Bile juice prevents deposition of cholesterol in the blood vessel, digests fat, and absorbs fatty acid and vitamins, and it plays a great role on metabolism. Recently, emotional stimulus and mentally over-depression cause a person to come to illness, and westernization of way of life makes more patients with cholelithiasis, resulting into without bile secretion after cholelithotomy. Ageing, and gastrectomy and kidney transplantation are also the causes of more cholelithiasis occurrences. To solve these medical problems, we studied how Fel Sus Scrofa, which is not different from human bile juice, was used in the traditional Korean medicine. We I researched flavor, property, efficacy of Fel Sus Scrofa and how it was used by folk medicine, and we studied the usage examples of Fel Sus Scrofa in Sanghanlon and Dongeuibogam. The property of Sus Scrofa is bitter and cold. Its efficacy is to deposit glycogen. So it is known that it has been widely used, with many edible forms, without any humoral loss, for the inflammatory disease from various fever, problems of urine and feces, cutaneous disease, pulmonary disease, opthalmopathy, fever, thirst from diabetes, hepatocystic duct disorder. Fel Sus Scrofa can be used internally and externally to prevent humoral loss, and to control cutaneous disease among various pediatric disorder full of fever. And as we have in mind that it also can be used to treat patients with cholestasis after cholecystectomy, it is expected that post study of it must be done.
The purpose of this study is to isolate probiotic lactic acid bacteria (LAB) that produced bile salts hydrolase. One hundred twenty strains were initially isolated from human feces. Based on their resistance of acid, tolerances of bile salts, and inhibitory activity against Escherichia coli, five strains were selected. A strain producing highest activity of bile salts hydrolase was identified as Lactoacillus plantarum using API carbohydrate fermentation pattern and 16S rRNA sequences, and named CK102. Lactobacillus plantarum CK102 survived at a level of 1.36${\times}$10$\^$8/ CFU/$m\ell$ in pH 2 buffer for 6 h and showed exhibited excellent bile tolerance. When L plantarum CK102 was cultured with E. coli in MRS broth, no viable cells of E. coli was detected after 18 h fermentation. These results suggest that Lactobacillus plantarum CK 102 may be commercially used for the probiotic culture.

Several microorganisms from rat and human feces and lumen fluid of cows were screened for their ability to decolorize the synthetic dyes. Consequently, a novel dye-degrading strain AB&J was isolated. Taxonomic identification including 165 rDNA sequencing and phylogenetic analysis indicated that the isolate had 99.9% homology in its 165 rDNA base sequence with Clostridium perfringens. After 27 h Incubation with the strain, brilliant blue R, bromophenol blue, crystal violet, malachite green, methyl green, and methyl orange were decolorized by about 69.3%, 97.7%, 96.3%, 97.9%, 75.1%, and 97.2%, respectively. The triphenlmethane dye, bromophenol blue, was decolorized extensively by growing Clostridium perfringens AB&J cells in liquid cultures under anaerobic condition, although their growth was strongly inhibited in the initial stage of incubation. This group of dyes is toxic, depending on the concentration used. The dye was significantly decolorized at a relatively lower concentration of below 50 $\mu g \;ml^{-1}$, however, the growth of the cells was mostly suppressed at a dye concentration of 100 $\mu g \;ml^{-1}$. The decolorization activity in cell-free extracts was much higher in cytoplasm than in periplasm and cytoplasmic membrane. Therefore, the enzyme related uptake of bromophenol blue seemed to be localized in cytoplasm. The optimal pH and temperature of bromophenol blue uptake fur decolorization activities were 7.0 and 4$0^{\circ}C$, respectively.

Diphyllobothrium nihonkaiense has been reported in Korea as Diphyllobothrium latum because of their close morphologic resemblance. We have identified a human case of D. nihonkaiense infection using the mitochondrial cytochrome c oxidase subunit I (cox1) gene sequence analysis. On 18 February 2012, a patient who had consumed raw fish a month earlier visited our outpatient clinic with a long tapeworm parasite excreted in the feces. The body of the segmented worm was 2 m long and divided into the scolex (head) and proglottids. It was morphologically close to D. nihonkaiense and D. latum. The cox1 gene analysis showed 99.4% (340/342 bp) homology with D. nihonkaiense but only 91.8% (314/342 bp) homology with D. latum. The present study suggested that the Diphyllobothrium spp. infection in Korea should be analyzed with specific DNA sequence for an accurate species identification.
For lactic acid fermentation of chestnut broth,10 strains of bacteria were isolated from human feces and commercial yogurt,6 of which were identified to be Bifidobacterium and the rest isolated from Acidities of the chestnut broths fermented by these strains were lower than yogurt, but more than two times higher than yogurts made from seeds or vegetables including soy milk. To stimulate acidity of the fermented broths, addition of yeast extract and tryptone peptone were the most effective at the concentration of 0.2 and 0.4%, respectively, while glucose addition above 0.5% up to 8% did not increased the acid production except a few strains of Lactoba- Cillus. Among the tested fruits and vegetables, carrot juice supplementation was the most effective in acid produc- tion by most of the tested strains. Saccharification of chestnut broth by hydrolyzing process greatly increased the acid production at 25% of cooked chestnut. However, compared to the results from the 8% of unhydrolyzed chest- nut, the net increase in acid production by hydrolysis was not much stimulative.

This study was conducted to isolate antilisterial strains of the Bifidobacterium isolates from the infant feces. The bifidobacteria were isolated anaerobically on BL agar and screened for their inhibitory activity on the MRS-cysteine medium against three foodborne pathogens: Listeria monocytogenes, Bacillus cereus, and Staphylococcus aureus. Among the 52 bifidobacterial isolates, 5 strains(A24, Bl, B6, B10, and Bl2) were finally selected based on their stronger antilisterial activity against Listeria monocytogenes than other isolates tested. Morphologically, all the isolates were typically shown Y-and V-shaped under electron microscopic examination. Each isolate was primarily subjected to identification by a polymerase chain reaction(PCR) using a genus-specific primer designed for targeting the 16S rRNA gene sequence, and confirmed the primary identification data using an API-kit(Biomeriuex, France), commercially available product for identification based on biochemical and physiological traits. Of the isolates with antilisterial activity, strain A24 was finally confirmed as the Bifidobacterium longum A24.
To evaluate the probiotic potential of Bacillus polyfermenticus CJ6 isolated from meju, a Korean traditional soybean fermentation starter, its functionality and safety were investigated. B. polyfermenticus CJ6 was sensitive to all antibiotics listed by the European Food Safety Authority. The strain was also non-hemolytic, carried no emetic toxin or enterotoxin genes, and produced no enterotoxins. The resistance of B. polyfermenticus CJ6 vegetative cells and spores to simulated gastrointestinal conditions was high (60-100% survival rate). B. polyfermenticus CJ6 produced high amounts (0.36 g as a purified lyophilized form) of ${\gamma}$-polyglutamic acid (PGA). We speculate that the improved cell viability and the production of ${\gamma}$-PGA have a significant correlation. Adhesion of the strain to Caco-2 and HT-29 cells was weaker than that of the reference strain (Lb. rhamnosus GG), but it was comparable to or stronger than those of reported Bacillus spp. When B. polyfermenticus CJ6 spores were given orally to mice, the number of cells excreted in the feces was 4-fold higher than the original inocula. This suggests the inoculated spores propagated within the intestinal tract of the mice. This idea was confirmed by field emission scanning electron microscopy, which revealed directly that B. polyfermenticus CJ6 cells germinated and adhered within the gastrointestinal tract of mice. Taken together, these findings suggest that B. polyfermenticus CJ6 has probiotic potential for both human consumption and use in animal feeds.
Endogenous zinc is important for maintaining zinc homeostasis because the size of endogenous zinc pool is almost 3-4 times bigger than that of dietary zinc. The purpose of this study was to examine the phytate effect on the reabsorption of endogenous zinc and the additional Ca effect on the phytate effect. Rats were fed a casein-based diet with added sodium phytate containing either high(1.6%) or low(0.8%) Ca concentrations for 4 weeks to reduce the body zinc pool. After the depletion period, $^{65}$ Zn was given by intraperitoneal injection to label the endogenous zinc pool. Rats were then assigned into phytate or non-phytate group within the same Ca group. feces were collected for 2 weeks of the initial collection period and 1 week after dietary crossover. The ratios of excreted fecal $^{65}$ Zn radioactivity of phytate group non-phytate group were determined as a measure of the phytate effect on the endogenous zinc. Mean fecal $^{65}$ Zn radioactivity was higher in the phytate group than in the non-phytate group during the entire 3 weeks of the collection period in the low Ca group, and during the initial collection period in the high Ca group(p <0.0001). This study showed an adverse phytate effect on endogenous zinc at both high and low dietary Ca levels. Elevated dietary Ca levels showed a synergistic effect on the phytate effect on endogenous zinc(p <0.05). These results imply greater phytate effect on zinc homeostasis rather than on zinc bioavailability through complexing with the endogenous zinc which is larger portion than the dietary zinc on zinc homeostasis.
Surface sediments (0$\~$4 cm) were sampled at 40 stations from Ulsan Bay and its adjacent areas of Korea, to evaluate the contamination by sewage-derived organic matters using fecal sterols. Some sterols were quantified by gas chromatography coupled to mass spectrometry (GC/MS). Total concentrations of eight sterols (coprostanol, cholesterol, cholestanol, epicholestanol, epicoprostanol, ${\beta}-sitosterol,$ brassicasterol, and stigmasterol) in the sediments from Ulsan Bay and its vicinity varied from 929 to 23,444 ng/g dry weight. The most predominant sterols were cholesterol and coprostanol, accounting for $33\~72{\%}$ of total sterols. The concentration of coprostanol known as the indicator of human feces ranged from 141 to 8,257 ng/g dry weight. In particular, the coprostanol concentrations in the sediments from Jansaengpo Harbor and Taehwa River exceeded the value of 1,000 ng/g dry weight, suggesting that these areas could be considered as the hot-spot zones by municipal sewage contamination. Some molecular indices and multivariate data analysis were used to assess the origin of these sterols in the sediments. Results showed that the major routes of sewage contamination in Ulsan Bay and its vicinity were the input through Jansaengpo Harbor and Taehwa River.

Metabolomics is the study of changes in the metabolic status of an organism as a consequence of drug treatment, environmental influences, nutrition, lifestyle, genetic variations, toxic exposure, disease, stress, etc, through global or comprehensive identification and quantification of every single metabolite in a biological system. Since most chronic diseases have been demonstrated to be linked to nutrition, nutritional metabolomics has great potential for improving our understanding of the relationship between disease and nutritional status, nutrient, or diet intake by exploring the metabolic effects of a specific food challenge in a more global manner, and improving individual health. In particular, metabolite profiling of biofluids, such as blood, urine, or feces, together with multivariate statistical analysis provides an effective strategy for monitoring human metabolic responses to dietary interventions and lifestyle habits. Therefore, studies of nutritional metabolomics have recently been performed to investigate nutrition-related metabolic pathways and biomarkers, along with their interactions with several diseases, based on animal-, individual-, and population-based criteria with the goal of achieving personalized health care in the future. This article introduces analytical technologies and their application to determination of nutritional phenotypes and nutrition-related diseases in nutritional metabolomics.
