The effects of n-hexane and tetrachlorodiphenyl on chicken (n-hexane 과 tetrachlorodiphenyl 이 닭에 미치는 영향)

  • The effects of n-hexane and tetrachlorodiphenyl on the growth rate, the blood picture and histopathological change induced were observed in chicken. A total of 350 chicken were fed various daily dosages of n-hexane and tetrachlorodiphenyl for the experimental period of 60 days. The results obtained were as follows. 1. The growth rate of chicken in the group fed 100 ppm of tetrachlorodiphenyl was reduced (p<0.05). However no significant results were observed in the other dosage groups. 2. The reduction of the growth rate was found only in the liver and the kidney but not found in the other orgns. 3. No significant results were found in the number of erythrocytes and hematocrit value. However, the number of leukcocytes of chicken in the group fed 100 ppm of tetrachlorodiphenyl was significantly low. 4. In both n-hexane and tetrachlorodiphenyl groups, the number of lymphocytes was found to decrease but the number of basophiles and eosinophiles were found to increase (p<0.05). 5. All the chicken fed 300 ppm of tetrachlorodiphenyl died between 7th and 9th days. 6. Fatty change of hepatic cells and cloudy swelling of epithelia of renal tubules were found in the group fed 0.5 ml of n-hexane. However, in the group fed more the 100 ppm, fatty change of hepatic cells was followed by necrosis and comparatively severe cloudy swelling was found in epithelia of renal tubules.

A Tandem Water and Hexane Washing Method for Economical Recovery of Paclitaxel from Biomass (바이오매스 유래 파클리탁셀의 경제적인 회수를 위한 물과 헥산의 순차적 세척 방법)

  • In this study, a tandem water and hexane washing process was developed to improve the recovery efficiency of paclitaxel derived from Taxus chinensis. The polar impurities contained in the sample were effectively removed by washing with water at a sample/water ratio of 1:40 (w/v) for 10 min. In addition, the non-polar impurities were effectively removed by washing with hexane at a sample/hexane ratio of 1:160 (w/v) for 20 min. A high purity of paclitaxel (>30.0%) was obtained in a short operating time (~30 min) by sequential washing with water and hexane.

Analysis of dioxin-like PCBs in Soil samples (토양 중 dioxin-like PCBs의 분석)

  • The analytical method of 14 kinds of coplanar - PCBs was established and applied the soil sample. The three kinds of extraction solvents (toluene, acetone: n-hexane, dichloromethane) were selected to apply the soil sample. The silica gel, florisil and alumina column cleanup also performed to compare the elution recovery. The average recovery of selected solvents in soil A, B and C was surveyed the 84.25%, 56.09% and 44.69% for toluene, 52.56%, 81.42% and 58.53% for acetone : n - hexane and 55.94%, 71.33% and 61.05% for dichloromethane. The average recovery is represented 49.99% for silica gel (n - hexane 100 mL), 69.65% for florisil (6% ether/n - hexane 100 mL), and 65.23% for alumina (2% DCM : n - hexane 100 mL, 50% DCM: n-hexane 150 mL). In silica gel (n - hexane) and florisil (6% ether : n - hexane) cleanup, the 14 kinds of coplanar PCBs eluted until 40 mL. In the silica gel and florisil columns cleanup, the amounts of elution solvent can be reduced from these results, but the researcher has to confirm the elution amounts before performing the experiments. In alumina cleanup process, the result was obtained to the 100 mL of elution solvents (2% DCM: n-hexane 100 mL and 50% DCM: n-hexane 40 mL), therefore the change of elution solvent is necessary to develop the simple procedure.

Whitening Effect of Angelica keiskei Koidzumi Extract by Inhibition of Endothelin-1 Production and Melanogenesis (Endothelin-1 생성 저해와 멜라닌생성 저해에 의한 신선초 추출물의 미백 효과)

  • In order to investigate the potential of a Angelica keiskei Koidzumi extract and its fractions as an active ingredient for whitening cosmetics, we prepared Angelica keiskei Koidzumi extract(70% ethanol), and the aqueous suspension was successively extracted with hexane, ethyl acetate(EA), and n-butanol fraction. We measured their inhibitory effects on mushroom tyrosinase and melanin synthesis in B16 melanoma cells and normal human kerationocytes in vitro. They did not show inhibitory activity against mushroom tyrosinase and the melanin synthesis except hexane and EA fractions. Hexane and EA fractions markedly inhibited cellular tyrosinase activity at a lower concentration(25 and 5 ${\mu}g/mL$, respectively) than arbutin(250 ${\mu}g/mL$). We also quantified the released amount of endothelin-1(ET-1), a mitogen of melanocyte, and interleukin-$1{\alpha}$(IL-$1{\alpha})$, a mediator of UVB-induced inflammation. Hexane and EA fractions did not affect IL-$1{\alpha}$ production, but they decreased UVB-induced ET-1 production in normal human keratinocytes in a dose dependant manner. As a result, hexane and EA fractions could inhibit the melanogenesis through the inhibition of UVB-induced ET-1 production in normal human keratinocytes. This result suggests that hexane and EA fractions of Angelica keiskei Koidzumi extract could be used as an active ingredient for cosmetics.

Inhibitory Effect of Major Food Components on the Activity of Antimicrobial Active Substance from n-Hexane Fraction of Mallotus japonicus Muell on Listeria monocytogenes (식품의 주요 성분이 예덕나무 헥산추출물의 Listeria monocytogenes에 대한 항균활성에 미치는 영향)

  • Inhibitory action of major food components on antimicrobial active substance (linolenic acid) was tested against Listeria monocytogenes ATCC 19111 for 72 hr at $30^{\circ}C$. Linolenic acid (50, 1000 ppm) and n-hexane fraction of Mallotus japonicus Muell (50, 1000 ppm) were added to the broth culture medium containing casein(1%, 3%), soybean oil(1%, 3%) and soluble starch (1%, 3%), respectively. Linolenic acid 1000 ppm and n-hexane fraction 1000 ppm exhibited strongly antimicrobial actions on L. monocytogenes which were not detected viable cell after 24 hr. But casein (3%) and soybean oil (3%) strongly diminished the antimicrobial action of 1000 ppm of n-hexane fraction. Soluble starch (1%, 3%) did not affect the antimicrobial action of 1000 ppm of linolenic acid and n-hexane fraction.

Regulation of Wnt/β-catenin Signal Transduction in HT-29 Colon Cancer Cells by a Rhododendron brachycarpum Fraction (홍만병초 분획물에 의한 HT-29대장암 세포의 Wnt/β-catenin 신호전달 조절)

  • The aim of this study was to determine the anticancer activities of an 80% methanol extract and various fractions of Rhododendron brachycarpum (RB). The n-hexane fraction of RB showed the highest inhibitory activity (Inhibit concentration $50%=20.2{\pm}1.2{\mu}g/ml$) in HT-29 cells. Colony- and sphereforming abilities were significantly correlated with a decrease in the cell count and size. A TOP/FOP flash reporter assay revealed that the inhibitory activity of the n-hexane fraction of RB ($0.22{\pm}0.02$ fold change) was lower than that of the 80% methanol extract and that of other fractions. The n-hexane and ethyl acetate fractions of RB were predominantly dependent on the expression levels of intracellular ${\beta}-catenin$. Western blotting using $p-GSK3{\beta}$ with only the n-hexane fraction of RB was conducted to examine whether these secondary metabolites reduced ${\beta}-catenin$ degradation. Intracellular ${\beta}-catenin$ regulation resulted in quantitative changes in the nucleus. In summary, these results demonstrate the potential of the n-hexane fraction of RB as a natural anticancer agent.

In vitro Antibacterial and Synergistic Activity of an Ecklonia cava Extract against Anti biotic-Resistant Streptococcus parauberis

  • In an effort to discover alternative phytotherapeutic antimicrobial agents to combat Streptococcus parauberis, a fish pathogenic bacterium, we evaluated the antibacterial activity of seaweed extracts in vitro. A methanolic extract of Ecklonia cava exhibited strong antibacterial activity against S. parauberis isolated from olive flounder Paralichthys olivaceus. Furthermore, the n-hexane soluble (Hexane) fraction of the E. cava methanolic extract exhibited the greatest antibacterial effect on S. parauberis strains with a minimum inhibitory concentration (MIC) ranging from 256 to $1,024{\mu}g/mL$. In addition, the MIC values of oxytetracycline against antibiotic-resistant S. parauberis were markedly reduced up to 64-fold in combination with the Hexane fraction, suggesting that the antibacterial activity of the antibiotic was restored when combined with the Hexane fraction. The interaction between both antibiotics and the Hexane fraction was assessed by the fractional inhibitory concentration (FIC) index. The Hexane fraction and oxytetracycline combination against antibiotic-resistant S. parauberis strains resulted in a median ${\sum}FIC$ range of 0.502 to 0.516. Thus, the synergistic ranges of median ${\sum}FIC$ < 1 were observed for all combinations of the Hexane fraction and oxytetracycline against S. parauberis. To the best of our knowledge, this is the first report indicating the efficacy of an E. cava extract against fish pathogenic bacterium S. parauberis.

Effect of Rosmarinus officinalis L. on Growth Inhibition and Apoptosis Induction in Cancer Cells (로즈마리(Rosmarinus offcinalis L.) 분획물의 암세포에 대한 성장억제 및 세포사멸 유도 효과)

  • The goal of this study was to evaluate the anticancer effect of Rosmarinus officinalis L. In this study induction of apoptosis by methanol extract of rosemary and their fractions were investigated in vitro. In examining the effect of rosemary methanol extract on the inhibition of growth of Hela, HepG2, A549, AGS cells and HT-29 cell, it was found that the methanol extract of rosemary and their fractions demonstrated a cytotoxic effect in a dose-dependent manner; in addition, hexane and chloroform fractions showed a particularly high cytotoxic effect on Hela and AGS cells. The results showed that the hexane and chloroform fractions of rosemary have cytotoxic effect which are related to the activity of the essential oil in the rosemary. Apoptosis in Hela and AGS cells mediated by the hexane and chloroform fractions was associated with the increase of cleaved caspase-3 levels and cleaved PARP. Therefore, with more researches on identification and action mechanism of active compound, the hexane and chloroform fractions are expected to be natural sources for the developments of functional food and medical agents to prevent gastric cancer and uterus cancer.

Extracts from Pinus densiflora Siebold et Zuccarini Sawdust Inhibit the Mycelial Growth of Lentinus edodes (Berk.) Sing., Edible Mushroom

  • The extracts of Pinus densiflora sawdust by n hexane, ethyl acetate and methanol solvent were investigated to identify their mycelial growth inhibition against Lentinus edodes. The yields of n hexane soluble fraction, ethyl acetate-soluble fraction, and methanol soluble fraction from P. densiflora sawdust were obtained 1.36%, 2.21% and 4.03% using organic solvent, respectively. The mycelial growth inhibition of L. edodes was the greatest for n hexane extract, ranging from 36.5% to 47.6% at concentrations of 125 ppm to 1,000 ppm, with the values for all concentrations significantly different from one another. After direct extraction of P. densiflora sawdust using n hexane, ethyl acetate and methanol, each extract was separated into three fractions by silica gel column chromatography and then the fractions were isolated on the values of $R_f$ by thin layer chromatography. The mycelial growth inhibition against L. edodes was recognized in the fractions II (33.5%) and III (37.6%) of n hexane extract, the fraction II (21.4%) of ethyl acetate extract and the fraction II (26.4%) of methanol extract. The fractions III of n-hexane extract showed the highest growth inhibition among the nine fractions of the organic solvent extract.

Antimicrobial Activities Against Oral Microbes and Growth-inhibitory Effect on Oral Tumor Cell by Extract of Paeonia lactiflora (작약 추출물의 구강병원균에 대한 항균성 및 구강암 세포 증식 억제효과)

  • Paeonia lactiflora was stepwise extracted with hexane, chloroform, ethyl acetate, butanol and water. Anti-microbial activity of each extract was investigated. Methanol extract of P. lactiflora revealed anti-microbial activity against S. mutans, C. albicans, and S. aureus. Also, hexane fraction revealed anti-bacterial activity against S. mutans and ethyl acetate fraction acted as potent anti-microbial agent on C. albicans and S. aureus. The relative growth ratio(RGR) of hexane fraction of P. lactiflora against S. mutans were determined as 77.8% in concentration of 0.125 mg/ml, 98.46% in 0.25 mg/ml and 100% in 0.5 mg/ml. The ethyl acetate fraction of P. lactiflora revealed RGR against C. albicans as 52.5% in concentration of 0.125 mg/ml, 60.83% in 0.25 mg/ml and 78.33% in 0.5 mg/ml. It indicate that increasing concentration increase RGR. The measured minimal inhibitory concentration(MIC) of hexane fraction on S. mutans KCTC 5316 strain was 0.5 mg/ml and MIC of ethyl acetate fraction on C. albicans KCTC 7270 was 2.0 mg/ml. The experiment of inhibition to growth of KB roll(oral squamous cell carcinoma) result 61.9% in butanol, 76.7% in hexane extract of P. lactiflora. The hexane extract exhibit potent inhibition effect to the growth of KB cell. These results suggest that the hexane extract of Paeonia lactiflora has antimicrobial activity against S. mutans and has preventive effect to dental caries in addition to potent inhibition to KB cell growth.
