자성 흰쥐의 파라치온 급성독성 및 대사에 미치는 페노바르비탈 및 SKF-525-A의 영향 (EFFECT OF PHENOBARBITAL AND / OR SKF 525-A ON THE METABOLISM AND ACUTE TOXICITY OF PARATHION IN ADULT FEMALE PATS)

  • Effects of altering hepatic mixed-function oxidase (MFO) enzyme activities on the metabolism and acute toxicity of parathio were investigated in adult female rats. In vitro hepatic metabolism of parathion to paraoxon was increased by phenobarbital pretreatment (50 mg/kg/day, ip, for 4 consecutive days) and SKF 525-A (50 mg/kg, ip, 1 hr prior to sacrifice) decreased paraoxon formation indicating that phenobarbital induces that form(s) of cytochrome P-450 catalyzing conversion of parathion to paraoxon. Degradation of paraoxon to p-nitrophenol was increased by phenobarbital pretreatment, but not affected by SKF 525-A suggesting that MFO activities play only a minor role in the detoxification of the active metabolite of this insecticide. The phenobarbital-induced increase in paraoxon formation was partially antagonized by SKF 525-A. Significant activity for both parathion activation and paraoxon degradation was also observed in the lung preparation, however, this extrahepatic parathion and paraoxon metabolizing activity was not induced by phenobarbital or inhibited by SKF 525-A pretreatment. Phenobarbital pretreatment increased paraoxon level in livers of rats when measured 3 hr following parathion injection (2 mg/kg, ip). SKF 525-A did not alter parathion or paraoxon levels in brain, blood and liver. Phenobarbital pretreatment decreased the toxicity of parathion (4mg/kg, ip) or paraoxon (1.5 mg/kg, ip) as determined by decreases in lethality and inhibition of brain and lung acetylcholinesterases. An additional SKF 525-A treatment failed to decrease the protective effects of phenobarbital against parathion or paraoxon toxicity. These results suggest that some unknown factors other than hepatic MFO induction are involved in the protective action of phenobarbital against parathion and paraoxon toxicity.

구기자 복합물 약침액이 간기능 개선에 미치는 영향 (Protective Effect of Herbal Mixture Including Lycii Fructus on Hepatotoxicity Induced by Thioacetamide in Mice)

  • Objectives : This study investigated the hepatoprotective effect of herbal mixture including Lycii fructus (HML) in thioacetamide (TAA)-induced hepatotoxicity in mice. Methods : To confirm the liver protective effect, induced by TAA for 3 days injection at 100 mg/kg mice, HML were treated for 8 weeks at 300 mg/kg/day, 1000 mg/kg/day. Positive control was treated silymarin 50 mg/kg/day after TAA injection. The changes of mortality rate, clinical signs, organ weight, relative liver, blood chemistry and histopathological findings were analyzed after experiment. Results : Body weight gain was observed in all groups, but TAA treated group at 4th week and all treated groups decreased weight compared to the untreated group. As a result of organ weight measurement, organ weight gain due to hepatic injury was observed statistically significantly in TAA-treated group and TAA+Silymarin treated group, and the herbal mixture-treated group showed a tendency to decrease compared to the TAA treated group. Blood biochemistry showed that total cholesterol and very low density lipoprotein cholesterol decreased statistically in TAA+low-dose and high dose herbal mixture treated group compared to the TAA-treated group. Histopathological examination showed that liver abnormalities were not observed in untreated group, liver fibrosis was observed in liver injury with TAA treated and herbal mixture treated group. And, TAA+high dose herbal mixture group showed relaxation tendency on liver calcification compared to the TAA treated group. Conclusions : According to the above results, HML provided hepatoprotective effects on the hepatic injury by reduction of inflammatory responses.

  • Increasing evidence regarding free radical generating agents and inflammatory processes suggests that accumulation of reactive oxygen species can cause hepatotoxicity. A short-chain analog of lipid hydroperoxide, t-butyl hydroperoxide (t-BHP), can be metabolized to free radical intermediates by cytochrome P-450 in hepatocytes. which in turn can initiate lipid peroxidation, affect cell integrity and result in cell injury.   • Methanolic extract of Cassia tora leaves was evaluated for its hepatoprotective activity in rats by inducing hepatotoxicity with paracetamol (acute model). The extract at a dose of 400 mg/kg orally exhibited significant protective effect by lowering the serum levels of transaminase (SGOT and SGPT), bilirubin, and alkaline phosphatase (ALP). The effects produced were comparable to that of a standard hepatoprotective agent.

  • This study was undertaken to investigate the inhibition effects of Auricularia auricula-judae methanol extract in edible mushroom on lipid peroxidation and liver damage in benzo(a)pyrene(B(a)P)-treated mice. The activities of serum aminotransferase, cytochrome P-450, superoxide dismutase, catalase, glutathione peroxidase and the hepatic content of lipid peroxide after B(a)P-treatment was markedly increased than control but those levels were significantly decreased by the treatment of Auricularia auricula-judae methanol extract. Glutathione S-transferase activity and the hepatic glutathione content were decreased by B(a)P-treatment than control, but those were also inhibited by the treament of Auricularia auricula-judae methanol extract. These results suggest that Auricularia auricula-judae methanol extract have a protective effect on liver damage by B(a)P.

  • Quercetin, one of the most abundant flavonoids in human diet has been reported to exhibit a wide range of pharmacological properties. In this study, we investigated the protective effect of quercetin on hepatic fibrosis induced by dimethylnitrosamine (DMN) in rats. Treatment with DMN caused a significant decrease in body and liver weight. Oral administration of quercetin (10 mg/kg daily for 4 weeks) remarkably prevented this DMN-induced loss in body and liver weight and inhibited the elevation of serum alanine transaminase. aspartate transaminase, and bilirubin levels.   • To investigate the effect of Sarcodon aspratus methanol extract on liver damage in benzo(a)pyrene (B(a)P)-treated mice, mice were divided into 4 groups of control, B(a)P, Sarcodon aspratus methanol extract and Sarcodon aspratus methanol extract-B(a)P. The activities of serum aminotransferase, cytochrome P-450 and hepatic content of lipid peroxide after B(a)P-treatment were increased than control, but those levels were significantly decreased by the treatment of Sarcodon aspratus methanol extract. On the other hand, the hepatic glutathione content and glutathione S-transferase activity were increased by the treatment of Sarcodon aspratus methanol extract. Also the activities of superoxide dismutase, catalase and glutathione peroxidase were decreased by the treatment of Sarcodon aspratus methanol extract. In addition cytochrome P-450 1A1 isozyme protein level, which was remarkably increased by B(a)P treatment from results of immuno blotting, was decreased by the treatment with methanol extract of Sarcodon aspratus. These results suggest that Sarcodon aspratus methanol extract have protective effect on liver damage by decreasing lipid peroxide and activities of free radical generating enzymes.

겨우살이 Lectin이 지질대사에 미치는 영향 (Effect of Lipid Metabolism in Viscum album Lectin on Rats)

  • This study was carried out to investigate effect of lipid metabolism in Viscum album lectin on rats. The lectin was purified by sepharose 4B affinity chromatography and gel filtration using sephadex G-150 with plant material from Viscum album collected in Mt. Duk Yui. After 72 h of $CCl_4$ injection (in olive oil, 1:1, 2 mg/kg) there was a significant increase in serum total cholesterol and triglycerige levels relative to the control group. However, treatment of both Viscum album and purified lectin were significantly decreased lipid parameters against the $CCl_4$-induced. Histological observation basically supported the result obtained from serum lipid assay. The livers of rats challenged with $CCl_4$ produced a marked increase of cytoplasmic vacuoles in number, while the number of necrotic cells and swollen hepatocytes did not change significnatly. Rats administered olive oil alone did not alter the normal hepatic architecture. Histological observation of the liver section in rats treated 72 h with either Viscum album purified lectin or $CCl_4$-induced liver lipogenesis showed decreased numbers of cytoplasmic vaculoes and necrotic cells. The normal hepatic architectural pattern was observed in Hematoxylin-eosin stain. These results suggest that Viscum album lectin has a possible protective effect of lipid metabolim in rats.

홍삼복합제의 보간 및 항산화 작용에 대한 연구 (Study on Hepatoprotective and Antioxidant Activities of Korean Red Ginseng-Mixed Formula)

  • GRF (Korean Red Ginseng mixed formula) consists of six herbs such as Ginseng Radix rubra Koreana, Lycii Fructus, Artemisiae Capillaris Herba, Poria, and Glycyrrhizae Radix and Hoveniae Fructus. For the evaluation of hepatoprotective effect of GRF, the study was performed on protective effect against hepatic damage induced by galactosamine in vitro and ccl4 in vivo and also elucidate antioxidant activity. In vitro assay with 1.1 mM galactosamine, protection (%) was 44% (GR), and 58% (GRF-A) at 50 ug/ml. GRF effectively protected fatty degenertion and necrosis in murine hepatic damage induced by ccl4. For the -antioxidant study, GRF inhibited hemolysis of erythrocyte and decolored DPPH (2,2-diphenyl-l-picrylhydrazyl) free radical in a dose dependent manner more effetively than GR alone in vitro. GRF and GR significantly suppressed the time course $(1\;hr{\sim}6\;hr)-level$ of MDA (malondialdehyde) following AAPH (2,2'-azo-bis-(2-amidino -propane) dihydrochloride) treatment in vivo as compared with control data. From the results it can be concluded GR and GRF exerted the hepatoprotective effect by dint of antioxidant activity.
