Interaction of Pyrazole - and Chloroacetamide Herbicide Combinations in Control of Echinochloa crusgalli (Pyrazole계(系)와 Chloroacetamide계(系) 제초제(除草劑)들의 혼합처리(混合處理)가 피(Echinochloa crusgalli)의 살초효과(殺草效果)에 미치는 상호작용(相互作用))

  • Three pyrazole-herbicides, pyrazolate, pyrazoxyfene and benzophenap, were evaluated for their interaction in controlling barnyardgrass (Echinochloa crusgalli) with two chloroacetamide-herbicides, butachlor and pretilachlor. Percent inhibition of barnyardgrass growth by pyrazolate, pyrazoxyfene, and benzophenap was 44%, 64%, and 0%, respectively, when each was applied at the 1.5 leaf-stage of barnyardgrass at a rate of 3㎏ ai per ㏊ as single treatment, and the benzophenap showed 60% inhibition when it was applied at the coleoptile stage. While the lowest rate controlling the 1.5 leaf-stage barnyardgrasses by 98 to 100% of the butachlor and pretilachlor was 1.5㎏ and 200g per ㏊, respectively. All of the combinations of pyrazolate with butachlor, pyrazoxyfene with pretilachlor, and benzophenap with butachlor have shown synergistic interaction in controlling barnyardgrass on the Chisaka's isobole of 90% growth inhibition as well as on the Colby`s interaction efficacy data; synergism indices were 2.44, 1.62 and 1.52 in order. The dose combinations shown the maximal synergism were 1870g of pyrazolate with 140g of butachlor (1:0.075), 33008 of pyrazoxyfene with 338 of pretilachlor (1:0.01), and 3350g of benzophenap with 520g of butachlor (1:0.15) on the ai/㏊ basis.

Studies on the Sprouting Characteristics of Scirpus planiculmis and Storage Methods of Corm for the Herbicide Screening (새섬매자기(Scirpus planiculmis)의 출아특성(出芽特性)과 제초제(除草劑) 스크리닝을 위한 구경(球莖)의 보관방법(保管方法))

  • The sprouting characteristics of Scirpus planiculmis and storage methods of corm for the herbicide screening were investigated under both laboratory and greenhouse conditions. Sprouting and elongation of S. planiculmis corms were continued during storage at low temperature ($5^{\circ}C$) and wet condition. However, sprouting of corm was failed when the surface water was removed from the corm, and elongation was reduced by the treatment of 0.5-1.0% sodium chloride(NaCl) solution. Alternate temperature was more effective on the sprouting of S. planiculmis corm than constant temperature. Optimum temperature for the sprouting of S. planiculmis corm was alternation between $25^{\circ}C$ to $35^{\circ}C$. Storage at $-20^{\circ}C$ for 10 min. or 14.7% dehydration made the corm of S. planiculmis to fail for sprouting. S. planiculmis corm was more tolerant to sodium chloride than Echinochloa crus-galli or Oryza sativa(Donginbyeo) seeds. Sprouting rate was decreased by cutting of the corm. Sprouting percent by vertical cuttings, horizontal cuttings, and vertical plus horizontal cuttings were 72, 56, and 28%, respectively.

Leaching behavior of the herbicide bentazon in soil column (Soil column중 제초제 bentazon의 용탈 행적)

  • In order to elucidate the leaching behaviour of the herbicide bentazon in soil, soil columns(5cm ID ${\times}$ 34 cm L) were packed with three different soils up to 30 cm height, followed by the treatment of [$^{14}C$]bentazon, and rice plants(Oryza sativa L.) were grown for 9 weeks on these columns, with the columns without growing rice plants as the control for comparison. The amounts of $^{14}C$ activities percolated were about 92% of the originally applied $^{14}C$ irrespective of the physicochemical properties of the soils in the absence of rice plants, whereas $21{\sim}50%$ of the originally applied $^{14}C$ was percolated in the presence of rice plants, suggesting that the amounts of $^{14}C$ leached decreased remarkably in rice-cultivating soils. Bentazon leached faster in soil with higher pH and with lower organic matter content in the presence of growing rice plants. The amounts of $^{14}CO_{2}$ evolved from the soil columns were less than 0.2% of the originally applied $^{14}C$. Smaller amounts of $^{14}C$ were translocated into shoots via roots in soils with higher organic matter content. $^{14}C$ activities distributed into the aqueous phase of the leachate collected from the soil columns increased with leaching period and by rice cultivation, whereas the physico-chemical properties of soils did not exhibit any effect. The amounts of soil-bound residues increased remarkably by cultivating rice plants.

A Study on the Case Analysis and Health Management of Patients with Pesticide Poisoning from Spraying Pesticide in Hospitals in the Chungnam, Korea (충남 일부 지역 병의원의 농약살포 중 중독 사례 분석 및 보건관리방안 연구)

  • Objectives: This study aims to examine patients who developed acute pesticide poisoning while spraying or using pesticide and presented to the emergency department in hospitals in Chungcheongnam-do Province. Based on the findings, this study will provide implications for safety and health management pertaining to the use of pesticides. Methods: Pesticide poisoning data collected by the Chungnam Center for Farmers' Safety and Health from 2014 to 2018 was cross-sectionally analyzed. A total of 331 patients with pesticide poisoning presented to one of hospitals and four of medical centers in the region(Dankuk University Hospital, Gongju and Hongsung, Cheongyang, Cheonan Medical Center). Seventeen of these patients (15 men and two women) developed poisoning while spraying pesticide. The patients' charts were reviewed to collect data on pesticide poisoning, namely currently working in farming-related occupation, means of transportation to the hospital, place of poisoning, symptoms of acute pesticide poisoning, treatment, pesticide used when poisoning occurred, and classification by technical ingredients. Results: Fifteen out of 17 patients who developed poisoning while spraying pesticide were men. Insecticide was used in 35.3% of the cases, and herbicide was used in 29.4% of the cases, which was different from cases of poisoning from ingestion poisoning. The major symptoms were vomiting (35.3%), nausea (29.4%), dizziness (29.4%), and headache (23.5%). A total 11 ingredients were identified in 12 patients, and the most common ingredient was glyphosate, which is an herbicide. Most patients showed a state of mild toxicity, but two patients showed a state of severe toxicity. These patients respectively used glyphosate and paraquat. Conclusions: Our findings can be useful for suggesting the need for a national healthcare system to manage occupational pesticide poisoning among farmers. Further, these findings can be used to increase the awareness of the risk of acute poisoning during pesticide spraying and suggest the need for a safety health education to increase farmers' awareness of pesticide poisoning.

Persistence and degradation of herbicide molinate in paddy-soil environment (논토양 환경 중 제초제 molinate의 잔류성과 분해특성)

  • The herbicide molinate has been detected with high frequency in the main river during the growing season in Korea. To elucidate the exposure of molinate in agricultural environment, the persistence and the degradation characteristics of molinate were investigated in paddy ecosystems. The half-lives of molinate were 4.1 days with soil aquatic system, and 4.2 days in only aquatic system. Initial dissipation rate of molinate in water was greater with soil aquatic system than that of only aquatic system. Photolysis of molinate was occurred about 31.0% of molinate treated in pure water, when irradiated at 5,530 $J/cm^2$ by the xenon lamp, but its hydrolysis was stable. For the accelerated photolysis of molinate in aqueous solution, several photosensitizers were screened, showing that the hydroperoxide($H_2O_2$) and acetone were prominent among the chemical tested. When hydroperoxide and zinkoxide(ZnO) were used as photosensitizer, their photolysis were accelerated greater than 98% and 58% in aqueous solution, respectively. Elution rate of molinate as granular formulations in aqueous system was more than 90% in 30 hour at $35^{\circ}C$. Molinate concentration pattern in paddy water was rapidly decrease from treatment till 7 days in paddy rice field and its half-lives were 3.7 days($Y=1.9258{\times}e^{-0.1865X}$(r=-0.9402)).

Herbicide Resistance in Plant Tissue Culture (식물체(植物體) 조직배양(組織培養)과 제초제(除草劑) 저항성(抵抗性))

  • This study was conducted to evaluate herbicide resistant plant through tissue culture. Callus was induced from embryos of Echinochloa crusgalli Beauv. (var, oryzicola Ohwi, var. caudata Kitagawa and var, crusgalli). An optium medium for callus induction and succinate dehydrogenase activity in inducted callus were detected and callus growth of various varieties of Echinochloa crusgalli was assessed under the treatment of various rates of butachlor[N-(butoxymethyl)-2-chloro-N-(2,6-diethylphenyl)acetamide]. MS medium seemed to be the most appropriate to induce callus from the embryos of varieties of E. crusgalli by using 2,4-D about 5.5mg/l as a hormone source. The activity of succinate dehydrogenase in inducted callus showed positive reaction against to TTC(2,3,5-triphenyltetrazolium chloride) regardless of concentrations of butachlor and varieties of E. crusgalli, indicating that all the callus induced were alive. The callus growths derived from seeds of E, cnesgalli were greatly affected by various rates of butachlor and were completely inhibited at the highest concentration of butachlor, $10^{-3}M$, regardless of varieties of E. crasgalli. $10^{-6}M$ of butachlor inhibited 24.6% of the callus growth of E. crusgalli Beauv, var. oryzicola Ohwi, while E. crusgalli Beauv. var. crusgalli showed 42% of inhibition, showing that there was difference in response of varieties of E. crusgalli Beauv. to butachlor.

Characteristics of Sensitive HIS1 Genes to the 4-HPPD Inhibiting Rice Herbicides Isolated from Several Rice Cultivars (몇 가지 벼 품종으로부터 분리한 4-HPPD저해 제초제에 감수성인 HIS1 유전자 특성)

  • This study was conducted to determine phytotoxicity of domestic rice varieties and characteristic of sensitive gene involved in herbicide reaction of 4-HPPD (4-hydroxy phenylpyruvate dioxygenase) inhibiting herbicides. The five rice varieties were grown for 2 to 3 leaf stage on seedling trays and then transplanted into plastic pot: 4-Japonica type (Sangjubyeo, Sambaekbyeo, Sanduljinmi and Kumyoung) varieties and 1-Indica type variety (IR8). We trialled standard (14 g a.i. $10a^{-1}$) and double fold (28 g a.i. $10a^{-1}$) dose of benzobicyclon treatment at 10 days after transplanting in order to investigate phytoxicity. The Japonica-type Sangjubyeo showed no rice injury but Indica-type IR8 show 4-5 (standard) and 5-6 (double fold) phytoxicity levels. In spite of Japonica-type, Sanduljinmi and kumyoung showed 3-4 and 4-5 levels. Target resistant gene, Hydroxyphenylpyruvate dioxygenase inhibitor sensitive gene No.1 (HIS1) have been isolated from five domestic rice cultivars (Sangjubyeo, Sambaekbyeo, Sanduljinmi, Kumyoung and IR8). Results of the sequence through PCR, all five tested rice cultivars had HIS1 gene regardless to rice cultivars. And the difference between rice varieties from sequence of HIS1 were identified some variation in genes.

Trait Variation and Molecular Characterization of Ds insertional rice lines (벼 Ds 삽입변이 계통의 특성변이 및 분자생물학적 특성)

  • The 1,874 rice lines were selected from 3,000 Ds insertional mutant pool by Basta herbicide treatment and were surveyed for trait variation and molecular characteristics of genes knocked out by Ds insertion. Compared with "Donjin", an original japonica cultivar used for transformation, Ds insertion mutant pool showed large variation in major agronomic traits including tiller, panicle, and heading etc. Southern blot analysis demonstrated that these lines on the average had two Ds copies in Donjin genome, resulting in 38.4% of one copy, 32.5% of two copies, 16.7% of three copies, and 11.3% of over four copies. GUS analysis showed that 3.9% of lines (73/1,860) had tissue-specific expression in leaves, nodal parts, floral organs such as stigma and pollen, and roots. Data set obtained from agricultural trait variation and molecular characteristics for individual Ds insertional lines would provide researchers with more information for understanding the function of unknown rice genes controlling economically important traits.

Molecular physiological inhibitory effects of chloroacetanilide herbicide pretilachlor on marine dinoflagellate Prorocentrum minimum (해양 와편모조류 Prorocentrum minimum에 대한 아세트아닐라이드계 제초제 프레틸라클로르의 분자 생물학적 저해 효과)

  • Pretilachlor (PRE) is a common acetanilide herbicide used worldwide. However, its effects on aquatic organisms, particularly marine photosynthetic life, are not sufficiently known. Herein, we evaluated the toxic effects of PRE by physiological and molecular parameters in the photosynthetic dinoflagellate Prorocentrum minimum. The cell density, pigment content, and photosynthetic parameters (Fv/Fm and PIABS) were considerably decreased with increased PRE exposure time and doses. In addition, photosynthesis-related genes, PmpsbA, PmpsaA, and PmatpB, were significantly upregulated when exposed to 1.0 mg L-1 of PRE for 24 h (p<0.001). In 72 h treatment, the relative gene expression was significantly increased (0.1 and 0.5 mg L-1; p<0.01). In contrast, PmrbcL was decreased or little changed compared to the controls. Reactive oxygen species (ROS) increased after 24 h exposure (p<0.001). However, the transcriptional fold-changes in glutathione S-transferase (GST) were significantly increased (0.5 and 1.0 mg L-1; p<0.001) at 72 h. These findings suggested that the PmGST might be involved in PRE detoxification in P. minimum. In addition, PRE may affect the photosystem function in phytoplankton similar to other acetanilides, causing severe damage or cell death.

Agrobacterium-mediated transformation produces transgenic oilseed rape with a high-yield trait (아그로박테리움 형질전환법을 이용한 수량증대 유채 식물체 개발)

  • This study includes the transformation of genes such as ORE7, the increase of gene expression, and the use of the bar gene as a selectable marker that shows herbicide resistance with Agrobacterium tumefaciens using hypocotyls from the oilseed rape "Youngsan" cultivar. To establish an Agrobacterium transformation system for the production of oilseed rape with a high-yield trait, infection time and co-cultivation period with Agrobacterium were tested. Therefore, when hypocotyls from the oilseed rape "Youngsan" cultivar were infected with Agrobacterium for 20 min and co-cultivated for 3 days, approximately 32-36 putatively transformed hypocotyls with shoots including roots survived from 100 inoculated hypocotyls after 4 weeks of transformation on a selection medium containing 20 mg/L of phosphinothricin (PPT) as a selectable agent. Additionally, a PCR assay was performed to confirm the insertion of target genes and showed the presence of the ORE7 gene as a high-yielding trait and the bar gene as a selectable marker. Treatment with 0.5% (v/v) Basta solution as a selectable agent for 6 days with leaves from transformed oilseed rape expressed the bar gene. Therefore, this study can contribute to the development of special oilseed rapes containing agriculturally useful traits such as herbicide resistance, drought tolerance, high yielding traits, and high oleic acid content.
