Characterization and Identification of Organic Selenium-enriched Bacteria Isolated from Rumen Fluid and Hot Spring Water

  • In the present study, the isolation of selenium (Se)-enriched bacteria from rumen fluid and hot spring water was carried out. Rumen fluid samples were taken from cannulated goats fed a basal diet and the water samples were collected from Selayang hot spring, Selangor-Malaysia. A total number of 140 Se-tolerant isolates were obtained aerobically using an Se-enriched medium and spread plate technique. All the isolates were initially screened for the ability to transform the Se-containing medium to a red-orange culture using a spectrophotometer. Twenty isolates of dark red-orange medium were selected for a screening of the highest Se-containing protein accumulating strains using the dialysis technique and icp.ms to measure the Se content. Four isolates, identified as Enterobacter cloacae (ADS1, ADS7, and ADS11), and Klebsiella pneumoniae (ADS2) from rumen fluid origin, as well as, one isolate from hot spring water (Stenotrophomonas maltophilia (ADS18)), were associated with the highest biomass organic Se-containing protein when grown in a medium enriched with $10{\mu}g/ml$ sodium selenite. In addition, around $50{\mu}g/100{\mu}g$ of the absorbed inorganic Se was accumulated as an organic form. Organic Se-containing protein in all the selected strains showed antioxidant properties in the range of 0.306 to 0.353 Trolox equivalent antioxidant capacity (TEAC) mg/ml. Therefore, these strains may offer a potential source of organic Se due to their Se-tolerant nature and higher biomass organic to inorganic Se ratio.

The Effect of Balneotherapy on Obesity Index and Body Composition on Obese Children (온천요법이 비만 아동의 비만지수와 체성분에 미치는 영향)

  • Objectives The purpose of this study is to evaluate the effect of hot spring bath in obese children. Methods The study was conducted with 20 obese children whose BMI were over 20 ($kg/m^2$). The hot spring bath program was performed in the hot-spring facilities with temperature of $32{\sim}36^{\circ}C$. The children bathed four times from June to July, 2012. Prior to their bath, their heights, weights, and body compositions were measured every time. They were also confirmed their obesity index and obesity degree during the program. Results After the bathing program, degree of obesity has changed, three obesity judgment index (BMI, RI and OI) of the children have decreased (BMI, RI, OI on the $1^{st}$ day: $25.99{\pm}3.47$, $177.63{\pm}17.43$, $37.74{\pm}13.42$; on the $4^{th}$ day: $25.06{\pm}3.08$, $168.4{\pm}14.8$, $30.60{\pm}12.12$), so have body fat mass and percent body fat (BFM, PBF on the $1^{st}$ day: $21.83{\pm}7.03$, $38.24{\pm}3.36$; on the $4^{th}$ day:$19.55{\pm}6.35$, $34.20{\pm}3.77$), but skeletal muscle mass has increased (SMM on the $1^{st}$ day: $18.37{\pm}4.24$ ; on the $4^{th}$ day: $19.80{\pm}4.44$). Conclusions This study shows that hot spring bath could be an effective way of managing and treating obesity.

An Analysis of the Relationships between Quantity and Drawdown at the Onyang Hot Spring Area (온양온천지구에서의 양수량-수위강하 관계 해석)

  • Most of hot springs don't spring out naturally but are pumped by submersible pumps in Korea. When pumped piezometric head in a well is dropped with proportion to quantity. This research investigates relationships between quantity and drawdown at the Onyang hot spring area. There are 38 wells at this area and the depths of wells range from 124 m to 303 m. Piezometric heads of 4 wells were observed for about 10 months. Fluctuation patterns of piezometric heads seem to be a sine curve with a you period by a high demand and a slack season. Drawdowns of fluctuations were about 98-139 m depth to water table when wells were pumped at $2,300-4,800m^3/day$. A equation was made through analyzing quantity and drawdown.

The Evaluation of Safe Yield Considered Interference Drawdown between Hot Spring Wells at Yecheon Hot Spring Area (예천온천지구내 온천공간 수위간섭을 고려한 적정양수량 평가)

  • There exist 5 wells drilled at the Yecheon hot spring area, the distance between the wells is about $159m{\sim}702.6m$. The safe yield of each well is controlled by interference drawdown between a pumping well and an observation well after pumping test. The well No. 2 and the well No. 5 are the closest, at the distance of 159 m; therefore interference drawdown between two wells was occurred considerably. The drawdown of the well No. 2 (an observation well) was 16.67 m; the drawdown of the well No. 5 (a pumping well) was 17.21 m. The degree of the interference is about 97% and the safe yield decreased from $750m^3/day\;to\;24m^3/day$. Significant interference between two wells is due to the fact that the aquifer has 1.5 dimensions.

A EPIDEMIOLOGICAL STUDY ON THE DENTAL CARIES OF THE DECLDUOUS TEETH OF KOREAN CHILDREN (지역별 유치우식경험도 조사연구)

  • The authors selected socio-geographically different sample areas. The dental caries on the deciduous teeth was observed and evaluated in each sample area. And then, dft rate, dft index, dt rate and ft rate were calculated and compared. The obtained results were as follows; 1. The dft rate of the deciduous teeth was 33.28% in the metropolitan area, 19.78% in city area, 18.21% in town area, 13.48% in field area, 15.26% in coastal area, 22.11% in mountain area, and 20.37% in hot spring area. 2. The dft index of the deciduous teeth was 4.51 in metropolitan area, 2.78 in city area, 2.59 in town area, 1.95 in field area, 2.30 in coastal area, 3.03 in mountain area, and 2.85 in hot spring area. 3. The dt rate of the deciduous teeth was 66.51% in metropolitan area, 95.38% in field area, 98.94% in hot spring area, and 100% in the other areas. 4. The ft rate of the deciduous teeth was 33.48% in metropolitan area, 4.61% in field area, 10.50% in hot spring area, and 0.00% in the other areas. 6. We could not find that the some dental cares for the dental caries on the deciduous teeth of Korean children were supplied in metropolitan area, and the track of dental cares was appreciated in the field area, but no trace was indentified in the coastal, mountain, city, and town areas.

Validation of Korean Water Quality Standards to Hot Springs for Agreement with Legionella-Incidence Risk (레지오넬라균 출현위해도에 대한 현행 온천수 수질기준의 적합성 분석)

  • Observed trends in climate change, globalization and an aging population have an effect on public health conditions in Korea, prompting a reevaluation of current environmental regulations. In this study, we evaluated the performance of the total coliform (TC) standard, which is the only microbiological standard in the current regulation regime for hot spring water, by estimating correlation with the presence/absence of Legionella, a non-fecal opportunistic pathogen with heat-tolerance. Microbiological data in 7 studies that surveyed Legionella in hot spring waters were subjected to meta-analyses with the odds ratio (OR) as the effect size. The presence/absence of Legionella was significantly correlated to TC levels [OR = 3.1(1.5–6.4, 95% CI), p = 0.002]. Due to there being no direct explanation as to the reason for the occurrence of TC, mesophilic fecal bacteria, being coupled with Legionella presence, the mechanism of the correlation between the two kinds of bacteria was further investigated. Legionella presence was more prevalent with a high heterotrophic plate count [HPC; 4.0(2.2–7.2); p < 0.001] and water temperature [4.3(1.4–13.6), p = 0.011] when the temperature range was <40℃. However, it was reverse-correlated with water temperature when the temperature was >50℃ [0.2(0.1–0.4), p < 0.001]. Therefore, bacterial standing crops in hot spring waters appear to be determined by water temperature in general, and this forces TC and Legionella levels be correlated. In accordance with this relationship, HPC rather than TC reflect the levels of non-fecal contamination better. Therefore, employing HPC as the sole microbiological standard, or adding HPC into the current standard of hot spring water assessment, is suggested as a proactive measure to prevent health issues arising from contamination.

Study of Volcanic Gases and Hot Spring Water to Evaluate the Volcanic Activity of Mt. Baekdu (백두산 화산활동 평가를 위한 화산가스 및 온천수에 대한 연구)

  • This study performed the analysis on the volcanic gases and hot spring waters from the Julong hot spring at Mt. Baekdu during the period from July 2015 to August 2016. Also, we confirmed the errors that $HCO_3{^-}$ concentrations of hot spring waters in the previous study (Lee et al. 2014) and tried to improve the problem. Dissolved $CO_2$ in hot spring waters was analyzed using gas chromatograph in Lee et al. (2014). Improving this, from 2015, we used TOC-IC to analysis dissolved $CO_2$. Also, we analyzed the $Na_2CO_3$ standard solutions of different concentrations using GC, and confirmed the correlation between the analytical concentrations and the real concentrations. However, because the analytical results of Julong hot spring water were in discord with the estimated values based on this correlation, we can't estimate the $HCO_3{^-}$ concentrations of 2014 samples. During the period of study, $CO_2/CH_4$ in volcanic gases are gradually decreased, and this can be interpreted in two different ways. The first interpretation is that the conditions inside the volcanic edifice are changing into more reduction conditions, and carbon in volcanic gases become more favorable to distribute into $CH_4$ or CO than $CO_2$. The second interpretation is that the interaction between volcanic gases and water becomes greater than past, and the concentrations of $CO_2$ which have much higher solubility in water decreased, relatively. In general, the effect of scrubbing of volcanic gas is strengthened during the quiet periods of volcanic activity rather than active periods. Meanwhile, the analysis of hot spring waters was done on the anion of acidic gases species, the major cation, and some trace elements (As, Cd, Re).

A Study on the Epilithic Algae in a stream drained from Hot Springs (온천 주변 소형하천에 서식하는 부착조류군집에 관한 연구)

  • Attached algal community and several physico-chemicai characters were investigated at Seokjong hot spring in Chollabukdo in February and in April, 1997. The results of physico-chemicai factors are as follows : water temperatures were varied from 6.5$^{\circ}$C to 32.0$^{\circ}$C; pH range was 6.9 to 7.7; The levels of electric conductivity were 105 $\mu$mhos/cm to 477 $\mu$mhos/cm; chemical oxygen demand(COD) range was 1.5 mg/l to 21.2 mg/l; ammonia concentration was 0.20 mg/l to 8.74 mg/l; nitrate concentrations were 0.36 mg/l to 2.43 mg/l; phosphate concentration was N.D. to 0.52 mg/l; sulfur concentration was 14.6 mg/l to 66.1 mg/l. Attacched algal species were identified. Totally, 63 taxa were classified and composed of 4 phylum, 4 class, 10 order, 17 family and 31 genera. The total biomass of attacched algae was $1.036 \times 10^{7}$ cells/l that composed of $5.39 \times 10^{6}$ cells/l of blue-green algae, $4.05 \times 10^{6}$ cells/l of diatom and $0.93 \times 10^{6}$ cells/l of green algae. The succession of dominant species was Synechocystis thermalis. This could be due to the thermal wastewaters. in biotic indices tests, the values of dominance index(DI) were 0.33 to 0.67 in winter, 0.18 to 0.68 in spring. The values of diversity index(H') were 1.44 to 2.69 in winter, 1.62 to 2.89 in spring. And the values of eveness index (J) were 0.31 to 0.61 in winter, 0.37 to 0.65 in spring.

A Study on the Improvement of Prior Environmental Review System(PERS) in a Administrative Plan & Program Including a Small-Scale Development - Focus on Hot Spring Hole Wildness - (소규모 개발이 수반되는 행정계획 사전환경성검토 발전방안에 관한 연구 - 온천공보호구역을 중심으로 -)

  • Korea has made the rapid economical development under the condition with the high population density and low available land. In this context, conservation of land environment, connected directly with living quality of human, has mostly ignored. Prior Environment Review System(PERS) is the one to investigate the locational validity, environmental impacts, harmony with surrounding environment, from the early stage of administrative plan or development project. PERS was amended to include SEA(Strategic Environment Assessment) system on June 1, 2006. But, several problems, including the loss of administrative ability, cost, and time, have been appeared, since the system is applied to all administrative plans without any distinction and plans or projects are delayed generally. In this study, the weaknesses of PERS were indicated through the comprehensive analysis including literature survey, study of legal and internet database, and the recently published PERS reports. Based on that, the improvement schemes were suggested to be harmonized with the nature of SEA. This study was mainly focused on a PERS for the designation of conservational area of hot-spring wells, conducted by authors.

Occurrences of Hot Spring and Potential for Epithermal Type Mineralization in Main Ethiopian Rift Valley (주 에티오피아 열곡대 내 온천수의 산출특성 및 천열수형 광상의 부존 잠재성)

  • The East African Rift System(EARS) is known to be hosted epithermal Au-Ag deposits, and the best-known example is Main Ethiopian Rift Valley(MER) related to Quaternary bimodal volcanism. Large horst-graben system during rifting provides open space for emplacement of bimodal magmas and flow channel of geothermal fluids. In recent, large hydrothermally altered zones(Shala, Langano, and Allalobeda) and hot spring related to deeply circulating geothermal water have been increasing their importance due to new discoveries in MER and Danakil depression. The hot springs in Shala and Allalobeda occur as boiling pool and geyser on the surface, whereas some areas didn't observe them due to decreasing ground water table. The host rocks are altered to quartz, kaolinite, illite, smectite, and chlorite due to interaction with rising geothermal water. The hot springs in MER are neutral to slightly alkaline pH(7.88~8.83) and mostly classified into $HCO_3{^-}$ type geothermal water. They are strongly depleted in Au, and Ag, but show a higher Se concentration of up to 26.7 ppm. In contrast, siliceous altered rocks around hot springs are strongly enriched in Pb(up to 33 ppm, Shala), Zn(up to 313 ppm, Shala), Cu(up to 53.1 ppm, Demaegona), and Mn(up to 0.18 wt%t, Shala). In conclusion, anomalous Se in hot spring water, Pb, Zn, Cu, and Mn in siliceous altered rocks, and new discoveries in MER have been increasing potential for epithermal gold mineralization.
