Simultaneous Determination of Liquiritin, Nodakenin, Hesperidin and Glycyrrhizin in Bojungikgi-tang Using HPLC-PDA (HPLC-PDA를 이용한 보중익기탕 중 Liquiritin, Nodakenin, Hesperidin 및 Glycyrrhizin의 동시분석)

  • Bojungikgi-tang has been widely used for enhancement of physical fitness in Korea. The convenient, simple, and accurate high-performance liquid chromatography (HPLC) method was established for simultaneous determination of four marker compounds, liquiritin, nodakenin, hesperidin, and glycyrrhizin in Bojungikgi-tang (Buzhongyiqi-tang in Chinese, Hochuekkito in Japanese), a traditional Korean herbal prescription. The column for optimizing HPLC separation was used a Gemini $C_{18}$ column at column oven temperature of $40^{\circ}C$ with 1.0% (v/v) aqueous acetic acid (A) and 1.0% (v/v) acetic acid in acetonitirle (B) by gradient flow. The flow rate was 1.0 ml/min and the detector was a photodiode array (PDA) set at 254 nm, 280 nm, and 335 nm. Calibration curves of four components were acquired with $r^2$ values ${\geq}0.9999$. The recoveries were found to range 92.11~105.68% with relative standard deviations (RSDs, %) value less than 2.50%. The RSD values of intraand inter-day precision were 0.07~2.50% and 0.16~1.99%, respectively. The contents of liquiritin, nodakenin, hesperidin and glycyrrhizin in Bojungikgi-tang were 3.85~3.92 mg/g, 2.27~2.32 mg/g, 4.14~4.19 mg/g, and 3.39~3.45 mg/g, respectively. The established simultaneous analysis method will be effective for quality control of Bojungikgi-tang.

Stabilization to Sunlight of Natural Coloring Matter by Soluble Methyl-Hesperidin (수용성 메틸-헤스페리딘에 의한 천연색소의 빛에 대한 안정화)

  • Soluble methyl-hesperidin (MH) powder was prepared from crude MH, and was investigated on the stabilization to sunlight of natural coloring matter. Soluble MH powder was added to riboflavin, bixin, and paprika oleoresin solutions at concentration of 0.1 or 0.2%(w/w), and the absorbance of respective solutions was analyzed by spectrophotometer after exposing to sunlight for $10{\sim}24$ hr. While coloring matter solutions without MH was faded completely with lapse of time, addition of MH retarded to fade natural coloring matters. According as amount of MH added increased, MH showed more excellent stabilizing effect on the coloring matters. Especially, vitamin C with soluble MH showed remarkable synergistic effect in paprika oleoresin solution during the exposure to sunlight for 8 hr. Consequently, it was found that MH plays a role as excellent stabilizer of natural coloring matters to ultraviolet of sunlight.

Hesperetin Stimulates Cholecystokinin Secretion in Enteroendocrine STC-1 Cells

  • Hesperetin (3',5,7-trihydroxy 4'-methoxyflavanone) and its glycoside hesperidin (hesperetin 7-rhamnoglucoside) in oranges have been reported to possess pharmacological effects related to anti-obesity. However, hesperetin and hesperidin have not been studied on suppressive effects on appetite. This study examined that hesperetin and hesperidin can stimulate the release of cholecystokinin (CCK), one of appetite-regulating hormones, from the enteroendocrine STC-1 cells, and then examined the mechanisms involved in the CCK release. Hesperetin significantly and dose-dependently stimulated CCK secretion with an $EC_{50}$ of 0.050 mM and increased the intracellular $Ca^{2+}$ concentrations ($[Ca^{2+}]_i$) compared to the untreated control. The stimulatory effect by hesperetin was mediated via the entry of extracellular $Ca^{2+}$ and the activation of TRP channels including TRPA1. These results suggest that hesperetin can be a candidate biomolecule for the suppression of appetite and eventually for the therapeutics of obesity.

Quantitative Analysis of Bioconversion Constituents of Insampeadock-san Using Various Fermented Bacteria (다양한 발효균주를 이용한 인삼패독산의 생물 전환 성분의 정량 분석)

  • Insampaedoksan (IS) is the decoction of medicinal herbs, which was commonly used for anti-inflammatory and anti-pyretic in the Korean traditional medicine (KTM). Several studies on improving efficiency or searching new efficiency by fermenting traditional herbal medicines are recently in progress. The bioconversion has been conducted on IS using various bacteria. Liquiritin, ferulic acid, hesperidin, liquiritigenin, and glycyrrhizin in IS before and after fermented IS were simultaneously analyzed. These compounds were qualitatively analyzed and quantitatively analyzed using the HPLC-DAD system. The identifications of liquiritin, ferulic acid, hesperidin, liquiritigenin and glycyrrhizin were achieved by comparing the HPLC retention time ($R_t$) and the UV absorption of five pure compounds in the IS. As a result, the increased constituents were identified to be liquiritin, liquiritigenin and glycyrrhizin, while the decreased constituent was ferulic acid and the constituent of hesperidin was similar to before and after fermentation. Insampeadock-san fermented by Lactobacillus plantarum KFRI 144 exhibited the most remarkable changes in all of fermentation.

Hesperidin Ameliorates TNF-${\alpha}$-Mediated Insulin Resistance in Differentiated 3T3-L1 Cells

  • Adipose inflammation is linked to the development of insulin resistance and type 2 diabetes. Hesperidin (HES) is a flavonoid with antioxidant, anti-inflammatory and anti-diabetic properties. However, whether HES improves inflammation-mediated insulin resistance in adipose tissues remains unclear. The purpose of this study was to investigate whether HES attenuates inflammation-mediated insulin resistance in adipose tissue. Herein, RAW 264.7 cells and differentiated 3T3-L1 adipocytes were pretreated with various concentrations of HES in complete media for 1 h and then cultured in the presence or absence of LPS or TNF-${\alpha}$. Our results demonstrated that HES remarkably inhibited LPS-induced production of IL-6, TNF-${\alpha}$, and NO by RAW 264.7 cells in a dose-dependent manner. Also, HES inhibited TNF-${\alpha}$-induced production of IL-6 and $PGE_2$ in differentiated 3T3-L1 cells, while upregulated TNF-${\alpha}$-suppressed expression of adiponectin and PPAR-${\gamma}$ mRNA. These findings suggest that HES may ameliorate inflammation-mediated insulin resistance in adipose tissue.

Changes of Principal Components and Microbial Population in Pyungwi-san Decoction according to the Preservation Temperature and Period (평위산 전탕액의 보관온도 및 기간에 따른 주요성분 및 미생물 군집 변화)

  • Objectives: To optimize the preservation method of herbal decoction, we investigated the content of principle components of Pyungwi-san, liquiritin, glycyrrhizin, and hesperidin according to preservation temperature and period. We also investigated the changing patterns of pH and microbial population in Pyungwi-san decoction as a model case. Methods: With samples preserved at different temperatures, the content of liquiritin, glycyrrhizin, and hesperidin was determined using HPLC and microbial population was determined as viable counting method up to 8 times every month. Identification of isolated bacteria was performed by 16S rDNA analysis. Results: The content of liquiritin and glycyrrhizin did not change according to the preservation temperature and period, but that of hesperidin was severely decreased at room temperature. The isolate from the decoction was identified as Bacillus licheniformis by 16S rDNA sequence analysis. Microbial population appeared after 3 months' preservation and reached maximum value at 4 months; at all tested temperatures, the pH showed the lowest value (4.4-4.5) simultaneously. Conclusion: From the results, it seems to be that the microbial growth affects the pH of preserved decoction but not the change of liquiritin and glycyrrhizin content.

Quality Characteristics of Bread with Citrus Peel Water Homogenate (감귤과피 물 균질액을 첨가한 빵의 품질특성)

  • Quality characteristics of bread added with water homogenate of citrus peel powder(WHCP, 1~5% of wheat flour) were investigated. The pH, the volume of dough, and the loaf volume index of bread added with WHCP were lower than those of the control. The contents of total carotenoids, hesperidin and naringin in the bread with 3% of WHCP were 0.14, 38.4 and 25.3 mg%, respectively. The bread with WHCP(3%) showed comparable cohesiveness and springiness, but higher hardness than the control product. The bread with WHCP(3%) showed the best quality evaluated by color and overall acceptability, taste, flavor and texture.

  • A high performance liquid chromatographic (HPLC) method for the simultaneous determination of hesperidin and glycyrrhizin was established for the quality control of traditional herbal medicinal preparation, Pyungwi-san (PWS). Separation and quantification were successfully achieved with a Waters XTerra RP18 column ($5{\mu}m$, 4.6 mm I.D. ${\times}$ 150 mm) by gradient elution of a mixture of acetonitrile and water containing 0.03% phosphoric acid (pH 2.03) at a flow rate of 1.0 ml/min. The diode-array UV/vis detector (DAD) was used for the detection and the wavelength for quantification was set at 230 nm. The presence of hesperidin and glycyrrhizin in this extract was ascertained by retention time, spiking with each authentic standard and UV spectrum. All four compounds showed good linearity $(r^2>0.995)$ in a relatively wide concentration ranges. The R.S.D. for intra-day and inter-day precision was less than 7.0% and the limits of detection (LOD) were less than 60 ng. The mean recovery of each compound was 99.0-105.6% with R.S.D. values less than 4.0%. This method was successfully applied to the determination of contents of hesperidin and glycyrrhizin in three commercial products of PWS. These results suggest that the developed HPLC method is simple, effective and could be readily utilized as a quality control method for commercial PWS products.

Characteristics of Flavonoids in Juice and Cluster Analysis of Satsuma Mandarin Germplasms (온주밀감 유전자원의 과즙 중 플라보노이드 특성 및 군집 분석)

  • This research was conducted to analyze flavonoids in juice and to investigate correlation between 60 germplasms of satsuma mandarin (Citrus unshiu Marc.) on the basis of the result of flavonoid analysis. Juice of satsuma mandarin contained 6 flavonoids, 2 flavanones such as narirutin and hesperidin, 1 flavone such as rutin and 3 polymethoxylated flavones such as sinensetin, tangeretin and nobiletin. Hesperidin content ranged from 41 to 196 mg/L. Narirutin content ranged from 25 to 230 mg/L. The average of rutin content was 2.2 mg/L but it was not detected in some cultivars. Polymethoxylated flavone which is known as a citrus specific flavonoid, was detected in all germplasms in small quantities. Cluster analysis using R program (Version 3.1.1) was carried out with the results of the flavonoid analysis and harvesting time of satsuma mandarin germplasms. All germplasms were grouped as A, B1 or B2. But all 3 groups contained very early-ripening type, early-ripening type and commonly-ripening type cultivars. This showed there was no correlation between flavonoids in juce and their harvest time.

Phytochemical Analysis of Viticis Fructus (만형자의 성분분석)

  • From the Viticis Fructus n-hydrocarbons, ${\beta}-sitosterol$ $3-O-{\beta}-_D-glucoside$ and hesperidin along with the known polyoxygenated flavonoids such as vitexicarpin, artemetin and luteolin, and vanillic acid were isolated and identified by means of spectroscopic methods. HPLC analysis of the flavonoid components from the MeOH extract was established. Phytochemical analyses of the domestic plant sample and the imported ones were conducted and the flavonoid compositions of the domestic samples were greatly different from those of the imported ones.
