VA내생균근이 뽕나무의 생장에 미치는 영향 (Growth Stimulation of Mulberry Trees in Unsterilized Soil under Field Conditions with VA Mycorrhizal Inoculation)

  • 뽕나무에 VA mycorrhizae의 1종인 Glomus mosseae, Mosse and frappe를 접종하여 소독을 하지 않은 토양에 심어 6월간 재배한 결과를 요약하면 다음과 같다. 가. VA mycorrhizae 접종 뽕나무는 전체 조장은 15%, 조경은 19%, 뽕잎량은 30%가 더 많았다. 나. VA mycorrhizae접종 뽕나무의 뽕잎 성분은 질소는 12%, 인산은 10%, CaO는 10%가 더 많았고 $K_2$O는 차이가 없었으며 MgO는 20%가 더 많았다. 다. VA mycorrhizae접종 뽕나무 뿌리는 질소는 20%가 더 적었고 인산은 21%가 더 많았다.

Bacillus vallismortis Strain EXTN-1 Mediated Systemic Resistance against Potato virus Y and X in the Field

  • Efficacy of plant growth promoting rhizobacteria(PGPR) Bacillus vallismortis strain EXTN-1 has been proved in eliciting induced systemic resistance(ISR) in several crops. The present paper described the beneficial effects of EXTN-1 in potato as increase in yield and chlorophyll content, and plant protection against Potato Virus Y and X(PVY & PVX). EXTN-1 induced systemic resistance to the plants resulting in significant disease suppression in the field. Also the plants under treatment with EXTN-1 had higher chlorophyll content. The bacterized plants had significantly higher yields over the untreated control plants. The strain induced activation of defense genes, PR-1a and PDF 1.2 in transgenic tobacco model, which indicated the possible role of both SA, and JA pathways in EXTN-1 mediated plant protection against crop diseases.

Development of Leaf Spot (Myrothecium roridum) and Dispersal of Inoculum in Mulberry (Morus spp.)

  • Studies were conducted on the effect of pruning time, host age, conidial dispersal and weather parameters on the incidence and severity of mulberry leaf spot (Myrothecium roridum). The disease severity (%) increased with increase in shoot age irrespective of pruning date. Maximum disease severity was observed in plants pruned during first week of April and minimum disease severity in plants pruned during first week of March. Significant (P < 0.01) influence of date of pruning, shoot age and their interaction was observed on severity of the disease. Apparent infection rate (r) was significantly higher during the plant growth period from day 48 to day 55. Average apparent yale was higher in plants pruned during first week of April and least in plants pruned during first week of July. The disease infection was negatively correlated to distance from the inoculum source. Leaf spot severity (%) was influenced by weather parameters. Multiple regression analysis revealed contribution of various combinations of weather parameters on the disease severity. Linear prediction model $(Y = -81.803+1.176x_2+0.765x_3) with significant $R^2$ was developed for prediction of the disease under natural epiphytotic condition.

PCBs의 대기-식물간 분배 특성 인자들 (Characteristic Factors of Air-Plant Partitioning of PCBs)

  • The concentrations of PCBs (polychlorinated biphenyls) in air and plants (MOrus allba, Allum turberosum) were measured every other week at Hankyong University located in Ansung, Kyoung-ki province, Korea from July to November in 1999. Total concentrations of PCBs in air ranged from 19.8 to 71.9 pg/ $m^3$. It was observed in air that the concentrations of tai-chlorinated biphenyls(CBs) were higher than those of other PCB homologs probably due to their higher vapor pressure. Total concentrations of PCBs in plants ranged from 24.5 to 1,287 pg/g dry weight for Morus allba and 26.5 to 337 pg/g dry weight for Allum turberosum. A positive linear correlation was observed between log plant-air partition coefficients ( $m^3$air/g plant dry weight-defined here as the scavenging coefficient[S.C]) and log octanol-air partition coefficients ( $K_{oa}$ ) for each plant. In this study, slope of log S.C and log $K_{oa}$ for Morus allba, Allum turberosum were 1.07 ($R^2$= 0.83, p<0.01), 0.84 ($R^2$=0.53, p<0.05), respectively. This means that these plants may approach to equilibrium for air-plant partitioning.

Transcriptional activation of anthocyanin structural genes in torenia cv. Kauai rose by overexpression of anthocyanin regulatory transcription factors

  • This study was conducted to examine the role of the transcription factors (TFs) (RsMYB1 and mPAP1+B-Peru) in the regulation of anthocyanin biosynthesis in the ornamental torenia cv. Kauai rose. In this study, we could produce several putative transgenic lines overexpressing the TFs via Agrobacterium-mediated transformation, and presence of the TFs in the randomly selected five transgenic lines was confirmed using polymerase chain reaction (PCR). According to results of reverse transcription-PCR analysis (RT-PCR), the expression of the TFs in all transgenic lines and of the anthocyanin structural genes (CHS, F3H, DFR, and ANS) in all transgenic lines and WT plants were distinctly detectable. However, transcript levels of the structural genes expressed in the transgenic lines overexpressing TFs were significantly higher than those expressed in WT plants. Therefore, it is suggested that anthocyanin content in flowers of the transgenic torenia would be significantly higher than that in flowers of WT plants. Moreover, these results indicate that the TFs (RsMYB1 and mPAP1+B-Peru) could be exploited as potential anthocyanin regulatory TFs to enhance anthocyanin content in the other horticultural plants.

Tissue culture of medicinal plants: micropropagation, transformation and production of useful secondary metabolites

  • Plant tissue culture studies have been done for the preservation of medicinal plant resources and efficient production of pharmaceutically important secondary metabolites. Micropropagation methods for Cephaelis ipecacuanha have been established and these methods enabled much more efficient propagation of the plants than the conventional methods using seedling or layering. The C. ipecacuanha plants derived from tissue culture grew uniformly in the field and they showed higher alkaloid contents compared to the plants grown from seedlings. Hairy root cultures of C. ipecacuanha and Panax ginseng have been established by infection with Agrobacterium rhizogenes, and the production of important pharmaceuticals by these cultures have been successfully demonstrated. In the case of C. ipecacuanha, the highest alkaloid yields from the hairy roots cultured for 8 weeks were 2.75-fold cephaeline (5.5 mg) and one third emetine (0.7 mg) compared with those from the roots of one-year old plant propagated through shoot-tip culture and cultivated in a greenhouse (2.0 mg cephaeline and 2.0 mg emetine). In the case of P. ginseng, ginsenoside contents in the hairy roots optimally cultured for 4 weeks were much higher than those in the roots of 4-year old field-grown plant. Thus our medicinal plant tissue cultures demonstrate desirable properties. However, they are always exposed to danger of microbial contamination or unexpected trouble of culture facilities. Cryopreservation of plant tissue cultures is a reliable method for long-term preservation. Cryopreservation studies on these cultures are also presented.

Current Understanding of the Mechanism of qE, a Major Component of Non-photochemical Quenching in Green Plants

  • Plants dissipate excess excitation energy from their photosynthetic apparatus by a process called non-photochemical quenching (NPQ). The major part of NPQ is energy dependent quenching (qE) which is dependent on the thylakoid pH and regulated by xanthophyll cycle carotenoids associated with photosystem (PS) II of higher plants. The acidification of the lumen leads to protonation and thus conformational change of light harvesting complex (LHC) proteins as well as PsbS protein of PSII, which results in the induction of qE. Although physiological importance of qE has been well established, the mechanistic understanding is rather insufficient. However, recent finding of crystal structure of LHCII trimer and identification of qE mutants in higher plants and algae enrich and sharpen our understanding of this process. This review summarizes our current knowledge on the qE mechanism. The nature of quenching sites and components involved in this process, and their contribution and interaction for the generation of qE appeared in the proposed models for the qE mechanism are discussed.

Suppression of Nitrate Accumulation in Lettuce by Application of Mg and Micronutrients

  • High rates of nitrogen fertilization dangerously increase the nitrate content of vegetable crops, and the accumulation of nitrate in edible crops is undesirable because of potential risks to human health. Micronutrient solution containing Cu, Mn, Mo, Zn was tested for the suppression of nitrate accumulation in lettuce grown in pots treated with Mg fertilizer under a greenhouse condition. The micronutrient solution was sprayed on leaves at 3 and 4 weeks after transplanting of 20-day old seedlings. Plants were harvested after 5-week growth, and yield, contents of chlorophyll, sugar, micronutrient and nitrate, and also nitrate reductase activity were measured. Fresh weight of lettuce was significantly increased by the application of Mg and micronutrients, and the effect was the most significant in the Mg+micronutrient treatment. Also contents of chlorophyll and micronutrients were higher in the plants of micronutrient treatments. Contents of nitrate were reduced by about 14-18% in lettuce with Mg and/or micronutrient applications. Compared to the plants of control treatment, nitrate reductase activity was also higher in those plants treated with micronutrients, and in the treatment of Mg+micronutrients the enzyme activity was six times as high as that of control treatment. Although the effect of mineral nutrients on the suppression of nitrate accumulation in lettuce was relatively small in this study, an appropriate supply of mineral nutrients could be one of the solutions for the nitrate accumulation in vegetables.

A linear prediction model [$Y=66.05+(-1.39)x_1+(-0.219)x_4$] with significant $R^2$ was developed for prediction of the disease under natural epiphytotic condition.

Assessing the potential invasiveness of transgenic plants in South Korea: a three-year case study on sunflowers

  • Background: The introduction of new living modified (LM) crops may pose a latent threat to the biodiversity of each country. Here, we used sunflowers (Helianthus annuus L.) as a study system to investigate the potential for invasiveness of LM crops under different environmental conditions when released into a natural ecosystem in South Korea. We examined the seed germination, survival, and flowering of sunflowers under competition with wild plants at different sowing dates (March-December) and plot sizes (1 m × 1 m and 2 m × 2 m). Results: The germination rate showed a significant difference according to the sowing date. In addition, several sunflowers survived in plots with a high germination rate, which also led to a higher flowering rate. We found that the smaller the plot, the smaller the area available for inter-species competition, and the higher the number of surviving sunflower plants. The relative dominance and importance value of the species varied significantly between the sowing dates; in particular, sunflowers sown in March could compete with wild plants for longer than those sown on other sowing dates. Conclusions: These observations indicate that the potential for invasiveness of sunflowers differs depending on the environmental conditions and seed density at the time of release.
