This study was carried out to examine whether kimchi has hypolipidemic effect and to know how it exert lipid-lowering effect in rats. Male Sprague-Dawley rats were fed with kimchi-fee diet, or 3%, or 5%, or 10% kimchi diets for 6 weeks. Plasma cholesterol level was lowered in rats fed all concentrations of kimchi diets, and plasma triglyceride(TG) level was lowered in 10% kimchi diet group compared with that of control significantly(p<0.05). Th intake of kimchi lowered VLDL-cholesterol and VLDL-TG levels, whereas increased HDL-cholesterol level significantly(p<0.05). LDL-cholesterol level was lowered only in 5% kimchi diet group and LDL-TG level was lowered in all kimchi diet groups compared with those of control significantly (p<0.05). the intake of 5% and 10% kimchi diets also lowered the levels of hepatic cholesterol, TG, total lipid, and apolipoprotein B, whereas increased the levels of fecal total fat, cholesterol, TG, and apolipoprotein A-1 significantly(p<0.05). Triiodothyronine(T$_3$) level was elevated in rats fed kimchi diet, whereas thyroxine(T$_4$) level was not affected by kimchi treatment. These observations support that the intake of kimchin in rats loweres plasma and hepatic lipid levels by increasing the excretion of TG and cholesterol through feces, by the elevation of T$_3$ level, and by the altered lipoprotein metabolism.
Diethylnitrosamine (DEN) is known as a potential hepatic carcinogen by single administration. This study was designed to measure the effects of DEN-induced cell damage on the triglyceride and cholesterol concentration in the liver, excluding dietary effects. Fertilized chicken eggs, 10 days before hatching, were randomly divided into three groups (n=20) and each egg was injected 10 ${mu}ell$ of corn oil (vehicle control), 5 $\mu\textrm{g}$ of DEN/10 ${mu}ell$ of DEN/10 ${mu}ell$ into yolk via air sac. After 48 hr and 96 hr incubation, the damage of the chick-embryo liver cell was investigated by electron microscopy and by measuring the concentration of lipid components (total cholesterol, free cholesterol, phospholipid and triglyceride). For eggs administered 10 $\mu\textrm{g}$ of DEN and incuvated 96 hr, in hepatocyte, the nucleus membrane was roughed, the size of nucleolus was apparently increased and euchromatin was accumulated. Mitochondria were condensed and cristae, located mitochondiral inner membrane, were obscured. Additionally, the leaves of triglyceride and cholesterol classes were significantly increased depend on the amount treated with 10 $\mu\textrm{g}$ DEN at 96 hr, but phospholipids component of cell membrane, were decreased with significance. As a conclusion, carcinogen induced hepatic lesion was correlated with the changes in lipid component of liver.
Lipid peroxidation mediated by hydroxyl radicals which are generated during myocardial ischermia has suggested as a possible mechanism of ischemic myocardial damage. Recently, it has been reported that anti-arrhythmic action of lidocaine, a local anesthetic, is attributed to its "membrane-stabilizing" properties through scavenging free radicals, thus, inhibiting lipid peroxidation. Aldehyde oxidase and xanthine oxidase which catalyze the oxidation of many purine, pyrimidine and pteridine derivatives are known as free radical generating systems. In this experiment, we studied the effect of lidocaine and procainamide on the hepatic aldehyde and xanthine oxidase activity and antioxidative activities. It was found that lidocaine and procainamide inhibited both NADPH-dependent and independent lipid peroxidation. Both of tested compounds were found to be ineffective in inhibiting xanthine oxidase. Lidocaine and procainamide, however, inhibited aldehyde oxidase activity in vitro as well as in vivo. Based on the above results, lidocaine and procainamide could be employed as a therapeutic agent for aldehyde oxidaserelated disease.d disease.
Objectives : In this study, we investigated the effect and the underlying mechanism of ethanol extract of Benincasa seeds on a cellular model of non-alcoholic fatty liver disease (NAFLD) established by treating HepG2 cells with palmitate. Methods : We evaluated ethanol extract of Benincasa seeds (EEBS) for its hepatic lipid-lowering potential in fatty acid overloaded HepG2 cells. After incubation in palmitate containing media with or without EEBS, intracellular neutral lipid accumulations were quantified by Nile red staining. We also investigated the effect of EEBS on lipogenesis and ${\beta}$-oxidation. $LXR{\alpha}$-dependent SREBP-1c activation, expression of lipogenic genes, and expression of ${\beta}$-oxidation related genes were determined with or without pretreatment of EEBS. Results : EEBS significantly attenuated palmitate-induced intracellular neutral lipid accumulation in HepG2 cells. EEBS suppressed fatty acid synthesis by inhibiting $LXR{\alpha}$-dependent SREBP-1c activation. EEBS also repressed SREBP-1c mediated induction of lipogenic genes, including ACC, FAS, and SCD-1. However, EEBS had no effect on ${\beta}$-oxidation related CPT-1 and $PPAR{\alpha}$ gene expression. Conclusions : Our results suggest that EEBS has an efficacy to decrease hepatic lipid accumulation, and this effect was mediated by inhibiting the $LXR{\alpha}$-SREBP-1c pathway that leads to expression of lipogenic genes and hepatic steatosis. Therefore, the Benincasa seeds may have a potential clinical application for treatment of this chronic liver disease.
This study was designed to examine the effects of Salicornia herbacea L. (glasswort: GW) on the lipid peroxidation and mineral levels in diabetic rats. Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-220 g by an injection of streptozotocin (STZ) dissolved in a citrate buffer into the tail vein at a dose of 45 mg/kg of body weight. Sprague-Dawley rats were fed an AIN-93 recommended diet and the experimental groups were fed a modified diet containing 10% and 20% of glasswort powder for 4 weeks. The experimental groups were divided into 6 groups which consisted of normal (N)-control group, N-GW 10% and N-GW 20% treated groups, STZ-control, STZ-GW 10% and STZ-GW 20% treated groups. The rats' liver and muscle glycogen, liver and kidney protein, cholesterol and triglyceride (TG) in liver, malondialdehyde (MDA) in liver and kidney values were measured, along with the hepatic of chromium (Cr), iron (Fe), and zinc (Zn) content. The liver glycogen levels was significantly affected in N-GW 20% group among all the experimental groups. The liver MDA levels of the STZ-GW 10% and STZ-GW 20% groups were significantly lower than for the STZ-control group. There were significant differences between the N-control group and the STZ-control group in the hepatic of Zn levels. The hepatic of Cr levels in the N-GW 20% and STZ-GW 10% and STZ-GW 20% groups were significanly higher than for the each control groups. These results exhibited dose related effect of glasswort and it may have favorable influence on lipid peroxidation in the liver.
The effects of Cordyceps militaris on the weight gain, the food intake, the food efficient ratio, and the lipid concentrations of serum, liver, pancreas, heart and aorta in male rats fed cholesterol diet were studied. Male Sprague-Dawley rats were received 4 types of diets for 4 weeks, respectively : a normal diet(without cholesterol), a control diet(hypercholesterolemic diet), a hypercholesterolemic diet supplemented with 3% fruiting body of Cordyceps militaris(CF), a hypercholesterolemic diet supplemented with 3% mycelium of Cordyceps militaris(CM). The body weight gain of rats fed the control diet were significantly increased compared to those of the rats fed diets with CF or CM. But, the hepatic and pancreatic weights of rats fed diets with CF or CM were heavier than those of rats fed the normal diet. In serum of rats fed the CM diet, the concentrations of total cholesterol, triglyceride, LDL-cholesterol and the atherogenic index were significantly lower than those of rats fed the control diet. Also, serum HDL-cholesterol concentration and HDL-cholesterol/total cholesterol ratio were significantly lower in rats fed the control, CF or CM diets than those in rats fed the normal diet. Whereas, the hepatic concentrations of the total lipid, the total cholesterol and the triglyceride were significantly higher in rats fed 3 hypercholesterolemic diets than those in the rats fed normal diet. The heart and pancreatic concentrations of total cholesterol were the highest in the rats fed diet with the CR among rats fed experimental diets. In conclusion, the CM feeding decreased the concentrations of the cholesterol, triglyceride, and LDL-cholesterol and the atherogenic index in the serum of the rats. But the CF or CM feeding could not decrease the hepatic concentrations of the total lipid, cholesterol, triglyceride in the rats fed hypercholesterolemic diets. Effects of garlic powder supplementation on blood lipid profile and antioxidant system were investigated in rats with and without swimming exercise. Sprague-Dawley rats of four experimental groups were fed for 4 weeks diets containing $15\%$ beef tallow and $1\%$ cholesterol; control without garlic and exercise, Go with $2\%$ garlic alone, Ex with exercise alone, GoEx with $2\%$ garlic and exercise. Rats were trained 40 min a days a days a week. Group Ex and GoEx showed significant lowering in body weight gain and fat accumulation. In Go, Ex and GoEx, plasm TG and LDL-C were lower and HDL-C was higher, although not significantly, compared to levels in control. Total cholesterol was significantly reduced in group Go, and Ex and GoEx were lower than control. The total/HDL cholesterol ratio was also found to be significantly different, decreasing the ratios in Go, Ex and GoEx. The hepatic TBARS increased significantly in group Ex $(51.7{\pm}3.43nM/g\;liver)$, while TBARS in Go and GoEx were low $(35.68{\pm}3.61,\;39.30{\pm}5.55nM/g\;liver)$ and similar to control's one. The activity of hepatic SOD in Go and GoEx tended higher than control and Ex without garlic. The hepatic catalase showed significantly the highest activity in Go. Activity of GSH-px was significantly low in Ex with $0.14{\pm}0.03$ unit/mg protein, and control, Go and GoEx had higher activities of $0.23{\pm}0.08,\;0.20{\pm}0.07,\;0.22{\pm}0.01\;unit/mg$ protein, respectively. Lower activities of antioxidant enzymes in Ex are likely to associated with the highest level of TBARS. It seems that a decrease in TBARS in GoEx relative to Ex was related to the increase in GSHpx and SOD with garlic supplemented, which led to compensate the oxidative stress from exercise. The results suggests that exercise or garlic supplement exerts blood lipid attenuating effect. In adition, garlic supplementation could strengthen the antioxidant potential against exercise-induced oxidants, partly by modulating oxidant enzyme activity. These effects of garlic may make it a beneficial agent on CVD.
To evaluate hepatoprotective effects of G009, an hepatoprotective agent which was extracted from the mycelia of Ganoderma lucidum IY009, we were, studied using $CCl_4$-and galactosamine-induced hepatotoxicity in rats. The ratio of liver weight to body weight, the value of glutamic oxaloacetic transaminase(GOT) and glutamic pyruvic transaminase (GPT) activities, the change of a lipids in serum, and the inhibitory activity of malondialdehyde (MDA) formation in serum and liver homogenate were determined in rats. G009 was not significantly changed of the ratio of liver weight to body weight and the content of lipids in serum, but reduced the serum GOT and GPT values in $CCl_4$-and galactosamine-induced hepatotoxicity in rat. Especially, protective effect of G009 on rat hepatic injuries induced by galactosamine was significantly appeared. $CCl_4$ increased markedly the formation of lipid peroxides in the liver homogenate, and serum. The increase of lipid peroxides by $CCl_4$-induced hepatotoxicity was markedly reduced by the treatment with G009. These results suggest that the hepatoprotective effects of G009 may be correlated with its anti-lipid peroxidative activity, therefore, it may be potential agent for hepatic disease.

We evaluated the effects of newly developed functional food containing Bacillus polyfermenticus SCD as the main material on the lipid and antioxidant metabolisms of hypercholesterolemic rats. Thirty male SD rats were divided into three groups after a 1-week adaptation period and were fed with a high fat-cholesterol diet (control), or with a high fat-cholesterol diet supplemented with low or high doses ($3.1\;{\times}\;10^6\;cfu/day$ or $3.1\;{\times}\;10^8\;cfu/day$) of B. polyfermenticus SCD and other physiological active materials for 6 weeks. Both doses of B. polyfermenticus SCD significantly reduced hepatic total cholesterol and triglycerides, while increasing the fecal excretion rates of total lipid, total cholesterol and triglycerides. B. polyfermenticus SCD increased the total radical trapping antioxidant potential (TRAP). The erythrocytic glutathione peroxidase activity in the B. polyfermenticus groups was significantly lower than that in the control group. Plasma TRAP levels exhibited a highly significant negative correlation with hepatic total cholesterol while a significant positive correlation was detected between fecal total cholesterol and plasma TRAP. This hypolipidemic and antioxidative effect of B. polyfermenticus SCD seemed to be unrelated to its dosage. These results suggest that functional food containing B. polyfermenticus SCD can improve oxidative stress and hepatic lipid profiles by enhancing the excretion of cholesterol and triglycerides in feces of rats fed with high fat-high cholesterol diet.
콜레스테롤을 0.5% 수준으로 첨가하여 고콜레스테롤혈증을 유발시킨 상태에서 0.2% 클로로겐산을 병합 투여한 식이를 흰쥐에게 2주간 자유 급여시켜 혈청과 간장의 지질농도, 혈당치 및 뇨 단백질량에 미치는 영향을 검토하였다. 동물 실험군은 콜레스테롤과 클로로겐산을 무첨가한 정상 식이군, 콜레스테롤을 0.5% 수준으로 첨가한 콜레스테롤 식이군 및 콜레스테롤 식이군에 클로로겐산을 0.2% 수준으로 첨가한 클로로겐산 병합투여군으로 나누었다. 정상 식이군에 비해 콜레스레롤 식이군에서 혈청 총 콜레스테롤 농도의 유의적 증가는 클로로겐산 병합투여군에서 유의적으로 감소되었으며, 동시에 동맥경화 지수도 감소되었다. 간장의 지질 농도는 각 식이군에 의한 유의적인 차이는 없었다. 혈당치는 정상 식이군에 비해 콜레스테롤 식이군 및 클로로겐산 병합투여군에서 증가되었다. 뇨 단백질양은 정상 식이군과 콜레스테롤 식이군 간에는 유의적인 차이가 없었으나, 클로로겐산 병합 투여군에서는 유의적으로 증가하였다 이상의 결과로부터, 클로로겐산은 콜레스테롤 식이에 의한 고콜레스테롤혈증을 개선시키고 동맥경화 지수도 감소시켰다.
