The effect of active immunization with Acanthamoebn culbeksoni in mice born to immune mother (수동면역이 Acanthmoeba culbertsoni 능동면역 형성에 미치는 영향)

  • Acanthamoebn culbertsoni is a pathogenic free-living amoeba causing primary amoebic meningoencephalltls (PAME) in human and mouse. Several reports on the immune responses in mice with this amoebic infection have been published, but the effects of transferred passive Immunity on the active immunization In offspring mice have not been demonstrated. This experiment was done to observe the effect of active Acanthamoebn culbertsoni was cultured in the CGV medium axenlcally. Female BALB/c mice weighing about 20g were immunized through the intraperitoneal injection of Acanthamoeba cuLbensoni trophozoites 1 × 106 each three times at the interval of one week. Offspring mice were immunized two times. The mice were inoculated Intranasally with 1 × 104 trophozoites under secobarbital anesthesia. There was a statistical difference in mortality between the transferred immunity group and the active immunization group. Statistical differences were not demonstrated in antibody titer between both groups. But L3T4+ T ce11/Ly2+T cell ratio was increased in the transferred Immunity group more than active immunization group of the offspring mice at the age of 5 weeks. There was no differences statistically in mortality between both groups. It was recognized that active immunization in offspring mice born to immune mother could modulate the immune status according to the time of Immunization.

Impact on Inflammation and Recovery of Skin Barrier by Nordihydroguaiaretic Acid as a Protease-Activated Receptor 2 Antagonist

  • Atopic dermatitis is a chronic, inflammatory disease of the skin with increased transepidermal water loss. Both an abnormal inflammatory response and a defective skin barrier are known to be involved in the pathogenesis of atopic dermatitis. Protease activated receptor 2 (PAR2) belongs to a family of G-protein coupled receptors and is activated by both trypsin and a specific agonist peptide, SLIGKV-$NH_2$. PAR2 is expressed in suprabasal layers of the epidermis and regulates inflammatory responses and barrier homeostasis. In this study, we show that nordihydroguaiaretic acid (NDGA) inhibits the PAR2-mediated signal pathway and plays a role in skin barrier recovery in atopic dermatitis. Specifically, NDGA reduces the mobilization of intracellular $Ca^{2+}$ in HaCaT keratinocytes by down-regulating inflammatory mediators, such as interleukin-8, thymus and activation-regulated chemokine and intercellular cell adhesion molecule-1 in HaCaT keratinocytes. Also, NDGA decreases the protein expression of involucrin, a differentiation maker of keratinocyte, in both HaCaT keratinocytes and normal human epidermal keratinocytes. We examined NDGA-recovered skin barrier in atopic dermatitis by using an oxazolone-induced atopic dermatitis model in hairless mice. Topical application of NDGA produced an increase in transepidermal water loss recovery and a decrease in serum IgE level, without weight loss. Accordingly, we suggest that NDGA acts as a PAR2 antagonist and may be a possible therapeutic agent for atopic dermatitis.

Analysis on the Dynamic Responses of Fishing Vessels in a Seaway (파랑중 어선의 동력학 해석)

  • Ships in a seaway will encounter dangerous situation, such as slamming, stranding, and capsizing. The number of capsizing is small, but the loss due to them is very large from the viewpoint of human life, property, and the environmental pollution. The number of capsizing of fishing vessels is about 62% of total number of capsizing, and the half of them is originated from the operational mistake in a seaway. So the dynamics and the capsizing phenomena are to be studied, and the guide for the safe operation of a fishing vessel in a seaway are to be specified. The hydrodynamic forces consist of radiation forces (which are due to the motion of a ship), Froude-Krylov forces (which is due to the incoming waves), and diffraction forces (which is due to the wave and ship interaction). These forces are calculated by well-known strip method. Using the calculated forces, the motion of a ship in a regular sea is obtained. In the real seaway, the waves are very irregular, therefore the statistical analysis is very helpful. In this paper, using the results of the motion in a regular seaway and the wave spectrum, the motion in a irregular seaway are obtained and analyzed.

Gene Expression Profiling of Genotoxicity Induced by MNNG in TK6 Cell

  • Genotoxic stress triggers a variety of biological responses including the transcriptional activation of genes regulating DNA repair, cell survival and cell death. In this study, we investigated to examine gene expression profiles and genotoxic response in TK6 cells treated with DNA damaging agents MNNG (N-methyl-N'-nitrosoguanidine) and hydrogen peroxide $(H_2O_2)$. We extracted total RNA in three independent experiments and hybridized cRNA probes with oligo DNA chip (Applied Biosystems Human Genome Survey Microarray). We analyzed raw signal data with R program and AVADIS software and identified a number of deregulated genes with more than 1.5 log-scale fold change and statistical significancy. We indentified 14 genes including G protein alpha 12 showing deregulation by MNNG. The deregulated genes by MNNG represent the biological pathway regarding MAP kinase signaling pathway. Hydrogen peroxide altered 188 genes including sulfiredoxins. These results show that MNNG and $H_2O_2$ have both uniquely regulated genes that provide the potential to serve as biomarkers of exposure to DNA damaging agents.

Physiological Responses and subjective sensations of the human wearing three different materials of Taekwondo wears (태권도복 소재별 인체생리반응과 주관적 감각에 관한 연구)

  • The purpose of this study was to evaluate thermal properties of Taekwondo wears with three different materials. As a begging step, the questionnaire survey about the actual condition of Taekwondo wears was conducted. With the results of the questionnaire, cotton/nylon(70/30) blended fabric(CN) that was newly woven with sweat absorbent finishing and cotton/spandex(95/5) blended fabric with flexibility property were developed. The same designed 3 Taekwondo wears with 3 different materials which were two different materials(CN and CS) and a current material(cotton/PET, CP) were made. Four young males volunteered for this study, they kicked and punched as Taekwondo action for 20 minutes. Mean skin temperature was the highest in CS(33.1${\pm}$0.8$^{\circ}C$) and the lowest in CP(32.7${\pm}$0.6$^{\circ}C$). Increasing degree of rectal temperature didn't show any significant difference. Clothing microclimate temperature on the thigh was higher in CS(32.8${\pm}$17.4$^{\circ}C$) than in CN(29.4${\pm}$1.1$^{\circ}C$) and CP(29.4${\pm}$1.0$^{\circ}C$). Clothing microclimate temperature on the back and humidity on the thigh didn't show any significant differences. Clothing microclimate humidity on the back was higher in CP(65${\pm}$20%RH) than in CS(61${\pm}$17%RH). Heart rate, total body weight loss, and local sweating were not significantly different by materials. Most subject responded more hot in CN than in others, but there were no significant differences at the subjective sensation of thermal humidity. They answered more comfortable in CN than in others. Tectile sensations were the best in CN and the worst in CS. From those results, first of all, it is necessary to be weighted on Taekwondo wears made of CN in the aspects of the dignity of military arts uniform. Secondly, CS was required to be lighted and enhanced for the subjective sensation. Third, CP weaved honey comb was asked more various design to eliminate sweat high competition power within the scope of the dignity of military arts uniform.

Classification of Negative Emotions based on Arousal Score and Physiological Signals using Neural Network (신경망을 이용한 다중 심리-생체 정보 기반의 부정 감성 분류)

  • The mechanism of emotion is complex and influenced by a variety of factors, so that it is crucial to analyze emotion in broad and diversified perspectives. In this study, we classified neutral and negative emotions(sadness, fear, surprise) using arousal evaluation, which is one of the psychological evaluation scales, as well as physiological signals. We have not only revealed the difference between physiological signals coupled to the emotions, but also assessed how accurate these emotions can be classified by our emotional recognizer based on neural network algorithm. A total of 146 participants(mean age $20.1{\pm}4.0$, male 41%) were emotionally stimulated while their physiological signals of the electrocardiogram, blood flow, and dermal activity were recorded. In addition, the participants evaluated their psychological states on the emotional rating scale in response to the emotional stimuli. Heart rate(HR), standard deviation(SDNN), blood flow(BVP), pulse wave transmission time(PTT), skin conduction level(SCL) and skin conduction response(SCR) were calculated before and after the emotional stimulation. As a result, the difference between physiological responses was verified corresponding to the emotions, and the highest emotion classification performance of 86.9% was obtained using the combined analysis of arousal and physiological features. This study suggests that negative emotion can be categorized by psychological and physiological evaluation along with the application of machine learning algorithm, which can contribute to the science and technology of detecting human emotion.

  • The Purpose of This study is amid competition, High-quality human resources in food service industry make a lot of efforts. Especially, Work and Life Balance(WLB) is a considerable factor when modern workers decide and maintain the workplace and also it could be supposed as one strategic method that corporations could attract and maintain talented people. This study built a exploratory model that there is causal relationship of WLB(work and life balance) to employee's organizational commitment. Additionally this study examine the Moderating Effect of perceived value of work between WLB(work and life balance) and organizational commitment. This study utilized total 217 responses from workers of food service industrial employees and as analysis methods, it used descriptive statistic analysis, correlation analysis, multiple regression analysis, and hierarchical analysis. Results of analysis are as follows. First this study found that the work-life balance could give significant effects to the organizational commitment. work-growth balance and work-leisure balance has positive significance effect with the organizational commitment but work-family balance was not statistically significant. Second, The significance of this study indicates that perceived value of work plays a role as moderating variable influencing the relationship between work and growth balance and employee organizational commitment. Lastly, this study suggested the implication of the result, the limitation and the direction of future study. The results of this study provide suggestion to the company.

Effects of locally-delivered minocycline hcl on controlled periodontal disease (Minocycline 국소 약물 방출 제제의 치주질환 치료 효과)

  • The purpose of this study was to evaluate the clinical and microbiological outcomes following the use of 30% minocycline-loaded polycaprolacton film and 2% minocycline-loaded gel that was applied locally into pockets combined with scaling and root planing. 25 human subjects who were non-pregnant, non-lactating, aged 20-50 and diagnosed as moderate to advanced adult periodontitis were enrolled. Subjects were excluded if they had a history of severe acute or chronic systemic disease, if they required antibiotic prophylaxis for dental treatment for any reason, or if they reported a history suggestive of hypersensitivity reactions to minocycline or tetracycline. 4quadrants that had several teeth with a 5-8mm probing pocket depth and radiographic evidence of alveolar bone loss for each patient were selected and divided into test sites and control sites according to the split-mouth design. Scaling and root planing was done for each site at baseline(0week). Test sites received the minocycline gel and strip and control sites had saline irrigation. The patients received both treatments simyltaneously. Subgingival irrigation of sterile saline was applied to the control sites for approximately 30 seconds. Minocycline strip and gel was applied into the periodontal pocket at 1, 2, 3, 4 weeks each after scaling and root planing in the test sites. The clinical and microbiological measurements were made at baseline and at the follow-up visits 6, 10, 14, 20 weeks. The results of this study were as follows; 1. The sulcular bleeding index, probing pocket depth and Periocheck test was significantly reduced and the relative proportions of spirochetes and motile rods were significantly reduced and the proportion of cocci was correspondingly increased, in locally delivered minocycline strip group compared to saline irrigation group. 2. In locally delivered minocycline gel group, The effect was the same with minocycline strip group as compared with saline irrigation therapy. 3. There was no significant differences between minocycline strip group and minocycline gelgroup. In conclusion, minocycline HCl local drug delivery combined with scaling and root planing may provide added improvement of clinical and microbiological responses by inhibiting bacterial recolonization of treated sites. It is suggested that the local administration of minocycline-HCl in the periodontal pocket is effective when combined with subgingival mechanical debridement.

The Study on Dietary Habits and Nutrient Intakes of School-Aged Children Based on Their Hours in Extracurricular Studies (과외 학습을 받는 학령기 아동의 수강 시간에 따른 식습관 및 영양 섭취 상태에 관한 연구)

  • The purpose of this study was to examine the dietary habits and nutrient intakes of school-aged children participating in extracurricular classes, to with regards to class schedule. Three-hundred and two students from Seoul were selected and classified into two groups. One group attended extracurricular classes less than 12.43 hours per week, which was determined as the average number of hours per week(AELT, n=147). The other group attended extracurricular classes more than 12.43 hours per week(AEMT, n=155). The average student age in both the AELT and AEMT groups was 11.9 years, respectively. In terms of height and weight, there were no significant differences between the two groups. AELT received significantly more hours of sleep than AEMT(p<0.01), and there were significantly more responses with regard to lack of sleep in the AEMT group than in the AELT group(p<0.05). In a survey on 17 clinical symptoms regarding health, the AEMT group had significantly higher response rates than the AELT group in the following sections: no appetite (p<0.001), dizziness(p<0.001), headache(p<0.01), tiredness(p<0.01), cold symptoms(p<0.05), and sore eyes(p<0.05). Meal time irregularity was significantly higher than AEMT than in AELT(p<0.05). Approximately 50% of the subjects were picky eaters, and 40% and 20% skipped breakfast or dinner, respectively. The average energy intakes for AELT and AEMT were 2122.2 kcal and 2061.5 kcal, respectively, and there was no significant difference between the two groups. AEMT consumed significantly lower quantities of protein, animal fat, vitamin $B_6$, calcium, and phosphorus(each p<0.05) as compared to the AELT group. Beverage intake was significantly higher in AEMT than AELT(p<0.05). The number of hours in extracurricular classes showed a significant negative correlation with the intakes of protein, animal protein, animal fat, vitamin $B_6$, folate, calcium, plant calcium and phosphorus(each p<0.05). The number of hours in extracurricular classes also showed a significant negative correlation with soy foods intake(p<0.05). These results suggest that continuous and systematic nutritional education should be performed in order to prevent and improve the unbalanced diets and problematic of dietary habits that may result in students due to attendance in extracurricular classes.

Whole-genome Transcriptional Responses to Hypoxia in Respiration-proficient and Respiration-deficient Yeasts: Implication of the Mitochondrial Respiratory Chain in Oxygen-regulated Gene Expression (저산소 환경에 대한 전체 유전자 발현 반응에서 미토콘드리아 호흡계의 연루)

  • Cells sense, respond, and adapt to a low oxygen environment called hypoxia, which is widely involved in a variety of human diseases. Adaptation to low oxygen concentrations includes gene expression changes by inducing hypoxic genes and reducing aerobic genes. Recently, the mitochondrial respiratory chain has been implicated in the control of these oxygen-regulated genes when cells experience hypoxia. In order to obtain an insight into an effect of the mitochondrial respiratory chain on cellular response to hyxpoxia, we here examined whole genome transcript signatures of respiration-proficient and respiration-deficient budding yeasts exposed to hypoxia using DNA microarrays. By comparing whole transcriptomes to hypoxia in respiration-proficient and respiration-deficient yeasts, we found that there are several classes of oxygen-regulated genes. Some of them require the mitochondrial respiratory chain for their expression under hypoxia while others do not. We found that the majority of hypoxic genes and aerobic genes need the mitochondrial respiratory chain for their expression under hypoxia. However, we also found that there are some hypoxic and aerobic genes whose expression under hypoxia is independent of the mitochondrial respiratory chain. These results indicate a key involvement of the mitochondrial respiratory chain in oxygen-regulated gene expression and multiple mechanisms for controlling oxygen-regulated gene expression. In addition, we provided gene ontology analyses and computational promoter analyses for hypoxic genes identified in the study. Together with differentially regulated genes under hypoxia, these post-analysis data will be useful resources for understanding the biology of response to hypoxia.
