Effect of Houttuynia cordata on EtOH-induced Lipidperoxidation in Rat (알콜유도 지질과산화에 미치는 어성초 영향)

  • The antilipidperoxidative and hepatopreventive effects of Houttuynia cordata extract(HCE) were investigated at the level of liver homogenates and serum of SD rats intoxicated EtOH(3g/kg for 3days). We measured MDA(malondialdehyde) in the live. homogenate, AST(1-aspartate-2-oxoglutarate aminotransferase) and ALT( 1-alanine-2-oxoglutarate aminotransferse) in the serum. The analysis of the measurement indicated that HCE reduced MDA, ALT and AST significantly and their reduction was in relation to dose dependence. In rat liver homogenate intoxicated with EtOH, HCE treatment group inhibited lipidperoxidation by 50-75%. In rat serum intoxicated EtOH, HCE inhibited AST and ALT by 40-70%.

According to the records of the Chinese medicine, this plant has been used as folk medicine for analgesics, beriberi, edema, hepatitis, icterus, etc. TCDD (2, 3, 7, 8 -tetrachlorodibenzo-p-dioxin), one of the notorious toxic environmental pollutants, damages various organs including liver and is regarded as an endocrine disrupter. After 7 days from TCDD (1$\mu$g/kg) injection, HCT (200 mg/kg) was administered into rats intraperitoneally for 4 weeks. (omitted)

  • TCDD (2,3,7,8-tetrachlorodibenzo-p-dioxin), one of the notorious toxic environmental pollutants, damages various organs including liver and is regarded as an endocrine disrupter. To investigate the effects of Houttuynia cordata Thunb (HCT) on the biochemical parameters of function, liver and serum of TCDD-treated rats were used. After 7 days from TCDD (1 $\mu\textrm{g}$/kg) injection, HCT (200 mg/kg) was administered into rats intraperitoneally for 4 weeks. The lipidperoxide content was examined by measuring the level of total cholesterol, HDL-cholesterol, LDL-cholesterol, total lipid and triglyceride (TG) in serum, and malondialdehyde (MDA) in liver tissue of rats.   • The chemical composition of essential oil from the perennial herbaceous plants (Houttuynia cordata, Filipendula glaberrima, Peucedanum japonicum, and Ainsliaea acerifolia) was determined by GC/MS spectrometric analysis with the aid of NBS, Wiley Library and RI indice searches. The major constituents identified were $\alpha$-phellandrene (18.97%), $\gamma$-terpinene (12.32%), decanal (8.72%), 1-decanol (10.92%), decanoic acid (12.12%), and 2-undecanone (12.32%) from H. cordata, farnesol (2.83%), l-$\alpha$-terpineol (2.72%), benzenmethanol (2.03%), (Z)-3-hexen-1-ol (4.32%), and T-muurolol (2.07%) from F. glaberrima, $\alpha$-phellandrene (14.25%), endobornyl acetate (3.84%), heptanal (47.52%), octanal (2.65%), (E,E)-2,4-decadienal (2.75%), and octanoic acid (4.52%) from P. japonicum, and geyrene (9.74%), $\beta$-cubebene (11.15%), berkheyaradulen (22.32%), $\beta$-elemene (6.21%), (-)-A-selinene (4.85%), benzaldehyde (4.52%), and benzenacetaldehyde (3.40%) from A. acerifolia.

Simultaneous Determination of Seven Compounds in Houttuynia cordata using UPLC-PDA

  • A UPLC method for the simultaneous determination of seven compounds was established for the quality control in H. cordata. The UPLC was performed on a $C_{18}$ HSS T3 $2.1{\times}100$ mm, 1.8 ${\mu}m$ column during a 13 minute gradient elution of 0.2% aqueous acetic acid and acetonitrile with the flow rate of 0.2 mL/min at $30^{\circ}C$. The UPLC method was validated according to the ICH guideline of analytical procedures with respect to precision, accuracy, and linearity. The limit of determination and quantitation for the seven compounds were 0.01-0.09 and 0.03-0.28 ${\mu}g/mL$, respectively. The calibration curves of all seven compounds showed good linearity ($r^2$ > 0.999). The intra-day and inter-day the RSD values used to evaluate the precision of analysis were less than 0.9%. The recoveries of quantified compounds ranged from 98.63 to 103.85%. The developed UPLC method was found to be effective, convenient and sensitivity for quantitative analysis of seven compounds in H. cordata. This work could be provided a baseline source for quality control of H. cordata.

Effect of Houttuynia cordata Thunb and Herbs Mixture Extract on the Antioxidation in the LPS-induced Hepatotoxicity (LPS 유도 간독성에 대한 어성초 함유 혼합추출물의 항산화 효과)

  • The purpose of this study was to investigate the effects of Houttuynia cordata Thunb and Oriental Herb Mixture Extract (HCTM) through anti-oxidation against the hepatotoxicity-inducing lipopolysaccharide (LPS) in HCTM and LPS-treated rats. HCTM of 100 mg/kg concentration was intraperitoneally administered into rats at dose of 1.5 ml/kg for 20 days. On the day 21, 1.5 ml/kg of LPS was injected 5 hours before anesthetization. The activity of superoxide dismutase (SOD) was measured in mitochondrial fraction and malondialdehyde (MDA), catalase (CAT), glutathione peroxidase (GPx) were measured in liver homogenate. LPS-treatment markedly increased the levels of MDA and significantly decreased those of SOD, CAT and GPx. But HCTM pretreatment significantly increased those of SOD, CAT and GPx by 86.9%, 57.2% and 72.7% respectively. Moreover HCTM pretreatment decreased the levels of MDA. These results showed the HCTM had the effects against the hepatotoxicity-inducing LPS in the anti-oxidation. This suggested that HCTM could be used for functional beverage.

  • Lipopolysaccharide (LPS) induces the synthesis of several inflammatory cytokine, chemokine, NO and inflammation in the liver of rats. The purpose of this study was to investigate the preventive effects of Houttuynia cordata Thunb and Oriental herbs mixture extract (HCTM) against the hepatotoxicity-inducing LPS in the lipid metabolism. HCTM of 100 mg/kg concentration was intra-peritoneally administered into rats at dose of 1.5 ml/kg for 20 days. On the day 21, 1.5 ml/kg of LPS was injected 4 hours before anesthetization. The levels of glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), total lipid, triglyceride (TG) and HDL-cholesterol (HDL-C) were measured in serum. LPS-treatment markedly increased the levels of GOT, GPT, total lipid and TG, and decreased the level of HDL-C. However, HCTM pretreatment decreased the levels of GOT, GPT, total lipid and TG by 76.8%, 55.48%, 34.04% and 76.21% respectively, and increased the level of HDL-C by 97.84%. These results showed that HCTM had the protective effect against the hepatotoxicity-inducing LPS in the lipid metabolism, and it suggest that HCTM could be used for functional beverage.
