Breeding of Korean Raisin Tree (Hovenia dulcis var. koreana Nakai) for High Productivity in Korea

  • For the purpose of breeding a high-yield superior variety of Korean raisin tree (Hovenia dulcis var. koreana Nakai), whose value as an edible and medicinal resource is increasing, tree candidates for superior individuals were selected from its 11 habitats in Korea from 1996 to 1998. A clone bank preserve was created in 1998 with 70 clones proliferated by grafting; the fruition traits (e.g., the number of fructified laterals, the average number of bunches per fructified lateral, the average number of bunches per fruiting lateral, the fruitpetiole weight per individual, and the yield per individual) of 47 clones that had bloomed and borne fruit were investigated and analyzed in 2002; five upper-ranking clones whose yield per individual exhibited a 261% improvement against the total average were picked in 2005; and three clones, including 'Poong-Sung 1', that showed a difference in their fruit petiole ripening stage, were finally selected in 2007 as high-yield new cultivars of Korean raisin tree.

  • Fruits of Hovenia dulcis T. (Rhamnaceae) was called ‘jiguja’ in oriental medicine which has been used for diuresis, remove of hangover and leaves has been used for detoxified the alcohol. In order to investigate the efficacy of antioxidative activity from leaves of Hovenia dulcis, the activity guided fraction and isolation of physiologically active substance were performed. Its 20%, 40%, 60%, 80%, 100% MeOH, H2O, Acetone fractions were examined antioxidative activity by DPPH method and TBARS assay. (omitted)

  • 헛개나무(Hovenia dulcis var. koreana Nakai, Korea raisin tree)는 동아시아가 원산지이며, 식 약용 수종으로서 밀원수종으로도 가치가 증대되고 있는 수종이다. 본 연구는 헛개나무의 절접에 의한 접수의 채취시기와 연령 및 비닐하우스 설치에 따른 접목활착율의 효과를 조사하는데 목적이 있다. 헛개나무 신품종은 절접에 의한 접목 결과 클론간 유의적인 차이가 인정되지 않았다. 온대 북부 권역에서는 휴먼기인 춘분 전에 접수를 채취하는 것이 가장 적합하며, 비닐하우스를 설치하는 경우에는 86% 이상의 높은 접목 활착율을 얻을 수 있다. 접수 연령은 1년지가 가장 적합하며 접목 후 비닐터널을 설치하여 접목상을 관리하면 80% 이상의 높은 접목활착율을 얻을 수 있는 것으로 나타났다.

  • An efficient method for in vitro propagation of the medicinal plant Hovenia duleis, was established. Plantlets for micropropagation of H. dulcis were obtained from in vitro germinated seeds. The effectiveness of various levels of cytokinins (BAP, Kinetin and TDZ) on multiple shoot formation from stem nodes was tested. BAP (1.0 mg/L) treatment induced highest number of multiple shoots. The growth pattern of plantlet on various culture media was undertaken. The shoot elongation was optimal on 2MS basal medium without growth regulators. The in vitro rooting ability of H. dulcis shoots was examined with two-auxins IAA and IBA. The IAA (1.0 mg/L) treatments induced earliest rooting with maximum number of roots and root growth. Rooted shoots were transferred directly to small pots with artificial soil and such established plant exhibited a normal growth pattern similar to wild plantlet.

알코올 및 사염화탄소에 의해 유발된 급만성 간손상에 대한 지구자 열수추출물의 보호효과 (Hepatoprotective Effects of Hovenia dulcis Extract on Acute and Chronic Liver Injuries induced by Alcohol and Carbon Tetrachloride)

  • Objectives : The purpose of this study was to evaluate the hepatoprotective effect of Hovenia dulcis extract on acute and chronic liver injuries induced by alcohol and $CCl_4$ in mice and rats. Methods : In acute alcohol-induced liver injury, mice were administered Hovenia dulcis extracts (60 and 200 mg/kg) orally before and after alcohol administration. In chronic alcohol-induced liver injury, mice were administered alcohol containing liquid diet for 4 weeks. The mice were administered H. dulcis extracts (60 and 200 mg/kg) mixed with the liquid diet. In acute $CCl_4$-induced liver injury, rats received a single dose of $CCl_4$ (2 mL/kg in olive oil, intraperitoneally). Rats were administered H. dulcis extracts (30, 100 and 300 mg/kg) before and after $CCl_4$ administrations. After the ends of the administrations, the serum levels of AST and ALT were measured using chemical analyzer, and ${\gamma}$-GTP levels were measured using spectrophotometer. Results : In acute alcohol-induced liver injury, H. dulcis extracts treated group showed significant reduction in ALT levels compared to those of control group. In chronic alcohol-induced liver injury, it inhibited weight-loss compared to normal group and showed significant reduction in AST, ALT and ${\gamma}$-GTP levels compared to control group. In acute $CCl_4$-induced liver injury, it also showed significant reduction in AST, ALT levels compared to control group. Conclusions : The results show that H. dulcis extract has hepatoprotective effect in acute and chronic alcohol-induced liver injury and acute $CCl_4$-induced liver injury. These findings suggest that H. dulcis could be a potent hepatoprotective agent.

Dextrin과 β-cyclodextrin이 생체 내에서 헛개나무 추출물의 알코올성 손상으로부터 간보호에 미치는 영향 (Effects of Dextrin and β-cyclodextrin on Protective Effect of Hovenia dulcis Fruit Extract Against Alcohol-induced Liver Damage in vivo)

  • ${\beta}$-cyclodextrin has an ability to protect compounds from oxidative reaction by collecting them within its ring-like structure. So, In harsh condition ($40^{\circ}C$), marker compound, quercetin, was dramatically reduced in Hovenia dulcis fruit extract containing dextrin at 4 and 8 week compared to 0 week, but not that containing ${\beta}$-cyclodextrin. To evaluate the effects of dextrin and ${\beta}$-cyclodextrin on protective effect of H.dulcis fruit extract against alcohol-induced liver damage, The mice were orally injected alcohol, H. dulcis fruit extract/dextrin (HD) and H. dulcis fruit extract/${\beta}$-cyclodextrin (HCD), respectively, for 7 days. The mice orally administrated with alcohol significantly enhanced the serum concentration of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) and the activity of lactate dehydrogenase (LDH) in serum compared to the control group. HD and HCD significantly decreased the levels of serum ALT and AST and serum LDH activities compared to alcohol group. And also alcohol group significantly increased the level of total cholesterol compared to the control group, but HD and HCD significantly reduced it compared to the alcohol group. However, the levels of TG in blood were not significantly changed in all groups. The activities of alcohol dehydrogenase (ADH) were significantly increased in HD and HCD group although those of aldehyde dehydrogenase showed an increasing tendency. This data suggested that HD and HCD were able to induce alcohol degradation in the liver tissues. All together, the results showed that HCD demonstrated their ability to protect liver from alcohol-induced damage on equal terms with HD.

헛개나무 열매 분말을 첨가한 국수의 품질특성 (A Study on the Noodle Quality Made from Hovenia dulcis Composite Flour)

  • 숙취해소, 알콜성 간질환예방, 간의 해독작용 기능을 지닌 헛개나무 열매를 첨가한 국수를 제조한 후 품질특성을 살펴본 결과 다음과 같다. 유리아미노산 함량은 17종의 아미노산이 검출되었으며, leucine이 가장 높았다. 필수아미노산 함량은 헛개나무 열매 분말 첨가량이 증가할수록 높았으며 지미성분인 aspartic acid와 glutamic acid는 대조군보다 헛개나무 열매 분말 첨가군의 함량이 더 높았다. 조리국수의 중량(p < .001)과 부피 (p < .01), 수분흡수율(p < .001)은 헛개나무 열매 분말을 첨가할수록 낮았고, 탁도는 높았다(p < .001). 색도는 명도 L값은 대조군이 7.62로 가장 높고, 적색도 a값과 황색도 b값은 5$\%$ 첨가군이 가장 높았다. 기계적 검사의 texture는 경도(p < 0.05), 응집 성 (p < 0.05), 껌성과 파쇄성 (p < 0.001)은 대조군이 가장 높았고, 탄력성은 1$\%$ 첨가군이 85.03으로 가장 높았다. 전자현미경의 미세구조 관찰은 헛개나무 열매분말 첨가량이 증가할수록 전분입자들의 조직이 느슨해지고 입자들이 커지면서 형태가 부드러워지는 양상을 보였다. 관능검사의 색상과 허브향은 헛개나무 열매 분말의 함량이 높을수록 진하다고 평가하였으며 표면의 매끄러운 정도는 3$\%$ 첨가군이 가장 매끄럽다고 평가하였다(p < 0.01). 단맛과 구수한 맛은 헛개나무 열매 분말을 첨가할수록 높아 시료간에 유의적인 차이가 있었다(p < 0.05, p < 0.001). 맛의 기호도는 헛개나무 열매 분말 3$\%$ 첨가군이 가장 높고, 대조군이 가장 낮았다. 전반적인 기호도는 헛개나무 열매 분말 3$\%$ 첨가군에서 가장 좋았다(P < 0.01).

  • 지구자 물 추출물이 에탄올 중독된 흰쥐의 학습능력에 미치는 영향을 실험하기 위하여 정상군(normal), 25% 에탄올 투여군(control) 및 지구자 물 추출물과 에탄올을 함께 투여한 검액투여군(sample)으로 나누어 10일간 각 약물을 투여한 후 11일째 1일 3회 직선수로 실험을 시행하고, 다음날부터 3일간 1일 3회씩 실험한 T-미로실험을 시행한 결과, normal군은 직선 수로실험과 T-미로실험에서 학습에 의하여 목표점 도달 소요시간이 유의성 있게 단축되고, control군은 기억력쇠퇴로 인하여 학습능력이 현저히 감소하나, 지구자의 물 추출물을 계속적으로 투여한 sample군의 경우 에탄올 중독으로 저하된 학습 능력에 유의성 있는 회복을 나타냈다.

헛개(Hovenia dulcis Thunb)추출물 첨가 조미간장 개발을 위한 관능적 평가 (Sensory Evaluation of Hutgae(Hovenia dulcis Thunb) Extract for Soy Sauce Development)

  • The objective of this study was to investigate the sensory quality attributes for the development of soy sauce containing Hutgae(Hovenia dulcis Thunb). Aqueous extracts of Hutgae were prepared from different parts such as trunk, twig, and fruit. These extracts were used for determining the antioxidant effect by 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical scavenging activity and sensory evaluation. Hutgae twig and fruit extracts had a strong DPPH scavenging effect compared to Hutgae trunk extract. In sensory analysis, high intensities of roast smell, bitter taste, and astringent taste were observed in Hutgae twig extract, whereas those of sweet smell and sweet taste were predominated within Hutgae fruit extract. Hutgae trunk and fruit extracts obtained higher overall acceptability. Various seasoning items such as anchovy, dried-pollack, katsuobushi, shiitake, radish, and kelp were used to determine the suitable type of soy sauce containing Hutgae extracts regarding the different parts. Hutgae fruit and trunk extracts were evaluated for use as a good source of seasoned soy sauce, and the dried-pollack and radish among the seasoning items were well-matched with Hutgae extracts. From these results, soy sauce containing Hutgae trunk and fruit extracts added to dried-pollack soup may be used as a functional seasoning in order to remove hangovers.
