Elevated thyroid hormones caused by high concentrate diets participate in hepatic metabolic disorders in dairy cows

  • Objective: High concentrate diets are widely used to satisfy high-yielding dairy cows; however, long-term feeding of high concentrate diets can cause subacute ruminal acidosis (SARA). The endocrine disturbance is one of the important reasons for metabolic disorders caused by SARA. However, there is no current report about thyroid hormones involved in liver metabolic disorders induced by a high concentrate diet. Methods: In this study, 12 mid-lactating dairy cows were randomly assigned to HC (high concentrate) group (60% concentrate of dry matter, n = 6) and LC (low concentrate) group (40% concentrate of dry matter, n = 6). All cows were slaughtered on the 21st day, and the samples of blood and liver were collected to analyze the blood biochemistry, histological changes, thyroid hormones, and the expression of genes and proteins. Results: Compared with LC group, HC group showed decreased serum triglyceride, free fatty acid, total cholesterol, low-density lipoprotein cholesterol, increased hepatic glycogen, and glucose. For glucose metabolism, the gene and protein expression of glucose-6-phosphatase and phosphoenolpyruvate carboxykinase 1 in the liver were significantly up-regulated in HC group. For lipid metabolism, the expression of sterol regulatory element-binding protein 1, long-chain acyl-CoA synthetase 1, and fatty acid synthase in the liver was decreased in HC group, whereas carnitine palmitoyltransferase 1α and peroxisome proliferator activated receptor α were increased. Serum triiodothyronine, thyroxin, free triiodothyronine (FT3), and hepatic FT3 increased in HC group, accompanied by increased expression of thyroid hormone receptor (THR) in the liver. Conclusion: Taken together, thyroid hormones may increase hepatic gluconeogenesis, β-oxidation and reduce fatty acid synthesis through the THR pathway to participate in the metabolic disorders caused by a high concentrate diet.

Influence of Fiber Content and Concentrate Level on Chewing Activity, Ruminal Digestion, Digesta Passage Rate and Nutrient Digestibility in Dairy Cows in Late Lactation

  • The influence of fiber content of hay (low-fiber 47% NDF and high-fiber 62% NDF of DM) and concentrate level (high 50% and low 20% of ration DM) on chewing activity, passage rate and nutrient digestibility were tested on four restrict-fed (11.1 to 13.7 kg DM/d) Holstein cows in late lactation. Aspects of ruminal fermentation and digesta particle size distribution were also investigated on two ruminally cannulated (100 mm i.d.) cows of the same group of animals. All digestion parameters studied were more affected by the fiber content of the hay and its ratio to non structural carbohydrates than by the concentrate level. Giving a diet of high-fiber (62% NDF) hay and low concentrate level (20%) increased chewing activity but decreased solid passage rate and total digestibility of nutrients due to a limited availability of fermentable OM in the late cut fiber rich hay. A supplementation of high-fiber hay with 50% concentrate in the diet seems to improve the ruminal digestion of cell contents, whilst a depression of the ruminal fiber digestibility was not completely avoided. Giving a diet of low-fiber (47% NDF) hay and high concentrate level (50%) reduced markedly the chewing and rumination activity, affected negatively the rumen conditions and, consequently, the ruminal digestion of fiber. A reduction of the concentrate level from 50 to 20% in the diet of low-fiber hay improved the rumen conditions as reflected by an increase of the ruminal solid passage rate and of fiber digestibility and in a decrease of the concentration of large particles and of the mean particle size of the rumen digesta and of the faeces. Generally, it can be summarised that, (i) concentrate supplementation is not a strategy to overcome limitations of low quality (fiber-rich) hay, and (ii) increase of the roughage quality is an effective strategy in ruminant nutrition, especially when concentrate availability for ruminants is limited.

Effect of Synchronizing Starch Sources and Protein (NPN) in the Rumen on Feed Intake, Rumen Microbial Fermentation, Nutrient Utilization and Performance of Lactating Dairy Cows

  • Eight crossbred (75% Holstein Friesian) cows in mid-lactation were randomly assigned to a switchback design with a 2x2 factorial arrangement to evaluate two nonstructural carbohydrate (NSC) sources (corn meal and cassava chips) with different rumen degradability and used at two levels of NSC (55 vs. 75%) with protein source (supplied by urea in the concentrate mix). The treatments were 1) Low degradable low level of corn (55%) 2) Low degradable high level of corn (75%) 3) High degradable low level of cassava (55%) and 4) High degradable high level of cassava (75%). The cows were offered the treatment concentrate at a ratio to milk yield at 1:2. Urea-treated rice straw was offered ad libitum as the roughage and supplement with 1 kg/hd/d cassava hay. The results revealed that total DM intake, BW and digestion coefficients of DM were not affected by either level or source of energy. Rumen fermentation parameters; NH3-N, blood urea nitrogen and milk urea nitrogen were unaffected by source of energy, but were dramatically increased by level of NSC. Rumen microorganism populations were not affected (p>0.05) by source of energy, but fungal zoospores were greater for cassava-based concentrate than corn-based concentrate. Milk production and milk composition were not affected significantly by diets containing either source or level of NSC, however concentrate than corn-based concentrate averaging (4.4 and 4.2, respectively). Likewise, income over feed, as estimated from 3.5% FCM, was higher on cassava-based concentrate than corn-based concentrate averaging (54.0 and 51.4 US$/mo, respectively). These results indicate that feeding diets containing either cassava-based diets and/or a higher of oncentrates up to 75% of DM with NPN (supplied by urea up to 4.5% of DM) can be used in dairy rations without altering rumen ecology or animal performance compared with corn-based concentrate.

Nutrient Utilization and Compensatory Growth in Crossbred (Bos indicus×Bos taurus) Calves

  • A feeding trial was carried out over 238 days to determine the effect of compensatory growth in crossbred calves having 166 kg body weight. Fifteen crossbred calves were divided into two groups of five calves (G1 group) and ten calves (G2 group) as per randomized block design. Growth study was conducted on the feeding of wheat straw based diet containing 60 and 30 percent concentrate supplying equal amount of protein in group G1 and G2 respectively for 119 days (phase - I). At the end of phase-I, calves of G2 group were subdivided in to two groups (G3 and G4). One sub group (G4) received 60% concentrate in their diet (during 120 to 238 days of experiment) while other subgroup G3 received 30% concentrate in their diet (phase-II). The calves of G1 group continued to receive the same diet as during phase-I experiment. Mean DM intake was significantly higher in calves fed high level of concentrate (in G1 and G4 groups), which resulted in significantly higher digestibility of all nutrients except NDF. Nitrogen balance was positive in all the groups and showed significant differences in phase-II (higher nitrogen retention in G4 group than G1 group). ME intake was significantly affected by the level of dietary concentrate, being higher in high concentrate fed group (G1 and G4 than G2 and G3 group). Higher daily body weight gain in the calves of G4 group during phase-II than in G1 and G3 groups was due to compensatory growth on shifting animals from low concentrate to high concentrate based ration. Average daily body weight gain was higher in phase-I than in the phase-II. Protein and energy intake per unit body weight gain were significantly lower in calves fed high concentrate diet.

Strategic Supplementation with a High-Quality Feed Block on Roughage Intake, Milk Yield and Composition, and Economic Return in Lactating Dairy Cows

  • Twenty-four multiparous crossbred Friesian dairy cows (60-90 days in lactation) were randomly assigned into a $2{\times}2$ factorial arrangement in a randomized complete block design. Factors were two levels of concentrate supplementation (1:2, high vs 1:1.2, very high; concentrate:milk yield) and two levels of high-quality feed block (HQFB) supplementation (non vs ad libitum block licking). Ruzi grass (Brachiaria ruziziensis) was fed as a roughage throughout the 70 day feeding trial. High level of concentrate fed group resulted in higher roughage and HQFB intakes, compared with very high concentrate supplemented group. HQFB supplementation tended to increase roughage intake and significantly improved milk yield (2 kg/hd/d in high concentrate supplementation) and quality (% fat) which resulted in higher economical return. HQFB was recommended to be used as a strategic supplement in lactating dairy cows especially when fed on low-quality roughages or crop residues.

Effect of Dietary Concentrate on Fungal Zoosporogenesis in Sheep Rumen

  • Fluctuation of fungal zoospores on agar strips were observed in the rumen of sheep fed three different levels of dietary concentrate, timothy hay: concentrate = 3:0 (AF diet), timothy hay: concentrate = 2:1 (MC diet), timothy hay : concentrate = 1:2 (HC diet) respectively. The number of zoospores on the strip was drastically decreased after morning feed with AF diet. The number was the highest at 0 h ($1.34{\times}10^2/cm^2$), then declined to $2.0{\times}10^3/cm^2$ at 9 h after feeding. In the rumen of animals fed MC diet, the number of zoospores decreased with time after feeding, although the decrement was slower than that with AF diet. During 0-3 h after feeding, number of zoospores was $1.6{\times}10^4/cm^2$. Although the number slightly decreased at 6 and 9 h, relatively high levels were maintained. It seems that the inducers for zoospore-release were maintained at relatively high concentration throughout incubation period. The fluctuation pattern of number of germinated zoospores was different in the rumen of animals fed HC diet from those of AF and MC diets. The number of zoospores was constantly maintained at lower level ($1.0{\times}10^3/cm^2$) than the other diets. For MC diet, continuous high number of germinated zoospores may be due to the continuous release of zoospores by hemes in timothy hay and concentrate feed, and by unknown mechanisms. Unlike AF diet which promoted relatively rapid decline of zoosporogenesis, supplementation of concentrate feed to the timothy hay did not promote such rapid decline of zoosporogenesis. It was suggested that release of inducers for zoosporogenesis from concentrate feed persisted longer time than from timothy hay. HC diet promoted the lowest zoospore production, suggested the lowest fungal population size in this experiment. These results show that an appropriate amount of concentrate may support fungal growth and stimulate zoosporogenesis in the rumen.

친환경 고발포 소화약제 개발 및 성능 분석 연구 (Analysis of Performance and Development of Environment-friendly High Expansion Foam Concentrate)

  • Foam concentrate is used for fire-extinguishing purposes, occurring foam, mixed with air. In huge oil-fire or the efficiency of fire-extinguishing with small quantity of water, Foam concentrate is used widely. However, perfluorooc tanoicsulfonate and nonylpenol that are included in Foam concentrate could be exposure in danger while they were discharged. The purpose of the research is the development of environment-friendly high expansion foam that exclude the dangerous substances and improvement of danger of the existing foam concentrate. The developed foam is analyzed as the environment-friendly foam in indexes of the environment-toxicity, the biodegradation appraisal, and the dangerous substances-detecting. In addition, the performances of expansion rate, discharge rate, and fire-extinguishing are suitable in the domestic regulation, so it is useful for fire-extinguishing in real fire situation.

In-sacco Degradability of Dietary Combinations Formulated with Naturally Fermented Wheat Straw as Sole Roughage

  • Twelve dietary combinations were prepared using 70 parts of fermented wheat straw (FWS) as the sole roughage supplemented with 30 parts of either the low protein concentrate mixture (Conc.-I), high protein concentrate mixture (conc.-II), maize grains (M), solvent extracted mustard cake (DMC), deoiled rice bran (DRB), uromol bran mixture (UBM), deep stacked poultry litter (DSPL), dried poultry droppings (DPD), M-DMC mixture (50:50), M-UBM mixture (50:50), M-DPD mixture (50:50) or M-UBM-DPD mixture (50:25:25) and evaluated by in-sacco technique. The above dietary combinations were also evaluated by changing the roughage to concentrate ratio to 60:40. The digestion kinetics for DM and CP revealed that FWS:DPD had the highest, whereas, the FWS:M-DMC had the lowest rapidly soluble fraction. The potentially degradable fraction was found to be maximum in FWS:M and minimum in FWS:DPD dietary combinations. The higher degradation rate of FWS:DRB and FWS:UBM combinations was responsible for their significantly (p<0.05) higher effective degradability as compared to other combinations. The highest undegradable fraction noted in FWS:M-UBM-DPD followed by FWS:DMC was responsible for high rumen fill values. The FWS:DRB, FWS:UBM and FWS:DPD combinations had higher potential for DM intake. The dietary combination with higher concentrate level (60:40) was responsible for higher potentially degradable fraction, which was degraded at a faster rate resulting in significantly higher effective degradability as compared to the corresponding dietary combination with low concentrate level (70:30). The low undegradable fraction in the high concentrate diet was responsible for low rumen fill values, which predicted of high potential for DM intake. Out of 24 dietary combinations, FWS with either of UBM, DRB, DMC, Maize, M-DMC or DPD in 70:30 ratio supplemented with minerals and vitamin A in comparison to conventional feeding practice (roughage and concentrate mixture) could be exploited as complete feed for different categories of ruminants.

Effect of Levels of Supplementation of Concentrate Containing High Levels of Cassava Chip on Rumen Ecology, Microbial N Supply and Digestibility of Nutrients in Beef Cattle

  • The object of this study was to determine the influence of supplementation of concentrate containing high levels of cassava chip on rumen ecology, microbial protein and digestibility of nutrients. Four, rumen fistulated crossbred beef steers with initial body weight of 400${\pm}$10 kg were randomly assigned according to a 4${\times}$4 Latin square design. The dietary treatments were concentrate cassava chip based offering at 0, 1, 2 and 3% BW with urea-treated rice straw fed ad libitum. It was found that ruminal pH was significantly decreased with increase of concentrate. Volatile fatty acids (VFA) concentration in the rumen was significantly different among treatments. In addition, a molar proportion of propionate was higher in supplemented groups at 2 and 3% BW (p<0.05), leading to significantly decreased acetate:propionate ratio. Furthermore, microbial N supply was significantly improved and was highest at 2% BW supplementation. The efficiency of rumen microbial-N synthesis based on organic matter (OM) truly digested in the rumen was highest in level of concentrate supplementation at 2% BW (80% of cassava chip in diets). Moreover, bacterial populations such as amylolytic bacteria was linearly increased, while cellulolytic bacteria was linearly decreased (p<0.01) when cattle received concentrate supplementation in all levels. The total protozoal counts were significantly increased, while fungal zoospores were dramatically decreased in cattle receiving increased levels of concentrate. In conclusion, cassava chip can be use as energy source at 80% in concentrate and supplementation of concentrate at 2% BW with urea-treated rice straw as roughage could improve rumen fermentation efficiency in beef cattle.

이유 전 농후사료 (무조사료) 급여가 한우 송아지의 반추위 유두 발달에 미치는 영향 (Effects of Concentrate Feeding on Rumen Papillae Development in Hanwoo Calves before Weaning)

  • 본 연구는 이유 (생후 2개월) 전 농후사료 (무조사료) 급여가 한우 어린 송아지의 반추위 유두 발달에 미치는 영향을 조사하기 위하여 수행하였다. 총 24두 (수 12두, 암 12두)의 생후 6일령 한우 어린송아지를 3처리구에 배치하였고, 각각 어미젖을 포유할 수 있는 시설에서 사육하였다. 처리구는 크게 농후사료구 (무조사료구)와 조사료구로 구분하였고, 농후사료구는 농후사료의 섭취량에 따른 반추위 발달 양상을 조사하기 위하여, 사료섭취량을 기준으로 고농후사료급여구와 저농후사료급여구로 세분화하였다. 31~60일령의 1일 사료 섭취량은 고농후사료급여구 (380.2 g/d)가 저농후사료급여구 (76.9 g/d) 및 조사료구 (58.3 g/d)에 비하여 유의적 (P<0.05)으로 높았다. 고농후사료급여구와 저농후사료급여구의 1일 사료섭취량은 큰 차이를 보였지만 생시체중 및 60일령 체중은 비슷하게 나타나, 저농후사료급여구의 성장에 부족한 영양분은 어미소의 포유로부터 충족된 것으로 사료되었다. 60일령의 체중 대비 반추위 무게의 비율은 고농후사료급여구 (1.39%)가 저농후사료급여구 (1.06%) 및 조사료구 (0.85%) 보다 유의적 (P<0.05)으로 높게 나타났다. 뿐만 아니라 고농후사료급여구의 반추위 유두 길이 및 두께도 다른 처리구들에 비하여 유의적 (P<0.05)으로 높았다. 이는 육안으로도 그 차이를 뚜렷이 구분할 수 있었다. 60일령 혈중 ${\beta}$-hydroxybutyrate의 농도도 고농후사료급여구 (176.4 ${\mu}mol/L$)가 저농후사료급여구 (59.2 ${\mu}mol/L$) 및 조사료구 (58.9 ${\mu}mol/L$) 보다 유의적 (P<0.05)으로 높았다. 본 연구의 결과는 이유 전 (생후 2개월) 한우 송아지의 반추위 발달이 농후사료 섭취량에 의하여 크게 영향을 받으며, 우유와 조사료는 반추위 발달에 미치는 영향이 매우 낮음을 제시하였다.
