  • Equine herpesvirus myeloencephalopathy, out of symptoms by equine herpesvirus type 1 (EHV-1) infection, can cause devastating losses on individual farms. Although myeloencephalopathy syndromes of horses in Korea have been recognized for a couple of years in horse populations, there is little study regarding the occurrence of EHV-1 infections. The present study was performed to detect the viral infection of horses with neurological syndrome using real-time PCR. Fifteen horses (27.3%) out of 55 horses with neurological deficiency were positive for EHV-1 viral antigen. Among these 7 horses, 4 horses were detected genotype of A2254/N752 and 3 horses G2254/D752 strain, respectively.

한국에서의 canine herpesvirus감염실태에 대한 혈청역학적 조사 (Survey on the seroepidemiology of canine herpesvirus infection in Korea)

  • This study was carried out to investigate the actual condition of canine herpesvirus(CHV) infection in Korea. A total of 338 serum samples were collected randomly from the breeding and companion dogs in the local areas in Korea. The serum samples were used to determine the actual condition of the canine herpesvirus infection in Korea using enzyme linked immunosorbent assay(ELISA). The mean prevalence of CHV infection in dogs was 37% and that of the breeding and companion dogs was 58% and 28%, respectively. The prevalence of CHV infection in Seoul, Kyung-gi, Chung Nam, Cheon Nam and Pusan was detected 23%, 28%, 18%, 28% and 70%, respectively. The prevalence of CHV infection in less and more than 6 months old dogs, and in male and female dogs was 26% and 40%, 42% and 33%, respectively. These results indicate that the incidence of CHV infection is high in Korea, especially breeding dogs and older dogs.

굴의 Ostreid Herpesvirus 모니터링 (Monitering of Ostreid herpesvirus in Pacific oyster, Crassostrea gigas)

  • Monthly Screening of Pacific oyster, Crassostrea gigas, was surveyed by PCR for Ostreid herpesvirus(OsHV-1) infections from 2012 to 2013 in Korea. It was showed positive results in one hatchery in April and May 2012 and in oyster farming areas from August to November in 2013. The obtained sequences showed 99.3~99.5% homology with OsHV-1 ${\mu}var$(Accession Number: HQ842610) and 96.2~96.3% homology with OsHV isolated from Korea(Accession Number: AY509253).

Canine herpesvirus 감염증의 병리발생에 관한 연구 I. 병리조직학적 및 전자현미경적 관찰 (Study on the pathogenesis of canine herpesvirus infection I. Histopathological and electron microscopical observation)

  • This study was carried out to investigate the pathogenesis of canine herpesvirus(CHV) infection in dogs. The 17 puppies, one day old, delivered from CHV seronegative 3 dams were divided into two groups. The 13 puppies were inoculated intranasally with 1ml of CHV-KK inoculum($5{\times}10^{5.6}TCID_{50}/ml$) and 4 puppies were served as control. And then the puppies were sacrificed at 2, 4, 6 and 7 days after the treatment, and collected blood, nasal mucosa, trigeminal nerve, trigeminal ganglion, bone marrow, eye, brain and other major organs. These organs were examined histopathologically and electron microscopically. The platelets of puppies infected with CHV were dramatically decreased because of the damages of vascular endothelial cells. Histopathologically, necrotizing vasculitis and neuritis were proceeded the generalized focal necrosis of all organs. Necrotic changes in trigeminal ganglion, trigeminal nerve and ventroposteriomedial nucleus of thalamus were observed in 4 puppies infected with CHV. Herpesviral particles, various forms of maturation, were observed in endothelial cells of the alveolar capillary and hepatic sinusoid with electron microscopy. These results suggest that the generalized focal necrcsis of all organs including brain and eyes in canine herpesvirus infection were resulted from generalized vasculitis, and also the hemonecrotizing meningoencephalitis was related to the necrosis of trigeminal nerve pathway.

Genetic characterization of alloherpesvirus (cyprinid herpesvirus-2 and koi herpesvirus) and poxvirus (carp edema virus) identified from domestic and imported cyprinids in Korea

  • Cyprinids are popular species for aquaculture worldwide, with Asia being a significant contributor to their production. In Korea, common carp (Cyprinus carpio), koi carp (Cyprinus rubrofuscus), and goldfish (Carassius auratus) are cultivated domestically and imported for ornamental or human consumption purposes. Among the viruses that infect cyprinids, cyprinid herpesvirus-2 (CyHV-2), koi herpesvirus (KHV, also known as cyprinid herpesvirus-3), and carp edema virus (CEV) are of particular concern as they cause substantial economic losses to the aquaculture industry. In this study, we investigated these viruses in both of domestic and imported cyprinids. Our results revealed that CyHV-2 was only detected in imported goldfish from Thailand. To further investigate the genetic characteristics of them, the marker A region was analyzed. Despite belonging to the same cluster with isolates from China, France, Poland, and Israel, CyHV-2 detected in this study showed distinct differences in their repetitive sequence sizes. Furthermore, two different forms of KHV/CEV coinfection were identified from domestic koi carp, both of which exhibited typical symptoms. Phylogenetic analysis showed that one KHV isolate (ScKc-2105-K) was of the Asian type and closely related to isolates from Japan, Indonesia, Belgium, Taiwan, and China. Two CEV isolates (ScKc-2105-CE and GhKc-2207-CE) be- longed to the IIa type and showed high similarity with isolates from the USA, France, and Korea. Notably, koi carp injected with cultured KHV (ScKc-2105-K) showed 78.0% cumulative mortality within 14 days post-injection (dpi). Our findings support the importance of regular surveillance of viral diseases in cyprinids.

  • This study was carried out to investigate the pathogenesis of canine herpesvirus(CHV) infection in dogs. And then the puppies were sacrificed at 2, 4, 6 and 7 days after the treatment, and sampled nasal mucosa, trigeminal nerve, trigeminal ganglion, bone marrow, eye, brain and other major organs for the immunohistochemical examination. Distribution of CHV antigens was limited in cytoplasms and nuclei of necrotic nasal epthelia at 2 days after infection. At 4 days after infection, CHV antigens were detected in vascular walls and peripheral nerves of nasal lamina propria, reticuloendothelial cells of spleen, interstitium of kidney, leptomeningeal vascular walls and alveolar walls, At 6 and 7 days after infection, CHV antigens were detected in all of the necrotic area. CHV antigens were also detected in vascular endothelial cells of various organs and in blood leukocytes from 4 days after infection. Among the six puppies in which necrotic lesions of central nervous system were observed, CHV antigens were detected in trigeminal ganglion, trigeminal nerve and ventroposteriomedial nucleus of four puppies and in spinal trigeminal nucleus of three puppies. These results indicate that the generalized focal necrosis of all organs including brain and eyes in canine herpesvirus infection were resulted from generalized vasculitis with leukocyte-associated viremia, and also the hemonecrotizing meningoencephalitis was resulted from spreading of CHV via blood and nerve trunk.

  • Goldfish hematopoietic necrosis (GHN) affects hematopoietic organs and causes high mortality in goldfish. The causative agent of GHN is cyprinid herpesvirus 2 (CyHV-2). The purpose of this study was to detect CyHV-2 in ornamental cyprinid fish. CyHV-2 monitoring was conducted on a monthly basis for 1 year using goldfish Carassius auratus and pearlscale goldfish Carassius auratus that had been cultured at a domestic aquafarm and imported from Singapore, respectively. The results of polymerase chain reaction (PCR) assays using specific primers for the CyHV-2 helicase gene showed that 27.8% of goldfish and 33.3% of pearlscale goldfish were positive for CyHV-2. No cytophatic effects were detected in koi fin or common carp brain cells inoculated with tissue homogenates from either fish. Our data provides the useful data for establishing future quarantine and disinfection policies.

국내분리 말구진 원인병원체(Equine herpesvirus type 3)의 특징 (Characteristics on Equine Herpesvirus Type 3 from Korea)

  • Equine herpesvirus type 3 (EHV-3)는 말[마(馬)]에서 말구진을 유발하고 말산업에 치명적인 악영향을 끼치는 매우 중요한 병원체이다. 국내에서 처음으로 말구진이 발생하였고, 말 생산농가에 경제적 피해를 입혔으나 다행히 원인바이러스 분리에 성공하였다. 이에 원인바이러스의 생물학적 특징을 연구하여 다음과 같은 결과를 얻었다. EHV-3를 RK-13 cell에 접종한 결과 접종 48 시간부터 세포변성효과가 나타남을 확인하였고 접종 72 시간부터는 세포박리가 일어났다. 세포를 수확하여 바이러스 DNA를 추출한 후 PCR 검사를 수행하였다. 그 결과 EHV-3에만 양성이었고 EHV-1 및 EHV-4에는 음성이었다. Swab에서 바이러스 DNA를 추출하여 10배씩 serial dilution을 한 후 PCR 검사를 실시한 결과 $10^4$까지 밴드를 확인할 수 있었다. 국내분리 EHV-3의 gG 특정부위에 대한 염기에서 표준주인 EHV-3 334/74 strain과 397개가 일치하여 99.25%의 상동성을 나타내었고 국내분리주를 EHV-3 거로주(Georo strain)라 명명하였다. 전기영동 결과에서는 EHV-3가 EHV-1보다 적은 수의 밴드를 확인할 수 있었다. 국내 분리주 EHV-3 거로주의 단백질을 분석한 결과 145 kD, 60 kD, 45 kD, 40 kD에서 단백 밴드를 관찰하였다.

고려 수지침 요법의 항바이라스 및 세포재생 효과에 대한 탐색 (Exploration of Antiviral and Cell Regeneration Effects of the Korean Hand Acupuncture(Koryo Sooji Chim) Therapy)

  • 배경: 고려수지침을 이용한 항바이러스성 치료에 관한 연구는 전혀 없는 상태이다. 목적: Herpesvirus-2에 감염된 환자에 대한 수지침 자극에 대한 효과를 관찰하는 것이 목적이었다. 방법: 환자에 대한 수지침의 자극은 압봉 1호와 지압봉을 이용하여 수지 혈 자리에 부착하고, 5일간을 지나서 환부의 변화를 관찰하였다. 결과: 대상자 3명에 수지침혈 자리에 자극하면서 매일 관찰한 결과 20대의 환자는 3일째, 50대는 4일째, 그리고 70대는 5일째에 완치가 되었다. 결론: 수지침 자극으로 바이러스 감염으로 파괴된 세포가 재생되었고, 세포 내의 바이러스의 증식도 사라졌다. 수지침을 이용한 항바이러스성 치료가 유효하다는 것을 의미한다.

STP-C, an Oncoprotein of Herpesvirus saimiri Augments the Activation of NF-κB through Ubiquitination of TRAF6

  • Herpesvirus saimiri (HVS), a member of the $\delta$-herpesvirus family, encodes an oncoprotein called Saimiri Transforming Protein (STP) which is required for lymphoma induction in non-human primates. Previous study has shown that STP-C, an oncoprotein of HVS, activates NF-$\kappa$B signaling pathway. However, the detailed mechanism of STP-Cmediated NF-$\kappa$B activation has not been reported yet. We first report that STP-C interacts with TRAF6 protein in vivo and in vitro and further investigation shows that $Glu_{12}$ residue of STP-C is critical for binding to TRAF6. Introduction of ubiquitin together with STP-C augments NF-$\kappa$B activity compared to that of STP-C expression alone. STP-C expression further induces ubiquitination of endogenous TRAF6. In addition, either a deubiquitination enzyme, CYLD or a dominant negative E2-conjugation enzyme reduced NF-$\kappa$B activity in spite of the presence of STP-C, supporting that the interaction between STP-C and TRAF6 induces ubiquitination of TRAF6. NF-$\kappa$B activation by STP-C through the ubiquitinated TRAF6 causes the increased production of IL-8, an inflammatory chemokine and the enhanced expression of costimulatory molecule ICAM, which might ultimately contribute cellular transformation by the exposure of HVS-infected cells with inflammatory microenvironment and chronic activation.
