등검은말벌과 꿀벌의 장내 세균 군집 비교 (Intestine Bacterial Microbiota of Asian Hornet (Vespa Velutina Nigrithorax) and Honey Bee)

  • 한국에서 말벌은 양봉 산업에 큰 영향을 미치는 종이다. 특히 비교적 최근에 전국적으로 번식하는 말벌 종인 등검은말벌 (Vespa velutina nigrithorax)은 작은 체구에도 불구하고 높은 사냥 능력으로 꿀벌을 공격하여 양봉 업계에 큰 피해를 주고 있다. 그러나 이 새로운 침입 종에 대한 연구는 아직 시작단계에 있으며, 본 연구에서는 등검은말벌의 장내 박테리아 군집을 조사하고 이 결과를 꿀벌의 장내 박테리아 군집과 비교하여 등검은말벌의 방제를 위한 자료로 이용하고자 했다. 아시아 말벌과 꿀벌 장의 박테리아 게놈 DNA를 수집하여 16S rDNA를 증폭시켜 가변 부위인 V3, V4 염기서열을 판독 하였다. 계통별 분석결과 목 (order) 수준에서 Flavobacteriales는 등검은말벌에서 가장 우점인 박테리아였고, Aeromonadales와 Pseudomonadales가 그 뒤를 이었다. Flavobacteriaes는 꿀벌의 박테리아 군집과 비교했을 때 등검은말벌에서 증가하였으며, Aeromonadales와 Pseudomonadales는 우점종이지만 꿀벌에 비해 낮은 점유율을 보이는 등 두 개체 간의 차이를 확인할 수 있었다. 한편 이번 연구를 통해 이전에 동정되지 않은 등검은말벌 장 박테리아의 16S rDNA 염기 서열을 분석하였고, 이들 박테리아는 Thalassomonas, Caedobacter, Vampirovibrio, Alkaliphilus 및 Calothrix 속으로 분류되었다.

Cytotoxic activities of various fractions extracted from some pharmaceutical insect relatives

  • This research was performed to screen the cytotoxic activities of some pharmaceutical insect relatives. Cytotoxic activities of total extract and fractions of hexane, ethyl acetate, methanol, water and boiling water were extracted from four pharmaceutical insect relatives: the Chinese gall, the cicada slough, the hornet nest and the batryticated silkworm. These extracts were investigated against the cancer cell lines of L1210, P388 and SNU-1 in vitro tests. Results showed that, ED, , against the cancer cell lines of L1210, P388 and SNU-1 were 0.55, 0.50, and $0.83{\mu}g/ml$ in the ethyl acetate fraction from the Chinese gall; 1.07, 2.19, and $2.24{\mu}g/ml$in the ethyl acetate fraction, 1.51, 1.26, and $1.45{\mu}g/ml$ in the water fraction and 1.48, 2.29, and $1.29{\mu}g/ml$in the boiling water fraction from the cicada slough; 3.31, 2.00, and $6.61\mug/ml$ in the water fraction from the hornet nest and 13.80, 19.95, and $31.62{\mu}g/ml$in the hexane fraction and 33.88, 21.88, and $25.12{\mu}g/ml$in the ethyl acetate fraction from the batryticated silkworm, respectively. All of the fractions mentioned above showed high cytotoxic activities and could be suggested for further studies in vivo tests.

환경 독성 Peptide의 인지질과의 상호 작용 특성 분석 (Analysis of the Interactive Characteristic of Environmental Toxic Peptide and Phospholipid)

  • The interaction of mastoparan B, a cationic tetradecapeptide amide isolated from the hornet Vespa basalis, with phospholipid bilayers was studied with synthetic mastoparan B and its analogue with Ala instead of hydrophobic 12th amino acid residue in mastoparan B. MP-B and its derivative, [12-Ala]MP-B were synthesized by the solid-phase peptide synthesis method. MP-B and its analogue, [12-Ala]MP-B adopted an unordered structure in buffer solution. In the presence of neutral and acidic liposomes, the peptides took an $\alpha$-helical structure. The two peptides interacted with neutral and acidic lipid bilayers. These results indicated that the hydrophobic face in the amphipathic $\alpha$-helix of MP-B critically affected the biological activity and helical content.

  • The mode of action of mastoparan B, an antimicrobial cationic tetradecapeptide amide isolated from the hornet Vespa basalis, toward phospholipid bilayers was studied with synthetic mastoparan B and its analogs with individual Ala instead of hydrophobic amino acids (1-Ile, 3-Leu, 6-Leu, 7-Val, 9-Trp, 13-Val, 14-Leu) in mastoparan B. Mastoparan B and its analogs were synthesized by the solid-phase method. Circular dichroism spectra showed that mastoparan B and its analogs adopted an unordered structure in buffer solution. In the presence of neutral and acidic liposomes, most of the peptides took an α-helical structure. The calcein leakage experiment indicated that mastoparan B interacted strongly with neutral and acidic lipid bilayers than its analogs. Mastoparan B also showed a more or less highly antimicrobial activity and hemolytic activity for human erythrocytes than its analogs. These results indicate that the hydrophobic face in the amphipathic α-helix of mastoparan B critically affect biological activity and helical contents.

  • The interaction of mastoparan B, a tetradecapeptide toxin found in the hornet Vespa basalis, with phospholipid bilayers was investigated. Synthetic mastoparan B and its analogs, obtained by substituting one hydrophilic amino acid (2-Lys, 4-Lys, 5-Ser, 8-Ser, 11-Lys, or 12-Lys) in mastoparan B with Ala, were studied. Mastoparan B and its analogs were synthesized by the solid-phase method. As shown by circular dichroism spectra, mastoparan B and its analogs adopted an unordered structure in buffer solution. All peptides took an α-helical structure, and the α-helical content of its analogs increased in the presence of neutral and acidic liposomes as compared to that of mastoparan B. In the calcein leakage experiment, we observed that mastoparan B interacted more weakly with lipid bilayers in neutral and acidic media than its analogs. Mastoparan B also showed slightly lower antimicrobial activity and hemolytic activity towards human erythrocytes than its analogs. These results indicate that the greater hydrophobicity of the amphiphilic α-helix of mastoparan B by replacement with alamine residues results in the increased biological activity and helical content.

A study on the Beehive Door Opening and Closing System using a Hornet Sound Analysis

  • Recently, rapid climate change has had a significant impact on the ecosystem of honeybees. In addition, the problem of Vespa Hornets invasion of colonies has a fatal impact on the bee ecosystem, independent of climate change. Especially in late summer. This study relates to a method for preventing Vespa Hornets attack. In this study, we developed a Vespa Hornets sound detection device was developed by collecting and analyzing the sound of a Vespa Hornets and applying IoT technology. The developed device detects the sound of a Vespa Hornets when Vespa Hornets appears around the hive of the bees and sends a signal to automatically close the door of the beehive. The device that receives the signal drives the motor that controls the honeycomb door to close the beehive door. The Vespa Hornets sound detection device operates until no Vespa Hornets sound is detected. The system developed by us is expected to be installed in the beehives of actual beekeeping farms to dramatically reduce the damage caused by by Vespa Hornets.

  • Mastoparan B (MPB)는 벌독으로부터 정제된 양친매성 $\alpha$-helical 구조를 취하면서 14개의 아미노산 잔기로 구성된 염기성 항균성 펩타이드로서 여러가지 생물막과 작용한다. 본 연구에서는 우리 나라 연안의 적조 (HABs, harmful algal blooms)를 일으키는 4종의 적조생물 (Alexandrium tamarense, Chattonella marina, Cochlodinium polykrikoides 및 Gymnodinium catenatum)에 대한 MPB의 살조효과를 조사하였다. MPB의 4종의 적조생물에 대한 살조효과는 31.3 $\mu{g}$/mL부터 500 $\mu{g}$/mL에서 세포의 lysis 또는 ecdysis와 같은 형태로 현미경으로 관찰할 수 있었다. 또한 MPB는 C. marina 및 C. polykrikorides에 대해서 A. tamarense와 G. catenatum보다 더욱 강한 살조효과를 나타내었다. 이러한 MPB의 HABs에 대한 살조효과연구는 새로운 살조물질을 개발하기 위한 자료가 될 것으로 생각된다.

장수말벌 공격 조기 경보 시스템 프로토타입 설계 및 실내 시연 (Early Alert System of Vespa Attack to Honeybee Hive: Prototype Design and Testing in the Laboratory Condition)

  • 말벌류는 국내 양봉 산업에 가장 큰 피해를 입히는 생물군이다. 다양한 트랩 개발 및 포획 방법이 제시되고 있으나 아직까지 뚜렷한 해결책을 제시하지 못하고 있다. 본 연구팀은 장수말벌 특이 날갯짓 파동 프로파일 분석을 통하여, 장수말벌이 양봉장으로 공격해 들어올 때 이를 자동으로 탐지하고 그 정보를 사용자에게 전달할 수 있는 시스템을 개발하고 실험실 현장에서 검증하였다. 장수말벌 파동 프로파일은 명확하게 설정되었으며, 앉아서 날갯짓을 할 때와 실재 비행할 때의 파동 프로파일은 명확히 구분되었다. 이정보를 활용하여, 시스템을 구성하고 그 시스템은 장수말벌을 탐지하면 바로 트위터를 통하여 사용자에게 말벌 경보를 전송하는 방식으로 설계하였다. 시스템의 검증은 야외 온실 내 케이지에서 수행하였다. 수행결과 시스템은 성공적으로 말벌의 비행활동을 탐지하여 그 결과를 사용자에게 전달하였다. 비록 이 시스템은 실내 제한된 조건에서 검증되었지만, 향후 시스템의 개선 및 정확도가 향상된다면 양봉산업 현장에서 활용 가능성이 높다.

Social Wasp Diversity and Wasp Nest Removal Trends on Ulleungdo Island, South Korea

  • This study aimed to conduct a comprehensive survey of social wasp diversity and its impacts on remote Ulleungdo Island, South Korea. A total of 12 wasp traps were placed across the island, and specimens were collected monthly from May to October. Additional collection was conducted by thoroughly inspecting the areas surrounding the traps. The traps yielded a total of 5,068 individuals, representing two genera and two species. A total of 3,961 Vespa simillima simillima Smith individuals (78.2%) were collected, and 1,107 individuals (21.8%) were identified as Vespula flaviceps flaviceps Smith. Additionally, five individuals of Polistes snelleni de Saussure were collected through sweeping, bringing the total to 5,132 individuals of three genera and three species. It is worth noting that our findings indicate potential misidentifications in the species list of Vespidae found in the literature. Emergency services call centers reported the removal of 60 social wasp nests on Ulleungdo Island between 2018 and 2022, with the highest number of reports occurring from July to September, accounting for 87% of all cases. Dodong-ri and Jeodong-ri, with the highest populations, reported the most incidents, although fortunately, there have been no recent injuries. Past incidents involving the destruction of dried squid products by Vespula species have resulted in significant economic losses, underscoring the need for preventative measures. Furthermore, ongoing surveillance of hornet invasions is essential to safeguard the Jangwon bees being reared on Ulleungdo Island.
