Effect of Spent Mushroom Substrates of Hericium erinaceum on Plant Pathogens of Tomato (노루궁뎅이버섯 수확후 배지 추출물의 토마토에 발생하는 식물병원균에 대한 생육억제 효과)

  • Water extract from spent mushroom substrate of Hericium erinaceus inhibited the mycelial growth of seven strain of tomato pathogenic fungi including Phytophthora capsici and the growth of Ralstonia solanacearum. Control efficacy of tomato bacterial wilt by treatment of 33.3% and 50% water extract from spent mushroom substrate of Hericium erinaceus was showed 58.3%, 83.3%, respectively.

  • Mushrooms are considered not only as food but also for source of physiologically beneficial medicines. The culinary-medicinal mushrooms may important role in the prevention of age-associated neurological dysfunctions, including Alzheimer's and Parkinson's diseases. Hericium erinaceus (H. erinaceus), is edible mushrooms, is a parasitic fungus that grows hanging off of logs and trees and well established candidate for brain and nerve health. H. erinaceus contains high amounts of antioxidants, beta-glucan, polysaccharides and a potent catalyst for brain tissue regeneration and helps to improve memory and cognitive functions. Its fruiting bodies and the fungal mycelia exhibit various pharmacological activities, including the enhancement of the immune system, antitumor, hypoglycemic and anti-aging properties. H. erinaceus stimulates the synthesis of Nerve Growth Factor (NGF) which is the primary protein nutrient responsible for enhancing and repairing neurological disorders. Especially hericenones and erinacines isolated from its fruitin body stimulate NGF, synthesis. This fungus is also utilized to regulate blood levels of glucose, triglycerides and cholesterol. H. erinaceus can be considered as useful therapeutic agents in the management and/or treatment of neurodegeneration diseases. However, this review focuses on in vitro, in vivo and clinical trials for neurodegerative disease.

  • Vegetative growth of four different strains of Hericium erinaceus was observed. The temperature suitable for optimal mycelial growth was determined to be $25^{\circ}C$, with growth observed in the extend temperature range of $20{\sim}30^{\circ}C$. The different strains of this mushroom showed distinct pH requirements for their optimum vegetative growth, with the most favorable growth observed at pH 6. Considering vegetative mycelial growth, PDA, YM, Hennerberg, Hamada, and Glucose peptone were the most favorable media, and Czapek Dox, Hoppkins, Glucose tryptone, and Lilly were the most unfavorable media for these mushroom strains. With the exception of lactose, most of the carbon sources assayed demonstrated favorable vegetative growth of H. erinaceus. For mycelial growth, the most suitable nitrogen source was alanine and the most unsuitable was histidine. Oak sawdust medium supplemented with $10{\sim}20%$ rice bran was the best for mycelial growth of the mushroom.

Water Extract from Spent Mushroom Substrate of Hericium erinaceus Suppresses Bacterial Wilt Disease of Tomato

  • Culture filtrates of six different edible mushroom species were screened for antimicrobial activity against tomato wilt bacteria Ralstonia solanacearum B3. Hericium erinaceus, Lentinula edodes (Sanjo 701), Grifola frondosa, and Hypsizygus marmoreus showed antibacterial activity against the bacteria. Water, n-butanol, and ethyl acetate extracts of spent mushroom substrate (SMS) of H. erinaceus exhibited high antibacterial activity against different phytopathogenic bacteria: Pectobacterium carotovorum subsp. carotovorum, Agrobacterium tumefaciens, R. solanacearum, Xanthomonas oryzae pv. oryzae, X. campestris pv. campestris, X. axonopodis pv. vesicatoria, X. axonopodis pv. citiri, and X. axonopodis pv. glycine. Quantitative real-time PCR revealed that water extracts of SMS (WESMS) of H. erinaceus induced expressions of plant defense genes encoding ${\beta}$-1,3-glucanase (GluA) and pathogenesis-related protein-1a (PR-1a), associated with systemic acquired resistance. Furthermore, WESMS also suppressed tomato wilt disease caused by R. solanacearum by 85% in seedlings and promoted growth (height, leaf number, and fresh weight of the root and shoot) of tomato plants. These findings suggest the WESMS of H. erinaceus has the potential to suppress bacterial wilt disease of tomato through multiple effects including antibacterial activity, plant growth promotion, and defense gene induction.

  • In recent years, Hericium erinaceus became famous for comsumer because it contains some materials to assist in human health. This study was carried out to find the difference of occurring pattern of H. erinaceus by pinheading induction methods. Harvest period of T1, T2 treatments were longer than T3~T6. All treatments fresh weights were not significantly different, but T3, T6 treatments's dry weight in 2nd flush were lower than others. T5 treatment's 2nd flush's regeneration rate was the highest than others and looks best quality. Therefore, it is suggested that pinheading induction method would be changed not T1, T2 treatment which were used some farm as Pluerotus ostreatus cultivation, but to scratch surface method which is T5 treatment for cultivation of high quality H. erinaceus.

  • Hot water soluble extract was prepared from Hericium erinaceus and its neuritogenic activity on PC12h cells was analyzed, which is a clone originating from a rat pheochromocytomon. The moisture content of freeze dried hot water extract was 12.08%. The extract was mainly composed of carbohydrate (51.24%) followed by crude protein (24.04%), crude fat (0.26%), dietary fiber (5.09), and ash (12.18%). Fatty acids, glucan and inorganic constituents were found as minor components. The neuritogenic activity of hot water extract was evaluated under microscopic observation of neurite outgrowth in PC12h cells and by measuring the neurite length of induvidual cell. The extract exhibited strong effect of neurite outgrowth in a dose-dependent manner from 0.01 mg/mL to 1 mg/mL, in which longer neurite outgrowth was observed as the treatment dose increased.

  • The yields and the biological efficiency of Hericium erinaceus in the case of the bottle cultivation was 356 g/850 ml, 147.8% and in pot cultivation was 810 g/2,500 ml, 114.3% respectively. On the primordial formation the case of no removing inoculation spawn was well introduced, and the mushroom's yield and biological efficiency was to be high in the case of the bottle's cap was shut. The results in the use of the logs for Hericeum erinaceus's cultural media were to be fine in oak, alder, poplar, black locust in order. And in oak, the yield of the mushrooms were $1195.5\;g/0.1\;m^3$ biological efficiency was 17.3%, the period requirements for primordium was 69 days, and the mushroom's individual weight was 143 g.

Phylegenetic analysis of Hericium species based on ITS rDNA sequences (ITS 분석을 이용한 노루궁뎅이버섯 수집균주의 계통분류)

  • The aim of this study was to analyze the genetic diversity of Hericium species based on their rDNA ITS sequences. Hericium species were collected from various regions and the size of the ITS rRNA gene regions from different Hericium species varied from 450 to 500 bp. A phylogenetic trees based on the ITS region revealed that Hericium species could be classified into 4 different groups, H. erinaceus, H. coralloide, H. alpestre, H. americanum. Among them, ASI 48015 and ASI 48016 was identified as Sprassis and Lentinula genus, respectively, based on blast searches using their rDNA ITS sequences.

Control of Tomato Bacterial Wilt by the Prototypes Extracted from Spent Media Substrate of Hericium erinaceus (노루궁뎅이 수확 후 배지 추출한 시제품의 토마토 풋마름병 방제 효과)

  • The prototypes, Rto-x+$Ca(NO_3)_2$, K-Ca, and Rto-$x{\beta}$, which were extracted from spent mushroom substrate of Hericium erinaceus, strongly inhibited the growth of Ralstonia solanacearum. The control efficacies of tomato bacterial wilt by treatment with Rto-x+$Ca(NO_3)_2$, K-Ca, and Rto-$x{\beta}$ were 87.4~92.4%, 46.0~100%, and 65.0~91.9%, respectively. These results indicate that Rto-x+$Ca(NO_3)_2$, K-Ca, and Rto-$x{\beta}$ have a potential as eco-friendly antibacterial materials for the control of tomato bacterial wilt caused by R. solanacearum.

  • Water extracts from spent mushroom substrate (SMSE)of edible mushrooms, Pleurotus eryngii, Hericium erinaceus and Lentinula edodes promoted growth of pepper seedling. Mycellial growth rate of Phythopthora capsici and Fusarium oxysporum was dramatically inhibited by 100% and 70% on PDA added with SMSE of H. erinaceus. SMSEs from H. erinaceus, P. eryngii, and L. edodes effectively reduced the disease severity of Phytophthora blight of pepper caused by Phytophthora capsici to 75%, 10% and 35%, respectively. These results suggested that SMSE from the mushrooms have dual effects that suppress phythopthora blight disease and promote plant growth of pepper.
