The Effect of Microcurrent Stimulation on Histological Structure of Wound in Rat (미세전류전기자극이 흰쥐 창상의 조직학적 구조에 미치는 영향)

  • Purpose: The purpose of this study was to investigate the effect of the intensity of microcurrent stimulation (with currents of 50 ${\mu}A$, 100 ${\mu}A$ and 300 ${\mu}A$ using a pulse frequency of 5 pps) on wound healing in rats. Methods: Sixty male Korean rats were randomly divided into four groups of 15 rats that were subjected to four different treatment protocols (control group, no treatment; experimental groups, treated with currents of 50 ${\mu}A$, 100 ${\mu}A$ or 300 ${\mu}A$). An experimental 20 mm linear wound was made in each animal and all animals in the experimental groups received microcurrent stimulation once a day for 20 minutes until the day of sacrifice on day 1, day 3 and day 6. An optical microscope was used to determine any histological changes. Results: The experimental results were as follows. 1. In an examination with the naked eye, all groups showed similar changes until the first day. However, from the third day, a little intercellular fluid soaked through wound region in the control group rats. In the experimental group animals, little intercellular fluid soaked through wound region, and swelling and redness did not appear, from the third day. 2. In an examination with histological evaluation, more significant changes were observed in all of the experimental group rats than the control group animals. Especially, a stimulation intensity of 50 ${\mu}A$ caused a more significant effect than the use of the other intensities by day 6 of wound healing. In addition, rapid recovery was observed. Conclusion: It was determined that microcurrent stimulation had a positive effect on wound healing. A stimulation intensity of 50 ${\mu}A$ was more effective than the other intensities (100 ${\mu}A$ and 300 ${\mu}A$) utilized for wound healing. Furthermore, low-intensity microcurrent stimulation was more effective for the purpose of wound healing.

A Histological Study of Skin on Some Amphibia Inhabitated Chiri Mt. and Moodeung Mt. (지리산과 무등산에 서식하는 한국산 양서류의 피부에 관한 연구)

  • For the purpose of this study was accumulating histological data of skin some amphibia near Chiri Mt. and Moodeung Mt. Analyzed Anura and Caudata were Rana nigromaculata, Rana rugosa, Rana catesbeiana, Hynobius leechii. The histological prepared skin of frogs were compared, of which were selected from dorsal and belly. Excretory glands were identified granular glands, mucous glands, serous glands, vacuoles and excretory ducts in epidermal and dermal tissue. And developing excretory glands, well developed excretory glands and post developing excretory glands were identified also. These results were significantly as basal data on the comparative epidermal skin histology on some Korean Amphibia. Probably these glands of amphibian skin could be infered which were adaptable structure to ecological suffered condition. Following study of these results were more considerable data for comparative histology, comparative anatomy and comparative physiology and ecology of Amphibia.

Silymarin attenuates escitalopram (cipralex) induced pancreatic injury in adult male albino rats: a biochemical, histological, and immunohistochemical approach

  • Depression is a prevalent global problem since ages, predominately treated with SSRI. Cipralex, is an antidepressant of the SSRIs class used as a remedy for mood, depression and anxiety. Silymarin (SIL), a natural free radical scavenging, has an antioxidant and anti-inflammatory properties. This hypothesis evaluates, for the first time, the role of cipralex on the structure of the endocrine and exocrine components of the pancreas and assess the beneficial effects of SIL on these changes. Forty-five rats were divided into control, cipralex, and cipralex plus SIL groups. During sacrifice, all rats and pancreases were weighed and the ratio of pancreatic weight (PW) to rat weight (RW) was calculated, blood samples were collected to estimate fasting glucose, insulin and amylase levels, the specimens were prepared for histological, immunohistochemical (inducible nitric oxide synthase [iNOS], tumour necrosis factor-alpha [TNF-α], caspase 3, proliferating cell nuclear antigen [PCNA], and anti-insulin antibody), and morphometrical studies. Cipralex group exhibited marked destruction of the pancreatic architecture of the exocrine and endocrine parts, with a dense collagen fiber deposition. Also, there is highly significant decrease (P<0.001) of PW/RT ratio, insulin, and amylase levels, the number and diameter of islets of Langerhans, the number of PCNA positive immunoreactive cells, and the number of insulin positive β-cells. Furthermore, a highly significant increase of glucose level, iNOS, TNF-α, and caspase-3 positive immunoreactive cells in the islets of Langerhans and acinar cells were observed. SIL improves the pancreatic histological architecture, weight loss, biochemical, and immunohistochemical analyses. Administering SIL is advantageous in managing cipralex induced pancreatic injury via its anti-inflammatory, antioxidant, and anti-apoptotic qualities.

Histological Changes of Rat Preputial Gland during Normal Sexual Cycle and Sex Hormone Treatment (성주기(性週期) 및 성(性)호르몬 주사(注射)에 따른 Rat 포피선(包皮腺)의 조직학적변화(組織學的變化))

  • In order to know the histological changes of rat preputial gland during normal sexual cycle and sex hormone treatment, matured Wistar rats (B.W.about 200g) were used for the experiment. Rats were subcutaneously given $2{\mu}g$ 17-${\beta}$-estradiol (Sigma) and 2.5g progesterone (Nakarai Chem., Japan) daily in 0.5ml propylen glycol for ten days respectively. The results obtained are as follow: At the stage of estrus and metestrus, the eosinophilic crystalloid granules and large vacuoles in the acinar cells appeared numerously, and the excretory ducts were severely extended. The developed connective tissue between the acinus were also found. At the stage of proestrus and diestrus, however, the small vacuoles and a few eosinophilic crystalloid granules appeared in the acinar cell. In the estradiol treatment, on the other hand, severely extended excretory ducts and a small number of the eosinophilic crystalloid granules compared with the progesterone treatment were found. The cyst-like structure was found, and in that the acinus disappeared completely. In the progesterone treatment, remarkably extended excretory ducts and nomerous appearance of oval formed eosinophilic granules in the acinar cells were found. As these findings, it could be suggested that the secretion of rat preputial gland was active at the estrus and metestrus stage, and estrogen may concerned in the secretion, land progesterone in the formation of secretory products.

Histological Studies of Gonad in the Hybrid Species Cobitis. sinensis-longicorpus Complex(Pisces, Cobitidae) (잡종기원의 Cobitis sinensis-longicorpus complex(Pisces, Cobitidae)에 대한 생식소의 조직학적 연구)

  • Cobitis sinensis-longicorpus complex considered as hybrid origin between C. sinensis and C. longicorpus coccurred ommonly in the upper streams of the Nakdong River, Korea. Histological examinations of their gonad were accompanied with 272 individuals of C. sinensis-longicorpus complex collected. Most of fishes collected were females, however, only 6 individuals were found males. The ovarian tissues of females are completely fertile undergoing normal oogenensis. In the male gonads, testicular lobule structure with abnormal vacuolar tissues were observed. Spermatogonia and spermatocytes were also observed of their testis however spermatids or sperms were not shown in their developmental stages. From these facts, we infer that female population of C. sinensis-longicorpus complex may be unique reproductive hierarchy accomplishing their reproduction with participation of males of their closely related bisexual species.

Biodegradable Hydroxyapatite/Chitosan Composites on the Bone Defect of Canine Model

  • Composites of hydroxyapatite (HAp) and chitosan (CS) have been successfully used in bone healing in humans and animals. However, the characteristics of HAp and CS are different. Therefore, the effects of HAp/CS composites on canine bone formation could differ according to their ratio. This study investigated the therapeutic effects of different contents ratios (100, 80:20, 60:40 wt%) on bone defects in a canine model. Thirty intrabony cylindrical defects were created in the humeruses and femurs of 5 beagle dogs, and then the defects were implanted with different composites. The evaluations were performed using radiographs obtained at 10 weeks post-surgery and by histological findings. In radiographic evaluation including the grades of bone filling, periosteal and endosteal reactions, pure hydroxyapatite composite had a significant effect on bone filling, and chitosan containing the composites showed vigorous responses at the periosteum and endosteum. In histological findings, the defect implanted with pure hydroxyapatite had healed completely into mature bony tissue with an obvious osteon structure, and the defect implanted with chitosan containing the composites had the amount of fibrous connective tissue increased significantly within the cortical bone tissue. The results indicate that hydroxyapatite/chitosan composites are therapeutically useful, promoting effective bone healing in defects when the ratio of hydroxyapatite is high and enhanced fibrous connective tissue formation at the periosteum as more chitosan is added.

  • Histological studies on the spleen of mud turtle(Amyda sinensis) in Korea were made and the results obtained were as follows: 1. Smooth muscle cells are absent in the capsule of the spleen, although reticular, elastic and collagenous fibers are presnet as in the spleen of other mammals. 2. The trabeculs of the spleen tissue are more highly developed than those of frog and do not extend as deeply into the splenic pulp as the trabecula of mammals. 3. The structure of blood system in the spleen is found to be similar to that of mammals. Particularly, the central arteries of spleen are more highly developed, but the splenic sinus appears to be less developed than that of mammals. 4. The ratio of red pulp and white pulp in the splenic pulp appears to be 1. 5. More hemopoietic cells are seen in the spleen of turtle than in the spleen of mammals.

Re-Examination of Blood Vessels of Rat Tail Using Angiographic and Histological Methods (혈관조형술과 조직학적 방법을 이용한 랫드 꼬리의 혈관구조 규명)

  • Although the tail vessels are frequently used for serial blood sampling in rat, the definition of its vasculature remains in dispute. Herein, we investigated the number of blood vessels in Sprague-Dawley rat tail using angiographic and histological methods. Our results showed rat tail has one dorsal vein, two lateral veins, one ventral artery and one ventral vein. In the conclusion, when deciding which site is best for collecting blood, it is critical to consider the structure and features of the blood vessels to be used. This study will also be helpful for investigators to understand the structure of blood vessels in rat tail.

The Effect of Mitomycin-C on Preventing Adhesion of Injured Flexor Tendon in Rabbit Model (가토의 굴곡건 손상모델에서 Mitomycin-C가 인대 유착 방지에 미치는 영향)

  • Purpose: Adhesion after flexor tendon injury is a result of fibrosis between tendon and tendon sheath. This, finally interfere with gliding mechanism of tendon and results in functional problem of hands. Therefore, there have been many trials to reduce adhesion around the tendon. However, there is no standard procedure clinically practiced in hospitals. Mitomycin-C is an antineoplastic alkylating agent that decrease fibroblast proliferation and scar formation. It is commonly used in many surgery to reduce postoperative adhesion. This study was designed to observe the effect of Mitomycin-C on preventing adhesion in injured flexor tendon. Methods: The deep flexor tendon of digit 2 and 4 in the left forepaw of 15 New Zealand White rabbits were subjected to partial tenotomy. In study group, injury site was exposed to a single 5-minute application of Mitomycin-C, and in control group was left untreated. Digit 2 and 4 in the right forepaw of each rabbit were considered as nonadhesion control group. After 2 weeks, the animals were sacrificed and digits were amputated for biomechanical test and histological study. Results: In biomechanical study to measure yield point, mean yield point of non-adhesion control was $17.43{\pm}2.33$ and $25.07{\pm}4.03$ for adhesion control, which proves increase of adhesion in adhesion control group (p<0.05) in 95% confidence. In Mitomycin-C group, mean yield point was $12.71{\pm}4.97$. Compared with adhesion control, there was decrease in adhesiveness in Mitomycin-C group (p<0.05) in 95% confidence. In histological study, the result of adhesion control revealed massive adhesions of bony structure, fibrotic tissue and tendon structure with ablation of the border. However in Mitomycin-C group, we could find increased fibrotic tissue, but adhesion is much lesser than adhesion group and borders between structures remain intact. Conclusion: This study suggests that Mitomycin-C can significantly reduce adhesion of injured flexor tendon in rabbit model.

  • This study was purposed to observe the advantages obtained from iridodiagnostics and automatic iridoscope or oriental medical doctors. In order to study the iridodiagnostics, 5 case-researches were made as the first step. The results were obtained as follows. 1. First of all, the formation of darkening area must be paid attentionally with toxic spot and lacunain relation to the interpretation of present illness. 2. The theoretical concepts about structure in iridodiagnostics and oriental medicine were same. 3. The constitutional type of iridodiagnostics played a important role in concluding the nature and background of symptoms. 4. Most of iris signs reflected the histological changes of body structure equally, and successive study about the difference of visceral scheme between iridology and Oriental Medicine would be needed in the future.
