Hesperidin (HES), a flavonone glycoside isolated from the citrus fruits such as lemons and oranges, has been reported to have many biological properties including antiinflammatory, antioxidant, and antiallergy activities. In this study, we focused on the action of HES modulating Th2-associated cytokines such as IL-4 and IL-13 expression in PMA/ionomycin (PI)-stimulated rat basophilic leukemia (RBL-2H3) cells. The production of IL-4 and IL-13 was quantified by ELISA and the mRNA expression was detected by using RT-PCR assay. In addition, western blot analysis was performed to determine the transcription factors involved in the cytokine expression. We found that HES significantly decreased PI-induced IL-4 and IL-13 productions and also decreased the level of mRNA in a dose-dependent manner. Furthermore, western blot analysis of the transcription factors implied that HES down-regulated the protein level of c-Jun and c-Fos, which are the activating protein 1 (AP-1) family and nuclear factor-kappaB (NF-${\kappa}B$) characterized as a transcription factors related to the Th2-associated cytokine expression. Taken together, our data showed that the action of HES responsible for antiallergy activities is based on suppression of Th2-associated cytokines through inhibition of AP-1 and NF-${\kappa}B$ transcription factors.

A total of 96 pigs [(Landrace ${\times}$ Yorkshire) ${\times}$ Duroc] with an initial BW of $27.94{\pm}0.92kg$ were used in a 6-week experiment to determine the effects of dietary supplementation with fruit by-product extracts on growth performance and nutrient digestibility in growing pigs. Pigs were randomly allotted to four treatments : 1) CON (basal diet), 2) PRO (CON + 0.5% procyanidin), 3) HES (CON + 0.5% hesperidin), 4) TAN (CON + 0.5% tannin). There were six replications per treatment with four pigs per pen. Supplementation had no effect (p>0.05) on average daily gain, average daily feed intake, and G/F. The apparent total tract digestibility (ATTD) of dry matter and nitrogen was increased (p<0.05) in the HES treatment relative to the PRO treatment. Pigs fed the HES and TAN diets had greater (p<0.05) ATTD of energy than pigs fed PRO diet. The ATTD of ash was greatest (p<0.05) in HES treatments. In addition, the ATTD of calcium was greater (p<0.05) in HES treatments than in CON and PRO treatments. Overall, the results of this study indicated that dietary supplementation with 0.5% fruit by-products did not affect growth performance, but inclusion of 0.5% hesperidin increased nutrient digestibility in growing pigs.
Atopic dermatitis (AD) is an allergic disorder characterized by skin inflammation. It is well known that the activation of various inflammatory cells and the generation of inflammatory molecules are closely linked to the development of AD. There is accumulating evidence demonstrating the beneficial effects of herbal extracts (HEs) on the regulation of inflammatory response in both in vitro and in vivo studies of AD. This review summarizes the anti-atopic effects of HEs and its associated underlying mechanisms, with a brief introduction of in vitro and in vivo experiment models of AD based on previous and recent studies. Thus, this review confirms the utility of HEs for AD therapy.
Hypereosinophilic syndrome (HES) is characterized by the presense of hypereosinophilia with evidence of target organ damage. We report a patient diagnosed with eosinophilic cystitis and HES. A 7 year old boy had hematuria, dysuria, and increased urinary frequency for 1 day. Laboratory examinations revealed hypereosinophilia (eosinophils, $2,058/{\mu}L$), hematuria, and proteinuria. Abdominal sonography revealed diffuse and severe wall thickening of the bladder. The patient was treated initially with antibiotics. However, his symptoms did not improve after 7 days. A computed tomography scan demonstrated severe wall thickening of the bladder and the hypereosinophilia persisted (eosinophils, $2,985/{\mu}L$). The patient complained of chest discomfort, dyspnea, epigastric pain, and vomiting on hospital day 10. Parasitic, allergic, malignancy, rheumatologic, and immune workups revealed no abnormal findings. Chest X-rays, electrocardiography, and a pulmonary function test were normal; however, the hypereosinophilia was aggravated (eosinophils, $3,934/{\mu}L$). Oral deflazacort was administered. A cystoscopic biopsy showed chronic inflammation with eosinophilic infiltration. The patient's respiratory, gastrointestinal, and urinary symptoms improved after 6 days of steroids, and he was discharged. The eosinophil count decreased dramatically ($182/{\mu}L$). The hypereosinophilia waxed and waned for 7 months, and the oral steroids were tapered and stopped. This case describes a patient diagnosed with eosinophilic cystitis and HES.
두 개의 별로 구성되었을 것이라고 여겨진 공생별은 밝기변화는 궤도운동에 따라 밝기가 변하는 것으로 알려졌다. 분광 관측 자료에는 이러한 궤도 변화 외에도 다양한 요소가 밝기 변화에 관여하는 것이 특성으로 나타난다. 또한 공생별은 밝기가 급격하게 증가하고 혹은 감소하기도 하는데, 이는 폭발에 기인하는 것으로 판단된다. 이러한 변화, 궤도 운동에 따라 기하학적 변화와 폭발 현상을 모두 볼 수 있는 대표적인 공생별이 Z And이다. 우리는 선행연구(MIKOLAJEWSKA & KENYON, 1996)에서 발표한 저분산 분광 자료를 이용하여 위상별로 변하는 상대적 선세기 변화를 조사하였다. MIKOLAJEWSKA & KENYON (1996)의 자료는 저분산 기기로(${\Delta\lambda\sim}3{\AA}$), HeI, HII, [OIII]5007, [NeV]등이 관측되었는데, 이러한 선들의 세기를 광이온 모델을 이용하여 예측한 후, 공생별 가스를 이온화시킨 중심별(WD)의 물리적 특성을 연구하였다. 또한 Hyung & Aller가 2002년 8월 12일 Lick Observatory에서 Hamilton Echelle Spectrograph (HES)를 사용하여 3600초 노출 관측한 고 분산 분광자료(${\Delta\lambda\sim}0.1{\AA}$)도 분석하였다. HES 관측 자료는 공생별의 위상이 $\Phi$=0.22이며, HES자료는 $3470{\AA}-9775{\AA}$에서 HI, HII, HeI, HeII, NII, NIII, OII, [OI], [OII], [OIII] 등이 있었다. 이 선들의 선 윤곽을 IRAF와 StarLink/Dipso를 이용하여 분석하고, 각 성분이 위상($\Phi$=0.22)인 상태에서의 관측자에 대해 어떠한 기하학적인 구조를 가지고 있는지 연구하였다. CLOUDY를 사용하여 광 이온 모형성운을 만들어 화학원소 및 성운가스의 물리적 조건을 연구하였다. Z And의 수소의 수밀도($N_H$)는 $10^{8.5}/cm^3$으로 가정하였다. 중심별 온도는 약 110,000K, 광도는 태양의 2000배로 추정되었다.
Notch signaling is controlled at multiple levels. In particular, stabilized Notch receptor activation directly affects the transcriptional activations of Notch target genes. Although some progress has been made in terms of defining the regulatory mechanism that alters Notch stability, it has not been determined whether Notch1/NICD stability is regulated by $GSK-3{\alpha}$. Here, we show that Notch1/NICD levels are significantly regulated by $GSK-3{\beta}$ and by $GSK-3{\alpha}$. Treatment with LiCl (a specific GSK-3 inhibitor) or the overexpression of the kinase-inactive forms of $GSK-3{\alpha}/{\beta}$ significantly increased Notch1/NICD levels. Endogenous NICD levels were also increased by either $GSK-3{\alpha}/{\beta}$- or $GSK-3{\alpha}$-specific siRNA. Furthermore, it was found that $GSK-3{\alpha}$ binds to Notch1. Deletion analysis showed that at least three Thr residues in Notch1 (Thr-1851, 2123, and 2125) are critical for its response to LiCl, which increased not only the transcriptional activity of endogenous NICD but also Hes1 mRNA levels. Taken together, our results indicate that $GSK-3{\alpha}$ is a negative regulator of Notch1/NICD.
배초향은 항동맥경화나 항박테리아의 특성을 가지는 한약재에 널리 사용되는 영년생 약용식물이다. 연구의 목적은 수경재배에서 배양액의 종류와 PPFD값에 따른 배초향의 생장 및 항산화 물질의 변화를 조사하는 것이다. 배초향은 주야간 16:8 시간의 일장조건에서 150과 $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD 조건과 일본원시(HES), 서울시립대(UOS), 유럽채소연구소(EVR), 오오츠카 배양액(OTS)을 이용하여 6주간 재배하였다. OTS 배양액조건에서 자란 배초향의 지상부 및 지하부 건물중은 다른 배양액 처리구와 비교하여 유의적으로 높았다. 배초향의 틸리아닌 함량은 OTS 처리에서 가장 높았으며 다음으로 EVR, HES, UOS 순서로 낮아졌다. 총 아카세틴의 함량은 EVR처리에서 가장 높았으나 OTS처리와는 유의적 차이를 보이지 않았다. 또한 $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD 조건에서 자란 배초향은 PPFD 150처리구와 비교하여 유의적으로 생체중과 건물중이 증가하였으며 기능성 물질은 틸리아닌과 아카세틴의 함량도 높았다. 본 연구는 수경재배 방식을 이용하여 식물공장에서 배초향을 재배할 경우 $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD 조건과 OTS 배양액 조건에서 경제적인 광원조건으로 최적 바이오매스 생산량과 틸라아닌과 아카세틴의 함량을 증가시킬 수 있을 것으로 제안한다.
For the development of $^{99m}Tc-labelled$ antimony sulfide colloid and hydroxyethyl starch, various experiments such as preparation of colloid, control of the distribution of particle size, establishment of labelling conditions, determination of labelling yield and radiochemical purity, examination of stability, and organ imagings of rabbits etc. were carried out. 1) Antimony sulfide colloid was readily prepared by the reaction of aqueous solution of antimony potassium tartrate with hydrogen sulfide generated by treating ferrous sulfide with dilute sulfuric acid. The colloid could be stabilized by adding small amount of polyvinylpyrrolidone. 2) Electron microscopy analysis exhibited the distribution of colloid size in the range of $1\sim15nm$ with a major portion of 9 m. The colloid solution was sterilized by membrane filtration $(0.2{\mu}m)$ and then stored at $4^{\circ}C$. This sterilized colloid was so stable that it was usable at least for one year. 3) The antimony sulfide colloid was labelled by adding sodium $pertechnetate-^{99m}Tc$ solution to the reaction vial, followed by adding hydrochloric acid and then boiled for 30 min. The optimal pH of the reaction mixture was found to be in the range of $1.3\sim1.4$. Instant thin layer chromatography (ITLC) analysis showed high labelling yield of above 99.5%. This labelled colloid maintained high radio-chemical purity of above 99% until 10 hours after labelling. 4) Animal studies showed high uptake of $^{99m}Tc-Sb_2S_3$ colloid at lymph vessels and nodes indicating a suitable agent for lymphoscintigraphy. Satisfactory results were also abtained in other clinical studies. 5) Hydroxyethyl starch (HES $0.6\sim1.0%$) was labelled with $Na^{99m}TcO_4$ in the presence of $SnCl_2$ with high labelling yield of above 99.5%. The optimal pH of the reaction mixture was in the range of $1.8\sim2.0$. $^{99m}Tc-HES$ maintained high radiochemical purity of above 99% until 10 hours after labelling. 6) Animal studies showed that $^{99m}Tc-HES$ migrated more rapidly from the injection sites into the lymph vessels than $^{99m}Tc-Sb_2S_3$ colloid while less amount of the former was uptaken at lymph nodes than that of the latter. Similar phenomenon was also observed in other clinical studies. As a result, $^{99m}Tc-Sb_2S_3$ colloid was found to be more effective lymphoscintigraphic agent than $^{99m}Tc-HES$.
본 연구는 국내 쇼트트랙 선수에서 다빈도로 발생하는 스포츠 손상에 대해 무릎과 발목관절 및 허리의 근력강화 운동과 신경근 훈련을 기반으로 한 스포츠 손상 예방프로그램 적용이 비접촉성 스포츠 손상의 발생률과 그 특징들에 미치는 영향을 알아보고자 하였다. G시청에 소속된 25명의 쇼트트랙 선수를 대상으로 2017년 11월부터 2018년 7월까지 스포츠 손상에 대한 정보 수집과 함께 스포츠 손상 예방프로그램을 매 훈련 및 시합 전 준비운동 단계에서 적용하였고, OSTRC 과사용 손상 설문지(overuse injury questionnaire)와 Cumberland Ankle Instability Tool(CAIT)를 사용하여 스포츠 손상의 증상 수준을 평가하였다. 그 결과, 스포츠 손상 예방프로그램 적용 후의 손상 발생률은 훈련 및 시합 1,000시간 노출(1,000HEs)당 2.79건으로 적용 전(3.04건/1,000HEs)보다 감소하였고, 허리와 무릎관절의 증상 수준인 OSTRC 점수는 프로그램 적용 후(각각 30.89±28.34점과 23.84±23.61점)가 적용 전(각각 58.47±26.77점과 52.36±21.55점)보다 낮게 나타나 증상이 완화된 것을 확인할 수 있었으며, 발목관절의 증상 수준인 CAIT점수가 프로그램 적용 후(16.26±7.28점)가 적용 전(13.47±6.07점)보다 높게 나타나 증상이 완화되었음을 확인하였다. 본 연구에서 사용한 쇼트트랙 선수들의 움직임과 특성에 적합하게 맞춤 설계된 스포츠 손상 예방프로그램이 쇼트트랙 선수들의 비접촉성 손상 발생에 긍정적인 영향을 미치는 것으로 판단된다.
