Acaricidal Efficacy of Herbal Extracts against Tetranychus urticae (Acarina: Tetranychidae) (점박이응애, Tetranychus urticae (Acarina: Tetranychidae)에 대한 약용식물 추출물의 살비 효과)

  • Methanol and hexane extracts from 35 species in 27 families of herbal plants were evaluated for their acaricidal activities against two-spotted spider mite, Tetranychus urticae by leaf-dipping and spraying methods in laboratory, pot and field, respectively. Acaricidal activities were different depending on herbal plants. When T. urticae was released on leaves of Phaseolus vulgaris var. humilis that were dipped in the 1,000 ppm hexane extracts from fruits of Torreya nucifer and Daphan genkwa, seeds of Xanthium strumarium and Pharbitis nil at one minute they experienced 56.8, 47.8, 47.7, and 47.7% mortalities, respectively. Dipping time influenced acaricidal activity, that is, one minute dipping was more effective than 30 second dipping of Phaseolus vulgaris var. humilis leaves. However, extracts of X. strumarium and P. nil seeds had comparatively high activities. Concentration of hexane extract of X. strumarium and P. nil seeds revealing LC50 were 1,824 and 1,899 ppm, respectively. Extracts of both plants from hot and cold water were not effective representing <20% mortality at 1,000 ppm. However, hexane extracts of X. strumarium and P. nil seeds were effective against T. urticae on P. vulgaris var. humilis representing 76.3 and 71.3% mortalities in pot, respectively. Control effects of hexane extract of P. nil seed were 50.8 and 35.1% at 2,000 and 1,000 ppm against T. urticae on Chrysanthemum morifolium in greenhouse, respectively.

  • Yields of various solvent extracts for Zanthoxylum schinifolium and Z. piperitum leaves were higher in ethanol extract layer. Ethanol Extract in sancho and chopi leaves was fractionated with different solvents, such as hexane, chloroform, ethyl acetate, butanol and water, yields of hexane fraction was higher. In the solvent extracts using methanol, ethanol, hexane, chloroform, and water, ethanol extract showed the most effective antimicrobial activities. Ethanol extract in sancho and chopi leaves was fractionated, the most antimicrobial activities of sancho leaf were ethyl acetate fraction, and chopi leaf were chloroform fraction.

Synergistic Effects of Catechin or Ascorbic Acid on Antioxidative Activities of Hexane and Methanol Extracts from Rosemary, Sage, Oregano, and Ginger (Rosemary, Sage, Oregano와 Ginger의 메탄올과 헥산 추출물의 항산화 작용에 대한 카테킨과 아스코르브산의 상승 효과)

  • This study was peformed to investigate the synergistic effects of catechin and ascorbic acid on antioxidative activities of methanol and hexane extracts (500 ppm) from rosemary, sage, oregano, and ginger. Ascorbic acid (200 ppm) and (-)-catechin (200 ppm) could be solubilized in a rice bran oil via a reverse micelles using small amount of water and dioctyl sulfosuccinate as the sufactant. Methanol extracts from rosemary, sage, oregano, and ginger showed the synergistic effects by (-)-catechin. However, methanol extracts showed the synergistic effect by ascorbic acid except that of ginger. The synergistic effects of (-)-catechin on methanol extracts were higher than those of ascorbic acid. Hexane extracts of oregano and ginger showed the synergistic effects by (-)-catechin, and no synergistic effects by ascorbic acid. On the other hand, rosemary and sage showed the synergistic effects by ascorbic acid and no synergistic effects by (-)-catechin.

Inhibitory Effect of Mugwort Extracts on Tyrosinase Activity (쑥 추출물의 Tyrosinase 효소활성에 미치는 억제효과)

  • To determine the inhibition of mushroom tyrosinase activity, fresh and dried mugwort, Artemisia princeps was extracted initially with water and ethyl alcohol, and subsequently with hexane, chloroform and ethyl acetate in that order. The highest yield was obtained from the ethyl acetate (15.2%) and hexane (15.5%) fraction of the ethanolic extract of fresh and dried mugwort, respectively. For all fractions tested, the inhibition of tyrosinase activity by fresh mugwort was higher than that of dried mugwort, and the inhibition ratio of tyrosinase activity was 98.9% in the chloroform fraction and 96.7% in the hexane fraction.

  • To investigate the cytotoxicity of Korean traditional fermented soybean products using the MTT assay, we extracted soybean, Kanjang, Doenjang, Kochujang, and Chongkukjang with water, methanol, and hexane. Primary testing of cytotoxicity of 14 extracts was done for P388D1(mouse lympoid neoplasm) and L1210(mouse leukemia) cell lines. Doenjang methanol extract, Kochujang hexane extract, Chongkukjang methanol extract, and Chongkukjang hexane extract showed cytotoxicity of 86.1, 94.3, 83. 6, and 81.1%, respectively against P388D1, and showed cytotoxicity of 69.4, 96.9, 51.4, and 95.1%, respectively against L1210. All the other extracts showed less than 50% cytotoxicity. Methanol extracts of Doenjang and Chongkukjang showed dose-dependent cytotoxicity against P388DI, L1210, SNU-16 (human stomach cancer), HepG2(human hepatic cancer), WiDr(human colon cancer) cell lines, and IC$_{50}$ of Doenjang methanol extract was 67.7, 90.4, 1338.0, 706.4, and 371.2 $\mu$g/ml, respectively, and IC$_{50}$ of Chongkukjang methanol extract was 107.1, 228.3, 756.2, 1346.0, and 327.0 $\mu$g/ml, respectively.

Antibacterial Activities of Oenothera laciniata Extracts (애기달맞이꽃(Oenothera laciniata Hill) 추출물의 항균활성)

  • The solvent extracts of Oenothera laciniata, which were extracted by using several solvents with different polarities, were prepared for utility as a natural preservative. The O. laciniata extract by 80% ethanol was sequentially fractionated with n-hexane, dichloromethane, ethylacetate, and butanol. The antibacterial activities and cell growth inhibition were investigated on each strain with the different concentrations of O. laciniata extracts. Antibacterial activities were shown in ethanol, ethylacetate, and butanol fraction of O. laciniata. However n-hexane, dichloromethane and water fraction showed weak antibacterial activity against the tested microorganisms. Among the five fractions, ethylacetate fraction showed the highest antibacterial activities against microorganisms tested, such as Bacillus cereus, Listeria monocytogenes, Staphylococcus aureus, Escherichia coli, Salmonella Enteritidis and Salmonella Typhimurium. The polyphenolic compounds widely occurring in the traditional medicine plants have been reported to possess high antibacterial activity. The polyphenolic compounds from ethanol, n-hexane, dichloromethane, ethylacetate, butanol, and water fraction were 63.96 mg/g, 8.49 mg/g, 28.11 mg/g, 172.64 mg/g, 114.56 mg/g, and 34.91 mg/g, respectively. There are some relationships between antibacterial activity and polyphenol content in natural plant. The ethylacetate fraction could be suitable for the development of a food preservative.

  • In this study, hot water and ethanol, acetone, and hexane extracts of the steamed Deodeok were investigated for use as raw materials for natural cosmetics. Hot water and ethanol, acetone and hexane extracts of steamed Deodeok did not indicate cell toxicity up to 300 ㎍/mL concentration in the MTT assay. The tyrosinase inhibition effects of the ethanol extract were higher than the ascorbic acid. The efficacy of the steamed Deodeok's ethanol extract on the elastase inhibition was similar to that of quercetin at higher concentrations. In conclusion, water, ethanol, acetone and hexane extracts of the steamed Deodeok indicated strong possibility as a whitening and anti-wrinkle functional cosmetic ingredient. Among them, the steamed Deodeok's ethanol extract, which showed excellent activity, is highly available because it has high safety compared to other organic solvent extraction methods used in cosmetics, and has minimal irritation to the skin.

  • To investigate the inhibitory effect of soybean and Korean traditional fermented soybean products on the chemically induced mutagenesis, we extracted soybean, Kanjang, Doenjang, Kochujang, and Chongkukjang with water, methanol and hexane. Inhibitory effect of the extracts was assayed by the SOS chromotest using Escherichia coli PQ37 as a test strain. 4-nitroquinoline-1-oxide(4NQO), N-methyl-N'-nitro-N-nitrosoquanidine(MNNG), and aflatoxin B$_{1}$(AFB$_{1}$) were used as mutagens. Methanol extracts showed relatively higher inhibitory effect than water and hexane extracts. Methanol extracts of soybean, Doenjang, Kochujang, and Chonhkukjang showed inhibitory effect of 68.4, 96.3, 17.5, and 100.9% against MNNG, and 28.6, 109.1, 41.3, and 101.8% against AFB$_{1}$, respectively. Doenjang methanol extract showed inhibitory effect of 51.0, 96.3, and 109.1% against 4NQO, MNNG, and AFB$_{1}$, respectively. Methanol extract of Doenjang showed dose-dependent inhibitory effect against 4NQO, MNNG, and AFB$_{1}$. Inhibitory effect of heat-treated Doenjang and Chongkukjang methanol extracts on the mutagenecity of MNNG and AFB$_{1}$ was remained over 95% of the inhibitory effect of heat-untreated extracts, demonstrating the heat stability of the potent antimutagenic activity.

Studies on Antioxidative Substances of Ganoderma lucidum (영지의 항산화성 물질에 관한 연구)

  • To study antioxidative activities of Ganoderma lucidum, its extracts were fractionated by various organic solvents with different polarity the extracts were purified by thin chromatography, silicagel column chromatography and preparative liquid chromatography. In antioxidative activity tests using thiocyanate method, TBA method and weighing method, fraction 5 from the hexane extract and fraction II from the methanol extract showed antioxidative activity. When the antioxidative activities were expressed as TBA value using a homogeneous liver extracte of rats, the relative antioxidative activities of fraction 5 and fraction II were increased by 13.0% and 54.6%, respectively.

Antioxidative and Free Radical Scavenging Effects of Rhus javanica Linne (오배자의 항산화 및 자유라디칼 소거효과)

  • This study was carried out to investigate the antioxidative activities of medicinal plants. Through the examination of methanol extracts from 30 species for radical scavenging effects using DPPH method, the extracts from Rhus javanica Linne, Smilax china Linne and Polygonum cuspidatum Siebold et Zucarinii showed strong antioxidative activity. Because of its highest antioxidative activity among 30 medicinal plants, radical scavenging effects of 4 different extract compartments (n- Hexane, EtOAc, BuOH and $H_2O$ extracts) from Galla Rhois MeOH extract of Rhus javanica Linne were examined by DPPH method and antioxidant effects on the 4 different extract compartments were tested by Ferric-Thiocyanate method. Antioxidative activities of n- Hexane, EtOAc and BuOH extracts were similar or even higher than that of natural (tocopherol) or synthetic antioxidants (BHA), suggesting that major fractions for the antioxidative activity of Rhus javanica Linne were the n- Hexane, EtOAc and BuOH extract compartments.
