Regulation of CYP 1A1 gene expression by retinoic acid receptor, retinoid X receptor and constitutive androstane receptor in rainbow trout hepatoma cells(RTH 149)

  • Exposure of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) causes a variety of biological and toxicology effects, most of which are mediated by aryl hydrocarbon receptor (AhR). The ligand-bound AhR as a heterodimer with AhR nuclear translocator (ARNT) binds to its specific DNA recognition site, the dioxin-responsive element (DRE), and it results in increased transcription of CYP1A1 gene. Retinoic acid (RA) regulates the transcription of various genes for several essential functions through binding to two classes of nuclear receptors, the retinoic acid receptor (RAR) and retinoid X receptor (RXR). Constitutive androstane receptor (CAR) also regulates the transcription of gene. In this study, we have examined how RAR, RXR and CAR regulated CYP1A1 in rainbow trout hepatoma cell (RTH 149) using luciferase reporter gene assay system. We did transient transfection with CYP1A1 luciferase reporter gene and treated with TCDD, all-trans RA, 9-cis RA and phenobarbital. Treatment of all-trans RA, 9-cis RA or phenobarbital decreased the TCDD induced transcription of CYP1A1. When we did transient cotransfection with CYP1A1 luciferase reporter gene and RXR, as increase of RXR concentration, the TCDD induced transcription of CYP1A1 was decreased. Transfection with CAR also decreased the TCDD induced transcription of CYP1A1 in RTH 149 cells.

녹두 및 대두추출물의 항암 및 항염증 활성 (Anti-cancer and Anti-inflammatory Effects of Mung Bean and Soybean Extracts)

  • 본 연구에서는 녹두 및 대두의 용매추출물을 제조하고 hepatoma 1c1c7 세포 및 대식세포인 RAW264.7 배양을 이용하여 항암 및 항염증 효과를 비교분석하였다. 녹두추출물은 대두추출물 보다 ethylacetate 및 ethanol 추출물에서 모두 유의적으로 높은 QR 유도활성과 NO 및 $PGE_2$의 생성 억제 효과를 보였으며 농도 의존적 효과를 나타냈다. C18 silica flash column chromatography를 이용하여 회수된 녹두에탄올 추출물의 7개 분획 중 소수성이 가장 강한 분획은 24 ${\mu}g$/mL의 CD value와 783 ${\mu}g$/mL의 $IC_{50}$를 보였다. 현재 다수의 염증 억제물질의 작용기전이 prostaglandin 합성 억제이며 이는 COX-2의 생성 및 활성저해에 의한 것임을 근거로 할 때 녹두의 ethanol 추출물은 항염증 및 암 발생 예방 성분을 분리하기 위한 중요한 식품 자원으로 생각된다.

Histone Deacetylase Inhibitor Stimulate CYP3A4 Proximal Promoter Activity in HepG2 Cells

  • The expression of CYP3A4 gene is induced by a variety of structurally unrelated xenobiotics including the antibiotic rifampicin, pregnenolone 16-carbonitrile (PCN), and endogenous hormones, that might mediate through steroid and xenobiotic receptor (SXR) system. The molecular mechanisms underlying regulation of CYP3A4 gene expression have not been understood. In order to gain the insight of the molecular mechanism of CYP3A4 gene expression, study has been undertaken to investigate if the histone deacetylation is involved in the regulation of CYP3A4 gene expression by proximal promoter in human hepatoma HepG2 cells. Also we have investigated to see if SXR is involved in the regulation of CYP3A4 proximal promoter activity in human hepatoma HepG2 cells. HepG2 cells were transfected with a plasmid PCYP3A4-Luc containing ${\~}1kb$ of the CYP3A4 proximal promoter region (-863 to +64 bp) in front of a reporter gene, luciferase, in the presence or absence of pSAP-SXR. In HepG2 cells, CYP3A4 inducers, such as rifampicin, PCN and RU486 showed minimal stimulation of CYP3A4 proximal promoter activity in the absence of SXR and histone deacetylase (HDAC) inhibitors. 4-Dimethylamino-H-[4-(2-hydroxycarbamoylvinyl)benzyl]benzamide (IN2001), a new class HDAC inhibitor significantly increased CYP3A4 proximal promoter activity over untreated control cells and rifampicin concomitant treatment with IN2001 increased further CYP3A4 proximal promoter activity that was stimulated by IN2001 The results of this study demon-strated that both HDAC inhibitors and SXR are essential to increase of CYP3A4 proximal promoter activity by CYP3A4 inducers such as PCN, rifampicin, and RU486. Especially SXR seems to be important for the dose dependent response of CYP3A4 inducing chemicals to stimulate CYP3A4 proximal promoter activity. Also this data suggested that HDAC inhibitors seemed to facilitate the CYP3A4 proximal promoter to be activated by chemicals.

인간 간암세포주 HepG2에서 heme oxygenase-1 발현에 대한 diallyl disulfide의 효과 (Effect of Diallyl Disulfide on Heme Oxygenase-1 Expression in Human Hepatoma Cell Line HepG2)

  • Dially disulfide (DADS)는 마늘의 주요한 유기 황화합물 성분으로서 다양한 약리 작용을 나타낸다. 최근 DADS가 항염증과 항동맥경화 작용뿐만 아니라 암세포의 증식을 억제하고 사멸을 유도한다는 보고가 이어지고 있고, 이에 관련된 연구가 활발히 진행되고 있는 실정이다. 한편, DADS가 세포 내 항산화 인자인 glutathione을 증가시킨다는 연구결과와 세포 내 항산화 효소의 일종인 HO-1의 발현을 직접 유도한다는 결과가 보고되었다. 그래서, 본 연구에서는 논란이 되고 있는 DADS의 세포 내 항산화 효소인 HO-1의 발현에서의 효과 및 그 전사인자들의 작용에 관여하는지를 인간 간암세포주 HepG2에서 조사하였다. 배양 중인 HepG2 세포에서 DADS는 독성이 없는 농도에서 세포의 증식을 크게 억제하였고, 전사인자 Nrf2의 발현을 약하게 유도하였으나 HO-1의 발현에는 영향을 미치지 못하는 것으로 나타났다. 또한, DADS는 HO-1 유도제인 CoPP와 hemin에 의해 자극된 HepG2 세포의 HO-1 발현의 증가를 단백질 수준에서 강력하게 억제시키는 것으로 나타났다. 그러나 DADS는 CoPP에 의한 HO-1 유전자의 mRNA 수준의 전사에는 억제 효과를 보이지 않았으며, 또한 Nrf2와 small Maf의 발현을 증가시키고 핵 내에 축적시키는 것으로 나타났다. 이를 종합해 볼 때 DADS는 단독으로 HO-1 발현을 유도하지 못하고, HO-1 유도제에 의한 HO-1 유전자의 발현과정에서는 전사단계가 아닌 번역단계에서 역할을 함으로써 HO-1의 단백질 합성을 억제하는 것으로 보인다. 결론적으로, 항산화 효소인 HO-1의 활성은 외부 자극으로부터 세포를 보호하고 사멸에 저항하게 하는데, DADS는 인간 간암세포주 HepG2에서 이 효소의 발현을 억제함으로써 항암제 및 redox 변화에 따른 암세포주의 성장을 억제하고 세포사멸을 촉진시킬 수 있다고 여겨진다.

Different Immunology Mechanisms of Phellinus igniarius in Inhibiting Growth of Liver Cancer and Melanoma Cells

  • To assess inhibition mechanisms of a Phellinus igniarius (PI) extract on cancer, C57BL/6 mice were orally treated with PI extractive after or before implanting H22 (hepatocellular carcinoma ) or B16 (melanoma) cells. Mice were orally gavaged with different doses of PI for 36 days 24h after introduction of H22 or B16 cells. Mice in another group were orally treated as above daily for 42 days and implanted with H22 cells on day 7. Then the T lymphocyte, antibody, cytokine, LAK, NK cell activity in spleen, tumor cell apoptosis status and tumor inhibition in related organs, as well as the expression of iNOS and PCNA in tumor tissue were examined. The PI extract could improve animal immunity as well as inhibit cancer cell growth and metastasis with a dose-response relationship. Notably, PI's regulation with the two kinds of tumor appeared to occur in different ways, since the antibody profile and tumor metastasis demonstrated variation between animals implanted with hepatocellular carcinoma and melanoma cells.

Caffeine attenuates lipid accumulation via activation of AMP-activated protein kinase signaling pathway in HepG2 cells

  • The main purpose of this study is to examine the effect of caffeine on lipid accumulation in human hepatoma HepG2 cells. Significant decreases in the accumulation of hepatic lipids, such as triglyceride (TG), and cholesterol were observed when HepG2 cells were treated with caffeine as indicated. Caffeine decreased the mRNA level of lipogenesis-associated genes (SREBP1c, SREBP2, FAS, SCD1, HMGR and LDLR). In contrast, mRNA level of CD36, which is responsible for lipid uptake and catabolism, was increased. Next, the effect of caffeine on AMP-activated protein kinase (AMPK) signaling pathway was examined. Phosphorylation of AMPK and acetyl-CoA carboxylase were evidently increased when the cells were treated with caffeine as indicated for 24 h. These effects were all reversed in the presence of compound C, an AMPK inhibitor. In summary, these data indicate that caffeine effectively depleted TG and cholesterol levels by inhibition of lipogenesis and stimulation of lipolysis through modulating AMPK-SREBP signaling pathways.

PU.1 Is Identified as a Novel Metastasis Suppressor in Hepatocellular Carcinoma Regulating the miR-615-5p/IGF2 Axis

  • Invasion and metastasis is the major cause of tumor recurrence, difficulty for cure and low survival rate. Excavating key transcription factors, which can regulate tumor invasion and metastasis, are crucial to the development of therapeutic strategies for cancers. PU.1 is a master hematopoietic transcription factor and a vital regulator in life. Here, we report that, compared to adjacent non-cancerous tissues, expression of PU.1 mRNA in metastatic hepatocellular carcinoma (HCC), but not primary HCC, was significantly down-regulated. In addition, levels of PU.1 mRNA in metastatic hepatoma cell lines MHCC97L and MHCC97H were much lower than in non-metastatic Hep3B cells. Transwell invasion assays after PU.1 siRNA transfection showed that the invasion of hepatoma cell lines was increased markedly by PU.1 knockdown. Oppositely, overexpression of PU.1 suppressed the invasion of these cells. However, knockdown and overexpression of PU.1 did not influence proliferation. Finally, we tried to explore the potential mechanism of PU.1 suppressing hepatoma cell invasion. ChIP-qPCR analysis showed that PU.1 exhibited a high binding capacity with miR-615-5p promoter sequence. Overexpression of PU.1 caused a dramatic increase of pri-, pre- and mature miR-615-5p, as well as a marked decrease of miR-615-5p target gene IGF2. These data indicate that PU.1 inhibits invasion of human HCC through promoting miR-615-5p and suppressing IGF2. These findings improve our understanding of PU.1 regulatory roles and provided a potential target for metastatic HCC diagnosis and therapy.

포공영의 자유라디칼 소거 및 간세포 보호활성 (In Vitro Free Radical Scavenging and Hepatoprotective Activities of Taraxacum mongolicum)

  • The methanol (MeOH) extract and ist fractions of Taraxacum mongolicum (Compositae) were examined for their scavenging effects on 1,1-diphenyl-2- phenylhydrazyl (DPPH) and superoxide radicals, and hepatoprotective effects on tacrine-induced cytotoxicity in human hepatoma cell line, Hep G2 cells. Both methylene chloride $(CH_2Cl_2)$ and butanol (n-BuOH) soluble fractions of the MeOH extract showed the free radicals scavenging and hepatoprotective effects. From these results, it is suggested that hepatoprotective effect of these fractions partly relies on their free radical scavenging activity.

Induction of Quinone Reductase Activity by Stilbene Analogs in Mouse Hepa 1c1c7 Cells

  • Based on the potential cancer chemoprebentive activity of resveratrol, a trihydroxystilbene with the induction of quinone reductase activeity this study was designed to determine if stilbene-related compounds were inducers of phase ll detoxifying metabolic enzyme quinone reductase (QR) in the mouse hepatoma Hepa 1c1c7 cells. Among the thirteen compounds tested, several compounds including 3,4,5,3',5'-pentamethoxy-trans-stibene were found to potentially induce QR activity in this cell line. In addition, substitution with 3-thiofurane ring instead of phenyl ring in the stilbene skeleton also exhibited potential induction of QR activity. This result will give primary information to design the potential inducers of QR activity in the stilbene analogs.
