Correlation of p53 Protein Overexpression, Gene Mutation with Prognosis in Resected Non-Small Cell Lung Cancer(NSCLC) Patients (비소세포폐암에서 p53유전자의 구조적 이상 및 단백질 발현이 예후에 미치는 영향)

  • Background : The p53 gene codes for a DNA-binding nuclear phosphoprotein that appears to inhibit the progression of cells from the G1 to the S phase of the cell cycle. Mutations of the p53 gene are common in a wide variety of human cancers, including lung cancer. In lung cancers, point mutations of the p53 gene have been found in all histological types including approximately 45% of resected NSCLC and even more frequently in SCLC specimens. Mutant forms of the p53 protein have transforming activity and interfere with the cell-cycle regulatory function of the wild-type protein. The majority of p53 gene mutations produce proteins with altered conformation and prolonged half life; these mutant proteins accumulate in the cell nucleus and can be detected by immunohistochemical staining. But protein overexpression has been reported in the absence of mutation. p53 protein overexpression or gene mutation is reported poor prognostic factor in breast cancer, but in lung cancer, its prognostic significance is controversial. Method : We investigated the p53 abnormalities by nucleotide sequencing, polymerase chain reaction-single strand conformation polymorphism(PCR-SSCP), and immunohistochemical staining. We correlated these results with each other and survival in 75 patients with NSCLC resected with curative intent. Overexpression of the p53 protein was studied immunohistochemically in archival paraffin- embedded tumor samples using the D07(Novocastra, U.K.) antibody. Overexpression of p53 protein was defined by the nuclear staining of greater than 25% immunopositive cells in tumors. Detection of p53 gene mutation was done by PCR-SSCP and nucleotide sequencing from the exon 5-9 of p53 gene. Result: 1) Of the 75 patients, 36%(27/75) showed p53 overexpression by immunohistochemical stain. There was no survival difference between positive and negative p53 immunostaining(overall median survival of 26 months, disease free median survival of 13 months in both groups). 2) By PCR-SSCP, 27.6%(16/58) of the patients showed mobility shift. There was no significant difference in survival according to mobility shift(overall median survival of 27 in patients without mobility shift vs 20 months in patients with mobility shift, disease free median survival of 8 months vs 10 months respectively). 3) Nucleotide sequence was analysed from 29 patients, and 34.5%(10/29) had mutant p53 sequence. Patients with the presence of gene mutations showed tendency to shortened survival compared with the patients with no mutation(overall median survival of 22 vs 27 months, disease free median survival of 10 vs 20 months), but there was no statistical significance. 4) The sensitivity and specificity of immunostain based on PCR-SSCP was 67.0%, 74.0%, and that of the PCR-SSCP based on the nucleotide sequencing was 91.8%, 96.2% respectively. The concordance rate between the immunostain and PCR-SSCP was 62.5%, and the rate between the PCR-SSCP and nucleotide sequencing was 95.3%. Conclusion : In terms of detection of p53 gene mutation, PCR-SSCP was superior to immunostaining. p53 gene abnormalities either overexpression or mutation were not a significant prognostic factor in NSCLC patients resected with curative intent. However, patients with the mutated p53 gene showed the trends of early relapse.

Downregulation of $\textrm{NF}_{-{\kappa}}\textrm{B}$ activation in human keratinocytes by melanogenic inhibitors

  • Exposure of skin cells, particularly keratinocytes to various nuclear factor-kappaB ($\textrm{NF}_{-{\kappa}}\textrm{B}$) activators [e.g. tumor necrosis factor-$\alpha$, interleukin-1, lipopolysaccharides, and ultraviolet light] leads to phosphorylation and degradation of the inhibitory protein, $\textrm{I}_{{\kappa}}\textrm{B}$. Liberated $\textrm{NF}_{-{\kappa}}\textrm{B}$ is translocated into the nucleus where it can change or alter expression of target genes, resulting in the secretion of extracellular signaling molecules including melanotrophic factors affecting melanocyte. In order to demonstrate the possible role of $\textrm{NF}_{-{\kappa}}\textrm{B}$ activation on the synthesis of melanotrophic factors from the keratinocytes, the activities of $\textrm{NF}_{-{\kappa}}\textrm{B}$ induced by melanogenic inhibitors (MIs) were determined in human HaCaT keratinocytes transfected with $\textrm{pNF}_{-{\kappa}}\textrm{B}$-SEAP-NPT plasmid. Transfectant cells released the secretory alkaline phosphatase (SEAP) as a transcription reporter in response to the $\textrm{NF}_{-{\kappa}}\textrm{B}$ activity and contain the neomycin phosphotransferase (NPT) gene for the dominant selection marker for geneticin resistance. MIs such as niacinamide, kojic acid, hydroquinone, resorcinol, arbutin, and glycolic acid were preincubated with transfectant HaCaT cells for 3 h and then ultraviolet B (UVB) was irradiated. $\textrm{NF}_{-{\kappa}}\textrm{B}$ activation was measured with the SEAP reporter gene assay using a fluorescence detection method. Of the Mis tested, kojic acid ($IC_{50}$/ = 60 $\mu$M) was found to be the most potent inhibitor of UVB-upregulating $\textrm{NF}_{-{\kappa}}\textrm{B}$ activation in transfectant HaCaT cells, which is followed by niacinamide ($IC_{50}$/= 540 $\mu$M). Pretreatment of the transfectant HaCaT cells with the Mis, especially kojic acid and niacinamide, effectively lowered $\textrm{NF}_{-{\kappa}}\textrm{B}$ binding measured by electrophoretic mobility shift assay. Furthermore, these two inhibitors remarkably reduced the secretion level of IL-6, one of melanotrophic factors, triggered by UV-radiation of the HaCaT cells. These observations suggest that Mis working at the in vivo level might act partially through the modulation of the synthesis of melanotrophic factors in keratinocyte.

Nano-safety Management and Exposure Assessment of Nanomaterials Producing Facilities (나노물질 생산시설의 환경노출 평가와 안전관리)

  • With the development of nanotechnology, nano-consumer products have been popularized. For the past 10 years, potential risk of nanomaterials to human and environment have been raised carefully. Especially, workers, who directly handle nanomaterials in laboratories and manfacturers, will lead to direct exposure of nanomaterials. Therefore, direct exposure assessment and field monitoring of nanomaterials are required to assess and manage the nanomaterial exposure to human and environment. In this work, two nano-manufacturing companies, which had plasma and sol-gel processes, were selected to analyze the main exposure source and process with in-situ SMPS (scanning mobility particle sizer) and ex-situ TEM (transmission electron microscopy). The results showed that the colloidal nanoparticle in liquid phase was easily evaporated and monitored by SMPS. Most serious thing is that the workers does not know about the potential risk of nanomaterials, and thus they are not taking proper protection activities, such as PPE (personal protective equipment). Therefore, exposure assessment for nanomaterial handling facilities should be additionally carried out, and nano-safety management protocols are also provided.

  • Embedding the compact wearable units to monitor the health status of a person has been analysed as a convenient solution for the home health care. This paper presents a method to detect fall from the other activities of daily living and also to classify those activities. This kind of ambulatory monitoring of the elderly and people with limited mobility can not only provide their general health status but also alarms whenever an emergency such as fall or gait has been occurred and a help is needed. A timely assistance in such a situation can reduce the loss of life. This work shows a detailed analysis of the data received from a chest worn sensor unit embedding a 3-axis accelerometer and depicts which features are important for the classification of human activities. How to arrange and reduce the features to a new feature set so that it can be classified using a simple classifier and also improving the classification resolution. Principal component analysis (PCA) has been used for modifying the feature set and afterwards for reducing the size of the same. Finally a Neural network classifier has been used to analyse the classification accuracies. The accuracy for detection of fall events was found to be 86%. The overall accuracy for the classification of Activities or daily living (ADL) and fall was around 94%.

A Study on Installation of Maritime Passenger Service Environment for the Disabled Persons (장애인을 위한 해상교통환경 구축 방안에 관한 연구)

  • Modern countries have almost ratified the UN Convention on the rights of persons with disabilities and are creating environments to prevent discrimination against these individuals. In 1996, the International Maritime Organization presented a recommendation for the design and operation of passenger ships to respond to elderly and disabled persons' needs. In Korea, an act on the promotion of transportation convenience for mobility disadvantaged persons has been implemented, and facilities for the disabled have been rapidly installed in ships, vehicles, aircraft, railways and passenger terminals. However, the maritime transport environment is less concerned about disabled persons than safety. In this study, basic research was conducted to improve the maritime transport environment for disabled people in Korea through a field survey and literature survey. As a result, it was found that relevant laws and regulations have not been properly applied to excursion ships and ferries as well as small-scale ports, which are responsible for a large part of coastal passenger transportation. In addition to low installation rates of facilities for the transportation of persons with disabilities in ships and terminals, loose equipment was also considered. Therefore, it is necessary to protect the human rights of persons with disabilities, which are universal values of humanity, by complementing laws and introducing improved facilities.

The Effect of Fermented Extracts of Korean Dendropanax Morbifera Levéille on Hair Growth (황칠나무 발효 추출물의 육모효과)

  • In previous studies, we confirmed the effective antimicrobial activity of fermented Dendropanax morbifera leaf/branch extracts with Lactobacillus plantarum ilchiwhangchil 1785 and Lactobacillus plantarum ilchiwhangchil 2020. In this study, we investigated the hair growth effect of D. morbifera leaf/branch extracts fermented with L. plantarum ilchiwhangchil 1785 and L. plantarum ilchiwhangchil 2020 on human hair dermal papilla cells. The growth rate of human hair dermal papilla cells treated with fermented extracts in the range of 1 to $10{\mu}g/ml$ significantly increased in a concentration-dependent manner, without increasing cell death. Double staining studies showed that the growth of cells treated with fermented D. morbifera leaf/branch extracts was more active than that of control cells. Moreover, the cells treated with the fermented D. morbifera leaf/branch extracts exhibited a 18.84% and 23.31% increase in cell mobility, respectively, as compared with that of the untreated cells. High-performance liquid chromatography (HPLC) was used to determine the active agents responsible for hair growth. The results showed that the content of ${\beta}$-sitosterol, which is known to affect hair growth, increased about 10 times in the fermentation process of D. morbifera leaf/branch extracts. Taken together, the findings confirm that fermented Dendropanax morbifera leaf/branch extracts promote hair growth.

Angiogenesis-inhibiting Effects of Prunus mume Butanol Fractions on Human Umbilical Vein Endothelial Cells (매실 부탄올 분획물에 의한 혈관 신생 억제 효과)

  • Prunus mume Sieb. et Zucc is distributed throughout Asia and has traditionally been used as medicine and food. P. mume is known to contain large amounts of various organic acids, minerals, and phenol components. To date, the trend of P. mume research has focused only on the effects of antioxidant, anticancer and antibacterial, with only a few studies have focused on angiogenesis. Angiogenesis is a common characteristic of metastatic cancer through which oxygen and nutrients are delivered to the cells and tissues. In the present study, angiogenesis-inhibiting activity was investigated by evaluating the total polyphenol and flavonoid contents of the P. mume butanol fraction (PBF) and their ability to inhibit VEGF-induced human umbilical vein endothelial cells (HUVECs) proliferation, migration, invasion, and capillary formation. The polyphenols (12.81 mg GAE/g) and flavonoids (28.4 mg QE/g) of the PBF exhibited high antioxidant activity. The results of this study showed that PBF did not inhibit the proliferation of HUVECs at concentrations of 25-200 ㎍/ml and did not exhibit toxicity to normal cells. However, PBF inhibited the VEGF-induced mobility, invasion, and capillary formation of HUVECs. These results show that PBF inhibits the angiogenesis of HUVECs induced by VEGF. Therefore, PBF could serve as a therapeutic agent for the inhibition of angiogenesis.

The Concept of Continuity in Confucianism through filial piety(孝) Ethics (효(孝) 윤리를 통해 본 유가(儒家)의 연속적 사유)

  • In addition to the emphasis on filial piety ethics in everyday life, filial piety obtained a further significance in Confucianism which had the strong sense of ancestor worship. This paper focuses on filial piety as a mechanism of continuity within Confucianism and points out that it owed its development to its connection to agricultural culture. The sedentary life with less mobility forged a relative intimacy among people and filial piety was the actual expression of that kind of intimate affection. Yet, filial piety in Confucianism created a unique culture in terms that it not only stipulated material and emotional support for parents but also expected one's piety to the further ancestors through a memorial service and made its connection to the infinite posterity through sons. From the perspective of Confucianism that established filial piety at the turning point from life to death, the self existing in present was not an isolated self anymore. Yet, one can see another characteristic of Confucianism from that filial piety, based on blood bonds, could move beyond paternalism to broaden itself. It could be expanded to the care for strangers. The aged experience and wisdom through agricultural life begot the insight that the nature made its infinite connections with everything through circulation. As a stone thrown in a pond would enlarge its boundary by drawing larger and larger concentric circles, this thought enabled people to enlarge their affection to their parents to universal humanity. In this enlarged network, though it was natural to make distinctions between the closer and the farther, Confucianism sought to overcome it by establishing oneself upright. Confucianism emphasized the moral cultivation with its filial piety concept that contained the diachronic thought penetrating life and death and the broadened perspective relating everything around. In Confucianism, filial piety provided an important medium in forming a moral subject that penetrated life and death and related self and others. Inherent in it is the Confucius thought of continuity that searched for a paragon of a moral human being regardless of time and space.

Analysis and Exposure Assessment of Factors That Affect the Concentration of Ambient PM2.5 in Seoul Based on Population Movement (인구 유동에 따른 서울시 대기 중 초미세먼지 농도 변화 요인 분석 및 노출평가)

  • Background: People's activities have been restricted due to the COVID-19 pandemic. These changes in activity patterns may lead to a decrease in fine particulate matter (PM2.5) concentrations. Additionally, the level of population exposure to PM2.5 may be changed. Objectives: This study aimed to analyze the impact of population movement and meteorological factors on the distribution of PM2.5 concentrations before and after the outbreak of COVID-19. Methods: The study area was Guro-gu in Seoul. The research period was selected as January to March 2020, a period of significant population movement changes caused by COVID-19. The evaluation of the dynamic population was conducted by calculating the absolute difference in population numbers between consecutive hours and comparing them to determine the daily average. Ambient PM2.5 concentrations were estimated for each grid using ordinary kriging in Python. For the population exposure assessment, the population-weighted average concentration was calculated by determining the indoor to outdoor population for each grid and applying the indoor to outdoor ratio to the ambient PM2.5 concentration. To assess the factors influencing changes in the ambient PM2.5 concentration, a statistical analysis was conducted, incorporating population mobility and meteorological factors. Results: Through statistical analysis, the correlation between ambient PM2.5 concentration and population movement was positive on both weekends and weekdays (r=0.71, r=0.266). The results confirmed that most of the relationships were positive, suggesting that a decrease in human activity can lead to a decrease in PM2.5 concentrations. In addition, when population-weighted concentration averages were calculated and the exposure level of the population group was compared before and after the COVID-19 outbreak, the proportion of people exceeding the air quality standard decreased by approximately 15.5%. Conclusions: Human activities can impact ambient concentrations of PM2.5, potentially altering the levels of PM2.5 exposure in the population.

Psychology of the self-nature in platform sutra-focused on dynamics in essential and inclusive self-nature (『단경』에 나타난 자성의 심리학 - 본래적 자성과 포괄적 자성의 역동성을 중심으로)

  • Zen Psychology refers to the psychology of Zen Buddhism as a field of Buddhist psychology. This paper examines the psychology of human beings, focusing on the "Platform Sutra" written by Huineng, the founder of Patriarchal Zen, and the southern Zen school. According to the classification of Buddhist psychology, Zen psychology is divided into theory of Zen mind, theory of Zend mind function, and Zen psychotherapy. In theory of Zen mind, Huineng explains the mind based on self-nature, and explains the theory of dharma, theory of humanity, theory of dependent origination, and worldview though self-nature. Every human mind has its own original character of self-nature. When one's own mind with self-nature is revealed, it is the basic mind. When it is covered, it is the mind of common people. Self-nature is characterized by dharma construction, incompetence, non-naming, cleanliness, equality, and self-Buddhahood. In the theory of dharma, dharma is non-dual and is equal. The theory of humanity is divided into two groups according to the sharpness and dullness or capacity of the human being. From theory of dependent origination, you can see the mobility of cover and revealment, enlightment and delusion, sacred and common. In the worldview, the world and phenomena are the functions of self-nature and one's own mind. The theory of Zen mind function deals with wisdom and defilement of common people. The function of wisdom is to know the whole and one at the same time, and is intangible. Defilement of common people is an inner common people that cover the functions of wisdom. In Zen psychotherapy, in order to eliminate the gap between human and dharma, seeing into self-nature is suggested and the goal. In teleology, the goal is to reveal and see the self-nature covered by defilements. As a methodology from cover to revealment, Huineng interprets the traditional methodology as a new interpretation from the viewpoint of the mind, and presents Sammu as its own methodology. Based on the absence of attachment, Huineng proposed a methodology of No-abiding, No-thought and No-form, to gather mind on reality and treat all things equally. Based on this, Huineng proposed a new methodology for dealing with karma through repentance and vows, and a methodology for edification. In view of the Four Noble Truth, it is the phenomenon that self-nature is covered, the cause of defilement and the mind, the aim of seeing the self-nature which is covered, and the way of revealing the covered self-nature is presented. In this sense, the Zen Psychology of Platform Sutra is psychology of self-nature.
