This study evaluated the quality characteristics of crust derived from dry-aged Holstein and Hanwoo loins and their effects on food as additives. With respect to physicochemical properties, we examined the proximate composition, pH value, salinity, color, water and fat absorption, emulsifying capacity, and swelling yield. The protein and ash contents in the Holstein crust were significantly higher than those in the Hanwoo crust (p < 0.0001). The fat content in the Hanwoo crust was significantly higher than that in the Holstein crust (p < 0.01). The salinity, lightness, and yellowness of the Hanwoo crust were significantly lower than those of the Holstein crust (p < 0.001). Furthermore, the pH value and emulsifying capacity of the Hanwoo crust were significantly higher than those of the Holstein crust (p < 0.001). The fat absorption of the Holstein crust was significantly higher than that of the Hanwoo crust (p < 0.001). The swelling yield of the Holstein crust was significantly higher than that of the Hanwoo crust at pH 3 and 4 (p < 0.001), whereas the swelling yield of the Hanwoo crust was significantly higher than that of the Holstein crust at pH 7 (p < 0.001). Principal component analysis of dry-aged Hanwoo, Holstein, and non-aged Holstein showed different flavor patterns for each sample. Finally, the results showed that the crusts derived from dry-aged Hanwoo and Holstein loins were suitable flavor enhancers.
This study was carried out to investigate the characterizations of $\kappa$ -CN genes of milk protein. In order to find out the characterizations of $\kappa$ -CN genes, the nucleotide sequences of 5'-flanking regions of $\kappa$ -CN genes were analyzed by PCR(polymerase chain reaction) technique with specific primers in order to investigate the characterizations of these promoters in Korean Native cattle and Holstein. PCR products obtained 349bp fragment and the nucleotide sequences of Korean Native cattle and Holstein of $\kappa$ -CN gene S'-flanking region was analyzed to -82bp from -431bp. On the comparison of each breed, Holstein substituted T$\rightarrow$C at -386bp, and -241bp(T) and -192bp(C) existed, but Korean Native cattle was deleted. Also, Korean Native cattle was existed T at -183bp but Holstein was not. The homology analysis of between Korean Native cattle and Holstein was showed 98.9% homology for $\kappa$ -CN promoter.
This study was conducted to compare the meat qualities of loin (m. longissimus dorsi) and top round (m. semimembranosus) from Hanwoo and Holstein veal. Ten Hanwoo and Holstein calves were randomly selected from a local cattle farm and raised. They were slaughtered when they were 8 mon old and weighed. Weight and percentage in primal cuts and slaughter performance of Hanwoo and Holstein veal calves are obtained. Immediately after weighting, slices of loin and top round muscles were sampled. After vacuum packaging, the samples were subjected to proximate composition, physicochemical and microbiological analyses. Dressing weight and percentage were heavier and greater (p<0.05) in the Holstein than in the Hanwoo. Water contents of the top round muscle was higher in the Holstein than in the Hanwoo (p<0.05). Water-holding capacity, protein content and CIE L* (lightness) of both muscles were higher in the Holstein than in the Hanwoo veal, whereas fat content, pH, cooking loss, a* (redness), and b* (yellowness) were higher in the Hanwoo than in the Holstein veal (p<0.05). Thiobarbituric acid and volatile basic nitrogen values of both the muscles were lower in the Hanwoo than in the Holstein veal during the first 10 d of storage (p<0.05).
To investigate serum neutralizing antibodies against Bovine ephemeral fever (BEF) virus, Akabane virus and Ibaraki virus in southern area of Chungnam province, Holstein sera were collected from April-May(269 heads /37 farms) and October-November (226 heads /35 farms), 1993. The results were summarized as follows ; 1. Bovine ephemeral fever.-antibody positive rates to BEF virus were 46.1% (124 heads /269 Holstein) in April-May and 53.9%(122 heads /226 Holstein) in October-November. 2. Akabane disease.-antibody positive rates to Akabne virus were 34.2%(92 heads /269 holstein) in April-May and 51.3%(116 heads /226 Holstein) in October-Novermber. 3. Ibaraki disease.-antibody positive rates to Ibaraki virus were 57.6%(155 heads /269 Holstein) in April -May and 38.5%(87 heads /226 Holstein) in October-November.
The objective of this study was to investigate the relationship between behavioral signs and duration of estrous in Hanwoo and Holstein cattle for the prediction of optical insemination. Fifty two of 58 and 71 of 89 in Hanwoo and Holstein cows or heifers showed estrous exhibition within 72 h after 2 days following $PGF_{2}\alpha$ administration, respectively. The number of mounting and standing heat were 54.2 and 57.6, 59.4 and 53.5, 42.0 and 30.8 and 16.2 and 10.7 times in Hanwoo cows, Hanwoo heifers, Holstein cows and Holstein heifers, respectively. Duration of mounting were no significantly difference for Hanwoo cows ($21.2\pm11.3$), Hanwoo heifers ($19.9\pm11.4$), Holstein cows ($8.7\pm4.4$) and Holstein heifers ($16.9\pm8.0$). Duration of standing heat were significantly (p<0.05) shorter for Holstein cows ($5.4\pm3.4$) than for Hanwoo cows ($17.1\pm9.6$), Hanwoo heifers ($16.5\pm6.3$) and Holstein heifers ($15.0\pm7.2$). Time until mounting after injection of $PGF_{2}\alpha$ were significantly (p<0.05) longer for Holstein cows ($56.3\pm11.45$) than for Hanwoo cows ($42.71\pm10.44$), Hanwoo heifers ($36.6\pm8.21$) and Holstein heifers ($40.70\pm6.15$). Time until standing heat after injection of $PGF_{2}\alpha$ were significantly (p<0.05) longer for Holstein cows ($61.6\pm8.92$) than for Hanwoo cows ($46.2\pm11.49$), Hanwoo heifers ($42.7\pm6.06$) and Holstein heifers ($44.1\pm6.72$). In the results of this study, duration of standing heat was the shortest in Holstein cows ($5.4\pm3.4$). The estimation of estrous with Holstein cows has more difficulty because to significantly shorter duration of standing heat than for Hanwoo cows, Hanwoo heifers and Holstein heifers. The standing heat can be a good predictor for time of ovulation but it is concluded that mounting behavior could be the best predictor for time of ovulation.
A ten-year-old holstein cow was presented because of prolapse of the third eyelid and apparent hyperplasia of the right lower eyelid. Historical findings included increased appetite as well as polyuria and polydipsia for about two weeks. The most remarkable fadings on physical examination were a large periocular proliferative tissue and bleeding. Surgical incision was used both as a biopsy and therapeutic tool in holstein cow with mass. Histopathological examination of the mass revealed squamous cell carcinoma. Blood and milk tests of patient with squamous cell carcinomatosis were peformed, but normal values.
Present study was conducted to investigate the optimal feeding levels for producing the high quality meat on the basis of the information deriving from the comparison of the growth performance and carcass characteristics among breeds(Holstein vs F1, Holstein♀×Hanwoo♂), sex(steer vs heifer) and interaction between breed and sex. Thirty two animals on 4 treatments(i.e. eight head each) were used for 540 days from seven to 24 months of age. The results obtained are summarized as follows; the range of average daily gains was 0.733 to 1.018, 0.994 to 1.255, 0.947 to 1.259 and 0.736 to 0.824kg for the growing, the early-fattening, the mid- fattening and the finishing periods, respectively. The range of average daily gains for the entire period was 0.882 to 1.061kg. The gains were higher for Holstein(7.3%) and the steers(10.5%) than F1 and the heifers, respectively. Concentrates and total digestible nutrients intakes per kg gain were higher for Holstein and the heifers than F1 and the steers, respectively. These findings may indicate that feed utilization is higher for Holstein than F1, and higher for the steers than the heifers. In carcass characteristics, back fat thickness was thicker for Holstein than F1, and rib-eye area was smaller for Holstein than F1. The rib-eye area per kg carcass weight was larger for F1 and the heifers than Holstein and the steers, respectively. Meat color was better for Holstein than F1, but the sex distinction did not show any differences. In physicochemical properties of longissimus dorsi, shear force, cooking loss, water holding capacity and the panel test scores of juiciness, tenderness and flavor for F1 and the heifers were better than those for Holstein and the steers, respectively. According to the above results, we may conclude that F1 and heifers rather than Holstein and steers are recommended for high quality meat production. In steers and heifers of Holstein and F1, the optimal feeding levels may be 1.9% of apparent body weight for concentrates and 25% of concentrates intake for rice straw.
The need for refining dietary nutrient levels and limited information regarding Holstein steer and phosphorus supplementation led to the objective of examining the response of removing supplemental dietary phosphorus from a corn-based finishing diet offered to yearling Holstein steers. Two groups of yearling Holstein steers were utilized to study responses of increasing dietary phosphorus level during the finishing period. In Exp. 1, 96 Holstein steers (419 kg) were blocked into four weight groups. Dietary treatments included no additional phosphorus (NDC) or the inclusion of dicalcium phosphate (DCP) to achieve 0.30% phosphorus (P) in the complete diet. Daily gain, DMI and carcass traits were not different (p>0.05). Overall gain efficiency was slightly lower for NDC (p<0.05). Exp. 2 consisted of 78 Holstein steers (491 kg) blocked into two weight groups. Steers were harvested on d 84 and 112 d on test with carcass data collected following a 48-h chill. No differences (p>0.05) were detected for DMI, ADG, or gain efficiency. No differences (p>0.05) were detected in carcass characteristics in this experiment. Percent bone ash, calcium, P, and bending moment also did not differ among treatments (p>0.05). Removing supplemental phosphorus in the feedlot diet did not produce adverse effects on steer performance, carcass, or rib bone characteristics for yearling Holstein steers.
In Korean beef market, one of the major problems is mislabeling or fraudulent distribution of Holstein dairy meat or imported beef as domestic Hanwoo meat. Therefore, there has been a great need for a development of technology to identify beef breeds in meat and meat products. This study was carried out to develop the accurate and reliable method for the identification of beef breed using PCR-RFLP marker of MC1R, MGF and TYRPl genes affecting coat colors in cattle. A single base substitution (G\longrightarrowT transition) at the codon for amino acid position 104 of MC1R gene was identified between Hanwoo and Holstein and Angus breeds. The change at this position creates Msp I restriction site in Holstein and Angus, but not in Hanwoo. When the DNA amplified products (537 bp) was digested with Msp I, Hanwoo meat showed a single band of 537bp, while two fragments of 329bp and 208 bp were observed in Holstein meat and Angus breed, respectively. Thus, breed-specific RFLP marker in the MC1R gene can be used to distinguish between Hanwoo meat and Holstein and Angus meats. In the RFLP genotype of MGF gene, the frequency of r/r type was 75% in Manwoo, whereas the frequency of R/R was 80% in Hereford breed. Holstein and Angus breeds showed 100% for R/r type. Therefore, Hanwoo meat showed significant difference in the MGF genotype frequencies compared with those of Holstein meat and imported beef cattle breeds. However, TYRP1 gene showed the same genotype in all breeds examined. Thus, this TYRP1 gene can not be used as a molecular marker for breed identification. As a consequence, we suggest that RFLP markers of the MC1R and MGF coat color genes could be used as DNA marker for identification of Hanwoo meat from Holstein and imported meats.
Despite the importance of mutation rate, some difficulties exist in estimating it. Next-generation sequencing (NGS) data yields large numbers of single-nucleotide polymorphisms, which can make it feasible to estimate substitution rates. The genetic substitution rates of Hanwoo and Holstein cattle were estimated using NGS data. Our main findings was to calculate the gene's substitution rates. Through estimation of genetic substitution rates, we found: diving region of altered substitution density exists. This region may indicate a boundary between protected and unprotected genes. The protected region is mainly associated with the gene ontology terms of regulatory genes. The genes that distinguish Hanwoo from Holstein in terms of substitution rate predominantly have gene ontology terms related to blood and circulatory system. This might imply that Hanwoo and Holstein evolved with dissimilar mutation rates and processes after domestication. The difference in meat quality between Hanwoo and Holstein could originate from differential evolution of the genes related to these blood and circulatory system ontology terms.
