Application of methylene blue color test for the detection of inherited susceptibility to hemolysis of Korean native cattle (한우(韓牛)의 선천성용혈감수성검사(先天性溶血感受性檢査)에 대한 methylene blue 청색소실시험법(靑色消失試驗法)의 적용(適用))

  • Blood samples were obtained from Korean native cattle and dairy cattle of Holstein species in the slaughter house and methylene blue color tests were performed for the detection of the inherited susceptibitity to hemolysis. Glucose-6-phosphate dehydrogenase activities expressed as the optical density obtained by methylene blue color test were the highest as 0.54 in male Korean cattle, 0.62 in female Korean cattle and 0.72 in dairy cattle of Holstein species. Percent hemolysis, packed cell volume and plasma protein contents were measured and compaired with relation to the results of methylene blue color test and no correlation were observed in each.

  • The objective of this study was to investigate the relationship between behavioral signs and duration of estrous in Hanwoo and Holstein cattle for the prediction of optical insemination. Fifty two of 58 and 71 of 89 in Hanwoo and Holstein cows or heifers showed estrous exhibition within 72 h after 2 days following $PGF_{2}\alpha$ administration, respectively. The number of mounting and standing heat were 54.2 and 57.6, 59.4 and 53.5, 42.0 and 30.8 and 16.2 and 10.7 times in Hanwoo cows, Hanwoo heifers, Holstein cows and Holstein heifers, respectively. Duration of mounting were no significantly difference for Hanwoo cows ($21.2\pm11.3$), Hanwoo heifers ($19.9\pm11.4$), Holstein cows ($8.7\pm4.4$) and Holstein heifers ($16.9\pm8.0$). Duration of standing heat were significantly (p<0.05) shorter for Holstein cows ($5.4\pm3.4$) than for Hanwoo cows ($17.1\pm9.6$), Hanwoo heifers ($16.5\pm6.3$) and Holstein heifers ($15.0\pm7.2$). Time until mounting after injection of $PGF_{2}\alpha$ were significantly (p<0.05) longer for Holstein cows ($56.3\pm11.45$) than for Hanwoo cows ($42.71\pm10.44$), Hanwoo heifers ($36.6\pm8.21$) and Holstein heifers ($40.70\pm6.15$). Time until standing heat after injection of $PGF_{2}\alpha$ were significantly (p<0.05) longer for Holstein cows ($61.6\pm8.92$) than for Hanwoo cows ($46.2\pm11.49$), Hanwoo heifers ($42.7\pm6.06$) and Holstein heifers ($44.1\pm6.72$). In the results of this study, duration of standing heat was the shortest in Holstein cows ($5.4\pm3.4$). The estimation of estrous with Holstein cows has more difficulty because to significantly shorter duration of standing heat than for Hanwoo cows, Hanwoo heifers and Holstein heifers. The standing heat can be a good predictor for time of ovulation but it is concluded that mounting behavior could be the best predictor for time of ovulation.

Seroepidemiological Studies on Bovine Respiratory Syncytial Virus of Cattle in Chonnam Province (전남지방의 소 합포체성 폐렴바이러스(Bovine Respiratory Syncytial Virus) 감염에 관한 혈청학적 연구)

  • Sera from 425 Korean native and 203 Holstein cattle were collected from October 1994 to September 1995 from dairy farms and slaughterhouses in Chonnam province to study the exposure rate to bovine respiratory syncytial virus(BRSV). Serum antibody titers against BRSV were measured by neutralization test, and results were as follows: Overall prevalence of seropositive cattle to BRSV were 74.5%, and the exposure rate to BRSV was higher among the Holstein (77.3%) than among the Korean native cattle(73.2%). The serum antibody titers against BRSV ranged from 1:2~$\geq$1:256 in both species. Among Korean native cattle, the most frequent serum antibody titer was 1:4 against BRSV(19.3%), while only 1.4% of seropositive cattle had serum titer of $\geq$1: 256. Among Holstein cattle, 22.7% of examined cattle contained serum titer of 1:8, while 1.5% of seropositive cattle showed $\geq$ 1:256. Antibody titers against BRSV were higher among males than females in both Holstein (82.1% vs. 73.1%) and Korean native (74.5% vs. 69.2%) cattle. Prevalence of seropositive cattle by age in both species were evenly distributed, although the highest number (76.9%) of seropositive were at the age of 3 in Korean native cattle, while 83.5% of seropositive Holstein cattle were of 2 years old. The lowest seropositive rate was observed in cattle of less than 1 year old(25.0%). Seasonal occurrunce of BRSV was the highest in spring season in both Holstein (86.6%) and Korean native (81.0%) cattle (P<0.05).

Random Regression Models Using Legendre Polynomials to Estimate Genetic Parameters for Test-day Milk Protein Yields in Iranian Holstein Dairy Cattle

  • The objective of this study was to estimate the genetic parameters of milk protein yields in Iranian Holstein dairy cattle. A total of 1,112,082 test-day milk protein yield records of 167,269 first lactation Holstein cows, calved from 1990 to 2010, were analyzed. Estimates of the variance components, heritability, and genetic correlations for milk protein yields were obtained using a random regression test-day model. Milking times, herd, age of recording, year, and month of recording were included as fixed effects in the model. Additive genetic and permanent environmental random effects for the lactation curve were taken into account by applying orthogonal Legendre polynomials of the fourth order in the model. The lowest and highest additive genetic variances were estimated at the beginning and end of lactation, respectively. Permanent environmental variance was higher at both extremes. Residual variance was lowest at the middle of the lactation and contrarily, heritability increased during this period. Maximum heritability was found during the 12th lactation stage ($0.213{\pm}0.007$). Genetic, permanent, and phenotypic correlations among test-days decreased as the interval between consecutive test-days increased. A relatively large data set was used in this study; therefore, the estimated (co)variance components for random regression coefficients could be used for national genetic evaluation of dairy cattle in Iran.

Detection of genome-wide structural variations in the Shanghai Holstein cattle population using next-generation sequencing

  • Objective: The Shanghai Holstein cattle breed is susceptible to severe mastitis and other diseases due to the hot weather and long-term humidity in Shanghai, which is the main distribution centre for providing Holstein semen to various farms throughout China. Our objective was to determine the genetic mechanisms influencing economically important traits, especially diseases that have huge impact on the yield and quality of milk as well as reproduction. Methods: In our study, we detected the structural variations of 1,092 Shanghai Holstein cows by using next-generation sequencing. We used the DELLY software to identify deletions and insertions, cn.MOPS to identify copy-number variants (CNVs). Furthermore, we annotated these structural variations using different bioinformatics tools, such as gene ontology, cattle quantitative trait locus (QTL) database and ingenuity pathway analysis (IPA). Results: The average number of high-quality reads was 3,046,279. After filtering, a total of 16,831 deletions, 12,735 insertions and 490 CNVs were identified. The annotation results showed that these mapped genes were significantly enriched for specific biological functions, such as disease and reproduction. In addition, the enrichment results based on the cattle QTL database showed that the number of variants related to milk and reproduction was higher than the number of variants related to other traits. IPA core analysis found that the structural variations were related to reproduction, lipid metabolism, and inflammation. According to the functional analysis, structural variations were important factors affecting the variation of different traits in Shanghai Holstein cattle. Our results provide meaningful information about structural variations, which may be useful in future assessments of the associations between variations and important phenotypes in Shanghai Holstein cattle. Conclusion: Structural variations identified in this study were extremely different from those of previous studies. Many structural variations were found to be associated with mastitis and reproductive system diseases; these results are in accordance with the characteristics of the environment that Shanghai Holstein cattle experience.

  • This study was carried cut to determine the immunological response of uterus-induced by Lipopolysaccharides (LPS) in Holstein cows. The LPS isolated from Bacteroids helcogenes and Fusobacterium varium was injected at the rate of 100 ${\mu}g$ with 30 ml of phospahte buffer saline(PBS) in each cow(n=5). Three cows were acted as control. There was no difference in total polymorphonuclear leukocytes(PMNL) concentration in uterine fluid between control and LPS groups at 24, 48 and 72 hrs after LPS treatment. There was significant difference in rate of PMNL between control and LPS groups at 24(41.7% vs 72.1%), 48(41.0% vs 81.6%) and 72 hrs(44.3% vs 79.0%) after LPS treatment. There was no difference in PMNL viability between control and LPS groups at 24, 48 and 72 hrs after LPS treatment. There was significant difference in rate of phagocytic PMNL between control and LPS groups at 48 hr after LPS treatment(1.1% vs 7.7%).

Relationship between Resumption of Postpartum Ovarian Cyclicity and Plasma Urea Nitrogen (PUN) in Holstein Cows (젖소의 분만 후 난소 주기의 재개와 혈장 요소태 질소(PUN)와의 관계)

  • The objective of this study was to investigate the resumption of postpartum ovarian cyclicity, and to determine the relationship between concentrations of plasma urea nitrogen (PUN) and resumption of ovarian cyclicity in Holstein cows. The cows were considered to have resumed ovarian cyclicity on the day of ovulation, if followed by regular ovarian cycles. 58.8 percentage of the cows (114/194) had normal resumption of ovarian cyclicity (resumption within 40days after calving), and 41.2% (80/194) had delayed resumption (resumption did not occur until> 40days after calving). Delayed resumption Type I (one or more ovarian cycles with luteal phase> 20days, i.e. prolonged luteal phase; 17.5%) and delayed resumption Type II (first ovulation did not occur until ${\ge}40days$ after calving, i.e. delayed first ovulation 22.7%) were the most common types of delayed resumption. 18 percentage of the cows (35/194) did not resume their ovarian cyclicity until 60days postpartum. Prolonged luteal phase and delayed first ovulation were two important ovarian dysfunctions that delayed postpartum resumption of cyclicity in dairy cows. Cows with PUN of <15, $15{\sim}19.9\;and\;{\ge}20mg/dl$ had the likelihood ratios of normal ovarian cyclicity of 0.9, 1.14 and 0.55, respectively. Thus, PUN concentration of $15{\sim}19.9mg/dl$ had a favorable association with postpartum resumption of cyclicity, whereas lower or higher PUN had a negative association with postpartum resumption of cyclicity.

Prevalence of Lymphocyte Nuclear Pockets in Holstein-Friesian Dairy Cattle Infected with Bovine Leukemia Virus in Korea

  • The integral relationship between the occurrence of lymphocyte nuclear pockets (LNPs) and BLV-infection was examined in Holstein-Friesian dairy cattle in Korea. Transmission electron microscope (TEM) was used to detect LNP in peripheral blood lymphocytes. Morphologically, the membranes of LNP were composed of two layers of double nuclear membrane. The full thickness of LNP membranes including inner and outer nuclear membrane was 60 to 70 nm. LNP prevalence was different according to the bovine leukemia virus (BLV) infection status; in BLV-seropositive cattle, LNP prevalence was 48.4% and in BLV-seronegative cattle prevalence was 5.9%. Moreover, even in seropositive animals, leukemic group was the highest at 70% positive among the groups, followed by suspect group (42.4%) and aleukemic group (23.1%). Consequently, the numbers of LNP were increased in proportion to increase of the numbers of leukocytes among BLV-seropositive cattle. The numbers of LNP per lymphocyte were increased in BLVseropositive cattle compared with seronegative cattle. The mean numbers of LNP per 100-lymphocytes were 0.35, 0.77, 1.64 and 4.7 in BLV-seronegative, BLV-seropositive aleukemic, suspect and leukemic groups, respectively. Thus, it is reasonable that LNP test can be used as the one of the diagnostic criteria of BLV infection.

Study on the Analysis of β-lactoglobulin and κ-casein Genotypes of Cattle using Polymerase Chain Reaction (PCR 기법을 이용한 축우의 β-lactoglobulin 및 κ-casein 유전자형 분석에 관한 연구)

  • This study was performed to offer the basic and applicable data for improvement of Korean cattle and dairy cattle, according to finding the genetic construction obtained from analysis of genetic polymorphisms of ${\beta}$-lactoglobulin and ${\kappa}$-casein loci related Korean cattle and Holstein cows using PCR-RFLP. Genomic DNA used in this study was prepared from the blood of 253 individuals of Korean cattle in Korean Native Cattle Improvement Center, NLCF, and the blood of 113 individuals of Holstein cows in National Livestock Research Institute. The results obtained are summarized as follows : 1. This study confirmed amplified products of 530bp and 262bp fragments obtained from the amplification of ${\beta}$-lactoglobulin and ${\kappa}$-casein loci in Korean cattle and Holstein breed by PCR. 2. The ${\beta}$-lactoglobulin AA genotype showed 153bp and 109bp fragments, and ${\beta}$-lactoglobulin AB genotype showed 153bp, 109bp, 79bp and 74bp fragments, and BB genotype showed 109bp, 79bp and 74bp fragments in amplified products of ${\beta}$-lactoglobulin loci with the restricted enzyme digestion of Hae III. 3. The ${\kappa}$-casein AA genotype showed a 530bp fragment, and ${\kappa}$-casein AB genotype showed 530bp, 344bp and 186bp fragments, and BB genotype showed 344bp and 186bp fragments in amplified products of ${\kappa}$-casein loci with the restricted enzyme digestion of Taq I. 4. On ${\beta}$-lactoglobulin genotypes and gene frequencies, Korean cattle were 6.72%, 26.09% and 67.19% for AA, AB and BB genotypes, and ${\beta}$-lactoglobulin A and B alleles were 0.197 and 0.803, and Holstein were 35.40%, 56.64% and 7.96% for AA, AB and BB genotypes, and ${\beta}$-lactoglobulin A and B alleles were 0.637 and 0.363, respectively. 5. On ${\kappa}$-casein genotypes and gene frequencies, Korean cattle were 46.25%, 39.13% and 14.62% for AA, AB and BB genotypes, and ${\kappa}$-casein A and B alleles were 0.658 and 0.342, and Holstein were 60.18% and 38.94% and 0.88% for AA, AB and BB genotypes, and ${\kappa}$-casein A and B alleles were 0.796 and 0.204, respectively. 6. As a consequence, the gene frequency was 0.197 and 0.803 for ${\beta}$-lactoglobulin A and B alleles, and 0.658 and 0.342 for ${\kappa}$-casein A and B alleles in Korea cattle, but was 0.637 and 0.363 for ${\beta}$-lactoglobulin A and B alleles, and 0.796 and 0.204 for ${\kappa}$-casein A and B alleles in Holstein, respectively.
