Effects of Several Pre-treatments on Seed Germination or Sophora japonica L. (회화나무 종자발아에 미치는 전처리의 효과)

  • This experiment was conducted to study the effects of sulphuric acid, cutting, cold stratification and hot water on the germination of Sophora japonica seeds, which have difficulty to germinate because they have hard and thick seed coats. The seeds were immersed in 30, 60 and 90% sulphuric acid for 30 minutes as seed scarifications, and cut 1/8, 2/8 and 3/8 of the opposite parts of radicles. As cold stratification the seeds were wrapped in wet towel, and then stored in plastic bags in a refrigerator $(4^{\circ}C)$ for 3, 5 and 10 days. The seeds were immersed in hot water $(90{\sim}95^{\circ}C)$ for 2,5 and 10 minutes. Pretreated seeds represented different germination properties, respectively. After sulphuric acid treatment, the seeds showed an increase in germination (G) and germination index (GI). Compared to the other treatments, 90% sulphuric acid showed the highest G (31.7%) and GI (6.2). The G and GI of cut seeds decreased with the increase of seed cutting lengths. And G and GI of cold stratificated seeds were not significantly different among the days treated (p=0.258). Two minutes treatment of hot water showed lower G and Gl than control, and the seeds were not germinated in 5 and 10 minutes treatments of hot water. At the result of relative growth rate and T/R ratio of seedlings from pretreated seeds, the seedlings from seeds in 90% sulphuric acid treatment represented the highest relative growth rate and T/R ratio.

  • Low Calorific Gas Turbine (LCGT) has been developed as a next generation power system using landfill gas (LFG) and biogas made from various organic wastes, food Waste, waste water and Livestock biogas. Low calorific fuel purification by pretreatment system and carbon dioxide fixation by green house system are very important design target for the optimum applications of LCGT. Main troubles of Low Calorific Gas Turbine system was derived from the impurities such as hydro sulfide, siloxane, water contained in biogas. Even if the quality of the bio fuel is not better than natural gas, LCGT may take low quality gas fuel and environmental friendly power system. The mechanical characterisitics of LCGT system is a high energy efficiency (>70%), wide range of output power (30 kW - 30 MW class) and very clean emission from power system (low NOx). A green house has been designed for four different carbon dioxide concentration from ambient air to 2000 ppm by utilizing the exhaust gas and hot water from LCGT system. LCGT is expected to contribute achieving the target of Renewable Portfolio Standards (RPS).

  • Wastewoods (logging residues) generated in Korea were used to make lignocellulosic fillers for papermaking. Lignocellulosic fillers could play great roles to increase retention efficiency and thus decrease turbidity of white water in papermaking process. In addition, lignocellulosic fillers could be used to improve physical properties of paper through their high affinity to cellulosic fibers, leading to the less use of chemical additives like retention aids. Wastewoods including Pinus densiflora and Quercus variabilis were chemically and mechanically treated for making fine particles passing through 100 mesh wire. The newly generated fillers showed larger particle size distribution than ground calcium carbonates but similar distribution to talc. In particular, pretreatment by hot water was more effective to generate smaller particle size than by alkali treatment. Lignocellulosic particles mixed with ground calcium carbonates under intense hybridizing condition greatly contributed to surface coverage of organic fillers in addition to filling to lumen and pits.

  • Mushroom is very important to rural dwellers due to the various economic, health and nutritional benefits that can be derived from its consumption, sales and utilization. Hence, this experiment was carried out with a view to evaluating the influence of locally available substrate i.e., sawdust of different wood species on the cultivation of Pleurotus ostreatus (oyster mushroom). Pleurotus ostreatus was grown on sawdust (indigenous and exotic species) using two pretreatment methods, which included hot water treatment to remove (a water-soluble extract) and non-extracted sawdusts (substrate). The result of this study showed that there was a significant difference in the effect of different wood species selected. Parkia biglobosa produced a better yield (71.06 g), followed by Anogeissus leiocarpus (53.17 g) and Gmelina arborea (51.39 g) in a non-treated unextracted states. In the treated samples, Parkia biglobosa has the highest yield (50.61 g) followed by Anogeissus leiocarpus (49.28 g) and Gmelina arborea (45.00 g). Thus, Parkia biglobosa and Anogeissus leiocarpus produced a higher yield of mushroom either in a treated or non-treated state and they could be recommended for growing oyster mushroom (Pleurotus ostreatus).

Effects of Peeling and Sample types on Drying Time and Paeoniflorin Contents of Peony Root (작약근의 박피와 시료형태가 건조시간 및 Paeoniflorin 함량에 미치는 영향)

  • Effect of pretreatment processing (peeling time, drying temperature, sample type) on the quality of hot air drying peony roots was investigated. Peony roots were peeled for 0, 10, 20, 30, 60, 90 and 120 minute in the peeling machine after water washing. Sample types were whole or cut (thickness of 3mm). Temperatures of hot air drying were 30, 40, 50 and $60^{\circ}C$ The level of water contents in dried sample was $14{\sim}15%$. Drying time was muth reduced as the peeling time, cutting sample types and higher temperature. Contents of paeoniflorin were maintained highest amount (3.48%) in the non-peeling cut sample dried at $30^{\circ}C$ and lowest amount (2.29%) in the 120min-peeling whole one dried at $60^{\circ}C$. Skin color of dried peony roots was heavily discolored in the sample of non-peeling whole type dried at $60^{\circ}C$ and the color was dark brown $({\Delta}E\;was\;46.1)$. When considering shortening of drying time, preservation of active compounds and maintenance of color, the best pretreatment method of drying in peony roots was non-peeling, cut type processing.

Antioxidative and Hepatoprotective Effects of Acer tegmentosum M. Extracts (산겨릅나무 추출물의 항산화 및 간 기능 보호효과)

  • This study was carried out to investigate antioxidative and hepatoprotective effects of Acer tegmentosum M. (ATM) extracts. Content of total polyphenol of EtOAc fraction was the highest amount among fractions. Electron donating abilities of all fractions were increased as concentrations of each fraction were increased. ATM BuOH fraction showed the highest SOD like activity at low concentration (<$250\;{\mu}g/mL$). To investigate the protective effect of ATM on hepatotoxicity, ATM BuOH fraction was administered to mice for 7 consecutive days, and then lipopolysaccharide (LPS) was injected at a dosage of 1 mg/kg. The LPS led to increase of aspartate aminotransferase (AST), alanine aminotransferase (ALT), and blood urea nitrogen (BUN). However, pretreatment with BuOH fraction prior to LPS injection significantly decreased plasma hepatospecific enzyme levels. Histological findings demonstrated that pretreatment with BuOH fraction resulted in an attenuation of LPS induced liver damages.

Heavy Metal Accumulation in Cell of Heavy Metal-Tolerant Bacteria by Some Physical and Chemical Treatments (물리화학적 전처리에 의한 중금속 내성세균의 균체내 중금속 축적 변화)

  • Heavy metal-tolerant microorganisms, such as Pseudomonas putida, P. aeruginosa, P. chlororaphis and P. stutzeri which possessed the ability to accumulate cadmium, lead, zinc and copper, respectively, were isolated from industrial wastewaters and mine wasewaters polluted with various heavy metals. Metal binding sites in the cells were investigated by extracting the components of the cells through pretreatments with hot water, acid, alkli, chloroform-methanol or chloroform-methanol/concentrated alkali. The heavy metal accumulation was drastically decreased by pretreatment with alkali or chloroform-methanol/concentrated alkali, but the heavy metal accumulation was not changed by pretreatment with chloroform-methanol. The amount of heavy metal accumulation was remarkably decreased by decreasing crude protein remaining in the cell. These results suggested that proteins of cell components played an important role on the heavy metal accumulation.

Anti-inflammatory Effects of Naetakbaekryeom-san (내탁백렴산 추출물의 항염증 효능 연구)

  • Objectives This study was conducted to confirm the anti-inflammatory effect of Naetakbaekryeom-san (NTB), and whether it could be another treatment for inflammatory diseases. Methods The NTB water extract was extracted with hot water at 100℃ for 2 hours, concentrated at 80℃ under reduced pressure, and used. After 2 hours of pretreatment with NTB and positive control Bay11-7082, nitric oxide (NO), inducible NO synthase (iNOS), interleukin (IL)-6, IL-1𝛽, tumor necrosis factor alpha (TNF-𝛼) were measured in RAW264.7 cells activated with lipopolysaccharides (LPS) 500 ng/mL. After 2 hours of pretreatment with NTB, the anti-inflammatory effect of NTB was evaluated by measuring nuclear factor kappa-light-chain-enhancer of activated B cells (NF-𝜅B) in RAW264.7 cells and 293T cells activated with phorbol 12-myristate 13-acetic acid (PMA) 30 ng/mL. Results In RAW264.7 cells activated with LPS, NTB at concentrations of 0.1, 0.3, and 1.0 mg/mL showed no cytotoxicity, significantly inhibited NO production and inhibition of iNOS expression. TNF-𝛼 cytokine levels was not regulated, but NTB at each concentration inhibited the production of IL-1𝛽 and IL-6, and the effect was higher than that of the positive control Bay11-7082 (20 𝜇M). In PMA-activated RAW264.7 cells and 293T cells, each concentration of NBT decreased the NF-𝜅B transcriptional activity, with the greatest decrease at 1 mg/mL. Conclusions These results demonstrated the anti-inflammatory effect of NTB water extracts, but further studies such as comparison of anti-inflammatory effects and antioxidant effects by NTB component, comparison of effects according to extraction solvents, and clinical studies are needed.

  • This study was carried out to investigate the physiological activityof the water extract from EA. The present study was done to investigate the effects of EA on cultured hepatocyte cell system and lipid peroxidation in $A{\beta}$ treatment conditions. Pretreatment of EA attenuated in cell cytotoxicity enhanced by increasing concentrations of $A{\beta}$. MDA level induced by $A{\beta}$ treatment was significantly increased and the level was slightly reduced by pretreatment of EA. The ability of EA to reduce cell death and MDA level induced by $A{\beta}$ suggest that EA may be a protective agent against free radical generating compounds such as $A{\beta}$. EA exhibited anti oxidative activity at all concentration tested.The extract was as good as antioxidative activity of the synthetic antioxidants, butylated hydroxy toluene and ascorbic acid. Furthermore, this was superior to that of natural antioxidant, a-tocopherol. In the presence of heavy metal ions ($Fe^{2+},{\;}Zn^{2+}$), EA showed strong antioxidative activity. The extracts showed about 3075% in the nitrite scavenging effect under pH 1.2 and $37^{\circ}C$ for 1 hr. There was significant difference among concentration of extracts.

Main problems and accidents of Low Calorific Gas Turbine system was derived from bio fuel condition such as hydro sulfide concentration, siloxane level, moisture concentration and so on. Even if the quality of the bio fuel is not better than natural gas, LCGT system has the various fuel range and environmental friendly power system. The mechanical characterisitics of LCGT system is a high total efficiency (>70%), wide range of output power (30kW - 30MW class) and very clean emmission from power system (low NOx). Also, we can use co-generation system. A green house designed for four different carbon dioxide concentration from ambient air to 2000 ppm by utilizing the exhaust gas and hot water from LCGT system. We look forward to contribute the policy for Renewable Portfolio Standards(RPS) by using LCGT power system.
