Improved Procedure for Purification of Clostridium botulinum type B Toxin (Clostridium botulinum Type B 독소의 정제방법에 관한 연구)

  • The neurotoxin of Clostridium botulinum type B was purified from a liquid culture. The purification steps consist of ammonium sulfate precipitation of whole culture, treatment of Polymin P(0.15%, v/v), gel filtration on Sephadex G-100 at pH5.6 and DEAE-Sephadex charomatography at pH8.0. The procedure recovered 17% of the toxin assayed in the starting culture. The toxin was homogeneous by sodium dodecyl sulfate(SDS)-polyacrylamide gel electrophoresis and had a molecular weight of 163, 000. Subunits of 106, 000 and 56, 000 molecular weight were found when purified toxin was treated with a disulfide-reducing agent and electro phoresed on SDS-polyacrylamide gels.

Preparation of porous polymers by environmentally friend process in supercritical carbon dioxide (초임계 이산화탄소를 이용하는 친환경 공정에 의한 다공성 고분자의 제조)

  • An experimental study on the preparation of monolithic porous polymers by environmentally friend process in supercritical carbon dioxide has been carried out. Polymerization mixture composed of a cross-linking monomer, initiator and functional co-polymer was charged in the reactor with sapphire window. After the system was purged with a flow of $CO_2$ for 15 min, the reactor was pressurized with liquid $CO_2$ up to 100 bars. The reactor was isolated from and placed back to the system via quick connector for shaking until the mixture had become fully homogeneous. The reactor was then heated and pressurized to the required reaction conditions and left overnight. After cooling and $CO_2$ evacuation, the polymer was removed from the reactor as dry, white, continuous monoliths. The effect of experimental conditions on the physical properties of porous polymer was systematically examined, and it was found that monomer content had a major effect on the physical properties of the polymers.

  • LAS-system ceramic powder, spodumene ($Li_2O{\cdot}Al_2O_3{\cdot}4SiO_2$), was successfully synthesized by a chemical solution technique employing PVA(polyvinyl alcohol) as an organic carrier. The PVA content affected the microstructure of porous precursor gels and the crystalline development. The optimum PVA content contributed to homogeneous distribution of metal ions in the precursor gel and it resulted in the synthesis of glass free $\beta$-spodumene powder having a specific surface area of $7.57\;m^2/g$. The agglomerated $\beta$-spodumene powders were also enough soft to grind to fine powders by a simple ball milling process. The microstructures of the densified powder compacts were strongly dependant on the minor phases of spodumene solid solution and amount of liquid phase, which were formed from the inhomogeneous precursors.

  • The semi-solid metal forming process has been applied to realize a near-net shape fabrication of a high speed tool steel. A complicatedly shaped part out of SKH51 was successfully manufactured by introducing pertinent materials, tooling and processing conditions. A SKH51 billet with globular grains was heated at temperatures between 1300 and $1350^{\circ}C$ using high frequency induction heater to get semi-solid microstructure before high rate injection of mushy metal into a die cavity for the forming process. It was necessary to control the preheating of dies between 300 and $400^{\circ}C$ to maintain the homogeneous microstructure during the semi-solid metal forming process. Significant defects such as pores, high fraction of liquid fraction and segregation could be removed from the part by using air vents.

Synthesis and Properties of ITO Nano Powders by Spray Drying Process (분무건조법에 의한 ITO 나노분말의 합성과 특성)

  • The Indium Tin Oxide(ITO) nano powders were prepared by spray drying and heat treatment process. The liquid solution dissolved Indium and Tin salts was first spray dried to prepare chemically homogeneous recursor powders at the optimum spray drying conditions. Subsequently, the precursor powders were subjected to eat treatment process. The nano size ITO powders was synthesized from the previous precursor powders and the npuities also were decreased with increasing heat treatment temperature. Furthermore, the lattice parameter of TO nano powders was increased by doping Tin into Indium with increasing heat treatment temperature. The par icle size of the resultant ITO powders was about 20∼50nm and chemical composition was composed of In:Sn =86:10 wt.% at 80$0^{\circ}C$.

Synthesis of Two-Component Titanate Powders Using Ethylene Glycol Solution (에틸렌글리콜 용액을 이용한 2성분계 Titanate 분말의 합성)

  • Pure and fine, two-component titanate powders (barium titanate, calcium titanate etc.) were synthesized by an ethylene glycol method. Titanium isopropoxide and other metal ionic salts were dissolved in liquid-type ethylene glycol without any precipitation. In non-aqueous system, the amount of ethylene glycol affected the solubility and homogeneity of metal cation sources in the solution. At the optimum amount of the polymer, the metal ions were dispersed effectively in solution and a homogeneous polymeric network was formed. Most of the synthesized powders had sub-micron or nano-size primary particles after calcination and the agglomerated calcined powders were easily ground by ball milling process. All synthesized titanate powders had stable crystallization behavior at low temperature and high specific surface area after ball milling. The crystallization behavior and the microstructures of the calcined powders were affected on the ethylene glycol content.

  • We have studied the viewing angle control of a twist nematic liquid crystal display (TN-LCD). The TN-LCD exhibits a bad image quality along vertical direction, due to characteristics such as low contrast ratio and grey scale inversion. The proposed new TN-LCD has the function of switching between the wide viewing mode and narrow viewing angle mode using two tilted LC layers at both sides of a TN-cell Tilt angles of the two LC layers, $14^{\circ}$ and $60^{\circ}$ were required in both wide viewing angle and narrow viewing angle modes, respectively.

  • In this study, using the governing equation for thermohydrodyamic lubrication involving the homogeneous mixture of incompressible fluid derived by based on the principle of continuum mechanics, it is discussed the effects of water within turbine oil on the performance of high speed journal bearing of a steam turbine shipping engine. The governing equation is the general equation being able to be applied on the mixture of Newtonian fluid and non-Newtonian fluid. Here, the fluid viscosity index, n of power-law non-Newtonian fluid is supposed to be 1 for the application of the journal bearing in a steam turbine shipping engine lubricated with the mixture of two Newtonian fluid, for example, water within turbine oil. The results related with the bearing performance are showed.

  • It is the objective of this study to analyze the effect of various process variables on the quality of extruded product and extrusion force for semi-solid extrusion of Al2024 with solid phase structure of globular type by the finite element method. Process variables are initial solid fraction, ram speed, semi-angle of die, and reduction in area. The results of experiment are compared with those of simulation in order to verify the usefulness of the developed finite element program. The flow and deformation of semi-solid alloy are analyzed by coupling by coupling the deformation of porous skeleton and the flow of liquid phase. It is also assumed that initial solid fraction is homogeneous.

Fabrication of Mn-Zn Ferrite by Hot Petroleum Drying Method (석유증발 건조방법에 의한 Mn-Zn Ferrite의 제조에 관한 연구)

  • This study attempted to characterize the powder and sintered specimen of Mn-Zn ferrite that was prepared by Hot Petroleum Drying Method. The results of the experiment were as follows: 1. The mixed sulfate powder prepared by Hot Petroleum Drying Method was homogeneous and very reactive. The ferrite formation of this powder occurred at lower temperature than the one prepared by Sulfate Dry Mixing Method. 2. The calcined oxide powder prepared by Hot Petroleum Drying Method was found to be agglomerated, and therefore it was very difficult to compact this powder. 3. The sintered density was 4.95g/㎤, 97% of the theoretical density, when the specimen was prepared by Hot Petroleum Drying method, calcined at 90$0^{\circ}C$ in air for 3h, sintered at 1,30$0^{\circ}C$ in air for 3h, and then cooled in nitrogen. 4. The discontinuous grain growth occurred at lower temperature in the specimen prepared by Hot Petroleum Dyring Method than in the one prepared by Sulfate Dry Mixing Method. The discontinuous grain growth was considered to be due to the presence of liquid formed by addition of CaO and $SiO_2$.
