A Clinical Study on 2 Cases of Premature Ovarian Failure by Serum Hormone Assay (호르몬 검사를 통하여 확인한 조기난소부전 환자 2례의 임상 경과 보고)

  • Objectives: The purpose of this study is to report the effects of oriental medicine on 2 patients with premature ovarian failure. Methods: The patients who had amenorrhea caused by premature ovarian failure were treated by oriental medicine such as herb medication, acupuncture and moxibustion. We evaluated the results of treatments by change of symptoms, serum hormone assay, basal body temperature and Kupperman's Index. Results: After oriental medicine, the patients could have their normal menstrual cycle. Also serum hormone assay, basal body temperature and Kupperman's Index were improved. Conclusions: This study shows that oriental medicine has effects on patients with premature ovarian failure. And serum hormone assay, basal body temperature and Kupperman's Index would be valid measurement to evaluate the effect of treatments for premature ovarian failure.

  • The potency of yeast-derived methionyl-free human growth hormone (rhGH), which was obtained by removal of the N-terminal Met from methionyl-hGH, was estimated by in vitro and in vivo assays. In radio-receptor assay where the binding affinity of growth hormone to the receptor was estimated, the recombinant hGH showed 2.9 international units (IU) per mg of specific activity. In contrast, pitUitary-derived human growth hormone had a slightly lower receptor binding activity (2.5 IU/mg) compared with recombinant growth hormone. For the in vivo assay, efficacy of rhGH was tested by use of hypophysectomized rats, in which pituitary organs were surgically removed, resulting in the termination of growth hormone secretion. The weight-increase in rats by the injection of rhGH was almost identical to the result obtained by the injection of the same amount of pituitary-derived (international standard) hGH. A comparision of the secondary structures of rhGH and rMet-hGH by circular dichroism spectrophotometer demonstrated that the removal of the methionyl residue from rMet-hGH did not exert any effect on the structure of the growth hormone. In conclusion, methionyl-free human growth hormone produced from yeast was highly potent in biological activity and maintained a legitimate three dimensional structure.

Aequorin Based Functional Assessment of the Melanin Concentrating Hormone Receptor by Intracellular Calcium Mobilization

  • Melanin concentrating hormone is a neuropeptide highly expressed in the brain that regulates several physiological functions mediated by receptors in the G-protein coupled receptor family, especially plays an important role in the complex regulation of energy balance and body weight mediated by the melanin concentrating hormone receptor subtype 1 (MCH1). Compelling pharmacological evidence implicating MCH1 signaling in the regulation of food intake and energy expenditure has generated a great deal of interest by pharmaceutical companies as MCH1 antagonists may have potential therapeutic benefit in the treatment of obesity and metabolic syndrome. Although fluorescence-based calcium mobilization assay platform has been one of the most widely accepted tools for receptor research and drug discovery, fluorescence interference and shallow assay window limit their application in high throughput screening and have led to a growing interest in alternative, luminescence-based technologies. Herein, a luminescence-based functional assay system for the MCH1 receptor was developed and validated with the mitochondrial targeted aequorin. Aequorin based functional assay system for MCH1 presented excellent Z' factor (0.8983) and high signal-to-noise ratio (141.9). The nonpeptide MCH1 receptor antagonist, SNAP 7941 and GSK 803430, exhibited $IC_{50}$ values of 0.62 ${\pm}$ 0.11 and 12.29 ${\pm}$ 2.31 nM with excellent correlation coefficient. These results suggest that the aequorin based assay system for MCH1 is a strong alternative to the traditional GPCR related tools such as radioligand binding experiments and fluorescence functional determinations for the compound screening and receptor research.

Two Cases of Oriental Medicine on Premature Ovarian Failure by Checking Ultrasonography and Serum Hormone Assay (초음파기기를 통한 장부형상 및 호르몬검사로 확인한 조기난소부전 환자 치험 2례)

  • Objectives: The purpose of these cases is to report the effects of oriental medicine on two patients with premature ovarian failure. Methods: Two patients who had amenorrhea and hot flashes caused by premature ovarian failure were treated by oriental medicine such as herb medication. We estimated the effects of treatments by follow-up measurements of serum hormone level, menstrual cycle and symptoms. Besides, follicle and endometrium were checked by ultrasonography for academic research.Results After treatments, their symptoms of premature ovarian failure were improved and the menstrual cycle resumed. And the level of serum Follicle Stimulating Hormone decreased and the level of serum E2 increased.Conclusions Two cases shows that oriental medicine has effects on patients with premature ovarian failure by follow-up measurements of serum hormone level, menstrual cycle and symptoms.

  • We have identified 88 interactor candidates for human growth hormone (GH) by the yeast two-hybrid assay. Among those, we focused our efforts on carboxypeptidase E (CPE), which has been thought to play a key role in sorting prohormones, such as pro-opiomelanocortin (POMC), to regulated secretory vesicles. We found that CPE colocalizes with and interacts with GH in AtT20 pituitary cells. Downregulation of CPE led to decreased levels of GH secretion, consistent with involvement of CPE in GH sorting/secretion. Our binding assay in vitro with bacterially expressed proteins suggested that GH directly interacts with CPE but in a manner different from POMC.

  • Purpose: The purpose of this study is to assess the effects of oriental medicine on secondary amenorrhea & hypomenorrhea objectively. Methods: By serum hormone assay and history interview, we classified the causes of each patient having secondary amenorrhea & hypomenorrhea. We also diagnosed and treated each patient according to them. And then we estimated the results of treatment by follow-up measurements of serum hormone level. Results: 1. We diagnosed case I as hypothalamic-pituitary disfunction, case II, III as PCOS and case IV as ovarian failure by classifying the causes from western medical scientific angle. 2. We also diagnosed and treated the cases from oriental medical scientific angle. 3. We confirmed the improvement of cases by follow-up measurements of serum hormone level. Conclusion: These results of serum hormone assay showed how medically effective oriental medical therapies of secondary amenorrhea & hypomenorrhea were.

  • Juvenile honnone binding protein (JHBP) was identified in the last instar larval hemolymph of Lymantria dispar using dextran coated charcoal (DCC) binding assay and gel filtration. The p1 value of JHBP was estimated to be 5.3. JHBP was partially pudfied by polyethylene glycol(PEG) precipitation, DEAE-cellulose ion-exchange chromatography and gel filtration, and was confirmed by DCC binding assay.

Preparation and Characterization of an Antibody Antagonist That Targets the Porcine Growth Hormone Receptor

  • A series of antagonists specifically targeting growth hormone receptors (GHR) in different species, such as humans, rats, bovines, and mice, have been designed; however, there are currently no antagonists that target the porcine growth hormone (GH). Therefore, in this study, we developed and characterized a porcine GHR (pGHR) antibody antagonist (denoted by AN98) via the hybridoma technique. The results from enzyme-linked immunosorbent assay, fluorescence activated cell sorter, indirect immunoinfluscent assay, and competitive receptor binding analysis showed that AN98 could specifically recognize pGHR, and further experiments indicated that AN98 could effectively inhibit pGH-induced signalling in CHO-pGHR cells and porcine hepatocytes. In addition, AN98 also inhibited GH-induced insulin-like growth factor-1 (IGF-1) secretion in porcine hepatocytes. In summary, these findings indicated that AN98, as a pGHR-specific antagonist, has potential applications in pGH-pGHR-related research on domestic pigs.

A Case Report of a Woman in Menopausal Transition by Using Hormone Assay (폐경 이행기 환자 1례의 호르몬 검사를 통한 임상 경과 보고)

  • Purpose: This study was done to report the progress of treatment with oriental medicine on a woman who complained of irregular menstruation in menopausal transition. Methods: The patient in this case was treated with Chokyungjongoktanggamibang, acupuncture and moxibustion. And to assess the effectiveness of this treatment, we evaluated the ovarian function by using hormone assay. Results: During treatment period the patient had three times of uterine bleeding, two of the three seemed like normal menstrual bleeding. And analyzing the hormone assay, estraiol level was increased and FSH, LH levels were decreased significantly. However a month after treatment end, estradiol level was decreased and FSH, LH levels were incresead as the levels in menopausal transition or menopause state. And the patient had no more uterine bleeding since last one according to follow up by phone-call. Conclusion: In this study, the progress of ovarian function was specified with hormonal changes after treatment of oriental medicine during menopausal transition. And further studies are needed to find the more appropriate clinical approach for women in menopausal transition.

  • Objectives: The purpose of this study is to asses the effects of oriental medicine on premature ovarian failure. Methods: The patient in this case was 30-year-old female. The chief complain is oligomenorrhea. By serum hormone assay and history interview, we diagnosed premature ovarian failure. She was treated by oriental treatments, such as herbal medicine and acupuncture. And then we estimated the results of treatment by follow-up measurements of serum hormone level. Results: After treatments, symptoms of premature ovarian failure was improved, level of serum gonadotropin decreased and level of serum $E_2$ increased. We confirmed the improvement of case by follow-up measurements of serum hormone level. Conclusions: This results of serum hormone assay showed how medically effective oriental treatments of premature ovarian failure.
