Hwangryunhaedoktang, Chamaecyparis obtusa essential oil. We evaluated the antimicrobial and antioxidant activity by manufacturing mixed extracts as the materials for functional medicinal herb cosmetics. Methods : We performed antimicrobial were tested microbes (Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Candida albicans, Propionibacterium acnes) by disc diffusion method and measure clear zone. Antioxidant activities were measured by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity. Results : Antimicrobial activities of mixed extracts against S. epidermideis, P. acnes, S. aureus, E. coli, and Candida. A were $10.9{\pm}3.0mm$, $16.9{\pm}2.0mm$, $9.2{\pm}2.0mm$, $10.3{\pm}1.0mm$, $11.8{\pm}3.0mm$ respectively. The Chamaecyparis obtusa essential oil had the highest antimicrobial activities against S. epidermideis, P. acnes, S. aureus, E. coli, and Candia. A and clear zone of microbes ware $16.7{\pm}3.0mm$, $28.8{\pm}0.2mm$, $15.9{\pm}2.0mm$, $11.5{\pm}1.0mm$, $16.3{\pm}3.0mm$. Hwangryunhaedoktang extract showed antimicrobial activity but only P. acnes and S. aureus The antioxidant activities of the mixed extracts were tested through the evaluation of DPPH radical scavenging activity. The 100 % mixed extracts were found to have 90 % DPPH radical scavenging activity. The mixed extracts was presented similar antioxidant activities compared with that of ascorbic acid. As a result, A mixture extract is expected to have antimicrobial effects and free radical scavenging activity was found. Conclusions : Accordingly, It can be concluded that mixed extracts has the potential to cosmetic material.

Comparative Effectiveness of Hwangryunhaedok-tang Pharmacopuncture, Essential Bee Venom Pharmacopuncture and Jungsongouhyul Pharmacopuncture for Cervical Pain caused by Traffic Accidents: A Retrospective Observational Study (교통사고 후 발생한 경항통에 대한 황련해독탕약침, 봉독약침, 중성어혈약침 치료의 효과 비교 연구: 후향적 관찰 연구)

  • Objectives The purpose of this study is to compare the effectiveness of Hwangryunhaedok-tang Pharmacopuncture, Bee Venom Pharmacopuncture and Jungsongouhyul Pharmacopuncture on cervical pain caused by traffic accident retrospectively. Methods The retrospective observational chart review was conducted to 52 cases of traffic accident patients who had admitted to Daegu haany University Pohang Korean Hospital from December 1st, 2015 to May 31th, 2017. The patients were classified into three groups by which pharmacopuncture was used as Hwangryunhaedoktang pharmacopuncture treatment group, bee venom pharmacopuncture treatment group and Jungsongouhyul pharmacopuncture treatment group. Additionally, outcomes should include Neck disability index(NDI) and patient's global assessment(PGA) for evaluate the treatment effects. Results Each pharmacopuncture treatment group showed significant decrease of NDI and PGA. But there was no significant difference in NDI and PGA between three groups. Conclusions According to the results, we found out that 3 kinds of pharmacopunctures are considered to be effective and useful for cervical pain patients caused by traffic injury. Further well designed prospective studies are needed.

Effect of Herbal CP Soap on Facial Moisture (한약저온숙성비누가 얼굴수분에 미치는 영향)

  • Objectives : To maintain a healthy skin condition, the role of moisture in the stratum corneum is important. We use soap in our daily life, so we wanted to observe the changes of the skin moisture using herbal CP soap. Four kinds of herbal CP soaps were prepared and their effects on skin moisture were compared. Methods: We surveyed 39 students at A University to determine their subjective skin condition, lifestyle and washing habits. Then, a facial analysis test was performed. We made herbal CP soaps using Kyungohkgo, Hwangryunhaedoktang, Seosiokyongsan and Baeksoooh and distributed them to experiment participants. Prior to the experiment, their skin condition was checked and skin condition was assessed using Facial Diagnosis System three times in total. After the experiment, the change of skin was measured and analyzed. Results: As a result of examining the skin moisture according to the using of the herbal CP soap by the participants, the skin moisture was gradually increased compared with before use. The moisture content increased after 6 weeks of use compared to before using Seosiokyongsan, Hwangryunhaedoktang, Kyungohkgo CP soap compared to before use. But there was no significant difference between 3 weeks and 6 weeks of use compared to before use. As a result of using Baeksoo-oh CP soap, the skin moisture of the experimental participants was continuously increased. Conclusion: Natural CP soaps are believed to have a beneficial effect in increasing the moisture content of facial skin.

A Clinical Case Report on Grade Four Pressure Ulcer Patients Treated with Hwangryunhaedok-tang Pharmacopunctue and Herbal Medicine Taklisodok-eum (탁리소독음과 황련해독탕 약침을 병용하여 호전된 4도 욕창 1례)

  • ■ Objectives The purpose of this case study is to report the effectiveness of Korean medicine and pharmacopuncture in the treatment of pressure ulcer. ■ Methods The patient with pressure ulcer was treated with herbal medicine, Hwangryunhaedoktang pharmacopuncture, acu-moxi treatment. We started herbal medicine mainly Taklisodok-um for pressure ulcer, but changed to Paljung-san due to urinary tract infection on Day 67. During the daily dressing, we inject pharmacopuncture solution subcutaneous ulcer area and spray solution on the cavity of right pressure ulcer. The severity of pressure ulcer was assessed using[NPUAP] pressure ulcer stage, healing rate of pressure ulcer and depth of right side ulcer cavity on Day1, Day15, Day40, Day73, Day95. ■ Results After treatment, the total size of the pressure ulcer was reduced from 90(10*9) to 56(8*7)cm2, healing rate of pressure ulcer increased to 37.8%, depth of pressure ulcer was reduced from 3.5 to 2cm. Though NPUAP stage(Gr 4) was not changed, size and color of pressure ulcer definitely improved compared to Day1. ■ Conclusion This study suggested taht Korean medical treatment could be effective option for treating grade 4 pressure ulcer.

Two Case Report of Obesity with atopic dermatitis Treated with Hwangryunhaedoktang-gamibang (아토피 피부염을 겸한 20대 비만 여성 황련해독탕가미방 치험 2례)

  • Objectives : This study was performed1 to assess the effectiveness of HwangryunHaedokTang-gamibang on Obesity with atopic dermatitis. Methods : Patients were treated by HwangryunHaedokTang-gamibang. To measure treatment outcomes, BMI, weight, Fat mass, Fat %, VAS scale, SCORAD index were used. Results : After treatment, BMI, weight, Fat mass, Fat% were significantly decreased. Atopic dermatitis VAS and SCORAD index were also decreased at all cases. Conclusions: HwangryunHaedokTang-gamibang may be effective in the treatment of Obesity with atopic dermatitis patients.

Literature Review on Korean Medicine Treatment for Alopecia

  • This study aimed to analyze the use of Korean medicine treatments for alopecia in among clinical studies. We identified and analyzed 22 studies from Korean databases; Oriental Medicine Advanced Searching Integrated System (OASIS), Science ON, Korean Studies Information Service System (KISS), and Research Information Sharing Service (RISS) and international database; PubMed. We analyzed the Korean medical treatment in each case and determined the tendency to use each intervention. We analyzed 1,464 patients from 22 selected studies. Herbal medicine, acupuncture, external medicine or products, pharmacopuncture, and phototherapy were used for alopecia treatment. The herbal medicines mainly used to treat alopecia were Gagam Cheongyoung-tang, Gagam Hwajung-hwan, and Yukmijihwang-tang·hwan. The acupoints primarily used were GV20, EX-HN1, GB5, KI3, PC6, ST36, GV22, and A-shi. The most commonly used pharmacopuncture therapies were Hwangryunhaedoktang (HH), Carthami Fructus (CF), Bee Venom (BV), and Hominis placenta (HP). The Korean medical treatment for alopecia improved the condition of patients. However, seven studies reported the occurrence of side effects such as pruritus, dazed, drowsiness, headache, pain, and diarrhea. This study shows the potential of Korean medicine for the treatment of alopecia. Further studies with a large sample size and long-term follow-up are warranted to establish the primary treatment guidelines and objective outcome measures for alopecia.
